1Comprehensive Management of Daily Living Activities,behavioral and Psychological Symptoms,and Cognitive Function in Patients with Alzheimer's Disease:A Chinese Consensus on Alzheimer's Disease显示文摘Alzheimer's disease(AD)is the most common cognitive disorder in the elderly.Its main clinical manifestations are cognitive decline(C),behavioral and psychological symptoms(B),and a decline in the activities of daily living(A),also known as ABC symptoms.Early identification and evaluation of ABC symptoms are helpful for establishing the accurate diagnosis,comprehensive treatment,and prognosis of AD.To guide Chinese clinical practice for optimization of the comprehensive management of AD,in 2018,The Academy of Cognitive Disorder of China gathered 22 neurologists and gerontologists in China to build a consensus on the comprehensive management of AD.Based on a review of the evidence,the consensus summarizes the pathogenesis,pathological changes,clinical manifestations,evaluation,diagnosis,drug and non-drug treatment,and patient care for AD.Focus group discussion was used to establish a flowchart of comprehensive ABC management for AD patients.The new consensus provides a feasible AD management process for clinicians.Jianjun Jia Jun Xu Jun Liu Yongjun Wang Yanjiang Wang Yunpeng Cao Qihao Guo Qiuming Qu Cuibai Wei Wenshi Wei Junjian Zhang Enyan Yu 2021Neuroscience Bulletin2021,37,7:10
2Mechanosignaling activation of TGFβmaintains intervertebral disc homeostasis显示文摘Intervertebral disc(IVD) degeneration is the leading cause of disability with no disease-modifying treatment.IVD degeneration is associated with instable mechanical loading in the spine, but little is known about how mechanical stress regulates nucleus notochordal(NC) cells to maintain IVD homeostasis. Here we report that mechanical stress can result in excessive integrin α_vβ_6-mediated activation of transforming growth factor beta(TGFβ), decreased NC cell vacuoles, and increased matrix proteoglycan production, and results in degenerative disc disease(DDD). Knockout of TGFβ type II receptor(TβRII) or integrin α_v in the NC cells inhibited functional activity of postnatal NC cells and also resulted in DDD under mechanical loading.Administration of RGD peptide, TGFβ, and α_vβ_6-neutralizing antibodies attenuated IVD degeneration. Thus,integrin-mediated activation of TGFβ plays a critical role in mechanical signaling transduction to regulate IVD cell function and homeostasis. 5Cloning,expression and binding specificity analysis of odorant binding protein 3 of the lucerne plant bug,Adelphocoris lineolatus(Goeze)显示文摘Credible evidence shows that odorant binding proteins(OBPs)are required for insect olfaction perception and play a key role in transporting hydrophobic odorants across the sensillum lymph to the olfactory receptors(ORs).In the present study,a novel OBP(AlinOBP3)gene from the lucerne plant bug,Adelphocoris lineolatus,was cloned and expressed.The expression pattern of AlinOBP3 was evaluated by qPCR,which indicated that AlinOBP3 was dominantly expressed in antennae.The binding properties of AlinOBP3 with 9 cotton volatiles and 5 sex pheromone analogs were measured by fluorescence competitive binding assays with the fluorescence probe 1-NPN.The results revealed that of 9 cotton volatiles,Myrcene,β-Ocimene and α-Phellandrene can bind with AlinOBP3.α-Phellandrene especially bound to AlinOBP3 with a high binding affinity,with a dissociation constant of 56.68 μmol/L.Of the 5 sex pheromone analogs,Hexyl butyrate had the strongest binding affinity with AlinOBP3,with a dissociation constant as 59.53 μmol/L.Butyl butyrate,trans-2-Hexenyl butyrate and Ethyl butyrate had medium binding affinities with AlinOBP3,with dissociation constants of 227.39,108.77 and 143.47 μmol/L,respectively.The results suggest that AlinOBP3 might be a pheromone binding protein(PBP)with a dual-function for the perception of sex pheromones and plant volatiles.GU ShaoHua SUN Yang REN LiYan ZHANG XueYing ZHANG YongJun WU KongMing GUO YuYuan 2010Chinese Science Bulletin2010,55,34:7

