1Characteristics of typhoon disturbed gaps in an old-growth tropical montane rainforest in Hainan Island, China显示文摘Disturbances that create gaps can shape the structure and function of forests. However, such disturbance regimes in Asian tropical montane rainforests remain largely unquantified. Least studied are typhoon disturbances that are attributable to climate change. We investigated gap characteristics in terms of size, age, and gapmaker to quantify the gap disturbance regimes in an intact old-growth tropical montane rainforest on Hainan Island,China. The intensity of typhoons has increased since 1949,and typhoon winds blow mostly(45.5%) from the northeast corner of Hainan Island, resulting in a higher frequency of gaps in the northeast. A total of 221 gap-makers(trees that fell to create canopy gaps) and 53 gaps were observed in a 3.16 ha old-growth rainforest. Most canopy gaps(85%)were <200 m^2. The average size of canopy gaps was smaller in the rainforest than in other tropical forests, while the average size of expanded gaps was similar to those in other tropical forests. The maximum age of gaps was 23.5 years indicating that gaps had more rapid turnover than other parts of tropical forests. The frequency distribution of gap-makers followed a lognormal distribution with a distinctive peak at three gap-makers, which was different from the inverse J-shaped curve typical of other tropical forests. Gaps were recorded mainly on slopes between 20° and 35° and wood density of gap-makers was between 0.6 and 0.7 g cm^(-3). 2Development of a widely targeted volatilomics method for profiling volatilomes in plants显示文摘Volatile organic compounds play essential roles in plant environment interactions as well as determining the fragrance of plants.Although gas chromatography-mass spectrometry-based untargeted metabolo-mics is commonly used to assess plant volatiles,it suffers from high spectral convolution,low detection sensitivity,a limited number of annotated metabolites,and relatively poor reproducibility.Here,we report a widely targeted volatilomics(WTV)method that involves using a“targeted spectra extraction”algorithm to address spectral convolution,constructing a high-coverage MS2 spectral tag library to expand volatile annotation,adapting a multiple reaction monitoring mode to improve sensitivity,and using regression models to adjust for signal drift.The newly developed method was used to profile the volatilome of rice grains.Compared with the untargeted method,the newly developed WTV method shows higher sensitivity(for example,the signal-to-noise ratio of guaicol increased from 4.1 to 18.8),high annotation coverage(the number of annotated volatiles increased from 43 to 132),and better reproducibility(the number of volatiles in quality control samples with relative standard deviation value below 30.0%increased from 14 to 92 after normalization).Using the WTV method,we studied the metabolic responses of tomato to environmental stimuli and profiled the volatilomes of different rice accessions.The results identified benzothiazole as a potential airborne signal priming tomato plants for enhanced defense and 2-nonanone and 2-heptanone as novel aromatic compounds contributing to rice fragrance.These case studies suggest that the widely targeted volatilomics method is more efficient than those currently used and may considerably promote plant volatilomics studies.Honglun Yuan Guangping Cao Xiaodong Hou Menglan Huang Pengmeng Du Tingting Tan Youjin Zhang Haihong Zhou Xianqing Liu Ling Liu Yiding Jiangfang Yufei Li Zhenhuan Liu Chuanying Fang Liqing Zhao Alisdair R.Fernie Jie Luo 2022Molecular Plant2022,15,1:2

17Upregulation of Cartilage Oligomeric Matrix Protein and Bone Morphogenetic Protein-2 May Associate with Calcific Aortic Valve Disease显示文摘Objective:Calcific aortic valve disease(CAVD)affects millions of elderly people,and there is currently no effective way to stop or slow down its progression.Therefore,exploring the pathogenesis of CAVD is very important for prevention and treatment.Cartilage oligomeric matrix protein(COMP)have important role in cell phenotype change.This study is aimed to confirm whether COMP participate in CAVD and try to find the possible mechanisms.Methods:Human aortic valve tissues from Nanjing First Hospital(CAVD group,n=20;control group,n=11)were harvested.The expression level of COMP was tested by western blot and immunohistochemistry.Dual immunofluorescence staining was used for locating COMP.Bone morphogenetic protein-2(BMP2)signalling were tested by western blot.The animal model was also used to detect COMP level by immunohistochemistry.Results:The results showed that the expression level of COMP was significantly increased in the calcific valve samples when compared with that of the control valve(P<0.05);COMP was expressed near the calcific nodules and co-localized with a-smooth muscle actin(a-SMA).The protein levels of BMP2 and p-Smads 1/5/9 were markedly more highly expressed in the CAVD group than the control group(P<0.05).Furthermore,immunofluorescence detection showed that COMP and BMP2 were co-located in calcific valves.Conclusions:The above results suggested that upregulation of COMP and BMP2 may be associated with aortic valve calcification and that COMP may become a potential therapeutic target in human CAVD.Yueyue Xu Yide Cao Yafeng Liu Jingsong Wang Ganyi Chen Zhonghao Tao Yiwei Yao Yuchen Cai Yunzhang Wu Wen Chen 2021Cardiology Discovery2021,1,2:0

19Non-invasive photoacoustic computed tomography of rat heart anatomy and function显示文摘Complementary to mainstream cardiac imaging modalities for preclinical research,photoacoustic computed tomography(PACT)can provide functional optical contrast with high imaging speed and resolution.However,PACT has not been demonstrated to reveal the dynamics of whole cardiac anatomy or vascular system without surgical procedure(thoracotomy)for tissue penetration.Here,we achieved non-invasive imaging of rat hearts using the recently developed three-dimensional PACT(3D-PACT)platform,demonstrating the regulated illumination and detection schemes to reduce the effects of optical attenuation and acoustic distortion through the chest wall;thereby,enabling unimpeded visualization of the cardiac anatomy and intracardiac hemodynamics following rapidly scanning the heart within 10 s.We further applied 3D-PACT to reveal distinct cardiac structural and functional changes among the healthy,hypertensive,and obese rats,with optical contrast to uncover differences in cardiac chamber size,wall thickness,and hemodynamics.Accordingly,3D-PACT provides high imaging speed and nonionizing penetration to capture the whole heart for diagnosing the animal models,holding promises for clinical translation to cardiac imaging of human neonates.Li Lin Xin Tong Susana Cavallero Yide Zhang Shuai Na Rui Cao Tzung K.Hsiai Lihong V.Wang 2023Light(Science & Applications)2023,12,1:0
