1Genome-wide Identification and Classification of HSF Family in Grape, and Their Transcriptional Analysis under Heat Acclimation and Heat Stress显示文摘Heat stress is one of the most serious threats in viticulture. Heat shock transcription factors(HSFs) have been proven to be the most important transcriptional factor family in the plant response to heat. In this study, a total of 19 putative HSF genes(VHSFs) were identified from the 12 × Vitis vinifera Pinot Noir PN4002 genome, and their chromosomal location, phylogenetic tree, exon-intron structure and conserved motifs were also analyzed. In addition, these HSFs transcriptional profiles in Vitis vinifera 'Jingxiu'(weak heat tolerance) and Vitis davidii 'Tangwei'(strong heat tolerance) under heat stress(47℃) and heat acclimation(38℃) were presented. Only VHSF01, VHSF05, VHSF15 and VHSF18 among 19 VHSFs showed different transcriptional models in these two genotypes under the same treatment, suggesting that these four VHSFs may be important factors which result in a thermotolerance difference between 'Jingxiu' and 'Tangwei'. 2Power electronic transformer with adaptive PLL technique for voltage-disturbance ride through显示文摘The power electronic transformer(PET) has recently emerged as a type of power converter. It features the basic functions of power conversion and isolation as well as additional functions related to power quality control. A novel PET for a distribution grid called a flexible power distribution unit is proposed in this paper, and the energy exchange mechanism between the network and the load is revealed. A 30 kW 600 VAC/220 VAC/110 VDC medium-frequency isolated prototype is developed and demonstrated. This paper also presents key control strategies of the PET for electrical distribution grid applications,especially under grid voltage disturbance conditions.Moreover, stability issues related to the grid-connected three-phase PET are discussed and verified with an impedance-based analysis. The PET prototype is tested, and it passes the voltage-disturbance ride-through function. 3Cellular vesicles expressing PD-1-blocking scFv reinvigorate T cell immunity against cancer显示文摘Cancer cells aberrantly express immunosuppressive checkpoint ligands and produce certain metabolites that lead to T cell exhaustion.Immune checkpoint blockade(ICB)therapy that reinvigorates exhausted T cells have achieved impressive response in clinical cancer treatment.However,the limited clinical response rate and off-tumor toxicities restrict ICB therapy.Herein,cellular vesicles displaying anti-programmed cell death-1(PD-1)single-chain variable fragment antibody(aPD-1-scFv)were prepared to reinvigorate T cell immunity to counteract cancer.The nanovesicles displaying aPD-1-scFv(aPD-1-scFv NVs)could enhance the anti-tumor activation of T cells through PD-1 blockade.Furthermore,NVs loading the A_(2a)adenosine receptor(A_(2a)R)antagonist CPI-444 assisted T cells to antagonize adenosine,an immunosuppressive metabolite produced by cancer cells.Hence,CPI-444 loaded aPD-1-scFv NVs could intensively increase the density and activity of tumor infiltrating T cells,directly restraining tumor progress and metastasis.Tianyuan Xue Zhirang Zhang Tianliang Fang Baoqi Li Yuan Li Liyan Li Yanghua Jiang Fangfang Duan Fanqiang Meng Xin Liang Xudong Zhang 2022Nano Research2022,15,6:1

5Characterization of 3D DNA Assemblies Using Cryogenic Electron Microscopy显示文摘DNA nanotechnology utilizes DNA double strands as building units for self-assembly of DNA nanostructures.The specific base-pairing interaction between DNA molecules is the basis of these assemblies.After decades of development,this technology has been able to construct complex and programmable structures.With the increase in delicate nature and complexity of the synthesized nanostructures,a characterization technology that can observe these structures in three dimensions has become necessary,and developing such a technology is considerably challenging.DNA assemblies have been studied using different characterization methods including atomic force microscopy(AFM),scanning electron microscopy(SEM),and transmission electron microscopy(TEM).However,the three-dimensional(3D)DNA assemblies always collapse locally due to the dehydration during the drying process.Cryogenic electron microscopy(cryo-EM)can overcome the challenge by maintaining three-dimensional morphologies of the cryogenic samples and reconstruct the 3D models from cryogenic samples accordingly by collecting thousands of two-dimensional(2D)projection images,which can restore their original morphologies in solution.Here,we have reviewed several typical cases of 3D DNA-assemblies and highlighted the applications of cryo-EM in characterization of these assemblies.By comparing with some other characterization methods,we have shown how cryo-EM promoted the development of structural characterization in the field of DNA nanotechnology.WANG Mingyang DUAN Jialin DAI Lizhi XIN Xiaodong WANG Fangfang LI Zheng TIAN Ye 2020Chemical Research in Chinese Universities2020,36,2:0

