2Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 3Achieving over 16% efficiency for single-junction organic solar cells显示文摘To achieve high photovoltaic performance of bulk hetero-junction organic solar cells(OSCs), a range of critical factors including absorption profiles, energy level alignment, charge carrier mobility and miscibility of donor and acceptor materials should be carefully considered. For electron-donating materials, the deep highest occupied molecular orbital(HOMO) energy level that is beneficial for high open-circuit voltage is much appreciated. However, a new issue in charge transfer emerges when matching such a donor with an acceptor that has a shallower HOMO energy level. More to this point, the chemical strategies used to enhance the absorption coefficient of acceptors may lead to increased molecular crystallinity, and thus result in less controllable phase-separation of photoactive layer. Therefore, to realize balanced photovoltaic parameters, the donor-acceptor combinations should simultaneously address the absorption spectra, energy levels, and film morphologies. Here, we selected two non-fullerene acceptors, namely BTPT-4F and BTPTT-4F, to match with a wide-bandgap polymer donor P2F-EHp consisting of an imidefunctionalized benzotriazole moiety, as these materials presented complementary absorption and well-matched energy levels. 4Intravenous injection of mesenchymal stem cells is effective in treating liver fibrosis显示文摘AIM: To compare the influence of different transplant sites in bone marrow mesenchymal stem cell (MSC)-based therapy for liver fibrosis. METHODS: MSCs isolated from Sprague Dawley (SD) rats were induced into hepatocyte-like cells. Liver fibrosis in SD rats was induced with carbon tetrachloride. Following hepatocyte induction in vitro, 4',6-diamidino- 2-phenylindole (DAPI)-labeled MSCs were transplanted by intravenous, intrahepatic, and intraperitoneal injection. Histopathological staining, immunohistochemistry, and biochemical analysis were used to compare the morphological and functional liver regeneration among different MSC injection modalities. The expression differences of interleukins, growth factor, extracellular matrix, matrix metalloproteinases, and tissue inhibitor of metalloproteinase were examined by real-time reverse transcription-polymerase chain reaction (RT-PCR) andenzyme linked immunosorbent assay (ELISA). RESULTS: Four days after exposure to hepatocyte differentiation medium, MSCs that did not express hepatocyte markers could express α-fetoprotein, albumin, and cytokeratin 18. The results of histopathological staining, immunohistochemistry, and biochemical analysis indicated that intravenous injection is more effective at rescuing liver failure than other injection modalities. DAPI-labeled cells were found around liver lobules in all three injection site groups, but the intravenous group had the highest number of cells. PCR and ELISA analysis indicated that interleukin-10 (IL-10) was highest in the intravenous group, whereas il1β, il6, tnfα and tgfβ, which can be regulated by IL10 and are promoters of liver fibrosis, were significantly lower than in the other groups. CONCLUSION: MSC administration is able to protect against liver fibrosis. 5Orbit determination for Chang'E-2 lunar probe and evaluation of lunar gravity models显示文摘The Unified S-Band(USB) ranging/Doppler system and the Very Long Baseline Interferometry(VLBI) system as the ground tracking system jointly supported the lunar orbit capture of both Chang'E-2(CE-2) and Chang'E-1(CE-1) missions.The tracking system is also responsible for providing precise orbits for scientific data processing.New VLBI equipment and data processing strategies have been proposed based on CE-1 experiences and implemented for CE-2.In this work the role VLBI tracking data played was reassessed through precision orbit determination(POD) experiments for CE-2.Significant improvement in terms of both VLBI delay and delay rate data accuracy was achieved with the noise level of X-band band-width synthesis delay data reaching 0.2-0.3 ns.Short-arc orbit determination experiments showed that the combination of only 15 min's range and VLBI data was able to improve the accuracy of 3 h's orbit using range data only by a 1-1.5 order of magnitude,confirming a similar conclusion for CE-1.Moreover,because of the accuracy improvement,VLBI data was able to contribute to CE-2's long-arc POD especially in the along-track and orbital normal directions.Orbital accuracy was assessed through the orbital overlapping analysis(2 h arc overlapping for 18 h POD arc).Compared with about 100 m position error of CE-1's 200 km 200 km lunar orbit,for CE-2's 100 km 100 km lunar orbit,the position errors were better than 31 and 6 m in the radial direction,and for CE-2's 15 km 100 km