| 2 | A wheat integrative regulatory network from large-scale complementary functional datasets enables trait-associated gene discovery for crop improvement显示文摘Gene regulation is central to all aspects of organism growth,and understanding it using large-scale functional datasets can provide a whole view of biological processes controlling complex phenotypic traits in crops.However,the connection between massive functional datasets and trait-associated gene discovery for crop improvement is still lacking.In this study,we constructed a wheat integrative gene regulatory network(wGRN)by combining an updated genome annotation and diverse complementary functional datasets,including gene expression,sequence motif,transcription factor(TF)binding,chromatin accessibility,and evolutionarily conserved regulation.wGRN contains 7.2 million genome-wide interactions covering 5947 TFs and 127439 target genes,which were further verified using known regulatory relationships,condition-specific expression,gene functional information,and experiments.We used wGRN to assign genome-wide genes to 3891 specific biological pathways and accurately prioritize candidate genes associated with complex phenotypic traits in genome-wide association studies.In addition,wGRN was used to enhance the interpretation of a spike temporal transcriptome dataset to construct high-resolution networks.We further unveiled novel regulators that enhance the power of spike phenotypic trait prediction using machine learning and contribute to the spike phenotypic differences among modern wheat accessions.Finally,we developed an interactive webserver,wGRN(http://gffzzaeec8df70e7c4b21hnn0o9wwwfqk666un.ffgz.tsg.suse.edu.cn/wGRN),for the community to explore gene regulation and discover trait-associated genes.Collectively,this community resource establishes the foundation for using large-scale functional datasets to guide trait-associated gene discovery for crop improvement. | Yongming Chen Yiwen Guo Panfeng Guan Yongfa Wang Xiaobo Wang Zihao Wang Zhen Qin Shengwei Ma Mingming Xin Zhaorong Hu Yingyin Yao Zhongfu Ni Qixin Sun Weilong Guo Huiru Peng | 2023 | Molecular Plant2023,16,2: | 1 |

| 5 | The SACT Template:A Human Brain Diffusion Tensor Template for School-age Children显示文摘School-age children are in a specific development stage corresponding to juvenility,when the white matter of the brain experiences ongoing maturation.Dffusion-weighted magnetic resonance imaging(DWI),especially diffusion tensor imaging(DTI),is extensively used to characterize the maturation by assessing white matter properties in vivo.In the analysis of DWI data,spatial normalization is crucial for conducting inter-subject analyses or linking the individual space with the reference space.Using tensor-based registration with an appropriate diffusion tensor template presents high accuracy regarding spatial normalization.However,there is a lack of a standardized diffusion tensor template dedicated to school-age children with ongoing brain development.Here,we established the school-age children diffusion tensor(SACT)template by optimizing tensor reorientation on high-quality DTI data from a large sample of cognitively normal participants aged 6-12 years.With an age-balanced design,the SACT template represented the entire age range well by showing high similarity to the age-specific templates.Compared with the tensor template of adults,the SACT template revealed significantly higher spatial normalization accuracy and inter-subject coherence upon evaluation of subjects in two different datasets of schoolage children.A practical application regarding the age associations with the normalized DTI-derived data was conducted to further compare the SACT template and the adult template.Although similar spatial patterns were found,the SACT template showed significant effects on the distributions of the statistical results,which may be related to the performance of spatial normalization.Looking forward,the SACT template could contribute to future studies of white matter development in both healthy and clinical populations.The SACT template is publicly available now(tp://igshare com/aricles/dataseu'SACT_.template/14071283). | Congying Chu Haoran Guan Sangma Xie Yanpei Wang Jie Luo Gai Zhao Zhiying Pan Mingming Hu Weiwei Men Shuping Tan Jia-Hong Gao Shaozheng Qin Yong He Lingzhong Fan Qi Dong Sha Tao | 2022 | Neuroscience Bulletin2022,38,6: | 1 |

