| 6 | Inhibitory effects of matrine on electrical signals and amino acid neurotransmitters in hippocampal brain slices显示文摘<正>BACKGROUND:Studies on electrical signals of hippocampal brain slices in vivo have shown that matrine inhibits benzylpenicillin sodium-induced activation of neuronal signal transduction. OBJECTIVE:To verify the inhibition effect of matrine on activation of electrical signals in rat brain slices and the role matrine plays in hippocampal amino acid transmitter release. DESIGN,TIME AND SEFFING:The in vitro,neurophysiological,controlled experiment was performed in the Zhejiang Province Key Laboratory of Cardio-cerebrovascular Disease and Nerve System Drugs Appraisement and Chinese Traditional Medicine Screening and Research between July 2003 and May 2004.The in vivo,neuronal,biochemical experiment was performed in the Zhejiang Province Key Laboratory of Chinese Traditional Medicine Quality Standardization from July 2005 to December 2006. MATERIALS:Forty healthy,Sprague Dawley rats,7-8 weeks old,and 120 healthy,ICR mice,5-6 weeks old,were included in this study,irrespective of gender.Matrine powder was provided by the National Institute for the Control of Pharmaceutical and Biological Products,China.Matrine injection was purchased from Zhuhai Biochemical Pharmaceutical Factory,China.Penicillin was bought from Shijiazhuang Pharmaceutical Group Co.,Ltd.,China. METHODS:(1) Rats were randomly assigned to four groups:control,penicillin model,and matrine high-dose and low-dose,with 10 rats in each group.The control group was perfused with artificial cerebrospinal fluid.In the remaining three groups,hippocampal brain slices were perfused with normal artificial cerebrospinal fluid containing 1×10~6 U/L penicillin for the first 10 minutes.The penicillin model group received artificial cerebrospinal fluid for an additional 30 minutes,while the matrine high-dose and low-dose groups received 0.1 g/L and 0.05 g/L matrine,respectively,for an additional 30 minutes.(2) Mice were randomly assigned to four groups(n=30).The matrine high-, medium-,and low-dose groups were separately injected with 58.5,39.0,and 19.5 mg/kg matrine via caudal vein,respectively.No intervention was administered to the normal group. MAIN OUTCOME MEASURES:The field potential value in the CA1 region of penicillin-induced rat hippocampal brain slices was analyzed using the evoked field potential technique;chromatography was utilized to determine y-aminobutyric acid and glutamic acid content in the mouse hippocampus. RESULTS:(1) Both 0.1 g/L and 0.05 g/L matrine reduced the number of evoked field potentials in the penicillin-induced rat hippocampal brain slices(P<0.05 or P<0.01).In addition,0.1 g/L matrine led to a reduction of evoked field potential amplitude(P<0.05).(2) Compared with normal mice, y-aminobutyric levels were dramatically increased at 20 minutes after high-dose matrine treatment (P<0.05).In addition,significantly increased y-aminobutyric acid levels were observed at 40 minutes after medium-and low-dose matrine treatments(P<0.05 or P<0.01).The glutamic acid/y-aminobutyric acid ratio was significantly less at 20 minutes after high-dose matrine treatment compared with the normal mice group(P<0.05). | 7 | Bone Marrow Urokinase Plasminogen Activator Receptor Levels are Associated with the Progress of Multiple Myeloma显示文摘Objective To determine the mRNA and protein levels of urokinase plasminogen activator receptors(u PAR) in bone marrow fluid and bone marrow tissue from multiple myeloma(MM) patients and assess association of u PAR level with prognosis of MM.Methods u PAR levels in bone marrow fluid of 22 MM patients at the stable and progressive stages and 18 iron deficiency anemia patients with normal bone marrow(control) were examined by ELISA.Furthermore,u PAR expression in bone marrow tissue was investigated by RT-PCR and Western blot,respectively.The distribution of u PAR in MM cells was examined using immunofluorescence staining.The pathological changes in different stages of MM patients were studied by HE staining.Results u PAR level in bone marrow fluid of MM patients(1.52±0.32 μg/ml) was found to be higher than that in the control group(0.98±0.15 μg/ml).Interestingly,u PAR protein(0.686±0.075 vs.0.372±0.043,P<0.05) and m RNA(2.51±0.46 vs.4.46±1.15,P<0.05) expression levels of MM patients at the progressive stage were significantly higher than those at the stable stage.The expression of u PAR in MM bone marrow was confirmed by immunofluorescence staining.Moreover,HE staining revealed a great increased number of nucleated cells and severe impairment of hematopoietic function in the bone marrow of patients with progressive-stage myeloma.Conclusion Our study reveals that u PAR expression is positively correlated with the development and progress of MM. | Li-hong Shou Dan Cao Xiao-hui Dong Qiu Fang Bao-lian Xu Ju-ping Fei | 2016 | Chinese Medical Sciences Journal2016,31,3: | 0 |

