A New Evaluation Algorithm for the Influence of User in Social Network显示文摘Online social networks have gradually permeated into every aspect of people's life.As a research hotspot in social network, user influence is of theoretical and practical significant for information transmission, optimization and integration. A prominent application is a viral marketing campaign which aims to use a small number of targeted infl uence users to initiate cascades of infl uence that create a global increase in product adoption. In this paper, we analyze mainly evaluation methods of user infl uence based on IDM evaluation model, Page Rank evaluation model, use behavior model and some other popular influence evaluation models in currently social network. And then, we extract the core idea of these models to build our influence evaluation model from two aspects, relationship and activity. Finally, the proposed approach was validated on real world datasets, and the result of experiments shows that our method is both effective and stable. | JIANG Wei GAO Mengdi WANG Xiaoxi WU Xianda | 2016 | China Communications2016,13,2: | 5

Protein trafficking in plant cells:tools and markers显示文摘Eukaryotic cells consist of numerous membrane-bound organelles, which compartmentalize cellular materials to fulfil a variety of vital functions. In the post-genomic era,it is widely recognized that identification of the subcellular organelle localization and transport mechanisms of the encoded proteins are necessary for a fundamental understanding of their biological functions and theorganization of cellular activity.Multiple experimental approaches are now available to determine the subcellular localizations and dynamics of proteins. In this review,we provide an overview of the current methods and organelle markers for protein subcellular localization and trafficking studies in plants, with a focus on the organelles of the endomembrane system. We also discuss the limitations of each method in terms of protein colocalization studies. | Dongmei Zhu Mengdi Zhang Caiji Gao Jinbo Shen | 2020 | Science China(Life Sciences)2020,63,3: | 2
Insights into the phenomenon of an explosive growth and sharp decline in haze:A case study in Beijing显示文摘A severe haze episode occurred in winter in the North China Plain(NCP),and the phenomenon of an explosive growth and sharp decline in PM2.5(particulate matter with an aerodynamic diameter equal to or less than 2.5μm)concentration was observed. To study the systematic causes for this phenomenon, comprehensive observations were conducted in Beijing from November 26 to December 2,2015;during this period, meteorological parameters,LIDAR data, and the chemical compositions of aerosols were determined. The haze episode was characterized by rapidly varying PM2.5 concentration, and the highest PM2.5 concentration reached 667μg/m3.During the haze episode, the NCP was dominated by a weak high-pressure system and continuously low PBL(planetary boundary layer)heights, which are unfavorable conditions for the diffusion of pollutants. The large increases in the concentrations of SNA(SO42-,NO3-and NH4+)during the haze implied that the formation of SNA was the largest contribution.Water vapor also played a vital role in the formation of haze by promoting the chemical transformation of secondary pollutants, which led to higher PM2.5 concentrations. The spatial distributions of PM2.5 in Beijing at different times and the backward trajectories of the air masses also indicated that pollutants from surrounding provinces in particular, contributed to the higher PM2.5concentration. | Wenguang Li Xingang Liu Yuanhang Zhang Qinwen Tan Miao Feng Mengdi Song Lirong Hui Yu Qu Junling An Haokai Gao | 2019 | Journal of Environmental Sciences2019,31,10: | 1
Non-canonical STING-PERK pathway dependent epigenetic regulation of vascular endothelial dysfunction via integrating IRF3 and NF-κB in inflammatory response显示文摘Inflammation-driven endothelial dysfunction is the major initiating factor in atherosclerosis, while the underlying mechanism remains elusive. Here,we report that the non-canonical stimulator of interferon genes(STING)-PKR-like ER kinase(PERK)pathway was significantly activated in both human and mice atherosclerotic arteries.Typically,STING activation leads to the activation of interferon regulatory factor 3(IRF3)and nuclear factor-kappa B(NF-κB)/p65,thereby facilitating IFN signals and infammation. In contrast, our study reveals the activated non-canonical STING-PERK pathway increases scaffold