7Emerging role of natural products in cancer immunotherapy显示文摘Cancer immunotherapy has become a new generation of anti-tumor treatment,but its indications still focus on several types of tumors that are sensitive to the immune system.Therefore,effective strategies that can expand its indications and enhance its efficiency become the key element for the further development of cancer immunotherapy.Natural products are reported to have this effect on cancer immunotherapy,including cancer vaccines,immune-check points inhibitors,and adoptive immune-cells therapy.And the mechanism of that is mainly attributed to the remodeling of the tumorimmunosuppressive microenvironment,which is the key factor that assists tumor to avoid the recognition and attack from immune system and cancer immunotherapy.Therefore,this review summarizes and concludes the natural products that reportedly improve cancer immunotherapy and investigates the mechanism.And we found that saponins,polysaccharides,and flavonoids are mainly three categories of natural products,which reflected significant effects combined with cancer immunotherapy through reversing the tumor-immunosuppressive microenvironment.Besides,this review also collected the studies about nano-technology used to improve the disadvantages of natural products.All of these studies showed the great potential of natural products in cancer immunotherapy.Songtao Dong Xiangnan Guo Fei Han Zhonggui He Yongjun Wang 2022Acta Pharmaceutica Sinica B2022,12,3:5
8Hexagonal boron nitride nanosheet for effective ambient N2 fixation to NH3显示文摘Industrial production of NH3 from N2 and H2 significantly relies on Haber-Bosch process,which suffers from high energy consume and CO2 emission.As a sustainable and environmentally-benign alternative process,electrochemical artificial N2 fixation at ambient conditions,however,is highly required efficient electrocatalysts.In this study,we demonstrate that hexagonal boron nitride nanosheet (h-BNNS) is able to electrochemically catalyze N2 to NH3.In acidic solution,h-BNNS catalyst attains a high NH3 formation rate of 22.4 μg·h-1·mg-1cat.and a high Faradic efficiency of 4.7% at-0.75 V vs.reversible hydrogen electrode,with excellent stability and durability.Density functional theory calculations reveal that unsaturated boron at the edge site can activate inert N2 molecule and significantly reduce the energy barrier for NH3 fonmation.Ya Zhang Huitong Du Yongjun Ma Lei Ji Haoran Guo Ziqi Tian Hongyu Chen Hong Huang Guanwei Cui Abdullah M. 10Trends and predictors of myocardial infarction or vascular death after ischaemic stroke or TIA in China, 2007-2018: insights from China National Stroke Registries显示文摘Background Although stroke management,primary and secondary preventions have been improved in China last decades,the trends and predictors of major vascular events after ischaemic stroke or transient ischaemic attack(TIA)at national scale are less known.Methods Data were obtained from the three phases of China National Stroke Registry(CNSR),including CNSR-Ⅰ(years 2007-2008),CNSR-Ⅱ(years 2012-2013)and CNSR-III(years 2015-2018).For comparison,patients who were diagnosed as ischaemic stroke or TIA were included.Kaplan-Meier estimates of myocardial infarction(MI)or vascular death were calculated at 1 year.Independent predictors were further assessed with a Cox proportional hazards regression.Results From 2007 to 2018,a total of 50284 patients with ischaemic stroke or TIA were enrolled in this study.A declining trend was found in 1-year MI or vascular death(p for trend<0.001),while recurrent stroke depicted a U-shape curve with a nadir in 2012-2013 cohort.A similar trend was also observed in patients who were admitted to 26 hospitals in all three CNSRs.In 2015-2018 cohort,only 251(1.7%;95%CI 1.5%to 1.9%)MI or vascular death had occurred at 1 year.Older age,previous stroke or TIA,history of coronary artery disease and the National Institutes of Health Stroke Scale>6 were associated with both an increased risk of MI or vascular death and recurrent stroke.While early antiplatelet therapy and lipid-lowering agents at discharge predicted a reduced risk.Conclusion A declining trend and current low incidence of MI or vascular death,rather than recurrent stroke,after ischaemic stroke or TIA were observed in China.Traditional factors were found as independent predictors.These findings suggested there is still much room to improve for stroke management.Long Li Yuesong Pan Mengxing Wang Jing Jing Xia Meng Yong Jiang Caixia Guo Zening Jin Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:4

