1Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 2Enhancement of innate immune system in monocot rice by transferring the dicotyledonous elongation factor Tu receptor EFR显示文摘The elongation factor Tu(EF-Tu) receptor(EFR) in cruciferous plants specifically recognizes the N-terminal acetylated elf18 region of bacterial EF-Tu and thereby activates plant immunity.It has been demonstrated that Arabidopsis EFR confers broad-spectrum bacterial resistance in the EFR transgenic solanaceous plants.Here,the transgenic rice plants(Oryza sativa L.ssp.japonica cv.Zhonghua 17) and cell cultures with constitutive expression of AtEFR were developed to investigate whether AtEFR senses EF-Tu and thus enhances bacterial resistance in the monocot plants.We demonstrated that the Xanthomonas oryzae-derived elf18 peptide induced oxidative burst and mitogen-activated protein kinase activation in the AtEFR transgenic rice cells and plants,respectively.Pathogenesis-related genes,such as OsPBZ1,were upregulated dramatically in transgenic rice plant and cell lines in response to elf18 stimulation.Importantly,pretreatment with elf18 triggered strong resistance to X.oryzae pv.oryzae in the transgenic plants,which was largely dependent on the AtEFR expression level.These plants also exhibited enhanced resistance to rice bacterial brown stripe,but not to rice fungal blast.Collectively,the results indicate that the rice plants with heterologous expression of AtEFR recognize bacterial EF-Tu and exhibit enhanced broad-spectrum bacterial disease resistance and that pattern recognition receptor-mediated immunity may be manipulated across the two plant classes,dicots and monocots.Fen Lu Huiqin Wang Shanzhi Wang Wendi Jiang Changlin Shan Bin Li Jun Yang Shiyong Zhang Wenxian Sun 2015Journal of Integrative Plant Biology2015,57,7:6

4Neuro-protective effects of CNTF on hippocampal neurons via an unknown signal transduction pathway显示文摘In our previous study, we proposed that there may be an unknown pathway in the upper stream of the known signal transduction pathway of Ciliary neurotrophic factor (CNTF) that mediates the neuro-protective function of CNTF. In the present experiment, we observed that the neuro-protective function of the non-classic signal transduction path- way in a L-NMDA (a glutamic acid ion type receptor atagonist) induced hippocampal neuron injury model, using primary culture rat hippocampal neurons, con- tinuous photography and gp130 immunohistochemi- cal assay. The results showed that L-NMDA induced injurious reaction of hippocampal neurons, and CNTF was able to inhibit the toxic action of L-NMDA on hippocampal neurons. Additionally, when JAK/STATs in the known classic signal transduction pathway of CNTF were blocked by PTPi-2, the protective effect of CNTF against L-NMDA injury still existed. L-NMDA caused a rapid increase in the concentration of hip- pocampal intracellular free [Ca2+]i. 7The new sideway of CNTF sig-nal transduction pathway显示文摘The action of ciliary neurotrophic factor (CNTF) on intercellular free Ca2+ concentrations [Ca2+]1 induced by glutamate (Glu) in primary cultured hippocampal neurons were detected with Fura2/AM, a Ca2+-sensitive fluorophore. and the morphological influence of G-protein on it was ob-jected. Glu could induce rapid increase of [Ca2+]1 in hippo-campal neurons. CNTF had no significant action on [Ca2+]1 in resting hippocampal neurons. However, after incubation of CNTF for 5 min, the increase of [Ca2+]1 in hippocampal neurons rapidly induced by Glu was inhibited. Pretussis toxin (PTX)-sensitive G protein could block the action. 