| 9 | High-efficient removal of tetracycline in water via porous magnetic Ce/Fe photocomposite under visible light显示文摘Tetracycline is a typical antibiotic commonly used in various industries which is eco-toxic and quickly causes bacterial resistance.Therefo re,studying the efficient removal of tetracycline is necessary to protect the water environment.Herein,a novel Ce/Fe nanoparticle composite(1CCFO)was prepared by the sol-gel method and its removal effects of tetracycline under visible light were performed.The relationship between physicochemical properties of catalyst and photocatalytic degradation effects of tetracycline was analyzed based on a series of characterizations data such as X-ray diffraction(XRD),Raman spectro scopy,a vibrating sample magnetometer(VSM),scanning electron microscopy(SEM),Brunauer-Emmett-Teller(BET)method,X-ray photoelectron spectroscopy(XPS),and ultraviolet-visible(UV-Vis)spectroscopy.The results show that Ce/Fe photocatalyst possesses a large specific surface area,good visible light response,abundant oxygen vacancies and excellent redox performance,exhibiting good adsorption capacity,remarkable catalytic performance and stability.The optimal conditions for tetracycline removal were explored through orthogonal experiments.About 88%of tetracycline can be photodegraded in 1 h under optimal conditions.The possible decomposition pathways,main reactive oxygen species and suitable mechanism of the photocatalytic system were studied by liquid chromatography-mass spectro metry(LC-MS),an electron spin resonance(ESR)spectrometer and free radical quenching experiments.The results show that 1CCFO has an efficient degradation effect on tetracycline under visible light,which provides a feasible method to improve the performance of 1CCFO. | Weihang Han Jingxuan Shou Yifan Yang Liangchen Chen Luping Zhang Yutong Chen Xuewei Tu Dan Jin Shijie Zhang Yurong Chang Hui Zheng | 2023 | Journal of Rare Earths2023,41,10: | 0 |

| 13 | Prenatal inflammation exposure-programmed hypertension exhibits multi-generational inheritance via disrupting DNA methylome显示文摘The multi-generation heredity trait of hypertension in human has been reported,but the molecular mechanisms underlying multi-generational inheritance of hypertension remain obscure.Recent evidence shows that prenatal inflammatory exposure(PIE)results in increased incidence of cardiovascular diseases,including hypertension.In this study we investigated whether and how PIE contributed to multi-generational inheritance of hypertension in rats.PIE was induced in pregnant rats by intraperitoneal injection of LPS or Poly(I:C)either once on gestational day 10.5(transient stimulation,T)or three times on gestational day 8.5,10.5,and 12.5(persistent stimulation,P).Male offspring was chosen to study the paternal inheritance.We showed that PIE,irrespectively induced by LPS or Poly(I:C)stimulation during pregnancy,resulted in multi-generational inheritance of significantly increased blood pressure in rat descendants,and that prenatal LPS exposure led to vascular remodeling and vasoconstrictor dysfunction in both thoracic aorta and superior mesenteric artery of adult F2 offspring.Furthermore,we revealed that PIE resulted in global alteration of DNA methylome in thoracic aorta of F2 offspring.Specifically,PIE led to the DNA hypomethylation of G beta gamma(Gβγ)signaling genes in both the F1 sperm and the F2 thoracic aorta,and activation of PI3K/Akt signaling was implicated in the pathologic changes and dysregulated vascular tone of aortic tissue in F2 LPS-P offspring.Our data demonstrate that PIE reprogrammed DNA methylome of cells from the germline/mature gametes contributes to the development of hypertension in F2 PIE offspring.This study broadens the current knowledge regarding the multi-generation effect of the cumulative early life environmental factors on the development of hypertension. | Xiao Guan Guo-rong Dan Yao Yang Yan Ji Wen-jing Lai Fang-jie Wang Meng Meng Bang-hui Mo Pei Huang Ting-ting You Ya-fei Deng Liang Song Wei Guo Ping Yi Jian-hua Yu Yuan Gao Wei-nian Shou Bing-bo Chen You-cai Deng Xiao-hui Li | 2022 | Acta Pharmacologica Sinica2022,43,6: | 0 |
