| 1 | Nomogram to predict overall survival after gallbladder cancer resection in China显示文摘AIM To integrate clinically significant variables related to prognosis after curative resection for gallbladder carcinoma(GBC) into a predictive nomogram.METHODS One hundred and forty-two GBC patients who underwent curative intent surgical resection at Peking Union Medical College Hospital(PUMCH) were included. This retrospective case study was conducted at PUMCH of the Chinese Academy of Medical Sciences and Peking Union Medical College(CAMS & PUMC) in China from January 1, 2003 to January 1, 2018. The continuous variable carbohydrate antigen 19-9(CA19-9) was converted into a categorical variable(cCA19-9) based on the normal reference range. Stages 0 to IIIA were merged into one category, while the remaining stages were grouped into another category. Pathological grade X(GX) was treated as a missing value. A multivariate Cox proportional hazards model was used to select variables to construct a nomogram. Discrimination and calibration of the nomogram were performed via the concordance index(C-index) and calibration plots. The performance of the nomogram was estimated using the calibration curve. Receiver operating characteristic(ROC) curve analysis and decision curve analysis(DCA) were performed to evaluate the predictive accuracy and net benefit of the nomogram, respectively.RESULTS Of these 142 GBC patients, 55(38.7%) were male, and the median and mean age were 64 and 63.9 years, respectively. Forty-eight(33.8%) patients in this cohort were censored in the survival analysis. The median survival time was 20 months. A series of methods, including the likelihood ratio test and Akaike information criterion(AIC) as well as stepwise, forward, and backward analyses, were used to select the model, and all yielded identical results. Jaundice [hazard ratio(HR) = 2.9; 95% confidence interval(CI): 1.60-5.27], cCA19-9(HR = 3.2; 95%CI: 1.91-5.39), stage(HR = 1.89; 95%CI: 1.16-3.09), and resection(R)(HR = 2.82; 95%CI: 1.54-5.16) were selected as significant predictors and combined into a survival time predictive nomogram(C-index = 0.803; 95%CI: 0.766-0.839). High prediction accuracy(adjusted C-index = 0.797) was further verified via bootstrap validation. The calibration plot demonstrated good performance of the nomogram. ROC curve analysis revealed a high sensitivity and specificity. | 2 | Distributed Collaborative Response Surface Method for Mechanical Dynamic Assembly Reliability Design显示文摘Because of the randomness of many impact factors influencing the dynamic assembly relationship of complex machinery,the reliability analysis of dynamic assembly relationship needs to be accomplished considering the randomness from a probabilistic perspective.To improve the accuracy and efficiency of dynamic assembly relationship reliability analysis,the mechanical dynamic assembly reliability(MDAR)theory and a distributed collaborative response surface method(DCRSM)are proposed.The mathematic model of DCRSM is established based on the quadratic response surface function,and verified by the assembly relationship reliability analysis of aeroengine high pressure turbine(HPT)blade-tip radial running clearance(BTRRC).Through the comparison of the DCRSM,traditional response surface method(RSM)and Monte Carlo Method(MCM),the results show that the DCRSM is not able to accomplish the computational task which is impossible for the other methods when the number of simulation is more than 100 000times,but also the computational precision for the DCRSM is basically consistent with the MCM and improved by 0.40~4.63%to the RSM,furthermore,the computational efficiency of DCRSM is up to about 188 times of the MCM and 55 times of the RSM under10000 times simulations.The DCRSM is demonstrated to be a feasible and effective approach for markedly improving the computational efficiency and accuracy of MDAR analysis.Thus,the proposed research provides the promising theory and method for the MDAR design and optimization,and opens a novel research direction of probabilistic analysis for developing the high-performance and high-reliability of aeroengine. | BAI Guangchen FEI Chengwei | 2013 | Chinese Journal of Mechanical Engineering2013,26,6: | 23 |