8Expression of cyclin-dependent protein kinase 5 in the hippocampus of vascular dementia mice after cerebral ischemia and reperfusion显示文摘BACKGROUND:The p25-activated cyclin-dependent protein kinase 5(Cdk5) may induce neuronal cell death and cause the development of dementia following cerebral ischemia and reperfusion. OBJECTIVE:To observe changes in the expression of Cdk5 and p25 in hippocampal tissue of vascular dementia mice at different time points following cerebral ischemia and reperfusion. DESIGN,TIME AND SETTING:A randomized,controlled animal experiment was performed in the clinical trial center of Hebei Provincial People's Hospital between September 2007 and October 2008. MATERIALS:Cdk5 rabbit anti-mouse polyclonal antibody,p35 rabbit anti-mouse polyclonal antibody,andβ-actin mouse monoclonal antibody were purchased from Santa Cruz Biotechnology, Inc.,USA;horseradish peroxidase-labeled goat anti-rabbit IgG and horseradish peroxidase-labeled goat anti-mice IgG were offered by Beijing Zhongshan Goldenbridye Biotechnology Co.,Ltd.,China; the protein quantitative kit was produced by Applygen Gene Technology Corp.,Beijing,China;cDNA reverse transcription and PCR amplification reagents were products of TianGen&Biotech(Beijing) Co.,Ltd.,China. METHODS:One hundred and sixty male Kunming mice were randomly divided into two groups:a sham-operated group(n=65) and a model group(n=95).Vascular dementia was induced with three periods of transient ischemia and reperfusion of the bilateral common carotid arteries.In the sham-operated group,the bilateral common carotid arteries were not blocked. MAIN OUTCOME MEASURES:Behavioral tests were done at four and six weeks post surgery. Pathological changes in the hippocampal CA1 region were observed with hematoxylin-eosin staining Cdk5 mRNA expression was examined by RT-PCR,and Western blots were used to evaluate Cdk5 and p25 expression.Learning and memory performance were assayed using the Morris water maze. RESULTS:Vascular dementia reduced learning and memory performance at 4 and 6 weeks post surgery.Vascular dementia also caused severe,time-dependent neuronal damage and death in the hippocampal CA1 region.Dementia induction also increased mRNA and protein expression of Cdk5 and p25 at both 4 and 6 weeks after surgery. 12Investigation of the 12 orientations variants of nanoscale Al precipitates in eutectic Si of Al-7Si-0.6Mg alloy显示文摘Various orientations and diffraction patterns from nanoscale Al precipitates in eutectic Si were investigated by high-resolution transmission electron microscopy combined with transition matrix and stereographic projection.It was found that the Al precipitates had 12 variants,all orientation relationships can be expressed as:(001)Al//{111}Si,[110]Al//<110>Si.Further,a new diffraction pattern model from Al precipitates was established under[111]Si zone axis,which was in good agreement with the experiment data.The microstructure,adhesion strength and electronic structure of the interface between Al precipitate and Si matrix were studied by first-principles calculation and experimental observation.The results show that the covalent bonds are formed between interfacial Al and Si atoms,which play a key role in interfacial bind strength.Based on the Griffith fracture theory,the cracks tend to form and expand in the interior of Al precipitates firstly,and the interfaces can act as a protective layer to prevent crack propagation.Therefore,the nanoscale Al precipitates will improve the toughness of eutectic Si particles by releasing part of stress through lattice distortion.In addition,the stretched nanoscale Al precipitates can act as effective heterogeneous nucleation sites for high density deformation nanotwins in eutectic Si during deformation,which significantly improved the deformability of eutectic Si.Yifan Wang Yanli Lu Jing Zhang Wenchao Yang Changlin Yang Pan Wang Xiaoqing Song Zheng Chen 2021Journal of Materials Science & Technology2021,,8:1
13Different therapeutic proportion of patients undergone coronary angiography in the era of development in MSCT显示文摘Objective To study the different therapeutic proportion of the patient populations undergone coronary angiography (CAG) in the era of development in multislice spiral computed tomography(MSCT). Methods Two hundred and fifty four consecutive patients(mean age 59.24±10.65) , who underwent CAG at Daxing Hospital from February 2007 through October 2007, were enrolled, 160 patients were male and 94 were female. By evaluating from the coronary angiogram, the patients were not diagnosed to have coronary