2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation显示文摘ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. 5Terrestrial carbon sinks in China and around the world and their contribution to carbon neutrality显示文摘Enhancing the terrestrial ecosystem carbon sink(referred to as terrestrial C sink) is an important way to slow down the continuous increase in atmospheric carbon dioxide(CO_(2)) concentration and to achieve carbon neutrality target.To better understand the characteristics of terrestrial C sinks and their contribution to carbon neutrality,this review summarizes major progress in terrestrial C budget researches during the past decades,clarifies spatial patterns and drivers of terrestrial C sources and sinks in China and around the world,and examines the role of terrestrial C sinks in achieving carbon neutrality target.According to recent studies,the global terrestrial C sink has been increasing from a source of (-0.2±0.9) Pg C yr^(-1)(1 Pg=1015g)in the 1960s to a sink of (1.9±1.1) Pg C yr^(-1) in the 2010s.By synthesizing the published data,we estimate terrestrial C sink of 0.20–0.25 Pg C yr^(-1) in China during the past decades,and predict it to be 0.15–0.52 Pg C yr^(-1) by 2060.The terrestrial C sinks are mainly located in the mid-and high latitudes of the Northern Hemisphere,while tropical regions act as a weak C sink or source.The C balance differs much among ecosystem types:forest is the major C sink;shrubland,wetland and farmland soil act as C sinks;and whether the grassland functions as C sink or source remains unclear.Desert might be a C sink,but the magnitude and the associated mechanisms are still controversial.Elevated atmospheric CO_(2) concentration,nitrogen deposition,climate change,and land cover change are the main drivers of terrestrial C sinks,while other factors such as fires and aerosols would also affect ecosystem C balance.The driving factors of terrestrial C sink differ among regions.Elevated CO_(2) concentration and climate change are major drivers of the C sinks in North America and Europe,while afforestation and ecological restoration are additionally important forcing factors of terrestrial C sinks in China.For future studies,we recommend the necessity for intensive and long-term ecosystem C monitoring over broad geographic scale to improve terrestrial biosphere models for accurately evaluating terrestrial C budget and its dynamics under various climate change and policy scenarios.Yuanhe Yang Yue Shi Wenjuan Sun Jinfeng Chang Jianxiao Zhu Leiyi Chen Xin Wang Yanpei Guo Hongtu Zhang Lingfei Yu Shuqing Zhao Kang Xu Jiangling Zhu Haihua Shen Yuanyuan Wang Yunfeng Peng Xia Zhao Xiangping Wang Huifeng Hu Shiping Chen Mei Huang Xuefa Wen Shaopeng Wang Biao Zhu Shuli Niu Zhiyao Tang Lingli Liu Jingyun Fang 2022Science China(Life Sciences)2022,65,5:32
6Rock burst mechanism analysis in an advanced segment of gob-side entry under different dip angles of the seam and prevention technology显示文摘In order to investigate the frequent occurrences of rock burst in gob-side entry during the mining process of the mining zone No. 7, the mechanical model of main roof of fully-mechanized caving mining before breaking was established by the Winkler foundation beam theory, and the stress evolution law of surrounding rock with different dip angles of the seam during the mining process was analyzed by using FLAC3 D. The results show that: with the dip angle changing from 45° to 0°, the solid-coal side of gobside entry begins to form an L-shaped stress concentration zone at a dip angle of 30°, and the stress concentration degree goes to higher and higher levels. However, the stress concentration degree of the coalpillar side goes to lower and lower levels; the influence range and peak stress of the abutment at the lateral strata of adjacent gob increase with dip angle decreasing and reach a maximum value at a dip angle of 0°, but the tailgate is not affected; the abutment pressure superposition of two adjacent gobs leads to stress concentration further enhancing in both sides of gob-side entry. With the influence of strong mining disturbance, rock burst is easily induced by dynamic and static combined load in the advanced segment of gob-side entry. To achieve stability control similar to that in the roadway, the key control strategy is to reinforce surrounding rock and unload both sides. Accordingly, the large-diameter drilling and high-pressure water injection combined unloading and reinforced support cooperative control technology