1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 28-bromo-7-methoxychrysin-induced apoptosis of hepatocellular carcinoma cells involves ROS and JNK显示文摘AIM:To investigate whether the apoptotic activities of 8-bromo-7-methoxychrysin(BrMC) involve reactive oxygen species(ROS) generation and c-Jun N-terminal kinase(JNK) activation in human hepatocellular carcinoma cells(HCC).METHODS:HepG2,Bel-7402 and L-02 cell lines were cultured in vitro and the apoptotic effects of BrMC were evaluated by flow cytometry(FCM) after propidium iodide(PI) staining,caspase-3 activity using enzymelinked immunosorbent assay(ELISA),and DNA agarose gel electrophoresis.ROS production was evaluated by FCM after dichlorodihydrofluorescein diacetate(DCHFDA) probe labeling.The phosphorylation level of JNK and c-Jun protein was analyzed by Western blotting.RESULTS:FCM after PI staining showed a dose-dependent increase in the percentage of the sub-G1 cell pop-ulation(P < 0.05),reaching 39.0% ± 2.8% of HepG2 cells after 48 h of treatment with BrMC at 10 μmol/L.The potency of BrMC to HepG2 and Bel-7402(32.1% ± 2.6%) cells was found to be more effective than the lead compound,chrysin(16.2% ± 1.6% for HepG2 cells and 11.0% ± 1.3% for Bel-7402 cell) at 40 μmol/L and similar to 5-flurouracil(33.0% ± 2.1% for HepG2 cells and 29.3% ± 2.3% for Bel-7402 cells) at 10 μmol/L.BrMC had little effect on human embryo liver L-02 cells,with the percentage of sub-G1 cell population 5.4% ± 1.8%.Treatment of HepG2 cells with BrMC for 48 h also increased the levels of active caspase-3,in a concentration-dependent manner.z-DEVD-fmk,a caspase-3specific inhibitor,prevented the activation of caspase-3.Treatment with BrMC at 10 μmol/L for 48 h resulted in the formation of a DNA ladder.Treatment of cells with BrMC(10 μmol/L) increased mean fluorescence intensity of DCHF-DA in HepG2 cells from 7.2 ± 1.12 at 0 h to 79.8 ± 3.9 at 3 h and 89.7 ± 4.7 at 6 h.BrMC did not affect ROS generation in L-02 cells.BrMC treatment failed to induce cell death and caspase-3 activation in HepG2 cells pretreated with N-acetylcysteine(10 mmol/L).In addition,in HepG2 cells treated with BrMC(2.5,5.0,10.0 μmol/L) for 12 h,JNK activation was observed.Peak JNK activation occurred at 12 h post-treatment and this activation persisted for up to 24 h.The expression of phosphorylated JNK and c-Jun protein after 12 h with BrMC-treated cells was inhibited by N-acetylcysteine and SP600125 pre-treatment,but GW9662 had no effect.SP600125 substantially reduced BrMC-induced cell death and caspase-3 activation of HepG2 cells.N-acetylcysteine and GW9662 also attenuated induction of cell death and caspase-3 activation in HepG2 cells treated with BrMC.CONCLUSION:BrMC induces apoptosis of HCC cells by ROS generation and sustained JNK activation.Xiao-Hong Yang Jian-Guo Cao Hong-Lin Xiang Fei Liu Yuan Lv Hunan Province China Xing Zheng 2010World Journal of Gastroenterology2010,16,27:38
3The state-of-the-art of the China Seismo-Electromagnetic Satellite mission显示文摘The China Seismo-Electromagnetic Satellite(CSES) mission was proposed in 2003 and approved in 2013 after ten years' scientific and engineering demonstrations. To meet the requirement of scientific objectives, the satellite is designed to be in a sunsynchronous orbit with an altitude of 507 km and descending node time of 14:00 LT. The CSES satellite carries 8 instruments,including search-coil magnetometer(SCM), electric field detector(EFD), high precision magnetometer(HPM), GNSS occultation receiver(GOR), plasma analyzer package(PAP), langmuir probe(LAP), high energetic particle package(HEPP) and detector(HEPD), and tri-band beacon(TBB), among which HEPD is provided by Italian Space Agency. The CSES satellite was launched successfully on February 2, 2018, and is planned to operate for 5 years. 