| 3 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. | 4 | Genotypic Characterization of Methicillin-resistant Staphylococcus aureus Isolated from Pigs and Retail Foods in China显示文摘Objective To investigate the genotypic diversity of Methicillin-resistant Staphylococcus aureus(MRSA) isolated from pigs and retail foods from different geographical areas in China and further to study the routes and rates of transmission of this pathogen from animals to food. Methods Seventy-one MRSA isolates were obtained from pigs and retail foods and then characterized by multi-locus sequencing typing(MLST), spa typing, multiple-locus variable number of tandem repeat analysis(MLVA), pulsed-field gel electrophoresis(PFGE), and antimicrobial susceptibility testing. Results All isolated MRSA exhibited multi-drug resistance(MDR). Greater diversity was found in food-associated MRSA(7 STs, 8 spa types, and 10 MLVA patterns) compared to pig-associated MRSA(3 STs, 1 spa type, and 6 MLVA patterns). PFGE patterns were more diverse for pig-associated MRSA than those of food-associated isolates(40 vs. 11 pulse types). Among the pig-associated isolates, CC9-ST9-t899-MC2236 was the most prevalent clone(96.4%), and CC9-ST9-t437-MC621(20.0%) was the predominant clone among the food-associated isolates. The CC9-ST9 isolates showed significantly higher antimicrobial resistance than other clones. Interestingly, CC398-ST398-t034 clone was identified from both pig-and food-associated isolates. Of note, some community-and hospital-associated MRSA strains(t030, t172, t1244, and t4549) were also identified as food-associated isolates. Conclusion CC9-ST9-t899-MC2236-MDR was the most predominant clone in pigs, but significant genetic diversity was observed in food-associated MRSA. | 6 | Chinese physicians' perceptions of fecal microbiota transplantation显示文摘AIM: To explore Chinese physicians' perceptions towards fecal microbiota transplantation(FMT) and to provide information and an assessment of FMT development in China.METHODS: A self-administered questionnaire was developed according to the FMT practice guidelines and was distributed to physicians in hospitals via Internet Research Electronic Data Capture(REDcap) software and electronic mails to assess their attitudes toward and knowledge of FMT. The questionnaire included a brief introduction of FMT that was followed by 20 questions. The participants were required to respond voluntarily, under the condition of anonymity and without compensation. Except for the fill-in-the-blank questions, all of the other questions were required in the REDcap data collection systems, and the emailed questionnaires were completed based on eligibility.RESULTS: Up to December 9, 2014, 844 eligible questionnaires were received out of the 980 distributed questionnaires, with a response rate of 86.1%. Among the participants, 87.3% were from tertiary hospitals, and there were 647(76.7%) gastroenterologists and 197(23.3%) physicians in other departments(nongastroenterologists). Gastroenterologists' awareness of FMT prior to the survey was much higher than non-gastroenterologists'(54.3 vs 16.5%, P < 0.001); however, acceptance of FMT was not statistically different(92.4 vs 87.1%, P = 0.1603). Major concerns of FMT included the following: acceptability to patients(79.2%), absence of guidelines(56.9%), and administration and ethics(46.5%). On the basis of understanding, the FMT indications preferred byphysicians were recurrent Clostridium difficile infection(86.7%), inflammatory bowel disease combined with Clostridium difficile infection(78.6%), refractory ulcerative colitis(70.9%), ulcerative colitis(65.4%), Crohn's disease(59.4%), chronic constipation(43.7%), irritable bowel syndrome(39.1%), obesity(28.1%) and type 2 diabetes(23.9%). For donor selection, the majority of physicians preferred individuals with a similar gut flora environment to the recipients. 76.6% of physicians chose lower gastrointestinal tract as the administration approach. 