1Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192
2Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. 3Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 5Using nerve segment insert grafting to reconstruct neural pathways of brain-derived paralysis显示文摘Since 1992,task groups have used free nerve auto grafts to bridge partially transected nerves from the dominant area of the normal cerebral cortex to nerves that innervate spastic muscles from a diseased cerebral cortex,using transplanting sutures to alleviate the muscle spasm of cerebral palsy caused by different diseases. This has facilitated rebuilding of some of its neurological function. In this study,80 such patients were followed up,including 20 patients with traumatic brain injury,32 patients with stroke,and 28 pediatric patients with cerebral palsy. 6Investigation of high extraction efficiency flip-chip GaN-based light-emitting diodes显示文摘In order to obtain higher light output power, the flip-chip structure is used. We studied the ratio of the light of GaN sides before and after fabricating metal reflector on p-GaN. The SiO2/SiNx dielectric film reflectors were deposited through plasma enhance chemical vapor deposition following the fabrication of metal reflector, and then the dielectric film reflectors on the electrodes were etched in order to expose the electrodes to the air. It is found that comparing with the flip-chip GaN-LED without dielectric film reflectors, light output power can be increased by as high as 10.2% after the deposition of 2 pairs of SiO2/SiNx dielectric film reflectors on GaN-LEDs, which cover the sidewalls and the areas without the metal reflector. 7Single‐atomic Co‐B_(2)N_(2)sites anchored on carbon nanotube arrays promote lithium polysulfide conversion in lithium-sulfur batteries显示文摘Due to low cost,high capacity,and high energy density,lithium–sulfur(Li–S)batteries have attracted much attention;however,their cycling performance was largely limited by the poor redox kinetics and low sulfur utilization.Herein,predicted by density functional theory calculations,single‐atomic Co‐B2N2 site‐imbedded boron and nitrogen co‐doped carbon nanotubes(SA‐Co/BNC)were designed to accomplish high sulfur loading,fast kinetic,and long service period Li–S batteries.Experiments proved that Co‐B2N2 atomic sites can effectively catalyze lithium polysulfide conversion.Therefore,the electrodes delivered a specific capacity of 1106 mAh g−1 at 0.2 C after 100 cycles and exhibited an outstanding cycle performance over 1000 cycles at 1 C with a decay rate of 0.032%per cycle.Our study offers a new strategy to couple the combined effect of nanocarriers and single‐atomic catalysts in novel coordination environments for high‐performance Li–S batteries.Zhifeng Wang Yajing Yan Yongguang Zhang Yanxu Chen Xianyun Peng Xin Wang Weimin Zhao Chunling Qin Qian Liu Xijun Liu Zhongwei Chen 2023Carbon Energy2023,5,11:1

18Chemical characters and protective effect of Baqi Lingmao formula(巴芪灵猫方) on experimental liver injury显示文摘OBJECTIVE:To investigate the chemical characters of water-extract of Baqi Lingmao formula(巴芪灵猫方,BQLM formula)and its effects on anti-liver injury in model mice and live cells.METHODS:BQLM formula was composed of ten herbal medicines.We determined the contents of alkaloids,saponins,phenolic acids and flavonoid in BQLM formula by UV spectrophotometry.The active components of alkaloids and phenolic acids in BQLM formula were identified by HPLC chromatography.The anti-hepatic injury effects of BQLM formula were investigated with concanavalin A(Con A)-induced hepatitis model of mice,human liver LO2 and Hep G2.2.15 cells.RESULTS:BQLM formula(2 and 10 g/kg,orally)significantly improved the damages of liver tissues and functions caused by Con A in mice,reduced the infiltration of inflammatory cells into liver and inhibited the inflammatory cytokine secretion of interferon-γ,tumor necrosis factor-αand interleukin-6.BQLM formula simultaneously decreased the levels of alanine aminotransferase and aspartate aminotransferase of liver and serum,and recovered the superoxide dismutase and catalase activities of liver to normal levels in Con A-induced hepatic-injury mice.The serum of BQLM formula group stimulated the human liver LO2 cell proliferation in vitro.Further,BQLM formula obviously promoted the proliferation of normal hepatocytes(LO2 cells)and inhibited the hepatocytes death induced by Con A.It also significantly inhibited the proliferation of Hep G2.2.15 cells and decreased the secretion of HBs Ag and HBe Ag in vitro.CONCLUSIONS:BQLM formula has anti-inflammation and anti-hepatitis virus Beffects,and is capable of improving liver injury in vivo and in vitro.LIU Lulu CHEN Yanxu LIU Lulu LI Man XU Rui QU Wensheng YIN Jiye ZHANG Xin ZHOU Zhenhua GAO Yating NIE Dan SHAN Junjie GAO Yueqiu 2021Journal of Traditional Chinese Medicine2021,41,6:0

20Heavy metal gradients from rural to urban lakes in central China显示文摘Background:Limited information is available on heavy metal patterns in lakes under rapid watershed urbanization,especially considering a large spatial gradient with a long linear distance and great variations in topographic relief.To fill this gap,we studied concentrations of a series of heavy metals in both water and sediments from 20 lakes along a rural to urban gradient in central China,and we aimed to understand the effects of urban processes on heavy metal dynamics in lake ecosystems.Studied lakes were divided into five groups:A(rural reservoir group),B(rural commercial fishing group),C(urban park group),D(urban recreational fishing group),and E(urban commercial fishing group).An inductively coupled plasma optical emission spectrometer(ICP-OES)and an inductively coupled plasma-mass spectrometer(ICP-MS)were used to analyze the heavy metals in water and sediments.Results:An increasing trend of most heavy metals in water from rural to urban lakes was observed.Concentrations of cadmium(Cd),cobalt(Co),lead(Pb),chromium(Cr),arsenic(As),nickel(Ni),magnesium(Mn),iron(Fe),and aluminum(Al)in water were significantly lower in rural group A than those in other groups.Arsenic in sediments of rural group A was lower than those in other groups.No other heavy metal element in sediments was significantly different among groups.The enrichment factor analysis of selected heavy metals showed there were different degrees of enrichments of heavy metals in sediments.The potential ecological risk index showed a low level for heavy metals in sediments of all studied lakes.Conclusions:Results indicated that urban processes could have an impact on heavy metals in lake water.The sources of heavy metals in sediments were more likely from anthropogenic activities.These results could enhance our understanding of metal dynamics in lake ecosystems under urbanization and could help prevent heavy metal pollutions and promote sustainable management of urban ecosystems.Wentong Xia Rui Wang Bin Zhu Lars G.Rudstam Yinglong Liu Yanxue Xu Wei Xin Yushun Chen 2020Ecological Processes2020,9,1:0
