| 1 | Potential application of FoldX force field based protein modeling in zinc finger nucleases design显示文摘Engineered sequence-specific zinc finger nucleases (ZFNs) make the highly efficient modification of eukaryotic genomes possible.However,most current strategies for developing zinc finger nucleases with customized sequence specificities require the construction of numerous tandem arrays of zinc finger proteins (ZFPs),and subsequent largescale in vitro validation of their DNA binding affinities and specificities via bacterial selection.The labor and expertise required in this complex process limits the broad adoption of ZFN technology.An effective computational assisted design strategy will lower the complexity of the production of a pair of functional ZFNs.Here we used the FoldX force field to build 3D models of 420 ZFP-DNA complexes based on zinc finger arrays developed by the Zinc Finger Consortium using OPEN (oligomerized pool engineering).Using nonlinear and linear regression analysis,we found that the calculated protein-DNA binding energy in a modeled ZFP-DNA complex strongly correlates to the failure rate of the zinc finger array to show significant ZFN activity in human cells.In our models,less than 5% of the three-finger arrays with calculated protein-DNA binding energies lower than 13.132 kcal mol 1 fail to form active ZFNs in human cells.By contrast,for arrays with calculated protein-DNA binding energies higher than 5 kcal mol 1,as many as 40% lacked ZFN activity in human cells.Therefore,we suggest that the FoldX force field can be useful in reducing the failure rate and increasing efficiency in the design of ZFNs. | HE ZuYong MEI Gui ZHAO ChunPeng CHEN YaoSheng | 2011 | Science China(Life Sciences)2011,54,5: | 2 |

| 4 | Recycled Aggregate Pervious Concrete: Analysis of Influence of Water-Cement Ratio and Fly Ash under Single Action and Optimal Design of Mix Proportion显示文摘Pervious concrete is recommended,which is of great benefit to the ecological environment and human living environment.In this paper,the influences of five water-cement ratios and four fly ash contents to replace the cement by mass with a water-cement ratio of 0.30 on the properties of Recycled Aggregate Pervious Concrete(RAPC)were studied.Following this,based on the Grey relational-Technique for Order Preference by Similarity to an Ideal Solution(TOPSIS)optimization method,the strength,permeability,abrasion loss rate,and material costs of RAPC were adopted as evaluation indices to establish a mix proportion optimization model.The results show that the increase of water-cement ratio and fly ash replacement level of RAPC leads to decreased compres-sive strength while an increase in the permeability and abrasion loss rate.According to test results based on the optimal model 0.30 was identified as the best mix proportion.In addition,ecological-economic analysis of RAPC raw materials was carried out by comparing different natural aggregates.The results of EE(embodied energy)and ECO 2e(embodied CO_(2) emission)pointed out that the combination of recycled aggregate and fly ash leads to sig-nificant ecological and economic benefits. | Shoukai Chen Chunpeng Xing Mengdie Zhao Junfeng Zhang Lunyan Wang Qidong He | 2022 | Journal of Renewable Materials2022,10,3: | 1 |

| 6 | Study on the Effect of 'Glimmer' on Cadmium Absorption and Yield of Rice显示文摘[Objectives]The effects of'Glimmer'treatment on dry matter accumulation,yield and cadmium accumulation in different types of rice varieties were investigated,in order to provide a theoretical basis for the regulation of low cadmium accumulation in rice grains.[Methods]Under the treatment of cadmium contaminated soil with a concentration of 0.58 mg/kg,the effects of'Glimmer'treatment on dry matter accumulation,yield and cadmium content of different rice varieties were studied by pot experiment.[Results]Spraying'Glimmer'can increase the biological yield of rice,and the different types of rice showed the order of conventional rice>two-line hybrid rice>three-line hybrid rice.Among the conventional rice,most significant increase was observed in Huanghuazhan,and in two-line hybrid rice,C Liangyou 651 increased the most.The'Glimmer'treatment had an obvious yield increasing effect on conventional rice,among which such two varieties as Xiangwanxian 13 and Huanghuazhan had the largest increases,followed by the two-line hybrid rice,and a little effect was observed on the three-line hybrid rice.The'Glimmer'treatment resulted in a significant reduction in cadmium content in different types of rice,which decreased by 13.33%-26.09%,and seven varieties showed a reduction over 21.74%.[Conclusions]The'Glimmer'treatment can significantly reduce the cadmium content in rice,and generally promote the dry matter accumulation and yield increase of rice,of which conventional rice performed better. | Jing ZHOU Guoliang YU Chunpeng HE Guohui MA Guiyuan MENG | 2019 | Agricultural Biotechnology2019,8,4: | 0 |