7Evaluation of combined detection of nuclear factor erythroid 2-related factor 2 and glutathione peroxidase 4 in primary hepatic carcinoma and preliminary exploration of pathogenesis显示文摘This study aims to analyze the clinical significance and mechanism of nuclear factor erythroid 2-related factor 2(NRF2)and glutathione peroxidase 4(GPX4)in primary hepatic carcinoma(PHC).Methods:The expression of NRF2 and GPX4 in peripheral blood of patients with PHC was determined to analyze the diagnostic value of the two combined for PHC.The prognostic significance of NRF2 and GPX4 was evaluated by 3-year followup.Human liver epithelial cells THLE-2 and human hepatocellular carcinoma cells HepG2 were purchased,and the expression of NRF2 and GPX4 in the cells was determined.NRF2 and GPX4 aberrant expression vectors were constructed and transfected into HepG2,and changes in cell proliferation and invasion capabilities were observed.Results:The expression of NRF2 and GPX4 in patients with PHC was higher than that in patients with LC or VH(p<0.05),and the two indicators combined was excellent in diagnosing PHC.Moreover,patients with high expression of NRF2 and GPX4 had a higher risk of death(p<0.05).In in vitro experiments,both NRF2 and GPX4 expression was elevated in HepG2(p<0.05).HepG2 activity was enhanced by increasing the expression of the two,vice versa(p<0.05).Conclusion:NRF2 and GPX4 combined is excellent in diagnosing PHC,and promotes the malignant development of PHC.JIE DUAN AIDONG GU WEI CHEN CHANGHAO CHEN FANGNAN SONG FAXI CHEN FANGFANG JIANG HUIWEN XING 2023BIOCELL2023,47,12:0

13Rapid field testing of mercury pollution by designed fluorescent biosensor and its cells-alginate hydrogel-based paper assay显示文摘With increasing industrial activities,mercury has been largely discharged into environment and caused serious environmental problems.The growing level of mercury pollution has become a huge threat to human health due to its significant biotoxicity.Therefore,the simple and fast means for on-site monitoring discharged mercury pollution are highly necessary to protect human beings from its pernicious effects in time.Herein,a'turn off'fluorescent biosensor(mCherry L199 C)for sensing Hg2+was successfully designed based on direct modification of the chromophore environment of fluorescent protein mCherry.For rapid screening and characterization,the designed variant of mCherry(mCherry L199 C)was directly expressed on outer-membrane of Escherichia coli cells by cell surface display technique.The fluorescent biosensor was characterized to have favorable response to Hg2+at micromole level among other metal ions and over a broad pH range.Further,the cells of the fluorescent biosensor were encapsulated in alginate hydrogel to develop the cells-alginate hydrogel-based paper.The cells-alginate hydrogel-based paper could detect mercury pollution in 5 min with simple operation process and inexpensive equipment,and it could keep fluorescence and activity stable at 4℃ for 24 hr,which would be a high-throughput screening tool in preliminarily reporting the presence of mercury pollution in natural setting.Yanan Zheng Liudan Wei Linwei Duan Fangfang Yang Guixiang Huang Tianyi Xiao Min Wei Yanling Liang Huiting Yang Zhipeng Li Dan Wang 2021Journal of Environmental Sciences2021,33,8:0

15用于放射性束物理实验的探测硅阵列的研制显示文摘With the development of radioactive beam facilities,several exciting results were provided concerning the understanding of fundamental nuclear physics.However,typical-beam intensities are always low and the production is often in very small quantities,so,new generation detectors need to be developed with large solid angle and angular coverage,which can identify particle effectively as well as possesses high energy resolution,high spatial resolution and high geometric efficiency.The new detector based on Double Side Silicon Strip Detector(DSSD)is foreseen for the study of the reaction production in the experiment of direct nuclear reaction above the Coulomb barrier using RIBLL[1,2]at the HIRFL[3,4].However,the disadvantage of the RIBs is that it has a broad distribution in coordinate spaces.Therefore,new detector system with high resolution and a large solid angle was developed successfully for performing nuclear reaction experiments using RIBs.The geometric efficiency of the new detector array with 5 large area DSSD telescopes increases up to 70%.Duan Fangfang Yang Yanyun Gao Zhihao Liu Xingquan Ma Peng Wang Jiansong Bai Zhen Ma Junbing Hu Qiang Jin Shilun Jin Shuya Yu Gongming 2017IMP & HIRFL Annual Report2017,,1:0
