| 1 | Mesoporous silica nanoparticles for drug and gene delivery显示文摘Mesoporous silica nanoparticles(MSNs) are attracting increasing interest for potential biomedical applications. With tailored mesoporous structure, huge surface area and pore volume,selective surface functionality, as well as morphology control, MSNs exhibit high loading capacity for therapeutic agents and controlled release properties if modified with stimuli-responsive groups, polymers or proteins. In this review article, the applications of MSNs in pharmaceutics to improve drug bioavailability, reduce drug toxicity, and deliver with cellular targetability are summarized. | 3 | Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. | 4 | Synergistic immunoreaction of acupuncture-like dissolving microneedles containing thymopentin at acupoints in immune-suppressed rats显示文摘Dissolving microneedles carried drug molecules can effectively penetrate the stratum corneum of skin to improve the transdermal drug delivery. The traditional Chinese medicine acupuncture is based on the needle stimulation at a specific location(acupoint) to generate and transmit biochemical and physiological signals which alter the pathophysiological state of patients. However, the pain associated with conventional acupuncture needles and the requirement of highly trained professionals limit the development of acupuncture in non-Asian countries. The purpose of this study is to investigate whether the dissolving microneedles can be utilized as a self-administered painless replacement for acupuncture and locally released drug molecules can achieve expected therapeutic outcomes. Immunosuppressive rats were treated with acupuncture at Zusanli(ST36) acupoint using microneedles containing thymopentin.The immune functions and psychological mood of the immunosuppressed animals were examined. The proliferation of splenocytes was examined by CCK-8 assay. CD4 and CD8 expression patterns in spleen cells were detected by flow cytometry. The current study showed that use of either microneedles containing thymopentin or conventional acupuncture both resulted in immune cell proliferation, which was confirmed by flow cytometry. | 5 | Influence of physical properties of carrier on the performance of dry powder inhalers显示文摘Dry powder inhalers(DPIs) offer distinct advantages as a means of pulmonary drug delivery and have attracted much attention in the field of pharmaceutical science. DPIs commonly contain micronized drug particles which, because of their cohesiveness and strong propensity to aggregate, have poor aerosolization performance. Thus carriers with a larger particle size are added to address this problem. However, the performance of DPIs is profoundly influenced by the physical properties of the carrier, particularly their particle size, morphology/shape and surface roughness. | 6 | Multi-functional bismuth-doped bioglasses: combining bioactivity and photothermal response for bone tumor treatment and tissue repair显示文摘Treatment of large bone defects derived from bone tumor surgery is typically performed in multiple separate operations,such as hyperthermia to extinguish residual malignant cells or implanting bioactive materials to initiate apatite remineralization for tissue repair;it is very challenging to combine these functions into a material.Herein,we report the first photothermal(PT)effect in bismuth(Bi)-doped glasses.On the basis of this discovery,we have developed a new type of Bi-doped bioactive glass that integrates both functions,thus reducing the number of treatment cycles.We demonstrate that Bi-doped bioglasses(BGs)provide high PT efficiency,potentially facilitating photoinduced hyperthermia and bioactivity to allow bone tissue remineralization.The PT effect of Bi-doped BGs can be effectively controlled by managing radiative and non-radiative processes of the active Bi species by quenching photoluminescence(PL)or depolymerizing glass networks.In vitro studies demonstrate that such glasses are biocompatible to tumor and normal cells and that they can promote osteogenic cell proliferation,differentiation,and mineralization.Upon illumination with near-infrared(NIR)light,the bioglass(BG)can efficiently kill bone tumor cells,as demonstrated via in vitro and in vivo experiments.This indicates excellent potential for the integration of multiple functions within the new materials,which will aid in the development and application of novel biomaterials. | Liping Wang Nicholas J.Long Lihua Li Yao Lu Mei Li Jiangkun Cao Yu Zhang Qinyuan Zhang Shanhui Xu Zhongmin Yang Chuanbin Mao Mingying Peng | 2018 | Light(Science & Applications)2018,7,1: | 6 |
