2A Possible Dynamic Mechanism for Rapid Production of the Extreme Hourly Rainfall in Zhengzhou City on 20 July 2021显示文摘In this study,the unprecedented extreme rainfall event during 19-20 July 2021,which caused devastating flooding in Zhengzhou City and its nearby areas,is examined based on observational data analysis and WRF model 40-h simulations on 1-km horizontal resolution.The results show that the model successfully reproduces(i)major synopticscale weather systems(i.e.,the western Pacific subtropical high,the Tibetan high,two typhoons,and the Huang-Huai cyclone),(ii)convective initiation along the east to north edge of the Songshan Mountain,where orographic lifting is obvious,and(iii)subsequent formation of the convective storm producing the extreme rainfall in Zhengzhou.In particular,the model generates the maximum rainfall rate of 233 mm h^(-1)and 40-h accumulated rainfall of 704 mm,corresponding well to the observed extreme values of 201.9 mm h^(-1)and 818 mm,at nearly observed timing and location.Importantly,the model reproduces an intense quasi-stationary,well-organized meso-γ-scale convective system,surrounded by an arc-shaped convergence zone,allowing the development of convective updrafts in a three-quarter circle around the convective system,in a way similar to“multidirectional pumping,”attracting all associated precipitation overlaid and concentrated into the same trailing region to generate the extreme hourly rainfall over Zhengzhou.Our study emphasizes the significant contribution of the unique dynamic structure of the well-organized meso-γ-scale convective system to the record-high hourly rainfall.A possible dynamic mechanism for short-time extreme rainfall production is proposed.That is,the arc-shaped convergence zone of the mesoscale convective system,acting like multidirectional lifting pumps,transports precipitation from different directions into the same region,and thus produces the extreme rainfall.The results gained herein may shed new light on better understanding and forecasting of short-time extreme rainfall.Jinfang YIN Haodong GU Xudong LIANG Miao YU Jisong SUN Yanxin XIE Feng LI Chong WU 2022Journal of Meteorological Research2022,36,1:17
3Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). 6CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination.Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li 2019Cell Research2019,29,4:12

8Damage of nasal mucociliary movement after intensity-modulated radiation therapy of nasopharyngeal carcinoma显示文摘Background and Objective:Radiation usually results in paranasal sinusitis in patients with nasopharyngeal carcinoma (NPC), which influences patients' quality of life.This study aimed to determine the relationships between dose distribution in the nasal cavity and nasal mucous injury in patients with NPC treated by intensity-modulated radiation therapy (IMRT), and to find the tolerable radiation dose for the nasal mucous.Methods:Sixty-six patients with NPC treated by IMRT between October 2006 and November 2008 were enrolled.The irradiation dose in the nasal cavity was determined by the computer with the IMRT work platform.Mucociliary transport rate (MTR) was detected by modified saccharine test before IMRT, at the end of IMRT, and at 3, 6, and 12 months after IMRT.Results:The data were available for 129 nasal cavities.The cavities receiving a mean dose below or equal to 37 Gy showed substantial preservation of nasal mucous after IMRT.The MRT decreased to (62.82±38.59)%, (56.78±37.79)%, (64.05±39.37)%, and (71.13±39.55)% of pre-IMRT value at 4 time points after IMRT, with significant differences among the data (P<0.05).In contrast, when the cavities received a mean dose higher than 37 Gy, no significant differences in MTR among the time points were observed.At 3 months after IMRT, the MTR was the lowest (38.27% of pre-RT value).Conclusions:A mean radiation dose of ≤37 Gy for the nasal cavity is an optimal dose to protect the nasal cavity function.Gen-Di Yin Guan-Xia Xiong Chong Zhao Yuan-Yuan Chen 2010Chinese Journal of Cancer2010,29,9:10
