A novel superhard tungsten nitride predicted by machine-learning accelerated crystal structure search显示文摘Transition metal nitrides have been suggested to have both high hardness and good thermal stability with large potential application value, but so far stable superhard transition metal nitrides have not been synthesized. Here, with our newly developed machine-learning accelerated crystal structure searching method,we designed a superhard tungsten nitride,h-WN_6,which can be synthesized at pressure around65 GPa and quenchable to ambient pressure. This h-WN_6is constructed with single-bonded armchair-like N_6rings and presents ionic-like features, which can be formulated as W^(2.4+)N_6^(2.4à).It has a band gap of1.6 e V at 0 GPa and exhibits an abnormal gap broadening behavior under pressure.Excitingly, this h-WN_6is found to be the hardest among transition metal nitrides known so far(Vickers hardness around57 GPa)and also has a very high melting temperature(around 1,900 K).Additionally, the good gravimetric(3.1 k J/g)and volumetric(28.0 k J/cm^3)energy densities make this nitrogen-rich compound a potential high-energy-density material. These predictions support the designing rules and may stimulate future experiments to synthesize superhard and high-energy-density material. | Kang Xia Hao Gao Cong Liu Jianan Yuan Jian Sun Hui-Tian Wang Dingyu Xing | 2018 | Science Bulletin2018,63,13: | 11

Identification of biomarkers for hepatocellular carcinoma by semiquantitative immunocytochemistry显示文摘AIM:To investigate the expression of key biomarkers in hepatoma cell lines, tumor cells from patients’blood samples, and tumor tissues.METHODS:We performed the biomarker tests in two steps.First, cells plated on coverslips were used to assess biomarkers, and fluorescence intensities were calculated using the NIH Image J software. The measured values were analyzed using the SPSS19.0 software to make comparisons among eight cell lines.Second,eighty-four individual samples were used to assess the biomarkers’expression.Negative enrichment of the blood samples was performed, and karyocytes were isolated and dropped onto pretreated glass slides for further analysis by immunofluorescence staining.Fluorescence intensities were compared among hepatocellular carcinoma(HCC)patients, chronic HBV-infected patients, and healthy controls following methods similar to those used for cell lines. The relationships between the expression of biomarkers and clinical pathological parameters were analyzed by Spearman rank correlation tests. In addition,we studied the distinct biomarkers’expression with three-dimensional laser confocal microscopy reconstructions, and Kaplan-Meier survival analysis was performed to understand the clinical significance of these biomarkers.RESULTS:Microscopic examination and fluorescence intensity calculations indicated that cytokeratin 8/18/19(CK)expression was significantly higher in six of the seven HCC cell lines examined than in the control cells, and the expression levels of asialoglycoprotein receptor(ASGPR)and glypican-3(GPC3)were higher in all seven HCC cell lines than in the control.Cells obtained from HCC patients’blood samples also displayed significantly higher expression levels of ASGPR,GPC3,and CK than cells from chronic HBV-infected patients or healthy controls;these proteins may be valuable surface biomarkers for identifying HCC circulating tumor cells isolated and enriched from the blood samples. The stem cell-like and epithelial-mesenchymal transition-related biomarkers could be detected on the karyocyte slides.ASGPR and GPC3 were expressed at high levels, and thus three-dimensional reconstructions were used to observe their expression in detail. This analysis indicated that GPC3 was localized in the cytoplasm and membrane, but that ASGPR had a polar localization.Survival analyses showed that expression of GPC3 and ASGPR is associated with a patient’s overall survival(OS).CONCLUSION:ASGPR,GPC3,and CK may be valuable HCC biomarkers for CTC detection;the expression of ASGPR and GPC3 might be helpful for understanding patients’OS. | Hong Mu Kai-Xuan Lin Hong Zhao Shu Xing Cong Li Fang Liu Hai-Zhen Lu Ze Zhang Yu-Lin Sun Xi-Yun Yan Jian-Qiang Cai Xiao-Hang Zhao | 2014 | World Journal of Gastroenterology2014,20,19: | 8