protein bromodomain protein 4(BRD4)expression,which encourages the formation of super-enhancers on the proximal promoter regions of the proinflammatory cytokines, thereby enabling the transactivation of these cytokines by integrating activated IRF3 and NF-κB via a condensation process.Endothelium-specific STING and BRD4 deficiency significantly decreased the plaque area and inflammation.Mechanistically, this pathway is triggered by leaked mitochondrial DNA(mtDNA)via mitochondrial permeability transition pore(mPTP),formed by voltage-dependent anion channel 1(VDAC1)oligomer interaction with oxidized mtDNA upon cholesterol oxidation stimulation.Especially, compared to macrophages, endothelial STING activation plays a more pronounced role in atherosclerosis. We propose a non-canonical STING-PERK pathway-dependent epigenetic paradigm in atherosclerosis that integrates IRF3,NF-κB and BRD4 in inflammatory responses, which provides emerging therapeutic modalities for vascular endothelial dysfunction. | Xuesong Li Xiang Chen Longbin Zheng Minghong Chen Yunjia Zhang Ruigong Zhu Jiajing Chen Jiaming Gu Quanwen Yin Hong Jiang Xuan Wu Xian Ji Xin Tang Mengdie Dong Qingguo Li Yuanqing Gao Hongshan Chen | 2023 | Acta Pharmaceutica Sinica B2023,13,12: | 1
Rational construction of metal–base synergetic sites on Au/Mg-beta catalyst for selective aerobic oxidation of 5-hydroxymethylfurfural显示文摘The selective aerobic oxidation of biomass-derived 5-hydroxymethylfurfural to 2,5-furandicarboxylic acid (FDCA, a potential renewable substitution of fossil-based terephthalic acid to produce polyethylene 2,5-furandicarboxylate plastic) is an appealing transformation for constructing eco-friendly and sustainable chemical processes. Au supported catalysts have showed encouraging performances for this well-received conversion, whose catalytic behavior was greatly affected by the adopted support derived from the existence of metal-support interactions. Herein, a series of Mg-Beta zeolites were hydrothermally synthesized via developed structural reconstruction, which were employed as basic supports for Au catalysts to construct bifunctional catalysts. The relationship between structure (Au particle size, basicity within zeolites and Auδ+ contents) and FDCA yield was concretely established. The conclusion was made that the utilization of Mg-Beta zeolites with strong basicity as the support could not only improve the FDCA yield but also decrease the amount of additional base. Furthermore, the possible reaction mechanism was also proposed via tracking time-dependent variations of corresponding organics and controlled experiment. This work provides some guidance for rationally designing multifunctional catalysts in the view of integrating metal catalysts with metallosilicate zeolites, which was beneficial to the catalytic upgrading of organic compounds with multiple functional groups. | Zhiguo Zhu Xiongjie Gao Xiuming Wang Mengdie Yin Qingyao Wang Wanzhong Ren Bo Wang Hongying Lü Weiping Liao | 2021 | Journal of Energy Chemistry2021,30,11: | 1
Spatiotemporal dynamics of ecosystem fires and biomass burning-induced carbon emissions in China over the past two decades显示文摘Fire is a major type of disturbance that has important influences on ecosystem dynamics and carbon cycles.Yet our understanding of ecosystem fires and their carbon cycle consequences is still limited, largely due to the difficulty of large-scale fire monitoring and the complex interactions between fire, vegetation, climate, and anthropogenic factors. Here, using data from satellite-derived fire observations and ecosystem model simulations,we performed a comprehensive investigation of the spatial and temporal dynamics of China’s ecosystem fire disturbances and their carbon emissions over the past two decades(1997–2016).Satellite-derived results showed that on average about 3.47-4.53×10^(4) km^(2) of the land was burned annually during the past two decades, among which annual burned forest area was about 0.81-1.25×10^(4) km^(2),accounting for 0.33-0.51%of the forest area in China.Biomass burning emitted about 23.02 TgC per year.Compared to satellite products, simulations from the Energy Exascale Earth System Land Model(ELM)strongly overestimated China’s burned area and fire-induced carbon emissions.Annual burned area and fire-induced carbon emissions were high for boreal forest in Northeast China’s Daxing’anling region and subtropical dry forest in South Yunnan,as revealed by both the satellite product and the model simulations. Our results