| 1 | Association of polymorphism of tumor necrosis factor-alpha gene promoter region with outcome of hepatitis B virus infection显示文摘AIM: To determine whether -238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha (TNF-α), gene promoter and hepatitis B (HB) viral genotypes were associated with outcomes of HBV infection.METHODS: A total of 244 HBV self-limited infected subjects, 208 asymptomatic carriers, and 443 chronic HB patients were recruited to conduct a case-control study.TNF-α -238G/A and -857C/T gene promoter polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and HBV genotypes were examined by nested PCR.RESULTS: The positive rate of HBV DNA in asymptomatic carrier group and chronic HB group was 46.6% and 49.9%,respectively. HBV genotype proportion among the asymptomatic carriers was 2.1% for genotype A, 25.8% for genotype B, 68.0% for genotype C, and 4.1% for genotype B+C mixed infection, and 0.9% for genotype A,21.7% for genotype B, 71.5% for genotype C, 5.9% for genotype B+C mixed infection in chronic HB group. There was no significant difference in genotype distribution between the asymptomatic carrier group and chronic HB group (x2 = 1.66, P = 0.647). The frequency of -238GG genotype in self-limited group was 95.1%, significantly higher than 90.7% in chronic HB group and 89.0% in asymptomatic carrier group (P = 0.041 and P = 0.016,respectively).The frequency of TNF-α-857 CC in chronic HB group was 79.7%, significantly higher than 64.4% in asymptomatic carrier group and 70.9% in self-limited group (P<0.001 and P = 0.023, respectively). | 2 | WAVE-CURRENT INTERACTIONS WITH THREE-DIMENSIONAL FLOATING BODIES显示文摘A Time-domain Higher-Order Boundary Element Method(THOBEM) is developed for simulating wave-current interactions with 3-D floating bodies.Through a Taylor series expansion and a perturbation procedure,the model is formulated to the first-order in the wave steepness and in the current velocity,respectively.The boundary value problem is decomposed into a steady double-body flow problem and an unsteady wave problem.Higher-order boundary integral equation methods are then used to solve the proposed problems with a fourth-order Runge-Kutta method for the time marching.An artificial damping layer is adopted to dissipate the scattering waves.Different from the other time-domain numerical models,which are often focused on the wave-current interaction with restrained bodies,the present model deals with a floating hemisphere.The numerical results of wave forces,wave run-up and body response are all in a close agreement with those obtained by frequency-domain methods.The proposed numerical model is further applied to investigate wave-current interactions with a floating body of complicated geometry.In this work,the regular and focused wave combined with current interacting with a truss-spar platform is investigated. | LIU Zhen TENG Bin NING De-zhi GOU Ying | 2010 | Journal of Hydrodynamics2010,22,2: | 14 |

| 4 | SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19. | Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan | 2021 | Cell Research2021,31,12: | 6 |
| 5 | Draft effect on wave action with a semi-infinite elastic plate显示文摘A method for analyzing reflection and transmission of ocean waves from a semi-infinite elastic plate with a draft is developed. The relation of energy conservation for plates with three different edge conditions (free, simply supported and built-in) is also derived. It is found that the present method satisfies the energy relation very well. The effects of draft on wave reflection and transmission coefficients as well as on the vertical vibration of the plates are examined through numerical tests. | 6 | Multi-responsive nanotheranostics with enhanced tumor penetration and oxygen self-producing capacities for multimodal synergistic cancer therapy显示文摘Incorporation of multiple functions into one nanoplatform can improve cancer diagnostic efficacy and enhance anti-cancer outcomes. Here, we constructed doxorubicin(DOX)-loaded silk fibroinbased nanoparticles(NPs) with surface functionalization by photosensitizer(N770). The obtained nanotheranostics(N770-DOX@NPs) had desirable particle size(157 nm) and negative surface charge(-25 m V). These NPs presented excellent oxygen-generating capacity and responded to a quadruple of stimuli(acidic solution, reactive oxygen species, glutathione, and hyperthermia). Surface functionalization of DOX@NPs with N770 could endow them with active internalization by cancerous cell lines, but not by normal cells. Furthermore, the intracellular NPs were found to be preferentially retained in mitochondria, which were also efficient for near-infrared(NIR) fluorescence imaging, photothermal imaging,and photoacoustic imaging. Meanwhile, DOX could spontaneously accumulate in the nucleus. | 12 | Integrative Analyses of Lung Squamous Cell Carcinoma in Ten Chinese Patients with Transcriptome Sequencing显示文摘Few effective therapies have been developed for the treatment of lung squamous cell carcinoma(SQCC), in part due to a lack of understanding regarding the mechanisms underlying the initiation and development of this disease. Whole transcriptome sequencing not only provides insight into the expression of all transcribed genes, but offers an efficient approach for identifying genetic variations,including gene fusions, mutations and alternative splicing. In this study, we performed whole transcriptome sequencing of 10 patients with stage IIIA lung SQCC, and discovered a large number of single nucleotide variants(SNVs; mean of 12.2 SNVs/Mb), with C>T/G>A and A>G/T>C transitions being the most frequently observed. Additionally, a total of 132 gene fusions were identified based upon Top Hat alignments, 70.5%(93/132) of which occurred as a result of intra-chromosomal rearrangements. Based on the number of supporting reads for each fusion, we further validated 20 of the 26 top gene fusions by RT-PCR and Sanger sequencing. | 16 | Effect of pontoons on free surface elevation around a TLP platform显示文摘Tension leg platform(TLP)is a typical compliant offshore structure for oil/gas resources exploitation.In the design process,the prediction of the free surface elevation is of great importance for the determination of the air gap.So far,the existing researches for predicting the air gap of the TLPs focus on the supporting columns while pay little attention to the horizontal pontoons.For the second order diffraction problem or long incident wave condition,the velocity potential decays slowly with water depth and the effect of pontoons should not be neglected.Herein the effect of pontoons on the diffracted wave field in the vicinity of a TLP platform is investigated in this study.The diffraction of regular waves by a square array of truncated cylinders and a whole TLP structure is studied in detail by using both the linear and the second-order diffraction theory.Numerical calculation is performed for the free surface elevation and wave run up.Numerical results show that the near-trapping phenomenon can occur inside the TLP and leads to significantly increased wave height.To study the effect of pontoons on the free surface elevation comparisons are carried out between the results of these two structures with and without pontoons.It is found that pontoons have an appreciable effect on the diffracted wave field for long incident regular waves and increase the largest response notably when the near-trapping phenomenon occurs at the second-order. | CONG PeiWen GOU Ying TENG Bin | 2014 | Science China(Technological Sciences)2014,57,1: | 1 |

| 19 | Study of Thermal Treatment on Properties of Polycrystalline Silicon显示文摘The effects of thermal annealing on oxygen behavior and carrier lifetimes of mc-Si wafers were investigated by means of FTIR and QSSPCD during single step and two step heat treatments in N2 and O2 ambient. It reveals that interstitial oxygen concentration of mc-Si and CZ-Si has a slighter decrease in N2 and O2 ambient during single-step annealing, which means oxygen precipitates are not generated. But oxygen concentration greatly decreases and generates a number of oxygen precipitates during two-step annealing. Bulk lifetime of mc-Si increases in N2 ambient at 850, 950, 1150 ℃ respectively, and annealing in O2 shows better results than that in N2 and annealing in two-step reflected better consequence than annealing in single-step. But lifetime of CZ-Si annealed in N2 or O2 decreases rapidly. The reason of lifetime increase is probably considered due to the fact that interstitial Si atoms of Si/SiO2 interface fill vacancies or some recombination centers at high temperature annealing.