4Molecular screening of patients with nonsyndromic hearing loss from Nanjing city of China显示文摘Hearing loss is the most frequent sensory disorder involving a multitude of factors,and at least 50% of cases are due to genetic etiology.To further characterize the molecular etiology of hearing loss in the Chinese population,we recruited a total of 135 unrelated patients with nonsyndromic sensorineural hearing loss (NSHL) for mutational screening of GJB2,GJB3,GJB6,SLC26A4,SLC26A5 IVS2-2A>G and mitochondrial 12SrRNA,tRNA Ser(UCN) by PCR amplification and direct DNA sequencing.The carrier frequencies of deafness-causing mutations in these patients were 35.55% in GJB2,3.70% in GJB6,15.56% in SLC26A4 and 8.14% in mitochondrial 12SrRNA,respectively.The results indicate the necessity of genetic screening for mutations of these causative genes in Chinese population with nonsyndromic hearing loss.Yajie Lu Dachun Dai Zhibin Chen Xin Cao Xingkuan Bu Qinjun Wei Guangqian Xing 2011The Journal of Biomedical Research2011,25,5:30
5Self-tuning virtual synchronous generator control for improving frequency stability in autonomous photovoltaic-diesel microgrids显示文摘This paper investigates the use of a virtual synchronous generator(VSG) to improve frequency stability in an autonomous photovoltaic-diesel microgrid with energy storage. VSG control is designed to emulate inertial response and damping power via power injection from/to the energy storage system. The effect of a VSG with constant parameters(CP-VSG) on the system frequency is analyzed. Based on the case study, self-tuning algorithms are used to search for optimal parameters during the operation of the VSG in order to minimize the amplitude and rate of change of the frequency variations. The performances of the proposed self-tuning(ST)-VSG, the frequency droop method, and the CP-VSG are evaluated by comparing their effects on attenuating frequency variationsunder load variations. 7Induction of apoptosis of human gastric carcinoma SGC-7901 cell line by 5,7-dihydroxy-8-nitrochrysin in vitro显示文摘AIM: To investigate the effect of 5, 7-dihydroxy-8- nitrochrysin (NOChR) on apoptosis of human gastric carcinoma SGC-7901 cell line. METHODS: SGC-7901 cells were cultured in vitro and the inhibitory effect of NOChR on proliferation of SGC-7901 cells was measured by using an MTT assay. NOChR-induced apoptosis rate of SGC-7901 cells was detected using flow cytometry (FCM) with PI staining. DNA ladder bands were observed by DNA agarose gel electrophoresis. The influence of NOChR on the proxisome proliferator-activated receptor-γ (PPARγ), Bcl-2 and Bax protein expression of SGC-7901 cells was analyzed by Western blot. RESULTS: MTT assay showed that NOChR markedly inhibited proliferation of SGC-7901 cells in a dose- dependent manner, and when IC50 was 4.14 μmol/L, the potency of NOChR was 10 times than that of lead compound, chrysin (ChR, IC50 was 40.56 μmol/L), and was similar to 5-fluorouracil (5-FU, IC50 was 4.51 μmol/L). FCM with propidium iodide (PI) staining demonstrated that the apoptosis rates of SGC-7901 cells treated with 1.25, 5.00 and 20.00 μmol/L NOChR for 48 h were 9.8% ± 0.2%, 36.8% ± 1.9% and 45.5% ± 3.5%, respectively, and were significantly higher when treated with 5.00 and 20.00 μmol/L NOChR than that with 20.00 μmol/L ChR (12.9% ± 1.5%). DNA agarose gel electrophoresis showed that treatment of SGC-7901 cells with 20.00 μmol/L NOChR for 48 h resulted in typical DNA ladder bands of DNA of SGC-7901 cells, which could be eliminated by treating with 10.00 μmol/L GW9662, a blocker of PPARγ. Western blot analysis revealed that after 24 h of treatment with 20.00 μmol/L NOChR, PPARgamma and Bax protein expression of SGC-7901 cells increased but Bcl-2 expression decreased; however, pre-incubation with 10.00 μmol/L GW9662 could efficiently antagonize and weaken the regulatory effect of 20.00 μmol/L NOChR on Bax and Bcl-2 protein expression of SGC-7901 cells. 