| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | A rapid method for chemical fingerprint analysis of Panax notoginseng powders by ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry显示文摘A method coupling ultra-performance liquid chromatography(UPLC) with quadrupole time-of-flight mass spectrometer(Qtof MS) using the electrospray ionization(ESI) source was developed for the identification of the major saponins from Panax notoginseng powder(PNP). Ten different PNP samples were analyzed and evaluated for their quality by similarity evaluation and principle component analysis(PCA). Based on the accurate mass, summarized characteristic fragmentation behaviors, retention times of different types of saponins, related botanical biogenesis, and reported chromatographic behavior of saponins, fifty-one common peaks were effectively separated and identified, including 28 protopanaxadiol saponins and 18 protopanaxatriol saponins. Simultaneously, 15 significant discrepancy compounds were identified from the disqualified PNP samples. | 4 | Evolution of blood lipids and risk factors of dyslipidemia among people living with human immunodeficiency virus who had received first-line antiretroviral regimens for 3 years in Shenzhen显示文摘Background:Lipid abnormalities are prevalent among people living with human immunodeficiency virus(HIV)(PLWH)and contribute to increasing risk of cardiovascular events.This study aims to investigate the incidence of dyslipidemia and its risk factors in PLWH after receiving different first-line free antiretroviral regimens.Methods:PLWH who sought care at the Third People’s Hospital of Shenzhen from January 2014 to December 2018 were included,and the baseline characteristics and clinical data during the follow-up were collected,including total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C).The risk factors of dyslipidemia after antiretroviral therapy were analyzed with the generalized estimating equation model.Results:Among the 7623 PLWH included,the mean levels of TC,HDL-C and LDL-C were 4.23±0.85 mmol/L,1.27±0.29 mmol/L and 2.54±0.65 mmol/L,respectively,and the median TG was 1.17(IQR:0.85-1.68)mmol/L.Compared with that in PLWH receiving tenofovir disoproxil fumarate(TDF)+lamivudine(3TC)+ritonavir-boosted lopinavir(LPV/r),zidovudine(AZT)+3TC+efavirenz(EFV),and AZT+3TC+LPV/r,the incidence of dyslipidemia was lower in PLWH receiving TDF+3TC+EFV.In multivariate analysis,we found that the risks of elevations of TG,TC,and LDL-C were higher with TDF+3TC+LPV/r(TG:odds ratio[OR]=2.82,95%confidence interval[CI]:2.55-3.11,P<0.001;TC:OR=1.24,95%CI:1.14-1.35,P<0.001;LDL:OR=1.06,95%CI:1.00-1.12,P=0.041),AZT+3TC+EFV(TG:OR=1.41,95%CI:1.28-1.55,P<0.001;TC:OR=1.43,95%CI:1.31-1.56,P<0.001;LDL:OR=1.18,95%CI:1.12-1.25,P<0.001),and AZT+3TC+LPV/r(TG:OR=3.08,95%CI:2.65-3.59,P<0.001;TC:OR=2.40,95%CI:1.96-2.94,P<0.001;LDL:OR=1.52,95%CI:1.37-1.69,P<0.001)than with TDF+3TC+EFV,while treatment with TDF+3TC+LPV/r was less likely to restore HDL-C levels compared with TDF+3TC+EFV(OR=0.95,95%CI:0.92-0.97,P<0.001).In addition to antiretroviral regimens,antiretroviral therapy duration,older age,overweight,obesity and other traditional factors were also important risk factors for dyslipidemia.Conclusion:The incidence of dyslipidemia varies with different antiretroviral regimens,with TDF+3TC+EFV having lower risk for dyslipidemia than the other first-line free antiretroviral regimens in China. | Li-Qin Sun Jia-Ye Liu Yun He Yang Zhou Liu-Mei Xu Lu-Kun Zhang Fang Zhao Xiao-Ning Liu Ying Song Ting-Zhi Cao Yi-Mei Tian Man Rao Hui Wang | 2020 | Chinese Medical Journal2020,,23: | 9 |

| 8 | Rosmarinic acid ameliorates hypoxia/ischemia induced cognitive deficits and promotes remyelination显示文摘Rosmarinic acid,a common ester extracted from Rosemary,Perilla frutescens,and Salvia miltiorrhiza Bunge,has been shown to have protective effects against various diseases.This is an investigation into whether rosmarinic acid can also affect the changes of white matter fibers and cognitive deficits caused by hypoxic injury.The right common carotid artery of 3-day-old rats was ligated for 2 hours.The rats were then prewarmed in a plastic container with holes in the lid,which was placed in 37°C water bath for 30 minutes.Afterwards,the rats were exposed to an atmosphere with 8% O2 and 92% N2 for 30 minutes to establish the perinatal hypoxia/ischemia injury models.The rat models were intraperitoneally injected with rosmarinic acid 20 mg/kg for 5 consecutive days.At 22 days after birth,rosmarinic