suggest that climate and anthropogenic factors play critical roles in controlling the spatial and seasonal distribution of China’s ecosystem fire disturbances. Our findings highlight the importance of multiple complementary approaches in assessing ecosystem fire disturbance and its carbon consequences.Further studies are required to improve the methods of observing and modelling China’s ecosystem fire disturbances, which will provide valuable information for fire management and ecosystem sustainability in an era when both human activities and the natural environment are rapidly changing. | Anping Chen Rongyun Tang Jiafu Mao Chao Yue Xiran Li Mengdi Gao Xiaoying Shi Mingzhou Jin Daniel Ricciuto Sam Rabin Phillippe Ciais Shilong Piao | 2020 | Geography and Sustainability2020,1,1: | 1
Role of Elevated Thrombospondin-1 in Kainic Acid-Induced Status Epilepticus显示文摘Previous studies have suggested that thrombospondin-1(TSP-1) regulates the transforming growth factor beta 1(TGF-b1)/phosphorylated Smad2/3(p Smad2/3) pathway. Moreover, TSP-1 is closely associated with epilepsy. However, the role of the TSP-1-regulated TGFb1/p Smad2/3 pathway in seizures remains unclear. In this study, changes in this pathway were assessed following kainic acid(KA)-induced status epilepticus(SE) in rats. The results showed that increases in the TSP-1/TGF-b1/p Smad2/3 levels spatially and temporally matched the increases in glial fibrillary acidic protein(GFAP)/chondroitin sulfate(CS56) levels following KA administration.Inhibition of TSP-1 expression by small interfering RNA or inhibition of TGF-b1 activation with a Leu-Ser-Lys-Leu peptide significantly reduced the severity of KA-induced acute seizures. These anti-seizure effects were accompanied by decreased GFAP/CS56 expression and Smad2/3 phosphorylation. Moreover, inhibiting Smad2/3 phosphorylation with ponatinib or SIS3 also significantly reduced seizure severity, alongside reducing GFAP/CS56 immunoreactivity. These results suggest that the TSP-1-regulated TGF-b1/p Smad2/3 pathway plays a key role in KA-induced SE and astrogliosis, and that inhibiting this pathway may be a potential anti-seizure strategy. | Yurong Zhang Mengdi Zhang Wei Zhu Xiaohong Pan Qiaoyun Wang Xue Gao Chaoyun Wang Xiuli Zhang Yuxia Liu Shucui Li Hongliu Sun | 2020 | Neuroscience Bulletin2020,36,3: | 1
Molecular mechanisms for stemness maintenance of acute myeloid leukemia stem cells显示文摘Human acute myeloid leukemia(AML)is a fatal hematologic malignancy characterized with accumulation of myeloid blasts and differentiation arrest. The development of AML is associated with a serial of genetic and epigenetic alterations mainly occurred in hematopoietic stem and progenitor cells(HSPCs),which change HSPC state at the molecular and cellular levels and transform them into leukemia stem cells(LSCs).LSCs play critical roles in leukemia initiation, progression, and relapse, and need to be eradicated to achieve a cure in clinic.Key to successfully targeting LSCs is to fully understand the unique cellular and molecular mechanisms for maintaining their stemness. Here,we discuss LSCs in AML with a focus on identification of unique biological features of these stem cells to decipher the molecular mechanisms of LSC maintenance. | Jiazhen Wang Peipei Wang Tiantian Zhang Zhuying Gao Jing Wang Mengdie Feng Rong Yin Haojian Zhang | 2019 | Blood Science2019,1,1: | 0
Effects of Spatial Density of Farmland Shelterbelts on NDVI on the Northern Slope of Tianshan Mountains显示文摘[Objectives]To explore the effects of spatial density of farmland shelterbelts on NDVI on the northern slope of Tianshan Mountains.[Methods]Taking the economic belt of the northern slope of the Tianshan Mountains as the research area and using the grid method, the spatial density distribution of farmland shelterbelts can be known.Combining the grid method with NDVI data, the average value of normalized difference vegetation index(NDVI)during the growing period of crops can be obtained. In addition, the protection benefits of the shelterbelts on crops were analyzed through comparing the growth of crops within the protection zone with and without protection.[Results]The grids with shelterbelt distribution were better than the grids without shelterbelt distribution, and the shelterbelt played a great role in promoting crop growth. In the middle stage of crop growth, the protection benefit of shelterbelt was significant. The spatial density of shelterbelts was unevenly distributed in the range of 0.6 to 0.8,and the protection benefits were poor.[Conclusions]It is expected to explore the effects of shelterbelts on crop growth at a larger regional scale,so as to provide a basis for the management and design of shelterbelts in the future, and to provide a theoretical basis for studying the protective effect of farmland shelterbelts on crops. | Li CHEN Jie GAO Mengdi CHANG Xuan REN | 2022 | Asian Agricultural Research2022,14,10: | 0