69.2% of physicians considered FMT a safe treatment. CONCLUSION: Chinese physicians have awareness and a high acceptance of FMT, especially gastroenterologists, which provides the grounds and conditions for the development of this novel treatment in China. | 7 | Expression and mutation of c-kit gene in gastrointestinal stromal tumors显示文摘AIM: To investigate the expression and mutation of c-kit gene and its correlation with the clinical pathology and prognosis of gastrointestinal stromal tumors (GISTs).METHODS: A total of 94 cases of GISTs, 10 leiomyomas and 2 schwannomas were studied for the expression of KIT by immunohistochemistry. The c-kit gene mutations in exon 11 of these specimens were detected by PCR-SSCP technique.RESULTS: Of the 94 cases of GISTs, 91 (96.8 %) expressed the KIT protein. Leiomyomas and schwannomas were negative for KIT. The c-kit gene mutations of exon 11 were found in 38 out of the 94 cases of GISTs (40.4 %). The mutations involved point mutations (Va1560-Asp, Ile563-Met),del 557-559 and 579ins12. No mutations were detectable in benign GISTs, leiomyomas or schwannomas. | 8 | Land Use Changes in Northeast China Driven by Human Activities and Climatic Variation显示文摘Human-induced land use/cover change (LUCC) forms an important component of global environmental change. Therefore, it is important to study land use/cover and its change at local, regional and global scales. In this paper we conducted the study of land use change in Northeast China, one of the most important agricultural zones of the nation. From 1986 to 2000, according to the study results obtained from Landsat images, widespread changes in land use/cover took place in the study area. Grassland, marsh, water body and woodland decreased by 9864, 3973, 1367 and 10,052km2, respectively. By comparison, paddy field, dry farmland, and built-up land expanded by 7339, 17193 and 700km2, respectively. Those changes bore an interactive relationship with the environment, especially climate change. On the one hand, climate warming created a potential environment for grassland and marsh to be changed to farmland as more crops could thrive in the warmer climate, and for dry farmland to paddy field. On the other hand, the changed surface cover modified the local climate. Those changes, in turn, have adversely influenced the local environment by accelerating land degradation. | 11 | Fluorescence regional integration and differential fluorescence spectroscopy for analysis of structural characteristics and proton binding properties of fulvic acid sub-fractions显示文摘Structural characteristics and proton binding properties of sub-fractions(FA3–FA13) of fulvic acid(FA), eluted stepwise by pyrophosphate buffer were examined by use of fluorescence titration combined with fluorescence regional integration(FRI) and differential fluorescence spectroscopy(DFS). Humic-like(H-L) and fulvic-like(F-L) materials, which accounted for more than 80% of fluorescence response, were dominant in five sub-fractions of FA. Based on FRI analysis, except the response of F-L materials in FA9 and FA13, maximum changes in percent fluorescence response were less than 10% as pH was increased from 2.5 to 11.5.Contents of carboxylic and phenolic groups were compared for fluorescence peaks of FA sub-fractions based on pH-dependent fluorescence derived from DFS. Static quenching was the dominant mechanism for binding of protons by FA sub-fractions. Dissociation constants(p Ka) were calculated by use of results of DFS and the modified Stern-Volmer relationship. The p Kaof H-L, F-L, tryptophan-like and tyrosine-like materials of FA subfractions exhibited ranges of 3.17–4.06, 3.12–3.97, 4.14–4.45 and 4.25–4.76, respectively, for acidic pHs. At basic pHs, values of p Ka for corresponding materials were in ranges of 9.71–10.24, 9.62–10.99, 9.67–10.31 and 9.33–10.28, respectively. At acidic pH, protein-like(P-L)materials had greater affinities for protons than did either H-L or F-L materials. The dicarboxylic and phenolic groups were likely predominant sites of protonation for both H-L and F-L materials at both acidic and basic pHs. During its prolonged clonal propagation, numerous deleterious mutations have accumulated in the potato genome,leading to severe inbreeding depression; however,the shaping of this mutation burden during polyploidization and improvement is largely unknown.Here, we sequenced 20 diploid