1ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant. 3Research progress on permeability improvement mechanisms and technologies of coalbed deep-hole cumulative blasting显示文摘Coalbed gas extraction is an important means of exploiting and utilizing gas resources,as well as a means of preventing coal mine disasters.In view of the low gas extraction rate from coalbeds with high gas content and low permeability,a method of improving permeability through deep-hole cumulative blasting is applied to develop initial directional fractures using a jet flow.Under the action of the blasting stress wave and detonation gas wedge,the fractures extend over a large range within the coal,thereby improving coalbed permeability.This study focuses on the criteria of cumulative blasting-induced coalbed fracturing based on a literature review of the penetration effect of cumulative blasting.On this basis,we summarize the coal fracturing zone,crack extension process,and the key technologies of charging and hole sealing for cumulative blasting.In addition,the latest research progress in the optimization of field test drilling and blasting parameters for cumulative blasting is introduced.Research findings indicate that the permeability improvement mechanism of cumulative blasting could be further enhanced,and the technology and technical equipment are in urgent need of improvement.Finally,development trends in the cumulative blasting permeability improvement technique are identified.Deyong Guo Pengfei Lv Jiechao Zhao Chao Zhang 2020International Journal of Coal Science & Technology2020,7,2:5
4The pleiotropic ABNORMAL FLOWER AND DWARF1 affects plant height, floral development and grain yield in rice显示文摘Moderate plant height and successful establishment of reproductive organs play pivotal roles in rice grain production. The molecular mechanism that controls the two aspects remains unclear in rice. In the present study,we characterized a rice gene, ABNORMAL FLOWER AND DWARF1(AFD1) that determined plant height, floral development and grain yield. The afd1 mutant showed variable defects including the dwarfism, long panicle, low seed setting and reduced grain yield. In addition, abnormal floral organs were also observed in the afd1 mutant including slender and thick hulls, and hull-like lodicules.AFD1 encoded a DUF640 domain protein and was expressed in all tested tissues and organs. Subcellular localization showed AFD1-green fluorescent fusion protein(GFP) was localized in the nucleus. 6Rapid Creation of New Photoperiod-/Thermo-Sensitive Genic Male-Sterile Rice Materials by CRISPR/Cas9 System显示文摘Rice is an important food crop in China, and the development of hybrid rice is a crucial way to increase grain yield. The creation of dual-purpose nuclear-sterile lines for two-line hybrid breeding has become vital for commercial rice breeding. We constructed the pC1300-2 x35 S::Cas9-sgRNA^(PTGMS2-1) expression vector for editing the male fertility gene PTGMS2-1 in two widely compatible rice varieties, 93-11 and Huazhan, by using the CRISPR/Cas9 system. We obtained the marker-free photoperiod-/thermo-sensitive genic male-sterile(P/TGMS) lines in T_1 generation. According to the experiments in phytotron with four temperature and photoperiod treatments, we found the temperature is the main factor for restoring the pollen fertility of ptgms2-1 mutants in 93-11 and Huazhan, and the photoperiod also has some effects on pollen fertility in two different rice backgrounds. 7Experimental Research on Pore Structure and Gas Adsorption Characteristic of Deformed Coal显示文摘The pore structure and gas adsorption property of deformed coal with different degrees of metamorphism were tested by low-temperature nitrogen adsorption and isothermal adsorption experiments. The fractal theory and the Langmuir adsorption theory were used to analyze the experimental data. The test results showed that the deformed coal had more heterogeneous pore structures and open pores, and its specific surface area(SSA) and fractal dimension(D) were higher. There is a polynomial relationship between D and specific surface area as well as gas adsorption capacity(VL). The gas adsorption capacity of deformed coal is influenced by pore structure, coal rank, deformation and stress together, among which the pore structure is the main influencing factor for the adsorption capacity of deformed coal. 8Genome-wide