11Porphyrin-like Fe-N4 sites with sulfur adjustment on hierarchical porous carbon for different rate-determining steps in oxygen reduction reaction显示文摘We developed a strategy based on coordination polymer to synthesize singleatom site Fe/N and S-codoped hierarchical porous carbon (Fe1/N,S-PC).The as-obtained Fe1/N,S-PC exhibited superior oxygen reduction reaction (ORR) performance with a half-wave potential (Ev2,0.904 V vs.RHE) that was better than that of commercial Pt/C (E1/2,0.86 V vs.RHE),single-atom site Fe/N-doped hierarchical porous carbon (Fe1/N-PC) without S-doped (E1/2,0.85 V vs.RHE),and many other nonprecious metal catalysts in alkaline medium.Moreover,the Fe1/N,S-PC revealed high methanol tolerance and firm stability.The excellent electrocatalytic activity of Fe1/N,S-PC is attributed to the synergistic effects from the atomically dispersed porphyrin-like Fe-N4 active sites,the heteroatom codoping (N and S),and the hierarchical porous structure in the carbon materials.The calculation based on density functional theory further indicates that the catalytic performance of Fe1/N,S-PC is better than that of Fe1/N-PC owing to the sulfur doping that yielded different rate-determining steps.Konglin Wu Xin Chen Shoujie Liu Yuan Pan Weng-Chon Cheong Wei Zhu Xing Cao Rongan Shen Wenxing Chen Jun LUO Wensheng Yan Lirong Zheng Zheng Chen Dingsheng Wang Qing Peng Chen Chen Yadong Li 2018Nano Research2018,11,12:18

14Effects of residual organics in municipal wastewater on hydrogenotrophic denitrifying microbial communities显示文摘Hydrogenotrophic denitrification is promising for tertiary nitrogen removal from municipal wastewater. To reveal the influence of residual organics in municipal wastewater on hydrogenotrophic denitrifiers, we adopted high-throughput 16 S r RNA gene amplicon sequencing to examine microbial communities in hydrogenotrophic denitrification enrichments. Using effluent from a municipal wastewater treatment plant as water source, COD,nitrate and p H were controlled the same except for a gradient of biodegradable carbon(i.e., primary effluent(PE), secondary effluent(SE), or combined primary and secondary effluent(CE)). Inorganic synthetic water(IW) was used as a control. Hydrogenophaga, a major facultative autotroph, accounted for 17.1%, 5.3%, 32.7% and 12.9% of the sequences in PE, CE,SE and IW, respectively, implicating that Hydrogenophaga grew well with or without organics.Thauera, which contains likely obligate autotrophic denitrifiers, appeared to be the most dominant genera(23.6%) in IW and accounted for 2.5%, 4.6% and 8.9% in PE, CE and SE,respectively. Thermomonas, which is related to heterotrophic denitrification, accounted for 4.2% and 7.9% in PE and CE fed with a higher content of labile organics, respectively.In contrast, Thermomonas was not detected in IW and accounted for only 0.6% in SE. Our results suggest that Thermomonas are more competitive than Thauera in hydrogenotrophic denitrification with biodegradable organics. 15Surgical treatment for both-column acetabular fractures using pre-operative virtual simulation and three-dimensional printing techniques显示文摘Background:Surgical treatment of both-column acetabular fractures is challenging because of the complex acetabular fracture patterns and the curved surface of the acetabulum.Seldom study has compared the application of three-dimensional(3D)printing technology and traditional methods of contouring plates intra-operatively for the surgical treatment of both-column acetabular fractures.We presented the use of both 3D printing technology and a virtual simulation in pre-operative planning for both-column acetabular fractures.We hypothesized that 3D printing technology will assist orthopedic surgeons in shortening the surgical time and improving the clinical outcomes.Methods:Forty patients with both-column acetabular fractures were recruited in the randomized prospective case-control study from September 2013 to September 2017 for this prospective study(No.ChiCTR1900028230).We allocated the patients to two groups using block randomization(3D printing group,n=20;conventional method group,n=20).For the 