1Chinese consensus guidelines for diagnosis and management of gastrointestinal stromal tumor显示文摘In order to further promote the standardization of diagnosis and treatment of gastrointestinal stromal tumor(GIST) in China, the members of Chinese Society of Clinical Oncology(CSCO) Expert Committee on GIST thoroughly discussed the key contents of the consensus guidelines, and voted on the controversial issue. In final, the Chinese consensus guidelines for the diagnosis and management of GIST(2017 edition) was formed on the basis of 2013 edition consensus guidelines, which is hereby announced. 3Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. 4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5Determination of Interfacial Heat Transfer Behavior at the Metal/Shot Sleeve of High Pressure Die Casting Process of AZ91D Alloy显示文摘The interfacial heat transfer behavior at the metal/shot sleeve interface in the high pressure die casting(HPDC) process of AZ91 D alloy is carefully investigated.Based on the temperature measurements along the shot sleeve,inverse method has been developed to determine the interfacial heat transfer coefficient in the shot sleeve.Under static condition,Interfacial heat transfer coefficient(IHTC) peak values are 11.9,7.3,8.33 k W m^(-2)K^(-1)at pouring zone(S2),middle zone(S5),and end zone(S10),respectively.During the casting process,the IHTC curve displays a second peak of 6.1 k W m^(-2)K^(-1)at middle zone during the casting process at a slow speed of 0.3 m s^(-1).Subsequently,when the high speed started,the IHTC curve reached a second peak of 12.9 k W m^(-2)K^(-1)at end zone.Furthermore,under different slow casting speeds,both the calculated initial temperature(TIDS) and the maximum temperature(Tsimax) of shot sleeve surface first decrease from 0.1 m s^(-1)to 0.3 m s^(-1),but increase again from 0.3 m s^(-1)to 0.6 m s^(-1).This result agrees with the experimental results obtained in a series of 'plate-shape' casting experiments under different slow speeds,which reveals that the amount of ESCs decreases to the minimum values at 0.3 m s^(-1)and increase again with the increasing casting slow speed.Wenbo Yu Yongyou Cao Xiaobo Li Zhipeng Guo Shoumei Xiong 2017Journal of Materials Science & Technology2017,33,1:7

7Nrf2 epigenetic derepression induced by running exercise protects against osteoporosis显示文摘Osteoporosis(OP)is a common skeletal disease involving low bone mineral density(BMD)that often leads to fragility fracture,and its development is affected by multiple cellular pathologies and associated with marked epigenetic alterations of osteogenic genes.Proper physical exercise is beneficial for bone health and OP and reportedly possesses epigenetic modulating capacities;however,whether the protective effects of exercise on OP involve epigenetic mechanisms is unclear.Here,we report that epigenetic derepression of nuclear factor erythroid derived 2-related factor-2(Nrf2),a master regulator of oxidative stress critically involved in the pathogenesis of OP,mediates the significant osteoprotective effects of running exercise(RE)in a mouse model of OP induced by ovariectomy.We showed that Nrf2 gene knockout(Nfe2l2^(−/−))ovariectomized mice displayed a worse BMD reduction than the controls,identifying Nrf2 as a critical antiosteoporotic factor.Further,femoral Nrf2 was markedly repressed with concomitant DNA methyltransferase(Dnmt)1/Dnmt3a/Dnmt3b elevations and Nrf2 promoter hypermethylation in both patients with OP and ovariectomized mice.However,daily 1-h treadmill RE significantly corrected epigenetic alterations,recovered Nrf2 loss and improved the femur bone mass and trabecular microstructure.Consistently,RE also normalized the adverse expression of major osteogenic factors,including osteoblast/osteoclast markers,Nrf2 downstream antioxidant enzymes and proinflammatory cytokines.More importantly,the RE-conferred osteoprotective effects observed in the wild-type control mice were largely abolished in the Nfe2l2^(−/−)mice.Thus,Nrf2 repression due to aberrant Dnmt elevation and subsequent Nrf2 promoter hypermethylation is likely an important epigenetic feature of the pathogenesis of OP,and Nrf2 derepression is essential for the antiosteoporotic effects of RE.Xingren Chen Xiaobo Zhu Ai Wei Fang Chen Qi Gao Ke Lu Qing Jiang Wangsen Cao 2021Bone Research2021,9,2:6