Biology, invasion and management of the agricultural invader:Fall armyworm,Spodoptera frugiperda(Lepidoptera:Noctuidae)显示文摘The fall armyworm(FAW),Spodoptera frugiperda(J.E.Smith),is native to the Americas. It has rapidly invaded 47 African countries and 18 Asian countries since the first detection of invasion into Nigeria and Ghana in 2016.It is regarded as a super pest based on its host range(at least 353 host plants),its inherent ability to survive in a wide range of habitats, its strong migration ability, high fecundity, rapid development of resistance to insecticides/viruses and its gluttonous characteristics. The inherently superior biological characteristics of FAW contribute to its invasiveness.Integrated pest management(IPM)of FAW has relied on multiple applications of monitoring and scouting, agricultural control, chemical pesticides, viral insecticides, sex attractants,bio-control agents(parasitoids, predators and entomopathogens)and botanicals.Knowledge gaps remain to be filled to:(1)understand the invasive mechanisms of S.frugiperda;(2)understand how to prevent its further spread and(3)provide better management strategies. This review summarizes the biological characters of FAW,their association with its invasiveness and IPM strategies, which may provide further insights for future management. | Jing WAN HUANG Cong LI Chang-you ZHOU Hong-xu REN Yong-lin LI Zai-yuan XING Long-sheng ZHANG Bin QIAO Xi LIU Bo LIU Cong-hui XI Yu LIU Wan-xue WANG Wen-kai QIAN Wan-qiang Simon MCKIRDY WAN Fang-hao | 2021 | Journal of Integrative Agriculture2021,20,3: | 7
Toxoplasma ROP16Ⅰ/Ⅲ ameliorated inflammatory bowel diseases via inducing M2 phenotype of macrophages显示文摘BACKGROUND Inflammatory bowel disease(IBD)is characterized by chronic and non-specific inflammation of the intestinal mucosa and mainly includes ulcerative colitis and Crohn's disease.AIM To explore the beneficial effect of ToxoROP16I/III-induced M2 phynotype macrophages in homeostasis of IBDs through downregulation of M1 inflammatory cells.METHODS RAW264.7 macrophages stimulated by lipopolysaccharide(LPS)(M1 cells)were co-cultured with Caco-2 cells as an inflammatory model of IBD in vitro. The expression of ToxoROP16I/III was observed in RAW264.7 macrophages that were transfected with pEGFP-rop16I/III.The phenotypes of M2 and M1 macrophage cells were assessed by quantitative real-time reverse transcriptase polymerase chain reaction and the expression of tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,transforming growth factor(TGF)-β1,IL-10,inducible nitric oxide synthase(iNOS),and arginase-1(Arg-1)was detected. The expression of iNOS,Arg-1,signal transducer and activator of transcription 3(Stat3),p-Stat3,Stat6,p-Stat6,programmed death ligand-2(PD-L2),caspase-3,-8,and-9 was analyzed by Western blotting, and Griess assays were performed to detect nitric oxide(NO).TNF-α,IL-1β,IL-6,TGF-β1,and IL-10 expression in the supernatants was detected by enzyme-linked immunosorbent assay, and Caco-2 cell apoptosis was co-culture system.RESULTS M1 cells exhibited significantly increased production of iNOS,NO,TNF-α,IL-1β,and IL-6,while ToxoROP16I/III induced macrophage bias to M2 cells in vitro, showing increased expression of Arg-1,IL-10 and TGF-β1 and elevated production of p-Stat3 and p-Stat6.The mixed M1 and M2 cell culture induced by ToxoROP16I/III exhibited decreased production of NO and iNOS and upregulated expression of Arg-1 and PD-L2.Accordingly,Caco-2 cells became apoptotic, and apoptosis-associated proteins such as caspase-3,-8 and-9 