SYK-mediated epithelial cell state is associated with response to c-Met inhibitors in c-Met-overexpressing lung cancer显示文摘Genomic MET amplification and exon 14 skipping are currently clinically recognized biomarkers for stratifying subsets of non-small cell lung cancer(NSCLC)patients according to the predicted response to c-Met inhibitors(c-Metis),yet the overall clinical benefit of this strategy is quite limited.Notably,c-Met protein overexpression, which occurs in approximately 20–25%of NSCLC patients, has not yet been clearly defined as a clinically useful biomarker. An optimized strategy for accurately classifying patients with c-Met overexpression for decision-making regarding c-Meti treatment is lacking.Herein,we found that SYK regulates the plasticity of cells in an epithelial state and is associated with their sensitivity to c-Metis both in vitro and in vivo in PDX models with c-Met overexpression regardless of MET gene status.Furthermore,TGF-β1 treatment resulted in SYK transcriptional downregulation, increased Sp1-mediated transcription of FRA1,and restored the mesenchymal state, which conferred resistance to c-Metis.Clinically,a subpopulation of NSCLC patients with c-Met overexpression coupled with SYK overexpression exhibited a high response rate of 73.3%and longer progression-free survival with c-Meti treatment than other patients.SYK negativity coupled with TGF-β1 positivity conferred de novo and acquired resistance. In summary,SYK regulates cell plasticity toward a therapy-sensitive epithelial cell state.Furthermore, our findings showed that SYK overexpression can aid in precisely stratifying NSCLC patients with c-Met overexpression regardless of MET alterations and expand the population predicted to benefit from c-Met-targeted therapy. | Ji Zhou Xu-Chao Zhang Shan Xue Mengdi Dai Yueliang Wang Xia Peng Jianjiao Chen Xinyi Wang Yanyan Shen Hui Qin Bi Chen Yu Zheng Xiwen Gao Zuoquan Xie Jian Ding Handong Jiang Yi-Long Wu Meiyu Geng Jing Ai | 2023 | Signal Transduction and Targeted Therapy2023,8,6: | 0
Cellbow:a robust customizable cell segmentation program显示文摘Backgrounds Time-lapse live cell imaging of a growing cell population is routine in many biological investigations.A major challenge in imaging analysis is accurate segmentation,a process to define the boundaries of cells based on raw image data.Current segmentation methods relying on single boundary features have problems in robustness when dealing with inhomogeneous foci which invariably happens in cell population imaging. Methods:Combined with a multi-layer training set strategy,we developed a neural-network-based algorithm—Cellbow. Results'Cellbow can achieve accurate and robust segmentation of cells in broad and general settings. It can also facilitate long-term tracking of cell growth and division. To facilitate the application of Cellbow,we provide a website on which one can online test the software,as well as an I mage J plugin for the user to visualize the performance before software installation. Conclusions Cellbow is customizable and generalizable. It is broadly applicable to segmenting fluorescent images of diverse cell types with no further training needed. For bright-field images, only a small set of sample images of the specific cell type from the user may be needed for training. | Huixia Ren Mengdi Zhao Bo Liu Ruixiao Yao Qi liu Zhipeng Ren Zirui Wu Zongmao Gao Xiaojing Yang Chao Tang | 2020 | Quantitative Biology2020,8,3: | 0