8The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components.The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. 9Effect of ziprasidone on QTc interval in Chinese patients with schizophrenia显示文摘Objective: To investigate the effect of ziprasidone on QTc in Chinese patients with schizophrenia. Methods: The study involved 27 patients with schizophrenia. Ziprasidone was initiated with 40 mg/day. From day 3 to day 7, the dose was increased gradually to 120-160 mg/d according to the effectiveness and tolerability. QTc values were measured at the beginning and month 6. Results: At the beginning of the 6 months treatment, the mean QTc interval of patients was(387.5±19.0) ms. At the end of the study, it was(402.9±23.6) ms. The difference has statistically significance(P<0.05). The mean QTc interval changed significantly throughout 6-months period but no one exceed the QTc dangerous limits. Significant correlation was found between QTc interval and the dose. Conclusion: In summary, our results suggest that ziprasidone has good tolerance in patient with schizophrenia despite its dose-related QTc prolongation. 11Structural variation during dog domestication: insights from gray wolf and dhole genomes显示文摘Several processes like phenotypic evolution, disease susceptibility and environmental adaptations, which fashion the domestication of animals, are largely attributable to structural variations(SVs) in the genome.Here, we present high-quality draft genomes of the gray wolf(Canis lupus) and dhole(Cuon alpinus) with scaffold N50 of 6.04 Mb and 3.96 Mb, respectively. Sequence alignment comprising genomes of three canid species reveals SVs specific to the dog, particularly 16 315 insertions, 2565 deletions, 443 repeats, 16 inversions and 15 translocations. Functional annotation of the dog SVs associated with genes indicates their enrichments in energy metabolisms, neurological processes and immune systems. Interestingly, we identify and verify at population level an insertion fully covering a copy of the AKR1 B1(Aldo-Keto Reductase Family 1 Member B) transcript. Transcriptome analysis reveals a high level of expression of the new AKR1 B1 copy in the small intestine and liver, implying an increase in de novo fatty acid synthesis and antioxidant ability in dog compared to gray wolf, likely in response to dietary shifts during the agricultural revolution. For the first time, we report a comprehensive analysis of the evolutionary dynamics of SVs during the domestication step of dogs. 121,25-(OH)2D3 protects pancreatic beta cells against H2O2-induced apoptosis through inhibiting the PERK-ATF4-CHOP pathway显示文摘Endoplasmic reticulum(ER)stress plays a critical role in pancreaticβcell destruction which leads to the pathogenesis of type 1 diabetes mellitus(T1DM).Vitamin D(VD)has been reported to reduce the risk of T1DM;however,it remains unknown whether VD affects ER stress in pancreaticβcells.In this study,we investigated the role of the active form of VD,1,25-dihydroxyvitamin D3[1,25-(OH)2D3],in ER stress-inducedβcell apoptosis and explored its potential mechanism in mouse insulinoma cell line mouse insulinoma 6(MIN6).The results of cell counting kit-8(CCK8)and flow cytometric analyses showed that 1,25-(OH)2D3 caused a significant increase in the viability of MIN6 cells injured by H2O2.The protein kinase like ER kinase(PERK)signal pathway,one of the most conserved branches of ER stress,was found to be involved in this process.H2O2 activated the phosphorylation of PERK,upregulated the activating transcription factor 4(ATF4)and C/EBP homologous protein(CHOP)expression,and subsequently initiated cell apoptosis,which were significantly reversed by 1,25-(OH)2D3 pretreatment.In addition,GSK2606414,a specific inhibitor of PERK,suppressed PERK phosphorylation and reduced the expressions of ATF4 and CHOP,leading to a significant decrease inβcell apoptosis induced by H2O2.Taken together,the present findings firstly demonstrated that 1,25-(OH)2D3 could prevent MIN6 cells against ER stress-associated apoptosis by inhibiting the PERK-ATF4-CHOP pathway.Therefore,our results suggested that 1,25-(OH)2D3 might serve as a potential therapeutic target for preventing pancreaticβcell destruction in T1DM.Xiaobo Hu Cong Hu Jun Liu Zhuan Wu Tingting Duan Zhaohui Cao 2021Acta Biochimica et Biophysica Sinica2021,53,1:3
13Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 14Bias correction of sea surface temperature retrospective forecasts in the South China Sea显示文摘Offline bias correction of numerical marine forecast products is an effective post-processing means to improve forecast accuracy. Two offline bias correction methods for sea surface temperature(SST) forecasts have been developed in this study: a backpropagation neural network(BPNN) algorithm, and a hybrid algorithm of empirical orthogonal function(EOF) analysis and BPNN(named EOF-BPNN). The performances of these two methods are validated using bias correction experiments implemented in the South China Sea(SCS), in which the target dataset is a six-year(2003–2008) daily mean time series of SST retrospective forecasts for one-day in advance, obtained from a regional ocean forecast and analysis system called the China Ocean Reanalysis(CORA),and the reference time series is the gridded satellite-based SST. The bias-correction results show that the two methods have similar good skills;however, the EOF-BPNN method is more than five times faster than the BPNN method.