| 7 | TMEM16A inhibits angiotensin Ⅱ-induced basilar artery smooth muscle cell migration in a WNK1-dependent manner显示文摘Vascular smooth muscle cell(VSMC) migration plays a critical role in the pathogenesis of many cardiovascular diseases.We recently showed that TMEM16 A is involved in hypertensioninduced cerebrovascular remodeling.However,it is unclear whether this effect is related to the regulation of VSMC migration.Here,we investigated whether and how TMEM16 A contributes to migration in basilar artery smooth muscle cells(BASMCs).We observed that AngⅡ increased the migration of cultured BASMCs,which was markedly inhibited by overexpression of TMEM16 A.TMEM16 A overexpression inhibited AngⅡ-induced RhoA/ROCK2 activation,and myosin light chain phosphatase(MLCP)and myosin light chain(MLC20) phosphorylation.But AnsⅡ-induced myosin light chain kinase(MLCK)activation was not affected by TMEM16 A.Furthermore,a suppressed activation of integrinβ3/FAK pathway,determined by reduced integrinβ3 expression,FAK phosphorylation and F-actin rearrangement,was observed in TMEM16 A-overexpressing BASMCs upon AngⅡ stimulation.Contrary to the results of TMEM16 A overexpression,silencing of TMEM16 A showed the opposite effects.These in vitro results were further demonstrated in vivo in basilar arteries from VSMC-specific TMEM10 A transgenic mice during AngⅡ-induced hypertension.Moreover,we observed that the inhibitory effect of TMEM16 A on BASMC migration was mediated by decreasing the activation of WNK1,a Cl^(-)-sensitive serine/threonine kinase.In conclusion,this study demonstrated that TMEM16 A suppressed AnsⅡ-induced BASMC migration,thus contributing to the protection against cerebrovascular remodeling during AngⅡ-infused hypertension.TMEM16 A may exert this effect by suppressing the RhoA/ROCK2/MLCP/MLC20 and integrinβ3/FAK signaling pathways via inhibiting WNK1.Our results suggest that TMEM16 A may serve as a novel therapeutic target for VSMC migration-related diseases,such as vascular remodeling. | Huaqing Zheng Xiaolong Li Xin Zeng Chengcui Huang Mingming Ma Xiaofei Lv Yajuan Zhang Lu Sun Guanlei Wang Yanhua Du Yongyuan Guan | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 1 |

| 13 | Melatonin restores normal Bax and Bcl-2 protein expression in the subgranular zone of the dentate gyrus in pinealectomized rats显示文摘In this study, we sought to elucidate the effects of melatonin on learning and memory as well as apoptosis and expression of the Bax or Bcl-2 proteins in the subgranular zone of the dentate gyrus in pinealectomized rats. Using the Morris water maze and the olfactory memory tests, we found that the average escape latency in pinealectomized rats was clearly increased compared with sham-operated rats. Moreover, the average escape latency in the melatonin-treated and pinealectomized rats was longer than that in the sham-operated rats and shorter than that in the pinealectomized and untreated rats. Immunohistochemistry and terminal-deoxynucleoitidyl transferase mediated nick end labeling (TUNEL) showed that there were fewer Bax immunoreactive cells and TUNEL-positive (apoptotic) cells but more Bcl-2 immunoreactive cells in the melatonin-treated rats compared with the pinealectomized rats. The sham-operated rats showed numbers of these cells similar to the melatonin-treated rats. These experimental findings demonstrate that melatonin treatment may reduce abnormal apoptosis by promoting gene expression of Bax and suppressing gene expression of Bcl-2 in the subgranular zone of the dentate gyrus in pinealectomized rats. A comparison of the distributions of carbazoles and dibenzothiophenes has revealed the main results as follows: variations in the relative amounts ①of C0-, C1-, C2-dibenzothiophenes show a low correlation with that of the corresponding carbazoles, with the former's being much higher than the latter's; variations in the relative amounts of methyldibe② nzothiophene isomers also display a low correlation with that of the corresponding methylcarbazoles, with 4-/4- +1-methyldibenzothiophene ratio ranging from 0.52 to 0.96, while the corresponding carbazole ratio of 1-/1- +4-methylcarbazole only being 0.71±0.05; the mat③ urity parameter for 4,6-/4,6- +1,4-dimethyldibenzothiohene, ranging from 0.34 to 0.75, shows a remarkable linear correlation with the corresponding ratio of 1,8-/1,8- +1,4-dimethylcarbazole(R2>0.84). The un-correlation may indicate some different geological-geochemical fates for some isomers of dibenzothiophenes and carbazoles.