9Ginseng-Derived Panaxadiol Saponins Promote Hematopoiesis Recovery in Cyclophosphamide-Induced Myelosuppressive Mice:Potential Novel Treatment of Chemotherapy-Induced Cytopenias显示文摘Objective:To investigate the potential efficacy of panaxadiol saponins component(PDS-C),a biologically active fraction isolated from total ginsenosides,to reverse chemotherapy-induced myelosuppression and pancytopenia caused by cyclophamide(CTX).Methods:Mice with myelosuppression induced by CTX were treated with PDS-C at a low-(20 mg/kg),moderate-(40 mg/kg),or high-dose(80 mg/kg) for 7 consecutive days.The level of peripheral white blood cell(WBC),neutrophil(NEU) and platelet(PLT) were measured,the histopathology and colony formation were observed,the protein kinase and transcription factors in hematopoietic cells were determined by immunohistochemical staining and Western blot.Results:In response to PDS-C therapy,the peripheral WBC,NEU and PLT counts of CTX-induced myelosuppressed mice were significantly increased in a dose-dependent manner.Similarly,bone marrow histopathology examination showed reversal of CTX-induced myelosuppression with increase in overall bone marrow cellularity and the number of hematopoietic cells(P<0.01).PDS-C also promoted proliferation of granulocytic and megakaryocyte progenitor cells in CTX-treated mice,as evidenced by significantly increase in colony formation units-granulocytes/monocytes and-megakaryocytes(P<0.01).The enhancement of hematopoiesis by PDS-C appears to be mediated by an intracellular signaling pathway,this was evidenced by the up-regulation of phosphorylated mitogen-activated protein kinase(p-MEK) and extracellular signal-regulated kinases(p-ERK),and receptor tyrosine kinase(C-kit) and globin transcription factor 1(GATA-1) in hematopoietic cells of CTX-treated mice(P<0.05).Conclusions:PDS-C possesses hematopoietic growth factor-like activities that promote proliferation and also possibly differentiation of hematopoietic progenitor cells in myelosuppressed mice,probably mediated by a mechanism involving MEK and ERK protein kinases,and C-kit and GATA-1 transcription factors.PDS-C may potentially be a novel treatment of myelosuppression and pancytopenia caused by chemotherapy.SUN Xin ZHAO Yan-na QIAN Song GAO Rui-lan YIN Li-ming WANG Li-pei CHONG Beng-hock ZHANG Su-zhan 2018Chinese Journal of Integrative Medicine2018,24,3:9
10Panaxdiol Saponins Component Promotes Hematopoiesis and Modulates T Lymphocyte Dysregulation in Aplastic Anemia Model Mice显示文摘Objective: To investigate the potential efficacy of panaxadiol saponins component(PDS-C) in the treatment of aplastic anemia(AA) model mice. Methods: Totally 70 mice were divided into 7 groups as follows: normal, model, low-, medium-, high-dose PDS-C(20, 40, 80 mg/kg, namely L-, M-, H-PDS-C), cyclosporine(40 mg/kg), and andriol(25 mg/kg) groups, respectively. An immune-mediated AA mouse model was established in BALB/c mice by exposing to 5.0 Gy total body irradiation at 1.0 Gy/min, and injecting with lymphocytes from DBA mice. On day 4 after establishment of AA model, all drugs were intragastrically administered daily for 15 days, respectively, while the mice in the normal and model groups were administered with saline solution. After treatment, the peripheral blood counts, bone marrow pathological examination, colony forming assay of bone marrow culture, T lymphocyte subpopulation analysis, as well as T-bet, GATA-3 and Fox P3 proteins were detected by flow cytometry and Western blot. Results: The peripheral blood of white blood cell(WBC), platelet, neutrophil counts and hemoglobin(Hb) concentration were significantly decreased in the model group compared with the normal group(all P<0.01). In response to 3 dose PDS-C treatment, the WBC, platelet, neutrophil counts were significantly increased at a dose-dependent manner compared with the model group(all P<0.01). The myelosuppression status of AA was significantly reduced in M-, H-PDS-C groups, and hematopoietic cell quantity of bone marrow was more abundant than the model group. The colony numbers of myeloid, erythroid and megakaryocytic progenitor cells in the model group were less than those of the normal mice in bone marrow culture, while, PDS-C therapy enhanced proliferation of hematopoietic progenitor cells by significantly increasing colony numbers(all P<0.01). Furthermore,PDS-C therapy increased peripheral blood CD3^+ and CD3^+CD4^+ cells and reduced CD3^+CD8^+ cells(P<0.05 or P<0.01). Meanwhile, PDS-C treatment at medium-and high doses groups also increased CD4^+CD25^+Fox P3^+ cells, downregulated T-bet protein expression, and upregulated GATA-3 and Fox P3 protein expressions in spleen cells(P<0.05). 