2Position, Singularity and Workspace Analysis of 3-PSR-O Spatial Parallel Manipulator显示文摘Although the parallel mechanisms have the advantages of high accuracy, velocity, stiffness, and payload capacity, the shortcomings of the space utilization and workspace limit the applications in the confined space. A novel 3 degrees of freedom spatial parallel manipulator 3-PSR-O(prismatic-spherical-revolute) is proposed, which possesses a compact architecture and extended workspace while maintaining the inherent advantages of the parallel mechanisms. The direct-inverse position, singularity and workspace are investigated. The mapping method is adopted in the position analysis, and the closed form solution is derived in the form of a six order equation. The singularity analysis of the mechanism is also carried out based on the geometrical constraints, including six singularity boundaries. A feature boundary, which is independent of the prismatic joints' stroke limit, is obtained by integrating the six singularity boundaries. According to the formation of the reachable workspace, a concept of basic workspace is also introduced and presented in the analytical way. By demarcating the basic workspace along the central height with the feature boundary, the reachable workspace can be derived and analyzed more efficiently. Finally, a comparative study on the space utilization between the 3-PSP parallel mechanism and the new mechanism is also presented. The area of feature boundary of the new mechanism is about 140% of the 3-PSP parallel mechanism, while its installation radius is only 1/2 of the 3-PSP parallel mechanism. 3Rotational Swashplate Pulse Continuously Variable Transmission Based on Helical Gear Axial Meshing Transmission显示文摘The current research on pulse continuously variable transmission(CVT) is mainly focused on reducing the pulse degree and making pulse degrees a constant value. Current research mainly confined to find out new design parameters by using the method of optimization, and reduce the pulse degree of pulse CVT and its range of variation. But the fact is that the reduction of the pulse degree is not significant. This article presents a new structure of mechanical pulse CVT-the rotational swashplate pulse CVT with driven by helical gear axial meshing. This transmission is simple and compact in structure and low in pulsatile rate (it adopts 6 guide rods), and the pulsatile degree is irrelevant to the transmission ratio. Theoretically, pulsatile rate could be reduced to zero if appropriate curved surface of the swashplate is used. Compared with the connecting rod pulse CVT, the present structure uses helical gear mechanism as transmission part and it avoids unbalanced inertial force in the former model. This paper analyzes the principle of driving of this transmission, presents its mechanical structure, and discusses its motion characteristics. Experimental prototype of this type of CVT has been manufactured. Tests for the transmission efficiency(when the rotational speed of the output shaft is the maximum) and the angular velocity of the output shaft have been carried out, and data have been analyzed. The experimental results show that the speed of the output shaft for the experimental prototype is slightly lower than the theoretical value, and the transmission efficiency of the experimental prototype is about 70%. The pulse degree of the CVT discussed in this paper is less than the existing pulse CVT of other types, and it is irrelevant to the transmission ratio of the CVT. 4Additively manufactured biodegradable porous magnesium implants for elimination of implant-related infections:An in vitro and in vivo study显示文摘Magnesium(Mg)alloys that have both antibacterial and osteogenic properties are suitable candidates for orthopedic implants.However,the fabrication of ideal Mg implants suitable for bone repair remains challenging because it requires implants with interconnected pore structures and personalized geometric shapes.In this study,we fabricated a porous 3D-printed Mg-Nd-Zn-Zr(denoted as JDBM)implant with suitable mechanical properties using selective laser melting technology.The 3D-printed JDBM implant exhibited cytocompatibility in MC3T3-E1 and RAW267.4 cells and excellent osteoinductivity in vitro.Furthermore,the implant demonstrated excellent antibacterial ratios of 90.0% and 92.1% for methicillin-resistant S.aureus(MRSA)and Escherichia coli,respectively.The 3D-printed JDBM implant prevented MRSA-induced implant-related infection in a rabbit model and showed good in vivo biocompatibility based on the results of histological evaluation,blood tests,and Mg2+deposition detection.In addition,enhanced inflammatory response and TNF-α secretion were observed at the bone-implant interface of the 3D-printed JDBM implants during the early implantation stage.The high Mg^(2+)environment produced by the degradation of 3D-printed JDBM implants could promote M1 phenotype of macrophages(Tnf,iNOS,Ccl3,Ccl4,Ccl5,Cxcl10,and Cxcl2),and enhance the phagocytic ability of macrophages.The enhanced immunoregulatory effect generated by relatively fast Mg^(2+)release and implant degradation during the early implantation stage is a potential antibacterial mechanism of Mg-based implant.Our findings indicate that 3D-printed porous JDBM implants,having both antibacterial property and osteoinductivity,hold potential for future orthopedic applications.Kai Xie Nanqing Wang Yu Guo Shuang Zhao Jia Tan Lei Wang Guoyuan Li Junxiang Wu Yangzi Yang Wenyu Xu Juan Chen Wenbo Jiang Penghuai Fu Yongqiang Hao 2022Bioactive Materials2022,7,2:8