landraces of the Stenotomum group, eight tetraploid landraces, and 20 tetraploid modern cultivars, to analyze variations in their deleterious mutations. We show that deleterious mutations accumulated rapidly during the polyploidization of tetraploid potatoes. | 15 | The USDA cucumber (Cucumis sativus L.) collection: genetic diversity, population structure, genome-wide association studies, and core collection development显示文摘Germplasm collections are a crucial resource to conserve natural genetic diversity and provide a source of novel traits essential for sustained crop improvement.Optimal collection,preservation and utilization of these materials depends upon knowledge of the genetic variation present within the collection.Here we use the high-throughput genotyping-by-sequencing(GBS)technology to characterize the United States National Plant Germplasm System(NPGS)collection of cucumber(Cucumis sativus L.).The GBS data,derived from 1234 cucumber accessions,provided more than 23 K high-quality single-nucleotide polymorphisms(SNPs)that are well distributed at high density in the genome(~1 SNP/10.6 kb).The SNP markers were used to characterize genetic diversity,population structure,phylogenetic relationships,linkage disequilibrium,and population differentiation of the NPGS cucumber collection.These results,providing detailed genetic analysis of the U.S.cucumber collection,complement NPGS descriptive information regarding geographic origin and phenotypic characterization.We also identified genome regions significantly associated with 13 horticulturally important traits through genome-wide association studies(GWAS).Finally,we developed a molecularly informed,publicly accessible core collection of 395 accessions that represents at least 96%of the genetic variation present in the NPGS.Collectively,the information obtained from the GBS data enabled deep insight into the diversity present and genetic relationships among accessions within the collection,and will provide a valuable resource for genetic analyses,gene discovery,crop improvement,and germplasm preservation. | Xin Wang Kan Bao Umesh K.Reddy Yang Bai Sue A.Hammar Chen Jiao Todd C.Wehner Axel O.Ramírez-Madera Yiqun Weng Rebecca Grumet Zhangjun Fei | 2018 | Horticulture Research2018,5,1: | 7 |
| 16 | Mn-containing bioceramics inhibit osteoclastogenesis and promote osteoporotic bone regeneration via scavenging ROS显示文摘Osteoporosis is caused by an osteoclast activation mechanism.People suffering from osteoporosis are prone to bone defects.Increasing evidence indicates that scavenging reactive oxygen species(ROS)can inhibit receptor activator of nuclear factorκB ligand(RANKL)-induced osteoclastogenesis and suppress ovariectomy-induced osteoporosis.It is critical to develop biomaterials with antioxidant properties to modulate osteoclast activity for treating osteoporotic bone defects.Previous studies have shown that manganese(Mn)can improve bone regeneration,and Mn supplementation may treat osteoporosis.However,the effect of Mn on osteoclasts and the role of Mn in osteoporotic bone defects remain unclear.In present research,a model bioceramic,Mn-containedβ-tricalcium phosphate(Mn-TCP)was prepared by introducing Mn intoβ-TCP.The introduction of Mn intoβ-TCP significantly improved the scavenging of oxygen radicals and nitrogen radicals,demonstrating that Mn-TCP bioceramics might have antioxidant properties.The in vitro and in vivo findings revealed that Mn^(2+)ions released from Mn-TCP bioceramics could distinctly inhibit the formation and function of osteoclasts,promote the differentiation of osteoblasts,and accelerate bone regeneration under osteoporotic conditions in vivo.Mechanistically,Mn-TCP bioceramics inhibited osteoclastogenesis and promoted the regeneration of osteoporotic bone defects by scavenging ROS via Nrf2 activation.These results suggest that Mn-containing bioceramics with osteoconductivity,ROS scavenging and bone resorption inhibition abilities may be an ideal biomaterial for the treatment of osteoporotic bone defect. | Jianmei Li Cuijun Deng Wanyuan Liang Fei Kang Yun Bai Bing Ma Chengtie Wu Shiwu Dong | 2021 | Bioactive Materials2021,6,11: | 7 |
