| 1 | Recombinant rabies virus expressing IL-15 enhances immunogenicity through promoting the activation of dendritic cells in mice显示文摘Rabies remains a public health threat that kills approximately 59,000 people worldwide each year,most of which are from the developing countries of Africa and Asia where dog rabies are endemic.Therefore, developing an affordable and efficacious vaccine is crucial for rabies control in these countries. Interleukin(IL)-15, an immunoregulatory cytokine, is a pluripotent molecule with therapeutic potential, which targets many cell types and links the innate and adaptive immune system. In this study, IL-15 gene was cloned and inserted into the genome of a recombinant rabies virus(RABV) strain LBNSE(designated as LBNSE-IL15), and the effect of over-expression of IL-15 on the immunogenicity of RABV was investigated. It was found that mice vaccinated with LBNSEIL15 could induce significantly higher level of virus-neutralizing antibody(VNA) than those immunized with LBNSE, resulting in the higher protection after challenge. Further investigation was performed to find out the possible role of IL-15 plays in the process of antibody induction, and it was found that LBNSE-IL15 could enhance the maturation of dendritic cells(DCs) in immunized mice. Furthermore, the mice immunized with LBNSE-IL15 could promote the T_(FH) cells differentiation and the generation of germinal center B cells and plasma cells. | 3 | Production sharing contract: An analysis based on an oil price stochastic process显示文摘Assuming that oil price follows the stochastic processes of Geometric Brownian Motion (GBM) or the Mean-Reverting Process (MRP), this paper takes the net present value (NPV) as an economic index and models the PSC in 11 different scenarios by changing the value of each contract element (i.e. royalty, cost oil, profit oil as well as income tax). Then the NPVs are shown in probability density graphs to investigate the effect of different elements on contract economics. The results show that under oil price uncertainty the influence of profit oil and income tax on NPV are more significant than those of royalty and cost oil, while a tax holiday could improve the contractor's financial status remarkably. | 5 | A wheat integrative regulatory network from large-scale complementary functional datasets enables trait-associated gene discovery for crop improvement显示文摘Gene regulation is central to all aspects of organism growth,and understanding it using large-scale functional datasets can provide a whole view of biological processes controlling complex phenotypic traits in crops.However,the connection between massive functional datasets and trait-associated gene discovery for crop improvement is still lacking.In this study,we constructed a wheat integrative gene regulatory network(wGRN)by combining an updated genome annotation and diverse complementary functional datasets,including gene expression,sequence motif,transcription factor(TF)binding,chromatin accessibility,and evolutionarily conserved regulation.wGRN contains 7.2 million genome-wide interactions covering 5947 TFs and 127439 target genes,which were further verified using known regulatory relationships,condition-specific expression,gene functional information,and experiments.We used wGRN to assign genome-wide genes to 3891 specific biological pathways and accurately prioritize candidate genes associated with complex phenotypic traits in genome-wide association studies.In addition,wGRN was used to enhance the interpretation of a spike temporal transcriptome dataset to construct high-resolution networks.We further unveiled novel regulators that enhance the power of spike phenotypic trait prediction using machine learning and contribute to the spike phenotypic differences among modern wheat accessions.Finally,we developed an interactive webserver,wGRN(http://gffzzaeec8df70e7c4b21hvkp9cffwp5b66fvf.ffgz.tsg.suse.edu.cn/wGRN),for the community to explore gene regulation and discover trait-associated genes.Collectively,this community resource establishes the foundation for using large-scale functional datasets to guide trait-associated gene discovery for crop improvement. | Yongming Chen Yiwen Guo Panfeng Guan Yongfa Wang Xiaobo Wang Zihao Wang Zhen Qin Shengwei Ma Mingming Xin Zhaorong Hu Yingyin Yao Zhongfu Ni Qixin Sun Weilong Guo Huiru Peng | 2023 | Molecular Plant2023,16,2: | 1 |
| 6 | Cellular antioxidant activity and cytotoxicity assay of canolol显示文摘To better understand canolol as an efficient intracellular antioxidant agent in food related fields, potential cellular antioxidant activity and cytotoxicity were evaluated using multiple assays, including cellular antioxidant activity(CAA), 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide(MTT) and methylene blue assay. CAA assay was used to identify the half maximal effective concentration of canolol and other bioactive components, such as sinapic acid, resveratrol and sesamol in oil crops. Results showed that antioxidant capacity(without PBS wash) varies between different compounds. Capacity variation followed the order: sinapic acid > canolol > resveratrol > sesamol. While with PBS wash, the order changed to sinapic