13Rotifer community structure and its response to environmental factors in the Backshore Wetland of Expo Garden,Shanghai显示文摘We investigated rotifer community structure in the Backshore Wetland of the Expo Garden in Shanghai from September 2009 to August 2010.We identified 116 rotifer species,determined the dominant species and their relationships with environmental factors,and community diversity indices.Polyarthra trigla was the most common species over the four seasons and proportionally most dominant in spring and autumn,whereas Cephalodella exigua and Keratella cochlearis were the most dominant species in winter and summer,respectively.There was seasonal variability in species numbers,with the highest numbers in summer,followed by autumn and winter.The mean annual density was 815±1346 ind./L and the biomass was 0.3649±0.4940 mg/L.There were significant differences between months:the highest density(2723±2926 ind./L)and biomass(0.7938±0.8197 mg/L)occurred in June,while the lowest density(238±271 ind./L)and biomass(0.0476±0.0429 mg/L)occurred in May.Canonical correspondence analysis(CCA)indicated that temperature,pH,nitrite nitrogen(NO2-N),and total phosphorus(TP)were the most important abiotic factors affecting rotifer distribution.Our research showed that the adaptability of the rotifers to temperature was the important factor.Liping Yin Yu Ji Yinjiang Zhang Linxuan Chong Lijing Chen 2018Aquaculture and Fisheries2018,3,2:4
14Malnutrition Increases the Risk of Mortality in Hospitalized Lung Cancer Patients显示文摘Background The Global Leadership Initiative on Malnutrition(GLIM)recently developed a new set of diagnostic criteria for identifying patients with malnutrition.Because the GLIM criteria were only introduced a little over 3 years ago,additional validation and reliability testing are needed in a variety of populations.Methods We performed an observational,multicenter cohort study.From July 2013 to October 2018,lung cancer patients were recruited from the Daping Hospital of Army Medical University and the First Hospital of Jilin University as part of the INSCOC project.Previously-established cut-off values for the calf circumference(CC,male<30 cm,female<29.5 cm)were applied as the reduced muscal mass of phenotypic criteria to establish the GLIM diagnosis.Multivariate Cox regression analyses were performed to analyze the association between the GLIM criteria and survival.Results A total of 1219 patients with lung cancer were studied as subjects.Their age was 58.81±9.92 years old,and 820 were male and 399 were female.According to the GLIM diagnostic criteria using the CC as a muscle mass measurement,303 patients(24.9%)were categorized as malnourished,142 patients(23.1%)in the adult group(18≤age<60)and 161 patients(26.7%)in the older group(age≥60 years).The patients with malnutrition had a higher incidence of anemia than the nourished patients(P=0.012).The QLQ-C30 score and KPS score indicating that the malnourished patients had a consistently worse quality of life compared to the nourished group(all P<0.001).The median survival of the malnutrition group was 42(95%CI:34-50)months,which was much shorter than the 62(95%CI:57-66)months in the nourished group(P<0.001).In the adult group,the median survival decreased from 65(95%CI:55-72)months in nourished group to 34(95%CI:25-48)months in the patients with malnutrition(P<0.001).In the older group,it decreased from 61(95%CI:55-67)months to 48(95%CI:39-59)months(P=0.001).A Cox regression analysis showed that GLIM-diagnosed malnutrition was associated with an increased risk of death among adult group(HR=1.670,95%CI:1.29-2.16),older group(HR=1.332,95%CI:1.05-1.69)and overall(HR=1.453,95%CI:1.22-1.72).Conclusion All of these results demonstrate that GLIM-diagnosed malnutrition is associated with a poorer survival for all lung cancer patients,independent of age.Fei Fei Chong Liang Yu Yin Jie Liu Na Li Jing Guo Yang Fan Meng Yuan Zhang Ling Zhang Xiu Mei He Hong Mei Zhang Mu Li Shi Wei Li Hong Xia Xu 2022Journal of Nutritional Oncology2022,7,1:4
