| 1 | Restoration of electrical microenvironment enhances bone regeneration under diabetic conditions by modulating macrophage polarization显示文摘Macrophage-mediated inflammation compromises bone repair in diabetic patients.Electrical signaling cues are known to regulate macrophage functions.However,the biological effects of electrical microenvironment from charged biomaterials on the immune response for regulating osteogenesis under diabetic conditions remain to be elucidated.Herein the endogeneous electrical microenvironment of native bone tissue was recapitulated by fabricating a ferroelectric BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membrane.In vitro,the polarized BaTiO_(3)/poly(vinylidene fluoridetrifluoroethylene)(BTO/P(VDF-TrFE))nanocomposite membranes inhibited high glucose-induced M1-type inflammation,by effecting changes in cell morphology,M1 marker expression and pro-inflammatory cytokine secretion in macrophages.This led to enhanced osteogenic differentiation of human bone marrow mesenchymal stem cells(BM-MSCs).In vivo,the biomimetic electrical microenvironment recapitulated by the polarized nanocomposite membranes switched macrophage phenotype from the pro-inflammatory(M1)into the pro-healing(M2)phenotype,which in turn enhanced bone regeneration in rats with type 2 diabetes mellitus.Mechanistic studies revealed that the biomimetic electrical microenvironment attenuated pro-inflammatory M1 macrophage polarization under hyperglycemic conditions by suppressing expression of AKT2 and IRF5 within the PI3K-AKT signaling pathway,thereby inducing favorable osteo-immunomodulatory effects.Our study thus provides fundamental insights into the biological effects of restoring the electrical microenvironment conducive for osteogenesis under DM conditions,and offers an effective strategy to design functionalized biomaterials for bone regeneration therapy in diabetic patients. | Xiaohan Dai Boon Chin Heng Yunyang Bai Fuping You Xiaowen Sun Yiping Li Zhangui Tang Mingming Xu Xuehui Zhang Xuliang Deng | 2021 | Bioactive Materials2021,6,7: | 9 |
| 2 | On the mechanical behavior of the human biliary system显示文摘This paper reviews the progress made in understanding the mechanical behaviour of the biliary system. Gallstones and diseases of the biliary tract affect more than 10% of the adult population. The complications of gallstones, i.e. acute pancreatitis and obstructive jandice, can be lethal, and patients with acalculous gallbladder pain often pose diagnostic difficulties and undergo repeated ultrasound scans and oral cholecystograms. Moreover, surgery to remove the gallbladder in these patients, in an attempt to relieve the symptoms, gives variable results. Extensive research has been carried out to understand the physiological and pathological functions of the biliary system, but the mechanism of the pathogenesis of gallstones and pain production still remain poorly understood. It is believed that the mechanical factors play an essential role in the mechanisms of the gallstone formation and biliary diseases. However, despite the extensive literature in clinical studies, only limited work has been carried out to study the biliary system from the mechanical point of view. | 3 | Modified hyaluronic acid hydrogels with chemical groups that facilitate adhesion to host tissues enhance cartilage regeneration显示文摘Stable integration of hydrogel implants with host tissues is of critical importance to cartilage tissue engineering.Designing and fabricating hydrogels with high adhesive strength,stability and regeneration potential are major challenges to be overcome.This study fabricated injectable adhesive hyaluronic acid(HA)hydrogel modified by aldehyde groups and methacrylate(AHAMA)on the polysaccharide backbone with multiple anchoring mechanisms(amide bond through the dynamic Schiff base reaction,hydrogen bond and physical interpenetration).AHAMA hydrogel exhibited significantly improved durability and stability within a humid environment(at least 7 days),together with higher adhesive strength(43 KPa to skin and 52 KPa to glass),as compared to commercial fibrin glue(nearly 10 KPa)and HAMA hydrogel(nearly 20 KPa).The results showed that AHAMA hydrogel was biocompatible and could be easily and rapidly prepared in situ.In vitro cell culture experiments showed that AHAMA hydrogel could enhance proliferation(1.2-folds after 3 days)and migration(1.5-folds after 12 h)of bone marrow stem cells(BMSCs),as compared to cells cultured in a culture dish.Furthermore,in a rat osteochondral defect model,implanted AHAMA hydrogel significantly promoted integration between neo-cartilage and host tissues,and significantly improved cartilage regeneration(modified O’Driscoll histological scores of 16.0±4.1 and 18.3±4.6 after 4 and 12-weeks of post-implantation in AHAMA groups respectively,12.0±2.7 and 12.2±2.8 respectively in HAMA groups,9.8±2.4 and 11.5±2.1 respectively in untreated groups).Hence,AHAMA hydrogel is a promising adhesive biomaterial for clinical cartilage regeneration and other biomedical applications. | Jiaqing Chen Jiabei Yang Li Wang Xuewei Zhang Boon Chin Heng Dong-An Wang Zigang Ge | 2021 | Bioactive Materials2021,6,6: | 6 |

| 5 | Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers. | Feng Li Muthu K. | 7 | Tapering-induced enhancement of light extraction efficiency of nanowire deep ultraviolet LED by theoretical simulations显示文摘A nanowire(NW) structure provides an alternative scheme for deep ultraviolet light emitting diodes(DUV-LEDs)that promises high material quality and better light extraction efficiency(LEE). In this report, we investigate the influence of the tapering angle of closely packed Al Ga N NWs, which is found to exist naturally in molecular beam epitaxy(MBE) grown NW structures, on the LEE of NW DUV-LEDs. It is observed that, by having a small tapering angle, the vertical extraction is greatly enhanced for both transverse magnetic(TM) and transverse electric(TE) polarizations. Most notably, the vertical extraction of TM emission increased from 4.8% to 24.3%,which makes the LEE reasonably large to achieve high-performance DUV-LEDs. This is because the breaking of symmetry in the vertical direction changes the propagation of the light significantly to allow more coupling into radiation modes. Finally, we introduce errors to the NW positions to show the advantages of the tapered NW structures can be projected to random closely packed NW arrays.