were dampened during co-culture of M1 and M2 cells.Flow cytometry analysis showed that co-culture of M1 cells with Caco-2 cells facilitated the apoptosis of Caco-2 cells, but co-culture of M1 and M2 cells alleviated Caco-2 cell apoptosis.CONCLUSION ToxoROP16I/III-induced M2 macrophages inhibited apoptosis of Caco-2 cells caused by M1 macrophages. This finding may help gain a better understanding of the underlying mechanism and represent a promising therapeutic strategy for IBDs. | Yong-Wei Xu Rui-Xin Xing Wen-Hui Zhang Lu Li Yi Wu Jing Hu Cong Wang Qing-Li Luo Ji-Long Shen Xi Chen | 2019 | World Journal of Gastroenterology2019,25,45: | 7
Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China. To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines, namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics. Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines, some of which could be potential biomarkers of disease severity of 2019-n Co V infections. These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease. Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized. | Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang | 2020 | National Science Review2020,7,6: | 6
Role of PLC-PIP2 and cAMP-PKA Signal Pathways in Radiation-induced Immune-suppressing Effect显示文摘Objective The purpose of the present study was to observe the changes in CD4+CD25+Nrp1+Treg cells after irradiation with different doses and explore the possible molecular mechanisms involved. Methods ICR mice and mouse lymphoma cell line(EL-4 cells) was used. The expressions of CD4,CD25,Nrp1,calcineurin and PKC-α were detected by flow cytometry. The expressions of TGF-β1,IL-10,PKA and cAMP were estimated with ELISA. Results At 12 h after irradiation, the expression of Nrp1 increased significantly in 4.0 Gy group, compared with sham-irradiation group(P<0.05) in the spleen and thymus, respectively, when ICR mice received whole-body irradiation(WBI). Meanwhile the synthesis of Interleukin 10(IL-10) and transforming growth factor-β1(TGF-β1) increased significantly after high dose irradiation(HDR)(> or = 1.0 Gy). In addition, the expression of cAMP and PKA protein increased, while PKC-α,calcineurin decreased at 12h in thymus cells after 4.0 Gy X-irradiation. While TGF-β1 was clearly inhibited when the PLC-PIP2 signal pathway was stimulated or the cAMP-PKA signal pathway was blocked after 4.0 Gy X-irradiation,this did not limit the up-regulation of CD4+CD25+Nrp1+Treg cells after ionizing radiation. Conclusion These results indicated that HDR might induce CD4+CD25+Nrp1+Treg cells production and stimulate TGF-β1 secretion by regulating signal molecules in mice. | DONG Juan Cong CHENG Guang Hui SHAN Yu Xing WU Ning SHAO Ming Long LI Peng Wu JIN Shun Zi | 2014 | Biomedical and Environmental Sciences2014,27,1: | 5
Morphology and composition of particles emitted from a port fuel injection gasoline vehicle under real-world driving test cycles显示文摘Traffic vehicles, many of which are powered by port fuel injection(PFI) engines, are major sources of particulate matter in the urban atmosphere. We studied particles from the emission of a commercial PFI-engine vehicle when it was running under the states of cold start, hot start, hot stabilized running, idle and acceleration, using a transmission electron microscope and an energy-dispersive X-ray detector. Results showed that the particles were mainly composed of organic, soot, and Ca-rich particles, with a small amount of S-rich and metal-containing particles, and displayed a unimodal size distribution with the peak at 600 nm. The emissions were highest under the cold start running state, followed by the hot start, hot stabilized, acceleration, and idle running states. Organic particles under the hot start and hot stabilized running states were higher than those of other running states. Soot particles were highest under the cold start running state. Under