| 2 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 3 | Expression of reconstructive hF Ⅷ in the hrDNA by using hrDNA targeting vector显示文摘Our lab has constructed a new nonviral vec-tor—hrDNA targeting vector(pHrneo). pHrneo is a human derived vector that can target gene into human ribosomal DNA(hrDNA) locus. In this study, we inserted expression cassette of reconstructive hF Ⅷ (hFVIII-BDDAK39) to pHrneo to construct targeting vector: pHrneo-BDDAK39. Through electroporation of pHrneo-BDDAK39 into HT1080 cells, we identified the homologous recombinants by PCR and Southen blotting, and tested the expression of hFVIII- BDDAK39 in the hrDNA locus. The hFⅧ-BDDAK39 was successfully targeted into hrDNA locus of HT-1080 by pHrneo-BDDAK39, and the efficiency of site-specific inte-gration was 2.0×10?5. hFⅧ-BDDAK39 in hrDNA locus of HT-1080 is found to be able to express efficiently (32±5 ng·106 cells?1·24 h?1). | 4 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 6 | Seasonal variation of microelement contents in leaves of Cyclocarea paliurus among the provenances显示文摘Cyclocarya paliurus as a multiple function plant can accumulate biologically important microelement elements.To reveal the variation of selected microelement concentrations in leaves of C.paliurus provenances during the growing season,12 C.paliurus provenances in the field trial were sampled five times at approximately 1-month intervals.The method of inductively coupled plasma optical emission spectrometer(ICP-OES) was employed to determinate average concentrations of Fe,Mn,Zn,Cu and Se in leaves of 12 C.paliurus provenances.The results show that on average,the concentrations of five microelement in the leaves follows an order of Fe > Mn > Zn > Cu > Se.Variance analysis shows that there are significant differences in Fe,Mn and Zn concentrations among the twelve provenances(p<0.05),while there is no significant difference between Cu and Se concentrations.A significant difference was also observed in the concentrations of five microelements at the different sampling times(p<0.001),but the mean concentrations for each microelement showed different temporal dynamic patterns.Meanwhile,a significant correlation between concentrations of Se and other measured microelements was detected in the leaves of C.paliurus,except for Mn.Obtained results not only demonstrated that leaves of C.paliurus exhibited higher levels of microelements(Fe,Mn,Cu,Zn and Se),but also provided a basis for breeding strategies of superior provenances with rich content of microelements,and choosing optimum harvesting time for food industry in future. | FANG Sheng-zuo CHU Xiu-li SHANG Xu-lan YANG Wan-xia FU Xiang-xiang SHE Cheng-qi | 2011 | Journal of Forestry Research2011,22,2: | 3 |
| 7 | Protective effect of Sijunzi decoction on neuromuscular junction ultrastructure in autoimmune myasthenia gravis rats显示文摘OBJECTⅣE: To investigate the protective role of Sijunzi decoction in neuromuscular junction(NMJ)and muscle cell mitochondria ultrastructure; as well as its effects on the amount of adenosine triphosphate(ATP) and the activities of mitochondrial respiratory chain complexes I, Ⅱ, Ⅲ, and Ⅳ in autoimmune myasthenia gravis rats.METHODS: An experimental autoimmune myasthenia gravis(EAMG) rat model was established by inoculating rats with acetylcholine receptors extracted from Torpedo. Rats were divided into three groups: model, prednisone, and Sijunzi decoction, and were fed physiological saline, prednisone, or Sijunzi decoction, respectively. NMJ and muscle cell mitochondria ultrastructure were observed by transmission electron microscope. The amount of ATP was assessed by high performance liquid chromatography. The activities ofmitochondrial respiratory chain complexes I, Ⅱ, Ⅲ,and Ⅳ was determined using the Clark oxygen electrode method.RESULTS: In the model group, there were sparse muscle fibers, with decreased mitochondria, and sparse, diffluent, or absent NMJ folds. After intervention with Sijunzi decoction, the above pathology changes were improved: muscle fiber structure was clear and complete; the mitochondria count was higher; and the NMJ structure was close to normal. Gastrocnemius muscle mitochondria in the model group produced significantly less ATP than those in the prednisone group(P<0.01). Conversely, the ATP of Sijunzi decoction group was significantly higher than prednisone group(P<0.01). The activities of gastrocnemius muscle mitochondrial respiratory chain complexes I, Ⅱ, Ⅲ, and Ⅳ in both the prednisone and Sijunzi decoction groups was dramatically higher compared with the model group(P<0.05). | 8 | Tailoring of electromagnetic field localizations by twodimensional graphene nanostructures显示文摘Graphene has great potential for enhancing light−matter interactions in a two-dimensional regime due to surface plasmons with low loss and strong light confinement.Further utilization of graphene in nanophotonics relies on the precise control of light localization properties.Here,we demonstrate the tailoring of electromagnetic field localizations in the mid-infrared region by precisely shaping the graphene into nanostructures with different geometries.We generalize the phenomenological cavity model and employ nanoimaging techniques to quantitatively calculate and experimentally visualize the two-dimensional electromagnetic field distributions within the nanostructures,which indicate that the electromagnetic field can be shaped into specific patterns depending on the shapes and sizes of the nanostructures.Furthermore,we show that the light localization performance can be further improved by reducing the sizes of the nanostructures,where a lateral confinement of λ0/180 of the incidence light can be achieved.The electromagnetic field localizations within a nanostructure with a specific geometry can also be modulated by chemical doping.Our strategies can,in principle,be generalized to other two-dimensional materials,therefore providing new degrees of freedom for designing nanophotonic components capable of tailoring two-dimensional light confinement over a broad wavelength range. | Ze-Bo Zheng Jun-Tao Li Teng Ma Han-Lin Fang Wen-Cai Ren Jun Chen Jun-Cong She Yu Zhang Fei Liu Huan-Jun Chen Shao-Zhi Deng Ning-Sheng Xu | 2017 | Light(Science & Applications)2017,6,1: | 2 |