| 17 | A novel gain of function mutant in C-kit gene and its tumorigenesis in nude mice显示文摘AIM: To transfect mutant C-kit cDNA at codon 579 into human embryonic kidney cell line to observe its role in the pathogenesis of gastrointestinal stromal tumor (GIST).METHODS: Eukaryotic expression vectors of pcDNA3-Kit-NW and pcDNA3-Kit-W were constructed. Then pcDNA3-Kit-NW and pcDNA3-Kit-W plasmids were transfected into human embryonic kidney cell line by Lipofectamine. The resistant done was screened by G418filtration and identified by sequencing, Western blotting,and immunocytochemical staining. Human embryonic kidney cells were divided into three groups including pcDNA3-Kit-NW, pcDNA3-Kit-W, and vector control groups. Absorbency value with a wavelength of 574 nm was detected by MTT analysis. Mice were injected with three groups of cells. Volume, mass, and histological examinations of the tumors in different groups were measured and compared.RESULTS: The C-kit gene and mutant C-kit gene were successfully cloned into the eukaryotic expression vector pcDNA3. pcDNA3-Kit-NW and pcDNA3-Kit-W were successfully transfected into human embryonic kidney cell line and showed stable expression in this cell line.Cell proliferating activity had significant differences between pcDNA3-Kit-NW and pcDNA3, pcDNA3-KitNW and pcDNA3-Kit-W (P<0.05), respectively. | 18 | Low dose bisphenol A impairs spermatogenesis by suppressing reproductive hormone production and promoting germ cell apoptosis in adult rats显示文摘Bisphenol A (BPA), an estrogenic chemical, has been shown to reduce sperm count; however, the underlyingmechanisms remain unknown. Herein, we show that oral administration of BPA (2 μg/kg) for consecutive 14 daysin adult rats (BPA rats) significantly reduced the sperm count and the number of germ cells compared to controls.The serum levels of testosterone and follicle-stimulating hormone (FSH), as well as the level of GnRH mRNA inBPA rats were lower than those of control rats. Testosterone treatment could partially rescue the reduction of germcells in BPA rats. Notably, the number of apoptotic germ cells was significantly increased in BPA rats, which wasinsensitive to testosterone. Furthermore, the levels of Fas, FasL and caspase-3 mRNA in the testicle of BPA ratswere increased in comparison with controls. | 19 | A confinement strategy to in-situ prepare a peanut-like N-doped, C-wrapped TiO_(2) electrode with an enhanced desalination capacity and rate for capacitive deionization显示文摘Capacitive deionization(CDI)technology has been considered a promising desalination technique,especially for brackish water,because of its relatively low energy consumption,facile operation,and easy regeneration of electrodes.However,the desalination capacity,cost,fabrication method,electrochemical stability,and environmental unfriendliness of the electrodes have restricted the practical application of the CDI technique.Herein,we reported the one-step in situ preparation of nitrogen-doped and carbon-decorated MXene-derived TiO_(2)(termed N-TiO_(2−x)/C)through the confinement-growth strategy.The small particle size(∼25 nm)and uniform distribution of a peanut-like N-TiO_(2−x)/C material could be ascribed to the confined growth space created by the nanoporous structure of melamine foam.The defects produced by N doping provide an enhanced electrical conductivity and more adsorption sites,while wrapping with a carbon shell layer increases the conductivity and offers protection for N-TiO_(2−x) to achieve an excellent electrochemical stability.The prepared N-TiO_(2−x)/C electrode is hydrophilic due to the abundant oxygen-containing functional groups(e.g.,C-O,N-Ti-O,-NO_(x),and-OH)and exhibits a high salt removal capacity(33.4 mg·g^(−1)),desalination rate(1.5 mg·g^(−1)·min^(−1)),and remarkable cycling stability(without declining after 100 cycles),which might be ascribed to the synergistic effects of the short ion diffusion path,more active adsorption sites,enhanced conductivity,pseudocapacitive behavior,and protection of the carbon shell layer.This work provides a confined-growth strategy to develop MXene-derived oxide electrodes for electrochemical desalination. | Mingxing Liang Xueting Bai Fei Yu Jie Ma | 2021 | Nano Research2021,14,3: | 5 |