was proposed and applied in field test. 7BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 9The proliferation, differentiation, and mineralization effects of puerarin on osteoblasts in vitro显示文摘AIM: Osteoblasts are key functional cells in the process of bone metabolic balance. Phytoestrogens have an important influence on the proliferation and differentiation of osteoblasts. Puerarin, a plant estrogen, has a wide range concentration in vitro on the function of osteoblasts. The current study investigates the effect of the phytoestrogen puerarin on the proliferation, differentiation, and mineralization of osteoblasts in vitro. METHODS: The calvaria bone of eight-ten Wistar rats which were born within 24 h were obtained in aseptic condition. After enzyme digestion, isolation, purified osteoblasts of rats were cultured for further study. The cells of the first to third generation were divided into a control group and a puerarin-treated group with 10-3–10-10 mol·L-1 puerarin. The cells were exposed to the medium containing a low level of carbohydrates, 10%(V/V) FBS for 24 h. After 1 to 4 days of culture, the OD values on the proliferation of osteoblasts in each group were determined by microplate reader. The cells were cultured in the medium containing 50 μg·mL-1 vitamin C, 10-2 mol·L-1 sodium glycerophosphate, 10% FBS and the medium was changed every 3 to 4 days. After 2 to 8 days of culture, expression of alkaline phosphatase were tested and compared by microplate reader. The mineral nodes of osteoblasts were dyed using alizarin red or improved Von Kossa way after four weeks. RESULTS: Compared with those in the 10-5–10-9 mol·L-1 puerarin, the proliferation of osteoblasts, the expression of alkaline phosphatase, and the number of mineral nodes of osteoblasts were significantly decreased in the control group. The increase was the fastest in the third day, while on the fourth day it was decreased, and arrived at statistical significance compared with the alkaline phosphatase activities and control group. The 10-6 mol·L-1 group was the most distinct, and formed the most mineralized nodule. Compared with the 10-3 mol·L-1 puerarin group, those changes were markedly increased in the control group. CONCLUSIONS: Puerarin has proliferation, differentiation, and mineralization effects on osteoblasts in a dose-dependent manner, and has a double-way effect on the osteoblasts in vitro. 12Distribution of C_4 plants along the Northeast China transect and its correlation to the environmental factors显示文摘The distribution of C4 plants and the C4/C3 ratio along Northeast China transect (NECT) have been studied. A model to quantify their relation with environmental factors has been set up. 14Debris Flow Susceptibility Assessment Using a Probabilistic Approach:A Case Study in the Longchi Area, Sichuan Province, China显示文摘The Longchi area with the city of Dujiangyan, in the Sichuan province of China, is composed of Permian stone and diorites and Triassic sandstones and mudstones intercalated with slates. An abundance of loose co-seismic materials were present on the slopes after the May 12, 2008 Wenchuan earthquake, which in later years served as source material for rainfall-induced debris flows or shallow landslides. A total of 48 debris flows, all triggered by heavy rainfall on 13 th August 2010, are described in this paper. Field investigation, supported by remote sensing image interpretation, was conducted to interpret the co-seismic landslides in the debris flow gullies. Specific characteristics of the study area such as slope, aspect, elevation, channel gradient, lithology, and gully density were selected for the evaluation of debris flow susceptibility. A score was given to all the debris flow gullies based on the probability of debris flow occurrence for the selected factors. In order to get the contribution of the different factors, principal component analyses were applied. A comprehensive score was obtained for the 48 debris flow gullies which enabled us to make a susceptibility map for debris flows with three classes. 