acid > resveratrol > canolol > sesamol. Half maximal inhibitory concentration of canolol measured by MTT was 1887.08±1.22 μmol/L, implying that cytotoxicity of canolol was greatly lower than that of resveratrol, sesamol and sinapic acid. Methylene blue assay showed that cell viability was still up to 90% exposed to canolol at a high concentration of 4444 μmol/L. | 10 | China Crude Oil Imports and Oil Market-oriented Reform显示文摘Crude oil imports in China are mainly carried out by state-owned entities with non state-owned entities as compliment. Crude imported by 5 state-owned entities accounts for 90% of the total imports, while non state-owned entities are about given more freedom in using and importing crude. In 2015 only, there were 13 companies granted with access to imported crude oil and 6 were qualified to import rights. Currently, there are 29 non-state-owned companies engaging in crude import business. China oil market is faced with severe challenges. The growth rate of oil demand declined, and dependence upon imported oil increased and reached as high as 61.26% in 2015. Refined oil demand growth also slowed down, and oil refining overcapacity got prominent and completion would become fiercer in future. Overcapacity was about 140 million tons per year in 2015. Consumption ratio of diesel to gasoline went on declining, and the task of product structure adjustment was heavy. | 17 | Application of mannose-modified fullerene immunomodulator selectively polarizes tumor-associated macrophages potentiating antitumor immunity显示文摘Polarization of tumor associated macrophages(TAMs)has been a promising therapeutic paradigm for tumor.However,how to achieve precise regulation of TAMs and high efficiency of tumor immunotherapy is still a huge challenge.Here,we report dicarboxy fullerene modified with mannose(DCFM)as an immunomodulator to selectively polarize TAMs and prominently boost anti-tumor immunity.The dicarboxy fullerene molecule was synthesized through the Prato reaction and further covalently bonded with mannose,obtaining the DCFM with well-defined structure.Due to the exist of mannose in DCFM,it could accurately recognize mannose receptor in TAMs.Our cellular experiment results showed that mannose modification could notably promote the uptake of DCFM by the immunosuppressive M2-type macrophages that effectively reprogrammed M2-type macrophages into anti-tumor M1-type macrophages,leading to enhance the phagocytosis of tumor cells by macrophages and inhibiting tumor cells migration.Subsequently,we observed that DCFM could significantly distribute into tumor tissues by in vivo fluorescence imaging.Importantly,DCFM exhibited a superior anti-tumor efficiency in the subcutaneous colorectal tumor model.In addition,it showed that DCFM precisely polarized TAMs into M1-type macrophages and actively increased the infiltration of cytotoxic T lymphocytes(CTLs),inducing profound tumor growth inhibition. | Haoyu Wang Lei Li Xinran Cao Mingming Zhen Chunru Wang Chunli Bai | 2023 | Nano Research2023,16,11: | 0 |

The results showed that superoxide dismutase(SOD), catalase, glutathione reductase(GR), uric acid, vitamin E(VE) and vitamin C(VC) were more abundant in seminal plasma than in spermatozoa. The variation for each of them was specific. In seminal plasma, the activity of SOD and GR increased from November 15, November 30 to December 15, and then decreased on December 30. The concentrations of both VC and uric acid decreased during the first 3 sampling times and increased on December 30. The oxidants in seminal plasma accumulated to the highest on December 30. Lactic acid(LA) and ATP levels decreased to the lowest on December 30. | 20 | Prospective study on observations of γ-ray sources in the Galaxy using the HADAR experiment显示文摘The High Altitude Detection of Astronomical Radiation(HADAR)experiment is a refracting terrestrial telescope array based on the atmospheric Cherenkov imaging technique.It focuses the Cherenkov light emitted by extensive air showers through a large aperture water-lens system for observing very-high-energyγ-rays and cosmic rays.With the advantages of a large field-of-view(FOV)and low energy threshold,the HADAR experiment operates in a large-scale sky scanning mode to observe galactic sources.This study presents the prospects of using the HADAR experiment for the sky survey of TeVγ-ray sources from TeVCat and provids a one-year survey of statistical significance.Results from the simulation show that a total of 23 galactic point sources,including five supernova remnant sources and superbubbles,four pulsar wind nebula sources,and 14 unidentified sources,were detected in the HADAR FOV with a significance greater than 5 standard deviations(σ).The statistical significance for the Crab Nebula during one year of operation reached 346.0σand the one-year integral sensitivity of HADAR above 1 TeV was~1.3%–2.4%of the flux from the Crab Nebula. | Xiangli Qian Huiying Sun Tianlu Chen Danzengluobu Youliang Feng Qi Gao Quanbu Gou Yiqing Guo Hongbo Hu Mingming Kang Haijin Li Cheng Liu Maoyuan Liu Wei Liu Bingqiang Qiao Xu Wang Zhen Wang Guangguang Xin Yuhua Yao Qiang Yuan Yi Zhang | 2022 | Frontiers of physics2022,17,6: | 0 |