the idle running state, the relative number fraction of Ca-rich particles was high although their absolute number was low. These results indicate that PFI-engine vehicles emit substantial primary particles, which favor the formation of secondary aerosols via providing reaction sites and reaction catalysts, as well as supplying soot, organic, mineral and metal particles in the size range of the accumulation mode. In addition, the contents of Ca, P, and Zn in organic particles may serve as fingerprints for source apportionment of particles from PFI-engine vehicles. | Jiaoping Xing Longyi Shao Wenbin Zhang Jianfei Peng Wenhua Wang Cong Hou Shijin Shuai Min Hu Daizhou Zhang | 2019 | Journal of Environmental Sciences2019,31,2: | 4
Coexistence of plastic and partially diffusive phases in a helium-methane compound显示文摘Helium and methane are major components of gian icy planets and are abundant in the universe. However, helium is the most inert element in the periodic table and methane is one of the most hydrophobic molecules, thus whether they can react with each other is of fundamental importance. Here, our crystal structure searches and first-principles calculations predict that a He3CH4 compound is stable over a wide range of pressures from 55 to 155 GPa and a HeCH4 com pound becomes stable around 105 GPa.As nice examples of pure van der Waals crystals;the insertion of helium atoms changes the original packing of pure methane molecules and also largely hinders the polymerization of methane at higher pressures. After analyzing the diffusive properties during the melting of He3CH4 at high pressure and high temperature,in addition to a plastic methane phase,we have discovered an unusual phase which exhibits coexistence of diffusive helium and plastic methane. In addition, the range of the diffusive behavior within the helium-methane phase diagram is found to be much narrower compared to that of previously predicted helium-water compounds. This may be due to the weaker van der Waals interactions between methane molecules compared to those in helium-water compounds;and that the helium-methane compound melts more easily. | Hao Gao Cong Liu Andreas Hermann Richard J.Needs Chris J.Pickard Hui-Tian Wang Dingyu Xing Jian Sun | 2020 | National Science Review2020,7,10: | 4
The electrolyte comprising more robust water and superhalides transforms Zn-metal anode reversibly and dendrite-free显示文摘A great challenge for all aqueous batteries, including Zn-metal batteries,is the parasitic hydrogen evolution reaction on the low-potential anode.Herein,we report the formula of a highly concentrated aqueous electrolyte that mitigates hydrogen evolution by transforming water molecules more inert. The electrolyte comprises primarily ZnCl_(2) and LiCl as an additive, both of which are inexpensive salts. The O-H covalent bonds in water get strengthened in a chemical environment that has fewer hydrogen bonding interactions and a greater number of Zn-Cl superhalides,as suggested by integrated characterization and simulation. As a result, the average Coulombic efficiency of zincmetal anode is raised to an unprecedented>99.7%at 1mA cm^(−2).In the new electrolyte, the plating/stripping processes leave the zinc-metal anode dendrite-free,and the zinc-metal anode delivers stable plating/stripping cycles for 4000 hours with an areal capacity of 4 mAh cm^(−2) at 2mA cm^(−2).Furthermore,the high Coulombic efficiency of zinc-metal anode in the ZnCl_(2)-LiCl mixture electrolyte is demonstrated in full cells with a limited anode. The V_(2)O_(5)·H_(2)O||Zn full cell with an N/P mass ratio of 1.2 delivers a stable life of more than 2500 cycles, and the LiMn_(2)O_(4)||Zn hybrid cell with an N/P mass ratio of 0.6 exhibits 1500 cycles in its stable life. | Chong Zhang Woochul Shin Liangdong Zhu Cheng Chen Joerg C.Neuefeind Yunkai Xu Sarah I.Allec Cong Liu Zhixuan Wei Aigerim Daniyar Jia‐Xing Jiang Chong Fang P.Alex Greaney Xiulei Ji | 2021 | Carbon Energy2021,3,2: | 3