acid was found to improve motor,anxiety,learning and spatial memory impairments induced by hypoxia/ischemia injury.Furthermore,rosmarinic acid promoted the proliferation of oligodendrocyte progenitor cells in the subventricular zone.After hypoxia/ischemia injury,rosmarinic acid reversed to some extent the downregulation of myelin basic protein and the loss of myelin sheath in the corpus callosum of white matter structure.Rosmarinic acid partially slowed down the expression of oligodendrocyte marker Olig2 and myelin basic protein and the increase of oligodendrocyte apoptosis marker inhibitors of DNA binding 2.These data indicate that rosmarinic acid ameliorated the cognitive dysfunction after perinatal hypoxia/ischemia injury by improving remyelination in corpus callosum.This study was approved by the Animal Experimental Ethics Committee of Xuzhou Medical University,China (approval No.20161636721) on September 16,2017. | Man Li Miao-Miao Cui Nwobodo Alexander Kenechukwu Yi-Wei Gu Yu-Lin Chen Si-Jing Zhong Yu-Ting Gao Xue-Yan Cao Li Wang Fu-Min Liu Xiang-Ru Wen | 2020 | Neural Regeneration Research2020,15,5: | 3 |

| 11 | ACBD3 is required for FAPP2 transferring glucosylceramide through maintaining the Golgi integrity显示文摘Glycosphingolipid (GSL) metabolism is involved in various physiological processes, including all major cell signaling pathways, and its dysregulation is linked to some diseases. The four-phosphate adaptor protein FAPP2-mediated glucosylceramide (GlcCer) transport for complex GSL synthesis has been studied extensively. However, the molecular machinery of FAPP2 as a GlcCer-transferring protein remains poorly defined. Here, we identify a Golgi-resident protein, acyl-coenzyme A binding domain containing 3 (ACBD3), as an interacting partner of FAPP2. We find that ACBD3 knockdown leads to dramatic Golgi fragmentation, which subsequently causes FAPP2 dispersal throughout the cytoplasm and a decreased localization at trans-Golgi network. The further quantitative Upidomic analysis indicates that ACBD3 knockdown triggers abnormal sphingolipid metabolism. Interestingly, the expression of siRNA-resistant full-length ACBD3 can rescue these defects caused by ACBD3 knockdown. | 13 | Removing coarse sediment by sorting of reservoirs显示文摘Reservoir sedimentation is an unsolved problem.In this paper,based on measured data,theoretical analysis and numerical computations,we prove that a proportion of the sediment coarser than 0.1 mm(CS) is sorted and deposited in specific reaches in the upper backwaters or in run-of-river reservoirs.The ratio of CS is usually small but it impacts the slope of deposition delta greatly and raises the backwater in later stages for a river shaped reservoir(RSR).Based on these facts,we propose to remove such CS from a fixed basin(FB) in the upper backwater by dredging and we prove that the removal of CS is effective in reducing sediment deposition and in preserving the long-term capacity of reservoirs.A numerical model computation of the Three Gorges Project(TGP) reservoir indicates that dredging 30×106-50×106 t/a of CS could reduce 20% total deposition by the end of 100th year,so that the slope of deposition can be slowed down by 25%-30%.This would be remarkable for a long extended RSR.This method of removing CS can also be applied to the Xiaolangdi reservoir(XLD) on the Yellow River(YR) to not only limit reservoir deposition but also filter out the CS from entering the Lower Yellow River(LYR) to slow down the rise of the perched LYR.It provides a new alternative to postpone the continuous siltation of the LYR. | ZHOU JianJun ZHANG Man CAO HuiQun | 2011 | Science China(Technological Sciences)2011,54,4: | 2 |

| 16 | On the loss mechanisms of radiation belt electron dropouts during the 12 September 2014 geomagnetic storm显示文摘Radiation belt electron dropouts indicate electron flux decay to the background level during geomagnetic storms,which is commonly attributed to the effects of wave-induced pitch angle scattering and magnetopause shadowing.To investigate the loss mechanisms of radiation belt electron dropouts triggered by a solar wind dynamic pressure pulse event on 12 September 2014,we comprehensively analyzed the particle and wave measurements from Van Allen Probes.The dropout event was divided into three periods:before the storm,the initial phase of the storm,and the main phase of the storm.The electron pitch angle distributions(PADs)and electron flux dropouts during the initial and main phases of this storm were investigated,and