9Numerical Simulation and Experimental Research on Passive Hydrodynamic Bearing in a Blood Pump显示文摘The current research of hydrodynamic bearing in blood pump mainly focuses on the bearing structure design.Compared with the typical plane slider bearing and Rayleigh step bearing,spiral groove bearing has excellent performance in load-carrying capacity.However,the load-carrying capacity would decrease significantly with increasing flow rate in conventional designs.In this paper,the special treatment is made to the upper spiral groove bearing to make sure that both the circulatory flowing and load-carrying capacity are high.Three-dimensional computational fluid dynamics(CFD) models in the space between rotor and shaft are developed by using FLUENT software.Effects of groove number,film height and groove depth on load-carrying capacity of the spiral groove bearings are investigated by orthogonal experiment design.The experimental results show that film height is the most remarkable factor to the load-carrying capacity.The variation tendency of load-carrying capacity reveals that the best combination of geometry is the one with groove number of 8,film height 0.03 mm and groove depth 0.08 mm.The velocity and pressure distributions in spiral groove bearings are also analyzed,and the analysis result shows that the distributions are in conformity with the design of the blood pump based on the principle of hydrodynamic bearing.The displacement of the rotor with the best combination parameters is tested by using laser displacement sensors,the testing result shows that the suspending performance is satisfactory both in axial and radial directions.This research proposes a bearing design method which has sufficient load-carrying capacity to support rotor as an effective passive hydrodynamic bearing.HAN Qing RUAN Xiaodong CHEN Wenyu FU Xin 2013Chinese Journal of Mechanical Engineering2013,26,5:1

11Alp7-Mtol and Alpl4 synergize to promote interphase microtubule regrowth from the nuclear envelope显示文摘Microtubules grow not only from the centrosome but also from various noncentrosomal microtubule-organizing centers(MTOCs),including the nuclear envelope(NE)and pre-existing microtubules.The evolutionarily conserved proteins Mtol/CDK5RAP2 and Alpl4/TOG/XMAP215 have been shown to be involved in promoting microtubule nucleation.However,it has remained elusive as to how the microtubule nucleation promoting factors are specified to various noncentrosomal MTOCs,particularly the NE,and how these proteins coordinate to organize microtubule assembly.Here,we demonstrate that in the fission yeast Schizosaccharomyces pombe,efficient interphase microtubule growth from the NE requires Alp7/TACC,Alpl4/TOG/XMAP215,and Mtol/CDK5RAP2.The absence of Alp7,A lp l4 t or Mtol compromises microtubule regrowth on the NE in cells undergoing microtubule repolymerization.We further demonstrate that Alp7 and Mtol interdependently localize to the NE in cells without microtubules and that A lp l4 localizes to the NE in an Alp7 and Mtol-dependent manner.Tethering Mtol to the NE in cells lacking Alp7 partially restores microtubule number and the efficiency of microtubule generation from the NE.Hence,our study delineates that Alp7,A lpl4,and Mtol work in concert to regulate interphase microtubule regrowth on the NE.Wenyue Liu Fan Zheng Yucai Wang Chuanhai Fu 2019Journal of Molecular Cell Biology2019,11,11:1
12Applying circulating tumor DNA methylation in the diagnosis of lung cancer显示文摘Lung cancer is the leading cause of cancer-related deaths worldwide.Low dose computed tomography(LDCT)is commonly used for disease screening,with identified candidate cancerous regions further diagnosed using tissue biopsy.However,existing techniques are all invasive and unavoidably cause multiple complications.In contrast,liquid biopsy is a noninvasive,ideal surrogate for tissue biopsy that can identify circulating tumor DNA(ctDNA)containing tumorigenic signatures.It has been successfully implemented to assist treatment decisions and disease outcome prediction.ctDNA methylation,a type of lipid biopsy that profiles critical epigenetic alterations occurring during carcinogenesis,has gained increasing attention.Indeed,aberrant ctDNA methylation occurs at early stages in lung malignancy and therefore can be used as an alternative for the early diagnosis of lung cancer.In this review,we give a brief synopsis of the biological basis and detecting techniques of ctDNA methylation.We then summarize the latest progress in use of ctDNA methylation as a diagnosis biomarker.Lastly,we discuss the major issues that limit application of ctDNA methylation in the clinic,and propose possible solutions to enhance its usage.Lei Li Kai Fu Wenyu Zhou Michael Snyder 2019Precision Clinical Medicine2019,2,1:1

18An effective inducer of dopaminergic neuron-like differentiation显示文摘Rat bone marrow-derived mesenchymal stem cells were cultured and passaged in vitro. After induction with basic fibroblast growth factor for 24 hours, passage 3 bone marrow-derived mesenchymal stem cells were additionally induced into dopaminergic neurons using three different combinations with basic fibroblast growth factor as follows: 20% Xiangdan injection; all-trans retinoic acid + glial-derived neurotrophic factor; or sonic hedgehog + fibroblast growth factor 8. Results suggest that the bone marrow-derived mesenchymal stem cells showed typical neuronal morphological characteristics after induction. In particular, after treatment with sonic hedgehog + fibroblast growth factor 8, the expressions of nestin, neuron-specific enolase, microtubule- associated protein 2, tyrosine hydroxylase and vesicular monoamine transporter-2 in cells were significantly increased. Moreover, the levels of catecholamines in the culture supernatant were significantly increased. These findings indicate that Xiangdan injection, all-trans retinoic acid + glial-derived neurotrophic factor, and sonic hedgehog + fibroblast growth factor 8 can all induce dopaminergic neuronal differentiation from bone marrow-derived mesenchymal stem cells.