JARID2 coordinates with the NuRD complex to facilitate breast tumorigenesis through response to adipocyte-derived leptin显示文摘Background Proteins containing the Jumonji C(JmjC)domain participated in tumorigenesis and cancer progression. However, the mechanisms underlying this effect are still poorly understood. Our objective was to investigate the role of Jumonji and the AT-rich interaction domain-containing 2(JARID2)—a JmjC family protein—in breast cancer,as well as its latent association with obesity. Methods Immunohistochemistry,The Cancer Genome Atlas,Gene Expression Omnibus, and other databases were used to analyze the expression of JARID2 in breast cancer cells.Growth curve,5-ethynyl-2-deoxyuridine(EdU),colony formation, and cell invasion experiments were used to detect whether JARID2 affected breast cancer cell proliferation and invasion.Spheroidization-based experiments and xenotumor transplantation in NOD/SCID mice were used to examine the association between JARID2 and breast cancer stemness.RNA-sequencing,Kyoto Encyclopedia of Genes and Genomes, and Gene Set Enrichment Analysis were used to identify the cell processes in which JARID2 participates.Immunoaffinity purification and silver staining mass spectrometry were conducted to search for proteins that might interact with JARID2.The results were further verified using co-immunoprecipitation and glutathione S-transferase(GST)pull-down experiments.Using chromatin immunoprecipitation(ChIP)sequencing,we sought the target genes that JARID2 and metastasis-associated protein 1(MTA1)jointly regulated;the results were validated by ChIP-PCR,quantitative ChIP(qChIP)and ChIP-reChIP assays.A coculture experiment was used to explore the interactions between breast cancer cells and adipocytes. Results In this study,we found that JARID2 was highly expressed in multiple types of cancer including breast cancer.JARID2 promoted glycolysis, lipid metabolism, proliferation, invasion, and stemness of breast cancer cells.Furthermore,JARID2 physically interacted with the nucleosome remodeling and deacetylase(NuRD)complex, transcriptionally repressing a series of tumor suppressor genes such as BRCA2 DNA repair associated(BRCA2),RB transcriptional corepressor 1(RB1),and inositol polyphosphate-4-phosphatase type II B(INPP4B).Additionally,JARID2 expression was regulated by the obesity-associated adipokine leptin via Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3)pathway in the breast cancer microenvironment.Analysis of various online databases also indicated that JARID2/MTA1 was associated with a poor prognosis of breast cancer. Conclusion Our data indicated that JARID2 promoted breast tumorigenesis and development, confirming JARID2 as a target for cancer treatment. | Wei Liu Yi Zeng Xinhui Hao Xin Wang Jiaxiang Liu Tianyang Gao Mengdi Wang Jingyao Zhang Miaomiao Huo Ting Hu Tianyu Ma Die Zhang Xu Teng Hefen Yu Min Zhang Baowen Yuan Wei Huang Yunkai Yang Yan Wang | 2023 | Cancer Communications2023,43,10: | 0
Interrogating the impact of aggregation-induced emission nanoparticles on in vitro protein stability,ex vivo protein homeostasis, and in vivo biocompatibility显示文摘Aggregation-induced emission(AIE)materials offer promising perspectives in disease diagnosis and therapeutics given their unique optical and photochemical properties.A key step toward translational applications for AIE materials is to systematically and vigorously evaluate their biosafety and biocompatibility. While previous studies focus on cellular viability and toxicity, the impact of AIE materials on detailed stress responses manifesting cellular fitness has been less explored.Herein, this work provides the first piece of evidence to support amphiphilic functionalization of AIE nanoparticles minimizes the deterioration on proteome stability and cellular protein homeostasis(proteostasis). To this end, four scaffolds of AIE molecules were prepared, further functionalized into eight nanoparticles with two amphiphilic shells respectively, and characterized for their physicochemical properties.Thermal shift assay quantitatively demonstrates that AIE materials after amphiphilic functionalization into nanoparticles enhance proteome thermodynamic stability and ameliorate proteome aggregation propensity in cellular lysate, echoed by cell viability and fractionation experiments.Intriguingly, poor polydispersity index(PDI)of functionalized nanoparticles exaggerates their retention and aggregation in the cell.Comparative proteomic analysis uncovers that amphiphilic functionalization of AIE materials can minimize the deterioration of cellular protein homeostasis network. Finally, vigorous interrogation of functionalized AIE nanoparticles in mice model reveals the complexity of factors affecting the biocompatibility profiles in vivo, including materials’size,PDI,and treatment frequencies.Overall, amphiphilic functionalization of AIE materials into nanoparticles is necessary to maintain proteome stability and balance cellular protein homeostasis. | Wang Wan Qun Zhao Biao Jing Congcong Peng Mengdie Wang Yanan Huang Wenhan Jin Bowen Zhong Zhenduo Zhang Xuepeng Dong Zhenming Gao Lihua Zhang Yu Liu | 2022 | Aggregate2022,3,6: | 0