| 7 | Impairment of the autophagy-lysosomal pathway in Alzheimer’s diseases: Pathogenic mechanisms and therapeutic potential显示文摘Alzheimer’s disease(AD),the most common neurodegenerative disorder,is characterized by memory loss and cognitive dysfunction.The accumulation of misfolded protein aggregates including amyloid beta(Aβ)peptides and microtubule associated protein tau(MAPT/tau)in neuronal cells are hallmarks of AD.So far,the exact underlying mechanisms for the aetiologies of AD have not been fully understood and the effective treatment for AD is limited.Autophagy is an evolutionarily conserved cellular catabolic process by which damaged cellular organelles and protein aggregates are degraded via lysosomes.Recently,there is accumulating evidence linking the impairment of the autophagy-lysosomal pathway with AD pathogenesis.Interestingly,the enhancement of autophagy to remove protein aggregates has been proposed as a promising therapeutic strategy for AD.Here,we first summarize the recent genetic,pathological and experimental studies regarding the impairment of the autophagy-lysosomal pathway in AD.We then describe the interplay between the autophagy-lysosomal pathway and two pathological proteins,Aβand MAPT/tau,in AD.Finally,we discuss potential therapeutic strategies and small molecules that target the autophagy-lysosomal pathway for AD treatment both in animal models and in clinical trials.Overall,this article highlights the pivotal functions of the autophagy-lysosomal pathway in AD pathogenesis and potential druggable targets in the autophagy-lysosomal pathway for AD treatment. | Wei Zhang Chengchao Xu Jichao Sun Han-Ming Shen Jigang Wang Chuanbin Yang | 2022 | Acta Pharmaceutica Sinica B2022,12,3: | 6 |
| 8 | SNP-based QTL mapping for panicle traits in the japonica super rice cultivar Liaoxing 1显示文摘Panicle traits directly associated with yield are a target of selection in rice breeding.Although abundant QTL for panicle traits have been identified,there is little information about the genetic basis of panicle traits in japonica super rice(JSR)cultivars.In this study,we identified QTL for panicle traits in three environments using a population of recombinant inbred lines(RILs)derived from the JSR cultivar Liaoxing 1.A total of 197 RILs were genotyped with 285 polymorphic SNP markers.Phenotypic data and best linear unbiased prediction(BLUP)value of primary branch number(BNP),secondary branch number(BNS),grain number on primary branch(GNP),grain number on secondary branch(GNS),grain number per panicle(GN),panicle length(PL)and grain density(GD)were used for QTL mapping.A total of 105 QTL for seven panicle traits were detected in single environments using their BLUP values.Individual QTL explained 0.51%–52.22%of the phenotypic variation.Of the 105,49 were also detected by joint multi-environment analyses.Five stable QTL:qGD9,qPL9,qGNP9,qGN6,and qBNS6.2 were identified in multiple environments.qGD9,qGNP9,and qPL9,co-localizing on chromosome 9,likely correspond to the known gene DEP1.Importantly,qGN6 and qBNS6.2 in a co-localization region were identified as novel QTL,and their Liaoxing 1 alleles had a positive effect.Several RILs with the QTL allele combinations qGD9/qPL9/qGNP9 and qGN6/qBNS6.2 showed greater GN.Further investigation of the putative gene underlying qGN6/qBNS6.2 would shed light on the molecular mechanism of JSR. | Mingzhu Zhao Zuobin Ma Lili Wang Zhiqiang Tang Ting Mao Chuanbin Liang Hong Gao Liying Zhang Na He Liang Fu Changhua Wang Guomin Sui Wenjing Zheng | 2020 | The Crop Journal2020,8,5: | 4 |

| 16 | A real-time and modular approach for quick detection and mechanism exploration of DPIs with different carrier particle sizes显示文摘Dry powder inhalers(DPIs) had been widely used in lung diseases on account of direct pulmonary delivery, good drug stability and satisfactory patient compliance. However, an indistinct understanding of pulmonary delivery processes(PDPs) hindered the development of DPIs. Most current evaluation methods explored the PDPs with over-simplified models, leading to uncompleted investigations of the whole or partial PDPs. In the present research, an innovative modular process analysis platform(MPAP) was applied to investigate the detailed mechanisms of each PDP of DPIs with different carrier particle sizes(CPS). The MPAP was composed of a laser particle size analyzer, an inhaler device,an artificial throat and a pre-separator, to investigate the fluidization and dispersion, transportation,detachment and deposition process of DPIs. The release profiles of drug, drug aggregation and carrier were monitored in real-time. The influence of CPS on PDPs and corresponding mechanisms were explored. The powder properties of the carriers were investigated by the optical profiler and Freeman Technology four powder rheometer. The next generation impactor was employed to explore the aerosolization performance of DPIs.
