| 1 | Chemical composition and in vitro fermentation characteristics of high sugar forage sorghum as an alternative to forage maize for silage making in Tarim Basin, China显示文摘The chemical composition, mineral profile and in vitro fermentation characteristics of maize(MZ), high sugar forage sorghum(HS) and forage sorghum(FS), and silages made from each forage type were measured. The MZ and MZ silage(MZS) had higher crude protein, starch and ether extract contents than both sorghum forages and sorghum silages. HS had higher ash and water-soluble carbohydrate concentrations than FS and MZ. MZ, MZS, HS and HS silage(HSS) had lower neutral detergent fibre, acid detergent fibre and acid detergent lignin than FS and FS silage(FSS). FSS had higher dry matter(DM) and p H than MZS and HSS. HSS contained higher concentrations of P and K than FSS and MZS. MZS and HSS had higher in vitro dry matter and organic matter digestibility, CH4 production, total volatile fatty acids, acetate and propionate than FSS. p H was higher for FSS than for HSS, and ammonia was lower for HSS than for MZS and FSS. HSS had higher gas production than MZS and FSS after 2, 4, 6 and 8 h incubation. MZS had higher gas production than HSS and FSS after 26 and 28 h of incubation. The results indicate that HS may substitute for MZ to make good quality silage. | 2 | The adult human testis transcriptional cell atlas显示文摘Human adult spermatogenesis balances spermatogonial stem cell (SSC) self-renewal and differentiation,alongside complex germ cell-niche interactions,to ensure long-term fertility and faithful genome propagation. Here,we performed single-cell RNA sequencing of -6500 testicular cells from young adults. We found five niche/somatic cell types (Leydig,myoid,Sertoli,endothelial,macrophage),and observed germline-niche interactions and key human-mouse differences. Spermatogenesis,including meiosis,was reconstructed computationally,revealing sequential coding,non-coding,and repeat-element transcriptional signatures. Interestingly,we identified five discrete transcriptional/developmental spermatogonial states,including a novel early SSC state,termed State 0. Epigenetic features and nascent transcription analyses suggested developmental plasticity within spermatogonial States. To understand the or丨gin of State 0,we profiled testicular cells from infants,and identified distinct similarities between adult State 0 and infant SSCs. | 3 | Abnormal subchondral bone remodeling and its association with articular cartilage degradation in knees of type 2 diabetes patients显示文摘Type 2 diabetes(T2 D) is associated with systemic abnormal bone remodeling and bone loss. Meanwhile,abnormal subchondral bone remodeling induces cartilage degradation, resulting in osteoarthritis(OA).Accordingly, we investigated alterations in subchondral bone remodeling, microstructure and strength in knees from T2 D patients and their association with cartilage degradation. Tibial plateaus were collected from knee OA patients undergoing total knee arthroplasty and divided into non-diabetic(n = 70) and diabetes(n = 51) groups. Tibial plateaus were also collected from cadaver donors(n = 20) and used as controls.Subchondral bone microstructure was assessed using micro-computed tomography. Bone strength was evaluated by micro-finite-element analysis. Cartilage degradation was estimated using histology. The expression of tartrate-resistant acidic phosphatase(TRAP), osterix, and osteocalcin were calculated using immunohistochemistry. Osteoarthritis Research Society International(OARSI) scores of lateral tibial plateau did not differ between non-diabetic and diabetes groups, while higher OARSI scores on medial side were detected in diabetes group. Lower bone volume fraction and trabecular number and higher structure model index were found on both sides in diabetes group. These microstructural alterations translated into lower elastic modulus in diabetes group. Moreover, diabetes group had a larger number of TRAP^+ osteoclasts and lower number of Osterix^+ osteoprogenitors and Osteocalcin^+ osteoblasts. T2 D knees are characterized by abnormal subchondral bone remodeling and microstructural and mechanical impairments, which were associated with exacerbated cartilage degradation. | 4 | Mechanosignaling activation of TGFβmaintains intervertebral disc homeostasis显示文摘Intervertebral disc(IVD) degeneration is the leading cause of disability with no disease-modifying treatment.IVD degeneration is associated with instable mechanical loading in the spine, but little is known about how mechanical stress regulates nucleus notochordal(NC) cells to maintain IVD homeostasis. Here we report that mechanical stress can result in excessive integrin α_vβ_6-mediated activation of transforming growth factor beta(TGFβ), decreased NC cell vacuoles, and increased matrix proteoglycan production, and results in degenerative disc disease(DDD). Knockout of TGFβ type II receptor(TβRII) or integrin α_v in the NC cells inhibited functional activity of postnatal NC cells and also resulted in DDD under mechanical loading.Administration