Spatiotemporal change patterns of urban lakes in China’s major cities between 1990 and 2015显示文摘China has experienced unprecedented urbanization in the past decades, resulting in dramatic changes in the physical, limnological, and hydrological characteristics of lakes in urban landscapes. However, the spatiotemporal dynamics in distribution and abundance of urban lakes in China remain poorly understood. Here,we characterized the spatiotemporal change patterns of urban lakes in China’s major cities between 1990 and 2015 using remote-sensing data and landscape metrics. The results showed that the urban lake landscape patterns have experienced drastic changes over the past 25 years. The total surface area of the urban lakes has decreased by 17,620.02 ha,a decrease of 24.22%,with a significant increase in the landscape fragmentation and a reduction in shape complexity. We defined three lake-shrinkage types and found that vanishment was the most common lake-shrinkage pattern, followed by edge-shrinkage and tunneling in terms of lake area. Moreover,we also found that urban sprawl was the dominant driver of the lake shrinkage, accounting for 67.89%of the total area loss, and the transition from lakes to cropland was also an important factor(19.86%).This study has potential for providing critical baseline information for government decision-making in lake resources management and urban landscape design. | Cong Xie Xin Huang Leiguang Wang Xing Fang Wenzhi Liao | 2018 | International Journal of Digital Earth2018,11,11: | 2
The Msi1-mTOR pathway drives the pathogenesis of mammary and extramammary Paget’s disease显示文摘Mammary and extramammary Paget’s Diseases(PD)are a malignant skin cancer characterized by the appearance of Paget cells.Although easily diagnosed, its pathogenesis remains unknown.Here,single-cell RNA-sequencing identified distinct cellular states, novel biomarkers, and signaling pathways—including mTOR,associated with extramammary PD.Interestingly,we identified MSI1 ectopic overexpression in basal epithelial cells of human PD skin, and show that Msi1 overexpression in the epidermal basal layer of mice phenocopies human PD at histopathological,single-cell and molecular levels.Using this mouse model,we identified novel biomarkers of Paget-like cells that translated to human Paget cells.Furthermore,single-cell trajectory,RNA velocity and lineage-tracing analyses revealed a putative keratinocyte-to-Paget-like cell conversion, supporting the in situ transformation theory of disease pathogenesis.Mechanistically, the Msi1-mTOR pathway drives keratinocyte-Paget-like cell conversion, and suppression of mTOR signaling with Rapamycin significantly rescued the Paget-like phenotype in Msi1-overexpressing transgenic mice.Topical Rapamycin treatment improved extramammary PD-associated symptoms in humans, suggesting mTOR inhibition as a novel therapeutic treatment in PD. | Yongli Song Christian F.Guerrero-Juarez Zhongjian Chen Yichen Tang Xianghui Ma Cong Lv Xueyun Bi Min Deng Lina Bu Yuhua Tian Ruiqi Liu Ran Zhao Jiuzhi Xu Xiaole Sheng Sujuan Du Yeqiang Liu Yunlu Zhu Shi-jun Shan Hong-duo Chen Yiqiang Zhao Guangbiao Zhou Jianwei Shuai Fazheng Ren Lixiang Xue Zhaoxia Ying Xing Dai Christopher J.Lengner Bogi Andersen Maksim V.Plikus Qing Nie Zhengquan Yu-Show | 2020 | Cell Research2020,30,10: | 2