13Enhanced bioavailability of rebamipide nanocrystal tablets:Formulation and in vitro/in vivo evaluation显示文摘The purpose of this study was to formulate rebamipide nanocrystal tablets(REB-NTs)by wet-milling technique to enhance its dissolution rate and oral bioavailability.The formulation and preparation technology were screened by single factor tests with particle size and distribution as indicators.Rebamipide nanocrystals(REB-NSs)was then achieved by freeze-dry from the prepared nanosuspensions which were characterized by differential scanning calorimetry(DSC)and x-ray powder diffraction(XRD),while the vitro dissolution and the plasma drug concentration of the nanocrystal tablets were investigated.The results indicated that the prepared nanosuspensions got an average particle size of 286 nm,PI of 0.173 and the average Zeta potential of18.2 mv.The average particle size of obtained REB-NSs’redispersibility was 278 nm,and the crystalline of REB-NSs was the same as the rebamipide bulk drug as shown by DSC and XRD.The drug dissolution rate of self-made nanocrystal tablets in different dissolutions was slightly faster than that from the reference tablets,REB-MTs(Mucosta®),while the Cmax and AUC0e24 of REB-NTs were 1 and 1.57 times higher than that of REB-MTs,which means the nanotechnology could significantly improve the oral bioavailability of rebamipide.Yu Guo Yongjun Wang Lu Xu 2015Asian Journal of Pharmaceutical Sciences2015,10,3:2

16Dynamic changes of renal cortical blood perfusion before and after percutaneous transluminal renal artery stenting in patients with severe atherosclerotic renal artery stenosis显示文摘Background:This study aims to observe the dynamic changes of renal artery(RA)disease and cortical blood perfusion(CBP)evaluated by contrast-enhanced ultrasound(CEUS)after percutaneous transluminal renal artery stenting(PTRAS)in patients with severe atherosclerotic renal artery stenosis(ARAS)and to analyze the relationship between CBP and prognosis.Methods:This was a single-center retrospective cohort study.A total of 98 patients with unilateral severe ARAS after successful PTRAS in Beijing Hospital from September 2017 to September 2020 were included.According to renal glomerular filtration rate(GFR)detected by radionuclide imaging at 12 months after PTRAS,all patients were divided into the poor prognosis group(n=21,GFR decreased by≥20%compared with baseline)and the control group(n=77,GFR decreased by<20%or improved compared with baseline).Renal artery stenosis was diagnosed by digital subtraction angiography,and renal CBP was evaluated by CEUS using TomTec Imaging Systems(Germany)before PTRAS,at 6 months and 12 months after discharge.The receiver operating characteristic(ROC)curve with area under the curve(AUC)was used to analyze the predictive value of CBP parameters,including area under ascending curve(AUC1),area under the descending curve(AUC2),rising time(RT),time to peak intensity(TTP),maximum intensity(IMAX),and mean transit time(MTT)for poor prognosis.Results:Among the 98 patients,there were 52 males(53.1%),aged 55–74 years old,with an average age of 62.1±8.7 years,and an average artery stenosis of 82.3±12.9%.The poor prognosis group was associated with significantly increased incidence of diabetes(76.2%vs.41.6%),and lower levels of GFR of the stenotic kidney(21.8 mL/min vs.25.0 mL/min)and total GFR(57.6 mL/min vs.63.7 mL/min)(all P<0.05),compared with the control group(P<0.05).In addition,the rate of RA restenosis was significantly higher in the poor prognosis group than in the control group(9.5%vs.0,χ^(2)=9.462,P=0.002).Compared with the control group,the poor prognosis group was associated with significantly decreased baseline AUC1 and AUC2,and extended duration of TTP and MTT(P<0.05).At 6 months and 12 months of follow-up,patients in the control group were associated with markedly increased AUC1,AUC2,and IMAX,and shorter duration of RT and MTT(P<0.05).The ROC curve showed that the predictive values of AUC1,AUC2,RT,TTP,IMAX,and MTT for poor prognosis were 0.812(95%CI:0.698–0.945),0.752(95%CI:0.591–0.957),0.724(95%CI:0.569–0.961),0.720(95%CI:0.522–0.993),0.693(95%CI:0.507–0.947),and 0.786(95%CI:0.631–0.979),respectively.Conclusions:Preoperative renal CBP in severe ARAS patients with poor prognosis is significantly reduced,and does not show significant improvement after stent treatment over the first year of follow-up.The parameter AUC1 may be a good predictor for renal dysfunction after PTRAS in severe ARAS patients.Trial Registration:ChiCTR.org.cn,ChiCTR1800016252.Na Ma Yan Li Siyu Wang Mengpu Li Yongjun Li Hu Ai Hui Zhu Yang Wang Fajin Guo Junhong Ren 2022Chinese Medical Journal2022,,13:1