18The OsAMT1.1 gene functions in ammonium uptake and ammoniumepotassium homeostasis over low and high ammonium concentration ranges显示文摘Rice(Oryza sativa) grown in paddy fields is an ammonium(NH^+_4)-preferring crop;however,its AMT-type NH^+_4transporters that mediate root N acquisition have not been well characterized yet.In this study,we analyzed the expression pattern and physiological function of the OsAMT1.1 gene of the AMT1 subfamily in rice.Os AMT1.1 is located in the plasma membrane and is mainly expressed in the root epidermis,stele and mesophyll cells.Disruption of the Os AMT1.1 gene decreased the uptake of NH^+_4,and the growth of roots and shoots under both low NH^+_4and high NH^+_4conditions.Os AMT1.1 contributed to the short-term(5 min)^(15)NH^+_4influx rate by approximately one-quarter,irrespective of the NH^+_4concentration.Knockout of Os AMT1.1 significantly decreased the total N transport from roots to shoots under low NH^+_4conditions.Moreover,compared with the wild type,the osamt1.1 mutant showed an increase in the potassium(K)absorption rate under high NH^+_4conditions and a decrease under low NH^+_4conditions.The mutants contained a significantly high concentration of K in both the roots and shoots at a limited K(0.1 mmol/L)supply when NH^+_4was replete.Taken together,the results indicated that OsAMT1.1 significantly contributes to the NH^+_4uptake under both low and high NH^+_4conditions and plays an important role in Ne K homeostasis in rice.Chang Li Zhong Tang Jia Wei Hongye Qu Yanjie Xie Guohua Xu 2016Journal of Genetics and Genomics2016,43,11:12
19Detection for disease tipping points by landscape dynamic network biomarkers显示文摘A new model-free method has been developed and termed the landscape dynamic network biomarker(l-DNB) methodology. The method is based on bifurcation theory, which can identify tipping points prior to serious disease deterioration using only single-sample omics data. Here, we show that l-DNB provides early-warning signals of disease deterioration on a single-sample basis and also detects critical genes or network biomarkers(i.e. DNB members) that promote the transition from normal to disease states. As a case study, l-DNB was used to predict severe influenza symptoms prior to the actual symptomatic appearance in influenza virus infections. The l-DNB approach was then also applied to three tumor disease datasets from the TCGA and was used to detect critical stages prior to tumor deterioration using an individual DNB for each patient. The individual DNBs were further used as individual biomarkers in the analysis of physiological data, which led to the identification of two biomarker types that were surprisingly effective in predicting the prognosis of tumors. 20Covalently closed-circular hepatitis B virus DNA reduction with entecavir or lamivudine显示文摘AIM: To investigate the reduction in hepatitis B virus(HBV) covalently closed-circular DNA(ccc DNA) with entecavir(ETV) or lamivudine(LAM). METHODS: This analysis included patients who had participated in the randomized Phase Ⅲ study ETV-022 comparing ETV vs LAM in nucleos(t)ide-naive, HBe Agpositive patients. Patients received ETV(0.5 mg daily) or LAM(100 mg daily) for a minimum of 52 wk. Patients were eligible to participate in this sub-study if they had paired biopsies at baseline and week 48 with evaluable measurements for hepatic HBV ccc DNA and total hepatic HBV DNA. The main objective was to compare changes in hepatic HBV ccc DNA and total hepatic HBV DNA at week 48 of ETV or LAM treatment, which was a secondary endpoint of study ETV-022. Additional post hoc analyses included linear regression analyses to assess associations of baseline levels and on-treatment changes of ccc DNA with other baseline factors [sex,age, serum HBV DNA, alanine aminotransferase(ALT), Knodell necroinflammatory score, Ishak fibrosis score, total hepatic HBV DNA, and HBV genotype], or ontreatment factors(changes from baseline at week 48 in serum HBV DNA, ALT, Knodell necroinflammatory score, Ishak fibrosis score, total hepatic HBV DNA, and HBe Ag loss at week 48).RESULTS: Overall, 305 patients(ETV = 159; LAM = 146) of ETV-022 had paired baseline and week 48 liver biopsies with evaluable measurements for hepatic HBV ccc DNA and total hepatic HBV DNA, and were included in this analysis. Baseline demographics and disease characteristics were comparable between the two arms. After 48 wk, ETV resulted in significantly greater reductions in hepatic HBV ccc DNA [-0.9 log10 copies/human genome equivalent(HGEq) vs-0.7 log10 copies/HGEq; P = 0.0033] and total hepatic DNA levels(-2.1 log10 copies/HGEq vs-1.6 log10 copies/HGEq; P < 0.0001) than LAM. Virologic, biochemical, and histologic response rates at week 48 were also greater with ETV than with LAM. Baseline HBV ccc DNA levels were positively associated with baseline levels of serum HBV DNA and total hepatic HBV DNA, and negatively associated with HBV genotype F.