3D printing group,1:1 scaled pelvic models were created using 3D printing,and the plates were pre-contoured according to the pelvic models.The plates for the conventional method group were contoured during the operation without 3D printed pelvic models.The operation time,instrumentation time,time of intra-operative fluoroscopy,blood loss,number of times the approach was performed,blood transfusion,post-operative fracture reduction quality,hip joint function,and complications were recorded and compared between the two groups.Results:The operation and instrumentation times in the 3D printing group were significantly shorter(130.8±29.2 min,t=-7.5,P<0.001 and 32.1±9.5 min,t=-6.5,P<0.001,respectively)than those in the conventional method group.The amount of blood loss and blood transfusion in the 3D printing group were significantly lower(500[400,800]mL,Mann-Whitney U=74.5,P<0.001 and 0[0,400]mL,Mann-Whitney U=59.5,P<0.001,respectively)than those in the conventional method group.The number of the approach performed in the 3D printing group was significantly smaller than that in the conventional method group(pararectus+Kocher-Langenbeck[K-L]approach rate:35%vs.85%;x2=10.4,P<0.05).The time of intra-operative fluoroscopy in the 3D printing group was significantly shorter than that in the conventional method group(4.2±1.8 vs.7.7±2.6 s;t=-5.0,P<0.001).The post-operative fracture reduction quality in the 3D printing group was significantly better than that in the conventional method group(good reduction rate:80%vs.30%;x2=10.1,P<0.05).The hip joint function(based on the Harris score 1 year after the operation)in the 3D printing group was significantly better than that in the conventional method group(excellent/good rate:75%vs.30%;χ^2=8.1,P<0.05).The complication was similar in both groups(5.0%vs.25%;χ^2=3.1,P=0.182).Conclusions:The use of a pre-operative virtual simulation and 3D printing technology is a more effective method for treating bothcolumn acetabular fractures.This method can shorten the operation and instrumentation times,reduce blood loss,blood transfusion and the time of intra-operative fluoroscopy,and improve the post-operative fracture reduction quality.Ji-Hui Huang Hui Liao Xin-Yu Tan Wei-Rong Xing Qi Zhou Yu-Shi Zheng Hong-Yu Cao Can-Jun Zeng 2020Chinese Medical Journal2020,,4:15

18Effect of Quercetin on Atherosclerosis Based on Expressions of ABCA1, LXR-α and PCSK9 in ApoE-/- Mice显示文摘Objective:To investigate the effect of quercetin on ATP binding cassette transporter A1(ABCA1),liver X receptor(LXR),and proprotein convertase subtilisin/kexin type 9(PCSK9)expressions in apoE-knockout(ApoE-/-)mice.Methods:The high-fat diet-induced atherosclerosis(AS)in ApoE-/-mice was established.Thirtysix mice were divided into 3 groups using random number table method:model group(n=12),quercetin group(n=12),and atorvastatin group(n=12),with C57BL/6J mice of the same strain and age as the control group(n=12).Quercetin group and atorvastatin group were administrated with quercetin and atorvastatin by oral gavage,with doses of 12.5 and 4 mg/(kg・d),respectively.Animals in the control and model groups were given an equal volume of distilled water by oral gavage once per day for a total of 12 weeks.Western blot and immunohistochemical methods were employed to determine the aortic ABCA1,LXR-a and PCSK9 protein expressi on.En zyme linked imm uno sorbent assay method was used to detect the expressi on of serum total cholesterol(TC),triglyceride(TG),high density lipoprotein-cholesterol(HDL-C),low density lipoproteincholesterol(LDL-C),tumor necrosis factor-a(TNF-a),interleukin-6(IL-6),and IL-10,combined with tissue pathological exami nation.Results:ApoE-/-mice fed with a high-fat diet had no table atherosclerosis lesions,with reduced ABCA1,LXR-a and IL-10 levels(all P<0.01),elevated PCSK9,TNF-a and IL-6 expression,and increased TC and LDL-C con tents(all P<0.01).After querceti n in terventi on,the areas of AS plaques and the expressions of PCSK9,TNF-a and IL-6 were significantly reduced(all P<0.01),while the expressions of ABCA1 and LXR-a were increased significantly(all P<0.01).Conclusion:Quercetin effectively interfered with AS development by regulating the expressions of ABCA1,LXR-a and PCSK9 in ApoE,mice.LI Shan-shan CAO Hui SHEN Ding-zhu CHEN Chuan XING San-li DOU Fang-fang JIA Qing-ling 2020Chinese Journal of Integrative Medicine2020,26,2:13