the evolution of the radial profile of electron phase space density(PSD)and the(μ,K)dependence of electron PSD dropouts(whereμ,K,and L^*are the three adiabatic invariants)were analyzed.The energy-independent decay of electrons at L>4.5 was accompanied by butterfly PADs,suggesting that the magnetopause shadowing process may be the major loss mechanism during the initial phase of the storm at L>4.5.The features of electron dropouts and 90°-peaked PADs were observed only for>1 MeV electrons at L<4,indicating that the wave-induced scattering effect may dominate the electron loss processes at the lower L-shell during the main phase of the storm.Evaluations of the(μ,K)dependence of electron PSD drops and calculations of the minimum electron resonant energies of H+-band electromagnetic ion cyclotron(EMIC)waves support the scenario that the observed PSD drop peaks around L^*=3.9 may be caused mainly by the scattering of EMIC waves,whereas the drop peaks around L^*=4.6 may result from a combination of EMIC wave scattering and outward radial diffusion. | Xin Ma Zheng Xiang BinBin Ni Song Fu Xing Cao Man Hua DeYu Guo YingJie Guo XuDong Gu ZeYuan Liu Qi Zhu | 2020 | Earth and Planetary Physics2020,4,6: | 2 |
| 17 | Association between IgG N-glycans and Nonalcoholic Fatty Liver Disease in Han Chinese显示文摘Nonalcoholic fatty liver disease(NAFLD) is a major public health issue worldwide. Immunoglobulin G(IgG) N-glycans are associated with risk factors for NAFLD, such as obesity and diabetes. A cross-sectional study involving 500 Han Chinese adults recruited from a community in Beijing was carried out to explore the association between IgG N-glycans and NAFLD. IgG N-glycosylation was significantly associated with NAFLD, with the disease showing a negative correlation with galactosylation(GP14, GP14~n, and G2~n), positive correlation with fucosylation(FBG2~n/G2~n), and positive correlation with bisecting N-acetylglucosamine(Glc NAc) [FBG2~n/FG2~n and FBG2~n/(FG2~n+FBG2~n)], after controlling age, gender, and prevalence of obesity, type 2 diabetes mellitus, hypertension, and hyperlipidemia. In other words, the present study showed a possible association between NAFLD and the loss of galactose and elevations of fucose and bisecting GlcNAc. | 18 | Optimization of Hydrolysis Conditions for the Isolation of Angiotensin-I Converting Enzyme(ACE) Inhibitory Peptides from Rhopilema hispidum显示文摘To obtain the maximum angiotensin-I converting enzyme(ACE) inhibitory activity, the protein hydrolysis conditions of the jellyfish Rhopilema hispidum were optimized using response surface methodology(RSM). Trypsin was selected to produce R. hispidum protein hydrolysates(RPH) with ACE inhibitory activity. The optimal parameters for producing protein hydrolysates with the highest ACE inhibitory activity were as follows: hydrolysis time 5 h, hydrolysis temperature 50℃, and the enzyme-to-substrate ratio 6%. Under these conditions, the ACE inhibitory rate of RPH could reach 64.28% ± 5.72%. In addition, RPH contained high levels of Gly, Glu, Pro, Ala, Asp and Arg, with a molecular weight distribution range of 0.32–6.84 kDa. The following three novel ACE inhibitory peptides were isolated and identified: Ile-Gly-Glu-Thr-Gly-Pro, Gly-Ala-Thr-Gly-Pro-Ala-Gly-Tyr-Val and Gly-AlaPhe-Gly-Pro-Gly-Gly-Leu-Val-Gly-Arg-Pro. The IC_(50) values of the ACE inhibitory activity of these three purified peptides were 19.07, 27.42 and 31.26 μmol L^(-1), respectively. | 19 | EMC10 governs male fertility via maintaining sperm ion balance显示文摘Infertility is a severe public health problem worldwide that prevails up to 15% in reproductive-age couples,and male infertility accounts for half of total infertility.Studies on genetically modified animal models have identifed lots of genes involved in the pathogenesis of male infertility.The underlying causes,however,remain largely unclear.In this study,we provide evidence that EMCIO,one subunit of endoplasmic reticulum (ER)membrane protein complex (EMC),is required for male fertility.EMC10 is significantly decreased in spermatozoa from patients with asthenozoospermia and positively associated with human sperm motility. Male mice lacking Emc10 gene are completely sterile.Emc10-null spermatozoa exhibit multiple defects including abnormal morphology,decreased motility,impaired capacitation,and impotency of acrosome reaction,thereby which are incapable of fertilizing intact or ZP-free oocytes.However,intracytoplasmic sperm injection could rescue this defect caused by EMC10 deletion.