heart disease(CHD) with less than 50% diameter stenosis of coronary artery; the patients to have CHD with more than or equal to 50% stenosis of coronary artery; the patients were performed the procedure of percutaneous coronary intervention(PCI) with more than or equal to 70% stenosis; the patients were proposed to have coronary aortic bypass graft(CABG) surgery with left main coronary artery lesions or diffuse triple coronary artery lesions. Results In the 254 consecutive patients, 59 patients(23.2%) had not been diagnosed to have CHD; 195(76.8%)to have CHD, of these patients with CHD, 49 patients(19.3%)were not indicated for PCI (including the patients receiving follow-up coronary angiography after stenting), 81(31.9%)had been performed the procedure of stent implantation, 57(22.4%)proposed to have CABG, 8(3.1%)the procedure of PCI had not been successful, or had not been performed because of patients opposing to this therapy. 15Fluorescence lifetime based distance measurement illustrates conformation changes of PYL10-CL2 upon ABA binding in solution state显示文摘Forster resonance energy transfer(FRET)is a widely used distance measurement method to illustrate protein conformational dynamics.The FRET method relies on the distance between donor and acceptor,as well as the labelling efficiency,the size and the properties of the fluorophores.Here,we labelled a pair of small fluorophores and calculated the energy transferred efficiency through fluorescence lifetime analysis,which can provide more reliable distance measurement than intensity attenuation.The donor fluorophore,7-hydroxycoumarin-4-yl-ethylglycine(HC),was genetically incorporated into specific sites of PYL10,obtaining complete labelling efficiency.The acceptor fluorophore,Alexa488,was labelled through the disulfide bond,whose labelling efficiency was estimated through both absorption peaks and lifetime populations.Fluorescence lifetime and anisotropy analysis showed ABA-induced local conformation changes and dynamics of several HC incorporation sites of PYL10.The lifetime-based FRET distance measurement illustrated the conformation changes of PYL10 with or without ABA application,which is consistent with the previously reported crystal structures.Peng Zhou Pei Lv Lu Yu Sanling Liu Longhua Zhang Changlin Tian 2019Chinese Chemical Letters2019,30,5:0

19A direct spino-cortical circuit bypassing the thalamus modulates nociception显示文摘Nociceptive signals are usually transmitted to layer 4 neurons in somatosensory cortex via the spinothalamic-thalamocortical pathway.The layer 5 corticospinal neurons in sensorimotor cortex are reported to receive the output of neurons in superficial layers;and their descending axons innervate the spinal cord to regulate basic sensorimotor functions.Here,we show that a subset of layer 5 neurons receives spinal inputs through a direct spino-cortical circuit bypassing the thalamus,and thus define these neurons as spino-cortical recipient neurons(SCRNs).Morphological studies revealed that the branches from spinal ascending axons formed a kind of disciform structure with the descending axons from SCRNs in the basilar pontine nucleus(BPN).Electron microscopy and calcium imaging further confirmed that the axon terminals from spinal ascending neurons and SCRNs made functional synaptic contacts in the BPN,linking the ascending sensory pathway to the descending motor control pathway.Furthermore,behavioral tests indicated that the spino-cortical connection in the BPN was involved in nociceptive responses.In vivo calcium imaging showed that SCRNs responded to peripheral noxious stimuli faster than neighboring layer 4 cortical neurons in awake mice.Manipulating activities of SCRNs could modulate nociceptive behaviors.Therefore,this direct spino-cortical circuit represents a noncanonical pathway,allowing a fast sensory-motor transition of the brain in response to noxious stimuli.Bing Cai Dan Wu Hong Xie Yan Chen Huadong Wang Sen Jin Yuran Song Anan Li Shiqi Huang Sashuang Wang Yingjin Lu Lan Bao Fuqiang Xu Hui Gong Changlin Li Xu Zhang 2023Cell Research2023,33,10:0