Efficacy, safety and immunogenicity of hexavalent rotavirus vaccine in Chinese infants显示文摘A randomized,double-blind,placebo-controlled multicenter trial was conducted in healthy Chinese infants to assess the efficacy and safety of a hexavalent live human-bovine reassortant rotavirus vaccine(HRV)against rotavirus gastroenteritis(RVGE).A total of 6400 participants aged 6-12 weeks were enrolled and randomly assigned to either HRV(n?3200)or placebo(n?3200)group.All the subjects received three oral doses of vaccine four weeks apart. The vaccine efficacy(VE)against RVGE caused by rotavirus serotypes contained in HRV was evaluated from 14 days after three doses of administration up until the end of the second rotavirus season.VE against severe RVGE,VE against RVGE hospitalization caused by serotypes contained in HRV,and VE against RVGE,severe RVGE,and RVGE hospitalization caused by natural infection of any serotype of rotavirus were also investigated. All adverse events(AEs)were collected for 30 days after each dose.Serious AEs(SAEs)and intussusception cases were collected during the entire study. Our data showed that VE against RVGE caused by serotypes contained in HRV was 69.21%(95%CI:53.31-79.69).VE against severe RVGE and RVGE hospitalization caused by serotypes contained in HRV were 91.36%(95%CI:78.45-96.53)and 89.21%(95%CI:64.51-96.72)respectively.VE against RVGE,severe RVGE,and RVGE hospitalization caused by natural infection of any serotype of rotavirus were 62.88%(95%CI:49.11-72.92),85.51%(95%CI:72.74-92.30)and 83.68%(95%CI:61.34-93.11).Incidences of AEs from the first dose to one month post the third dose in HRV and placebo groups were comparable. There was no significant difference in incidences of SAEs in HRV and placebo groups. This study shows that this hexavalent reassortant rotavirus vaccine is an effective,well-tolerated,and safe vaccine for Chinese infants. | Zhiwei Wu Qingliang Li Yan Liu Huakun Lv Zhaojun Mo Fangjun Li Qingchuan Yu Fei Jin Wei Chen Yong Zhang Teng Huang Xiaosong Hu Wei Xia Jiamei Gao Haisong Zhou Xuan Bai Yueyue Liu Zhenzhen Liang Zhijun Jiang Yingping Chen Jiuwei Zhang Jialiang Du Biao Yang Bo Xing Yantao Xing Ben Dong Qinghai Yang Chen Shi Tingdong Yan Bo Ruan Haiyun Shi Xingliang Fan Dongyang Feng Weigang Lv Dong Zhang Xiangchu Kong Liuyifan Zhou Dinghong Que Hong Chen Zhongbing Chen Xiang Guo Weiwei Zhou Cong Wu Qingrong Zhou Yuqing Liu Jian Qiao Ying Wang Xinguo Li Kai Duan Yuliang Zhao Gelin Xu Xiaoming Yang | 2022 | Virologica Sinica2022,37,5: | 2

Structure-design and theoretical-calculation for ultrasmall Co_(3)O_(4) anchored into ionic liquid modified graphene as anode of flexible lithium-ion batteries显示文摘Cobalt oxide(Co_(3)O_(4))is currently suitable in energy storage applications because of its high capacity based on the conversion reaction mechanism. However, unmodified Co_(3)O_(4)suffers from distinctly inferior rate capability and poor cycling stability. On the basis of the aforementioned considerations and density functional theory(DFT)simulations, the three-dimensional hierarchical porous structure(HPS)ultrasmall Co_(3)O_(4)anchored into ionic liquid(IL)modified graphene oxide(GO)has been successfully prepared(ultrasmall/Co_(3)O_(4)-GA-IL).The ultrasmall/Co_(3)O_(4)-GA-IL consists of Co_(3)O_(4)co-assembled with IL modified GO to generate the HPS which can facilitate ion transfer channels through reduction of the electron and ion transportation path and transmission impedance. In addition,N-doping graphene can enhance the inherent electrical conductivity of Co_(3)O_(4),which is proved by the DFT calculations.By virtue of the novel superstructure, the ultrasmall/Co_(3)O_(4)-GA-IL electrode demonstrates a high reversible capacity of 1,304 mAh·g^(−1),an enhanced high-rate capability(715 mAh·g^(−1)at 5 C),and a capacity retention of 98.4%even after 500 cycles at 5 C rate, which corresponds to 0.0003%capacity loss per cycle.Pouch cells based on the cathode are further fabricated and demonstrate excellent mechanical and electrochemical properties under bent and folded state, highlighting the practical application of our deliberately designed electrode in wearable electronics. | Longda Cong Shichao Zhang Hengyao Zhu Weixin Chen Xueyan Huang Yalan Xing Jun Xia Puheng Yang Xia Lu | 2022 | Nano Research2022,15,3: | 1