15Ultrasound-guided fine needle aspiration of retropharyngeal lymph nodes after radiotherapy for nasopharyngeal carcinoma: a novel technique for accurate diagnosis显示文摘Background:Enlarged retropharyngeal lymph nodes(RLNs)are very common in patients with nasopharyngeal carcinoma(NPC)undergoing radiotherapy.The most suitable treatment option for enlarged RLNs depends on the pathological results.However,RLN sampling is difficult and imminent in the clinic setting.We recently developed a novel minimally invasive technique termed endoscopic ultrasound-guided fine needle aspiration(EUS-FNA)for sam-pling RLN tissues sufficient for pathological or cytological diagnosis.Methods:We enrolled 30 post-radiotherapy patients with NPC with suspected RLN metastasis detected via mag-netic resonance imaging(MRI).The EUS probe was introduced into the nasopharynx via the nostrils,and EUS was then used to scan the retropharyngeal space and locate the RLN in the anterior carotid sheath.EUS-FNA was subsequently performed.The safety and efficacy of using EUS-FNA to sample the RLN tissues were assessed.Results:Strips of tissue were successfully sampled from all patients using EUS-FNA.Of the 30 patients,23 were confirmed to have cancer cells in the biopsied tissues via pathology or cytology examinations with 1 EUS-FNA biopsy session.The seven cases without confirmed cancer cells were subsequently reanalyzed by using another EUS-FNA biopsy session,and two more cases were confirmed possessing cancer cells.The other five patients without con-firmed cancer cells were closely followed with MRI every month for 3 months.After follow-up for 3 months,three patients were still considered cancer-free due to the presence of RLNs with stable or shrinking diameters.The rest two patients who showed progressive disease underwent a third EUS-FNA biopsy procedure and were further confirmed to be cancer cell-positive.In the whole cohort reported here,the EUS-FNA procedure was not associated with any severe complications.Conclusion:EUS-FNA is a safe and effective diagnostic approach for sampling tissues from the RLNs in patients with suspected recurrent NPC.Long-Jun He Chuanbo Xie Yin Li Lin-Na Luo Ke Pan Xiao-Yan Gao Li-Zhi Liu Jian-Ming Gao Guang-Yu Luo Hong-Bo Shan Ming-Yuan Chen Chong Zhao Wei-Jun Fan Ping Yang Guo-Liang Xu Jian-Jun Li 2018Cancer Communications2018,38,1:3
16黑果枸杞生理指标测定、转录组分析以及分子标记开发研究(英文)显示文摘目的:以高耐盐碱多年生沙漠经济灌木黑果枸杞为研究材料,对其在盐碱胁迫处理下的生理指标进行测定,确定转录组测试的时间。通过转录组分析挖掘潜在抗逆基因,并挖掘全转录组水平的分子标记。旨在为黑果枸杞的优良基因资源利用、野生品种保护和新品种培育提供理论依据和实践指导。创新点:首次对黑果枸杞进行盐碱胁迫下的生理指标变化和全转录组水平的基因表达变化进行分析,并基于转录组进行大规模简单重复序列标记开发和验证,并将所获取的分子标记应用到9个野生群体进行遗传多样性分析。方法:采用双通道PAM-100荧光仪研究盐碱胁迫对黑果枸杞P700(PS I)和叶绿素荧光(PS II)的影响;通过盐碱胁迫下丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性变化选定转录组测序(RNA-seq)取样时间;采用Illumina高通量测序平台进行转录组从头测序;选取20个基因采用荧光定量聚合酶链式反应(PCR)法进行基因表达分析;基于转录组序列组装结果进行简单重复序列扫描;采用聚丙烯酰胺凝聚和毛细管电泳法鉴定引物多态性,选取其中11对多态性引物应用于遗传多样性分析。结论:通过对对照以及混合盐碱处理的黑果枸杞无菌苗进行生理和生化测试,结果选定处理6小时为取样点。RNA-seq结果共获得68 063个unigene,平均长度为877 bp,其中4096个基因在混合盐碱处理下表现为上调,4381个表现为下调。随机选取24个基因进行荧光定量表达分析,结果显示,荧光定量表达结果与RNA-seq结果呈显著正相关。基于转录组测试数据,在7940个基因中挖掘出9216个简单重复序列标记,对其中77个进行检测,显示有68个位点清晰存在,选取其中11个多态性位点对来自西北四个省份或自治区的9个野生种质资源进行遗传多样性分析,结果显示分析可靠。Jin-huan CHEN Dong-zhi ZHANG Chong ZHANG Mei-long XU Wei-lun YIN 2017Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2017,18,11:3

18Effect of bortezomib on migration and invasion in cervical carcinoma HeLa cell显示文摘Objective:To explore the effect of bortezomib on migration and invasion of cervical carcinoma HeLa cell and specific molecular mechanism.Methods:The effect of bortezomib on the viability of HeLa cell was measured by MTT assay.The effect of bortezomib on cell migration and invasion was measured by Transwell assay and invasion experiment respectively.The activation of Akt/mTOR signaling pathway and expression level of MMP2,MMP9 were assayed by western blot.Results:MTT assay indicated bortezomib(2.5 μM.5 μM,10 μM)could inhibit HeLa cell viability,and the inhibitory rate was highest at 48 h.Transwell assay and invasion experiment results showed that bortezomib inhibited HeLa cell migration and invasion.Western blotting assays presented bortezomib could suppress the phosphorylation of Akt and mTOR.and down-regulate the expression of MMP2 and MMP9.Conclusions:These results suggested bortezomib could inhibit migration and invasion in cervical carcinoma HeLa cell,which might be related to Akt/mTOR signal pathway.Chong Shi Guo-Bin Zhang Shu-Wang Yin 2015Asian Pacific Journal of Tropical Medicine2015,8,6:2