association study and transcriptome analysis reveal new QTL and candidate genes for nitrogen-deficiency tolerance in rice显示文摘The development of rice cultivars with improved nitrogen use efficiency(NUE)is desirable for sustainable agriculture.Achieving this goal depends in part on understanding how rice responds to low soil nitrogen(N)and identifying causative genes underlying this trait.To identify quantitative trait loci(QTL)or genes associated with low N response,we conducted a genome-wide association study(GWAS)using a diverse panel of 230 rice accessions and performed a transcriptomic investigation of rice accessions with differential responses to low N stress at two N levels.We detected 411 GWAS-associated genes in 5 QTL and 2722 differentially expressed genes in response to low N,of which 24 were identified by both methods and ranked according to gene annotations,literature queries,gene expression,and genetic diversity analysis.The large-scale datasets obtained from this study reveal low N-responsive characteristics and provide insights towards understanding the regulatory mechanisms of N-deficiency tolerance in rice,and the candidate genes or QTL would be valuable resources for increasing rice NUE via molecular biotechnology.Qing Li Xueli Lu Changjian Wang Lan Shen Liping Dai Jinli He Long Yang Peiyuan Li Yifeng Hong Qiang Zhang Guojun Dong Jiang Hu Guangheng Zhang Deyong Ren Zhenyu Gao Longbiao Guo Qian Qian Li Zhu Dali Zeng 2022The Crop Journal2022,10,4:3

13OsPPR9 encodes a DYW-type PPR protein that affects editing efficiency of multiple RNA editing sites and is essential for chloroplast development显示文摘Photosynthesis occurs mainly in chloroplasts,whose development is regulated by proteins encoded by nuclear genes.Among them,pentapeptide repeat(PPR)proteins participate in organelle RNA editing.Although there are more than 450 members of the PPR protein family in rice,only a few affect RNA editing in rice chloroplasts.Gene editing technology has created new rice germplasm and mutants,which could be used for rice breeding and gene function study.This study evaluated the functions of OsPPR9 in chloroplast RNA editing in rice.The osppr9 mutants were obtained by CRISPR/Cas9,which showed yellowing leaves and a lethal phenotype,with suppressed expression of genes associated with chloroplast development and accumulation of photosynthetic-related proteins.In addition,loss of OsPPR9 protein function reduces the editing efficiency of rps8-C182,rpoC2-C4106,rps14-C80,and ndhB-C611 RNA editing sites,which affects chloroplast growth and development in rice.Our data showed that OsPPR9 is highly expressed in rice leaves and encodes a DYW-PPR protein localized in chloroplasts.Besides,the OsPPR9 protein was shown to interact with OsMORF2 and OsMORF9.Together,our findings provide insights into the role of the PPR protein in regulating chloroplast development in rice.CHEN Chang-zhao WANG Ya-liang HE Meng-xing LI Zhi-wen SHEN Lan LI Qing REN Deyong HU Jiang ZHU Li ZHANG Guang-heng GAO Zhen-yu ZENG Da-li GUO Long-biao QIAN Qian ZHANG Qiang 2023Journal of Integrative Agriculture2023,22,4:0
14Transcriptomic analysis reveals the promotion of lymph node metastasis by Helicobacter pylori infection via upregulating chemokine (C-X-C motif) receptor 2 expression in gastric carcinoma显示文摘Gastric carcinoma (GC) progression is mainly caused by local aggression and lymph node metastasis. However, some patients with early T-stage disease have lymph node metastasis, whereas some patients with late T-stage disease do not have lymph node metastasis, which indicates that invasion and metastasis are not always sequential in some GC patients. In the present study, the data of 101 GC cases were acquired from TCGA and divided into T-late-N-negative and T-early-N-positive groups according to pathological stages. A total of 338 genes were identified as differential genes between the T-late-N-negative and T-early-N-positive groups. GSEA showed that epithelial cell signaling in the Helicobacter pylori (HP ) infection pathway was enriched in the T-early-N-positive group. MB staining indicated that the HP infection rate was 63% (39/62) in N-positive patients compared to 42% (16/38) in N-negative patients. To investigate the potential mechanism, we focused on the gene chemokine (C-X-C motif) receptor 2 (CXCR2), which was not only clustered in the gene set of epithelial cells signaling in the HP infection pathway but also significantly upregulated in T-early-N-positive GC by the analysis of the different genes based on the TCGA dataset. A meta-analysis showed that CXCR2 expression was positively correlated with N-stage but not with T-stage in GC. This study indicated that invasion and metastasis could be independent processes driven by different molecular mechanisms in some GC patients. 