orbit,the position errors were better than 45 and 12 m in the radial direction.In addition,in trying to analyze the Delta Differential One-Way Ranging(DOR) experiments data we concluded that the accuracy of DOR delay was dramatically improved with the noise level better than 0.1 ns and systematic errors better calibrated,and the Short-arc POD tests with DOR data showed excellent results.Although unable to support the development of an independent lunar gravity model,the tracking data of CE-2 provided evaluations of different lunar gravity models through POD.It is found that for the 100 km 100 km lunar orbit,with a degree and order expansion up to 165,JPL's gravity model LP165P did not show noticeable improvement over Japan's SGM series models(100×100),but for the 15 km×100 km lunar orbit,a higher degree-order model can significantly improve the orbit accuracy.Li PeiJia Hu XiaoGong Huang Yong Wang GuangLi Jiang DongRong Zhang XiuZhong Cao JianFeng Xin Nan 2012Science China(Physics,Mechanics & Astronomy)2012,55,3:30
6A study of the efficacy of bacterial biofilm cleanout for gastrointestinal endoscopes显示文摘AIM:To compare the influence and clearance effect of enzymatic and non-enzymatic detergents against Escherichia coli (E. coli) biofilm on the inner surface of gastroscopes.METHODS:Teflon tubes were incubated in a mixture of different detergents and E. coli culture (106 CFU/mL) for 72 h at 15℃,and biofilms on the inner surface of the teflon tubes were analyzed by bacterial count and scanning electron microscopy. To evaluate the clear-ance effect of detergents,after biofilms were formed on the inner surface of Teflon tubes by 72 h lavage with E. coli culture,tubes were lavaged by enzymatic and non-enzymatic detergents at a speed of 250 mL/min,then biofilms on the inner surface were analyzed by bacterial count and scanning electron microscopy.RESULTS:Non-enzymatic detergent had a better inhi-bition function on biofilm formation than enzymatic de-tergent as it reduced bacterial burden by 2.4 log compared with the control samples (P = 0.00). Inhibition function of enzymatic detergent was not significantly different to that of control samples and reduced bac-terial burden by 0.2 log on average (P > 0.05). After lavaging at 250 mL/min for 3 min,no living bacteria were left in the tubes. Scanning electron microscopy observation showed biofi lms became very loose by the high shear force effect. CONCLUSION:Non-enzymatic detergent has a better inhibition effect on biofilm formation at room temperature. 7Late Triassic sedimentary records in the northern Tethyan Himalaya:Tectonic link with Greater India显示文摘The Upper Triassic flysch sediments(Nieru Formation and Langjiexue Group)exposed in the Eastern Tethyan Himalayan Sequence are crucial for unraveling the controversial paleogeography and paleotectonics of the Himalayan orogen.This work reports new detrital zircon U-Pb ages and whole-rock geochemical data for clastic rocks from flysch strata in the Shannan area.The mineral modal composition data suggest that these units were mainly sourced from recycled orogen provenances.The chemical compositions of the sandstones in the strata are similar to the chemical composition of upper continental crust.These rocks have relatively low Chemical Index of Alteration values(with an average of 62)and Index of Compositional Variability values(0.69),indicating that they experienced weak weathering and were mainly derived from a mature source.The geochemical compositions of the Upper Triassic strata are similar to those of graywackes from continental island arcs and are indicative of an acidicintermediate igneous source.Furthermore,hornblende and feldspar experienced decomposition in the provenance,and the sediment became enriched in zircon and monazite during sediment transport.The detrital zircons in the strata feature two main age peaks at 225-275 Ma and 500-600 Ma,nearly continuous Paleoproterozoic to Neoproterozoic ages,and a broad inconspicuous cluster in the Tonian-Stenian(800-1200 Ma).The detrital zircons from the Upper Triassic sandstones in the study area lack peaks at 300-325 Ma(characteristic of the Lhasa block)and 1150-1200 Ma(characteristic of the Lhasa and West Australia blocks).Therefore,neither the Lhasa block nor the West Australia blocks likely acted as the main provenance of the Upper Triassic strata.Newly discovered Permian-Triassic basalt and mafic dikes in the Himalayas could have provided the 225-275 Ma detrital zircons.Therefore,Indian and Himalayan units were the main provenances of the flysch strata.The Tethyan Himalaya was part of the northern passive margin and was not an exotic terrane separated from India during the Permian to Early Cretaceous.This evidence suggests that the Neo-Tethyan ocean opened prior to the Late Triassic and that the Upper Triassic deposits were derived from continental crustal fragments adjacent to the northern passive continental margin of Greater India.Huawen Cao Yong Huang Guangming Li Linkui Zhang Jianyang Wu Lei Dong Zuowen Dai Liu Lu 2018Geoscience Frontiers2018,9,1:27