of RGD peptide, TGFβ, and α_vβ_6-neutralizing antibodies attenuated IVD degeneration. Thus,integrin-mediated activation of TGFβ plays a critical role in mechanical signaling transduction to regulate IVD cell function and homeostasis. | 5 | Application of Avaatech X-ray fluorescence core-scanning in Sr/Ca analysis of speleothems显示文摘Application of X-ray fluorescence core-scanning(XRF-CS) on both marine and lake sediments has achieved remarkable results. However, its application has not been widely extended to the research on speleothems. In this study, we measure the Sr abundance and the Sr/Ca ratios of three stalagmites(two aragonite stalagmites, one calcite stalagmite) using the state-of-the-art fourth-generation Avaatech high-resolution XRF core scanner. Through comparisons among different scan paths and among different scan resolutions, as well as comparisons with inductively coupled plasma optical emission spectrometer(ICP-OES), Itrax XRF, and Artax XRF results, we confirm that the Avaatech XRF core scanner could precisely, quickly, and nondestructively analyze the high-resolution Sr abundance of speleothems. | 6 | Influence of(FeO+Ti O_2) abundance on the microwave thermal emissions of lunar regolith显示文摘One of the essential controls on the microwave thermal emissions(MTE) of the lunar regolith is the abundance of Fe O and TiO_2, known as the(Fe O+Ti O_2) abundance(FTA). In this paper, a radiative transfer simulation is employed first to study the change in the brightness temperature(T_B) with FTA under a range of frequencies and surface temperatures. Then, we analyze the influence of FTA on the MTE of the lunar regolith using microwave sounder(CELMS) data from the Chang'E-2 lunar orbiter, Clementine UV-VIS data, and lunar samples recovered from the Apollo and Surveyor projects. We conclude that:(1) FTA strongly influences the MTE of the lunar regolith, but it is not the decisive control, and(2) FTA decreases slightly with depth. | 7 | Challenges and needs for China to eliminate rabies显示文摘In China,rabies is a significant public health concern where dogs remain the main reservoir of disease transmission to humans;rabies-related mortality ranks second in the world.We compiled all published articles and official documents on rabies in China's Mainland to examine challenges and needs to eliminate rabies in the country.The Chinese authorities have identified rabies as a priority,recognized rabies control in dogs as key to control rabies in humans and required intersectoral collaborations.Efforts have been made to respond effectively to the latest re-emergence of rabies,which peaked in 2007 with>3,300 cases.Despite these outcomes and the increasing volume of publications and regulations in the recent years,our review points to some major information gaps to improve rabies control activities and envisage elimination program.An emphasis on laboratory or pathogen-associated and basic epidemiology research in the literature has contrasted with the absence of information to monitor various systems in humans and animals(e.g.quality of surveillance,response and post-exposure prophylaxis).Information is also lacking to appropriately inform policymakers(e.g.economic disease burden,impact of policies)and assist program managers(e.g.comprehensive and strategic guidance for cost-effective prevention and control activities,public education and dog population management).In conclusion,strategic planning is needed to provide a sense of direction,demonstrate feasibility of elimination in China,and develop a research agenda,addressing country’s operational needs and constraints.The planning should be a multisectoral effort. | Wenwu Yin Jie Dong Changchun Tu John Edwards Fusheng Guo Hang Zhou Hongjie Yu Sirenda Vong | 2013 | Infectious Diseases of Poverty2013，2，1： | 3 |
| 8 | Tracking domestic ducks:A novel approach for documenting poultry market chains in the context of avian influenza transmission显示文摘Agro-ecological conditions associated with the spread and persistence of highly pathogenic avian influenza(HPAI) are not well understood, but the trade of live poultry is suspected to be a major pathway. Although market chains of live bird trade have been studied through indirect means including interviews and questionnaires, direct methods have not been used to identify movements of individual poultry. To bridge the knowledge gap on quantitative movement and transportation of poultry, we introduced a novel approach for applying telemetry to document domestic duck movements from source farms at Poyang Lake, China. We deployed recently developed transmitters that record Global Positioning System(GPS) locations and send them through the Groupe Spécial Mobile(GSM) cellular telephone system. For the first time, we were able to track individually marked ducks from 3 to 396 km from their origin to other farms, distribution facilities, or live bird markets. Our proof of concept test showed that the use of GPS-GSM transmitters may provide direct, quantitative information to document the movement of poultry and reveal their market chains.