15Experimental study on influence of operating parameters on tar components from corn straw gasification in fluidized bed显示文摘Gasification is a promising approach for converting solid fuel sources, including renewable ones like biomass, for use. The main problem in biomass gasification is the formation of condensable tars, including polycyclic aromatic hydrocarbons (PAHs). This paper investigated the conversion of tar components during corn straw gasification. It analyzed collected tar components using a gas chromatograph-mass spectrograph (GC-MS). Experimental results indicate that, with increasing temperature from 700°C to 900°C, the concentrations of benzene, indene, phenanthrene, naphthalene, acenaphthylene, fluorene, and pyrene increased whereas those of toluene, phenol, 1-methylnaphthalene, and 2-methylnaphthalene decreased. As the equivalence ratio (ER) increased from 0.21 to 0.34, the concentrations of indene and phenanthrene increased from 0.148% and 0.087% to 0.232% and 0.223%, respectively. Further, the phenol content increased as ER increased from 0.21 to 0.26 and then decreased as the ER increased further to 0.34. Other parameters like the steam/biomass (S/B) ratio and catalyst also played a critical role in tar reduction. 16Functional Analysis of Three Rice Chloroplast Transit Peptides显示文摘Chloroplast transit peptides(CTPs) can be used to transport non-chloroplastic proteins into the chloroplasts. Here, we studied the CTPs of three rice(Oryza sativa L.) chloroplast-localized proteins and found that their CTPs could be used to transport non-chloroplast-localized proteins into the chloroplasts. Fusion proteins lacking the CTP remained located in the cytoplasm. Furthermore, we constructed green fluorescent protein fusion vectors with the three CTPs and three non-chloroplast-localized proteins, Ghd10, MULTI-FLORET SPIKELET1(MFS1), and SHORTENED UPPERMOST INTERNODE 1(SUI1). After transforming these constructs into rice protoplasts, the fusion proteins all localized in the chloroplasts. 17Mild Renal Dysfunction and Risk of Thromboembolism and Bleeding in Patients with Atrial Fibrillation:The Chinese Atrial Fibrillation Registry Study显示文摘Objective:Previous studies indicated that patients with atrial fibrillation(AF)and moderate-to-severe chronic kidney disease(CKD)are at a higher risk of thromboembolism and bleeding during anticoagulation.Whether mild CKD is associated with an increased risk of thromboembolism and bleeding in AF patients remains unknown.This study aimed to evaluate the impact of mild CKD on thromboembolism and major bleeding among patients with AF.Methods:Baseline serum creatinine was available in 17,559 of 25,512 patients enrolled in the China-AF study between August 2011 and December 2018.After excluding those who underwent AF ablation or with moderate-to-severe CKD,7191 non-valvular AF patients(2059 with mild CKD and 5132 with normal renal function)with regular follow-up for at least 6 months were included.Primary outcomes were the time to the first occurrence of thromboembolic and major bleeding events.Results:Over a mean follow-up of(44.4±23.4)months,639 thromboembolism and 231 major bleeding events occurred.The crude incidence rates of thromboembolism were higher in the mild CKD group than that of the normal renal function group(3.0/100 person-years vs.2.2/100 person-years,P<0.0001),while the crude incidence rates of major bleeding were comparable between the two groups(1.0/100 person-years vs.0.8/100 person-years,P=0.076).After multivariate analyses,mild CKD was not associated with an increased risk of thromboembolism(HR=1.05,95%CI:0.89-1.25,P=0.547)or major bleeding(HR=1.11,95%CI:0.84–1.47,P=0.476).Conclusions:Mild CKD was not an independent risk factor of thromboembolism or major bleeding in patients with AF.Jingye Li Sitong Li Chao Jiang Jing Du Xueyuan Guo Songnan Li Chenxi Jiang Ribo Tang Caihua Sang Deyong Long Xin Du Jianzeng Dong Changsheng Ma 2021Cardiology Discovery2021,1,1:0