9A comprehensive review of Energy Internet: basic concept,operation and planning methods, and research prospects显示文摘With the intensifying energy crisis and environmental pollution, the Energy Internet and corresponding patterns of energy use have been attracting more and more attention. In this paper, the basic concept and characteristics of the Energy Internet are summarized, and its basic structural framework is analyzed in detail. On this basis,couplings between the electric power system and other systems such as the cooling and heating system, the natural gas system, and the traffic system are analyzed, and the operation and planning of integrated energy systems in both deterministic and uncertain environments are comprehensively reviewed. 10Induction of apoptosis of human gastric carcinoma SGC-7901 cell line by 5,7-dihydroxy-8-nitrochrysin in vitro显示文摘AIM: To investigate the effect of 5, 7-dihydroxy-8- nitrochrysin (NOChR) on apoptosis of human gastric carcinoma SGC-7901 cell line. METHODS: SGC-7901 cells were cultured in vitro and the inhibitory effect of NOChR on proliferation of SGC-7901 cells was measured by using an MTT assay. NOChR-induced apoptosis rate of SGC-7901 cells was detected using flow cytometry (FCM) with PI staining. DNA ladder bands were observed by DNA agarose gel electrophoresis. The influence of NOChR on the proxisome proliferator-activated receptor-γ (PPARγ), Bcl-2 and Bax protein expression of SGC-7901 cells was analyzed by Western blot. RESULTS: MTT assay showed that NOChR markedly inhibited proliferation of SGC-7901 cells in a dose- dependent manner, and when IC50 was 4.14 μmol/L, the potency of NOChR was 10 times than that of lead compound, chrysin (ChR, IC50 was 40.56 μmol/L), and was similar to 5-fluorouracil (5-FU, IC50 was 4.51 μmol/L). FCM with propidium iodide (PI) staining demonstrated that the apoptosis rates of SGC-7901 cells treated with 1.25, 5.00 and 20.00 μmol/L NOChR for 48 h were 9.8% ± 0.2%, 36.8% ± 1.9% and 45.5% ± 3.5%, respectively, and were significantly higher when treated with 5.00 and 20.00 μmol/L NOChR than that with 20.00 μmol/L ChR (12.9% ± 1.5%). DNA agarose gel electrophoresis showed that treatment of SGC-7901 cells with 20.00 μmol/L NOChR for 48 h resulted in typical DNA ladder bands of DNA of SGC-7901 cells, which could be eliminated by treating with 10.00 μmol/L GW9662, a blocker of PPARγ. Western blot analysis revealed that after 24 h of treatment with 20.00 μmol/L NOChR, PPARgamma and Bax protein expression of SGC-7901 cells increased but Bcl-2 expression decreased; however, pre-incubation with 10.00 μmol/L GW9662 could efficiently antagonize and weaken the regulatory effect of 20.00 μmol/L NOChR on Bax and Bcl-2 protein expression of SGC-7901 cells. 13Effects of Short-Term Forest Bathing on Human Health in a Broad-Leaved Evergreen Forest in Zhejiang Province,China显示文摘Objective To investigate the effects of short-term forest bathing on human health.Methods Twenty healthy male university students participated as subjects and were randomly divided into two groups of 10.One group was sent on a two-night trip to a broad-leaved evergreen forest,and the other was sent to a city area.Serum cytokine levels reflecting inflammatory and stress response,indicators reflecting oxidative stress,the distribution of leukocyte subsets,and plasma endothelin-1 (ET-1) concentrations were measured before and after the experiment to evaluate the positive health effects of forest environments.A profile of mood states (POMS) evaluation was used to assess changes in mood states.Results No significant differences in the baseline values of the indicators were observed between the two groups before the experiment.Subjects exposed to the forest environment showed reduced oxidative stress and pro-inflammatory level,as evidenced by decreased malondialdehyde,interleukin-6,and tumor necrosis factor α levels compared with the urban group.Serum cortisol levels were also lower than in the urban group.Notably,the concentration of plasma ET-1 was much lower in subjects exposed to the forest environment.The POMS evaluation showed that after exposure to the forest environment,subjects had lower scores in the negative subscales,and the score for vigor was increased.Conclusion Forest bathing is beneficial to human health,perhaps through preventive effects related to several pathological factors.MAO Gen Xiang LAN Xiao Guang CAO Yong Bao CHEN Zhuo Mei HE Zhi Huat LV Yuan Dong WANG Ya Zhen HU Xi Lian WANG Guo Fu YAN Jing 2012Biomedical and Environmental Sciences2012,25,3:20