Piceatannol enhances Beclin-1 activity to suppress tumor progression and its combination therapy strategy with everolimus in gastric cancer显示文摘The effects and regulation of Beclin-1-an autophagy-related protein-have not been fully defined, however, a negative correlation has been reported between Beclin-1 expression and carcinogenesis. Meanwhile, no compound has been shown to directly inhibit its activity. Here, we evaluate piceatannol, a naturally occurring polyphenolic compound, as a potential targeting agonist of Beclin-1, to assess its efficacy as an antitumor agent against gastric cancer. More specifically, we determine the effects of piceatannol treatment on cell viability using a monitoring system and colony forming assay. Piceatannol was found to efficiently inhibit the proliferation of several human gastric cancer cell lines. Autophagic flux is increased by piceatannol treatment, and correlates with inhibition of cell proliferation and colony formation. Additionally, microscale thermophoresis and surface plasmon resonance results show a direct interaction between piceatannol and Beclin-1, which reduces the phosphorylation activity of Beclin-1 at the Ser-295 site. Notably, piceatannol impairs the binding of Beclin-1 to Bcl-2 and enhances the recruitment of binding of UV radiation resistance-associated gene protein, which further triggers Beclin-1-dependent autophagy signaling. An increase in autophagic activity via treatment with the mTOR inhibitor, everolimus, effectively sensitizes piceatannol-induced antitumor effects. Xenograft models confirmed that piceatannol inhibits tumor development and elicits a potent synergistic effect with everolimus in vivo. Taken together, the findings of this study strongly support the application of combinatorial piceatannol and everolimus therapy in future clinical trials for gastric cancer patients. | Longtao Huangfu Xiaoyang Wang Shanshan Tian Junbing Chen Xueying Wang Biao Fan Qian Yao Gangjian Wang Cong Chen Jing Han Xiaofang Xing Jiafu Ji | 2023 | Science China(Life Sciences)2023,66,2: | 1

Changes of paired-pulse evoked responses during the development of epileptic activity in the hippocampus显示文摘Dysfunction of inhibitory synaptic transmission can destroy the balance between excitatory and inhibitory synaptic inputs in neurons, thereby inducing epileptic activity. The aim of the paper is to investigate the effects of successive excitatory inputs on the epileptic activity induced in the absence of inhibitions.Paired-pulse orthodromic and antidromic stimulations were used to test the changes in the evoked responses in the hippocampus.Picrotoxin(PTX),γ-aminobutyric acid(GABA) type A(GABA A) receptor antagonist, was added to block the inhibitory synaptic transmission and to establish the epileptic model.Extracellular evoked population spike(PS) was recorded in the CA1 region of the hippocampus. The results showed that the application of PTX induced a biphasic change in the paired-pulse ratio of PS amplitude.A short latency increase of the second PS(PS2) was later followed by a reappearance of PS2 depression. This type of depression was observed in both orthodromic and antidromic paired-pulse responses, whereas the GABAergic PS2 depression [called paired-pulse depression(PPD)] during baseline recordings only appeared in orthodromic-evoked responses. In addition, the depression duration at approximately 100 ms was consistent with a relative silent period observed within spontaneous burst discharges induced by prolonged application of PTX.In conclusion, the neurons may ignore the excitatory inputs and intrinsically generate bursts during epileptic activity. The depolarization block could be the mechanisms underlying the PPD in the absence of GABA A inhibitions. The distinct neuronal responses to stimulations during different epileptic stages may implicate the different antiepileptic effects of electrical stimulation. | Zhou-yan FENG Xiao-jing ZHENG Cong TIAN Yang WANG Hao-yu XING | 2011 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,9: | 1