19Advances in Vision-Based Lane Detection:Algorithms,Integration,Assessment,and Perspectives on ACP-Based Parallel Vision显示文摘Lane detection is a fundamental aspect of most current advanced driver assistance systems(ADASs). A large number of existing results focus on the study of vision-based lane detection methods due to the extensive knowledge background and the low-cost of camera devices. In this paper, previous visionbased lane detection studies are reviewed in terms of three aspects, which are lane detection algorithms, integration, and evaluation methods. Next, considering the inevitable limitations that exist in the camera-based lane detection system, the system integration methodologies for constructing more robust detection systems are reviewed and analyzed. The integration methods are further divided into three levels, namely, algorithm, system,and sensor. Algorithm level combines different lane detection algorithms while system level integrates other object detection systems to comprehensively detect lane positions. Sensor level uses multi-modal sensors to build a robust lane recognition system. In view of the complexity of evaluating the detection system, and the lack of common evaluation procedure and uniform metrics in past studies, the existing evaluation methods and metrics are analyzed and classified to propose a better evaluation of the lane detection system. Next, a comparison of representative studies is performed. 20Substitution of chemical fertilizer by Chinese milk vetch improves the sustainability of yield and accumulation of soil organic carbon in a double-rice cropping system显示文摘The double-rice cropping system is a very important intensive cropping system for food security in China. There have been few studies of the sustainability of yield and accumulation of soil organic carbon (SOC) in the double-rice cropping system following a partial substitution of chemical fertilizer by Chinese milk vetch (Mv). We conducted a 10-year (2008–2017) field experiment in Nan County, South-Central China, to examine the double-rice productivity and SOC accumulation in a paddy soil in response to different fertilization levels and Mv application (22.5 Mg ha^–1). Fertilizer and Mv were applied both individually and in combination (sole chemical fertilizers, Mv plus 100, 80, 60, 40, and 0% of the recommended dose of chemical fertilizers, labeled as F100, MF100, MF80, MF60, MF40, and MF0, respectively). It was found that the grain yields of double-rice crop in treatments receiving Mv were reduced when the dose of chemical fertilizer was reduced, while the change in SOC stock displayed a double peak curve. The MF100 produced the highest double-rice yield and SOC stock, with the value higher by 13.5 and 26.8% than that in the F100. However, the grain yields increased in the MF80 (by 8.4% compared to the F100), while the SOC stock only increased by 8.4%. Analogous to the change of grain yield, the sustainable yield index (SYI) of double rice were improved significantly in the MF100 and MF80 compared to the F100, while there was a slight increase in the MF60 and MF40. After a certain amount of Mv input (22.5 Mg ha^–1), the carbon sequestration rate was affected by the nutrient input due to the stimulation of microbial biomass. Compared with the MF0, the MF100 and MF40 resulted in a dramatically higher carbon sequestration rate (with the value higher by 71.6 and 70.1%), whereas the MF80 induced a lower carbon sequestration rate with the value lower by 70.1% compared to the MF0.
