1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Characteristics of strong earthquake evolution around the eastern boundary faults of the Sichuan-Yunnan rhombic block显示文摘Based on the existing materials of fault segmentation,characteristic earthquakes,and their empirical relationships,we calculated the parameters of the fault segments,such as length,width,magnitudes of characteristic earthquakes,etc.Constrained by GPS velocity field,the slip rates of these fault segments in depth were inversed using the 3-D half-space elastic dislocation model.As not all of the recurrence periods and co-seismic displacements of characteristic earthquakes are known,we selected the fault segments with these two parameters known and calculated the accumulation rate of average co-seismic displacement,which shows the faults' slip rate in seismogenic layer.Then,the slip rate in depth was compared with that in seismogenic layer,the relationship between them was obtained,and this relationship was used to get the recurrence periods and co-seismic displacements of all fault segments.After the studies above,we calculated the co-seismic deformation field of all the earthquakes larger than M s 6.8 from AD 1700 one by one and inversed the potential displacement in the co-seismic deformation field.Then,we divided the potential displacement by the slip rate from GPS inversion to get the influences of these fault segments,added the influences into the elapsed time of the characteristic earthquakes,and obtained the earthquake hazard degree of all the segments we studied in the form of the ratio of elapsed time to recurrence period;so,we name the ratio as the Impending Earthquake Risk (IER).Historical earthquake cases show that the fault segment is in safety when the IER is less than 1 but in danger after the IER becomes larger than 1.In 2009,the IER is larger than 1 on the following segments,1.35 on the Tagong segment of Xianshuihe fault,1 on the Menggu-Dongchuan segment,1.04 on the Dongchuan-Xundian segment,and 1.09 on the Yiliang-Chengjiang segment of Xiaojiang fault.CHENG Jia LIU Jie GAN WeiJun YU HuaiZhong LI Gang 2011Science China Earth Sciences2011,54,11:15
4Restorative effect and mechanism of mecobalamin on sciatic nerve crush injury in mice显示文摘Mecobalamin,a form of vitamin B12 containing a central metal element(cobalt),is one of the most important mediators of nervous system function.In the clinic,it is often used to accelerate recovery of peripheral nerves,but its molecular mechanism remains unclear.In the present study,we performed sciatic nerve crush injury in mice,followed by daily intraperitoneal administration of mecobalamin(65 μg/kg or 130 μg/kg) or saline(negative control).Walking track analysis,histomorphological examination,and quantitative real-time PCR showed that mecobalamin significantly improved functional recovery of the sciatic nerve,thickened the myelin sheath in myelinated nerve fibers,and increased the cross-sectional area of target muscle cells.Furthermore,mecobalamin upregulated m RNA expression of growth associated protein 43 in nerve tissue ipsilateral to the injury,and of neurotrophic factors(nerve growth factor,brain-derived nerve growth factor and ciliary neurotrophic factor) in the L4–6 dorsal root ganglia.Our findings indicate that the molecular mechanism underlying the therapeutic effect of mecobalamin after sciatic nerve injury involves the upregulation of multiple neurotrophic factor genes.Lin Gan Minquan Qian Keqin Shi Gang Chen Yanglin Gu Wei Du Guoxing Zhu 2014Neural Regeneration Research2014,9,22:11

9Mesoporous Ag nanocubes synthesized via selectively oxidative etching at room temperature for surface- enhanced Raman spectroscopy显示文摘Silver nanocubes enriched with {100} facets have been extensively used for surface-enhanced Raman scattering. Herein, we report a new water-phase synthesis method for weU-defined Ag nanocubes with tunable sizes via a two-step procedure at room temperature. First, irregularly shaped Ag nanoparticles (INPs) were prepared by reducing silver ammonia solution using ethylal. Second, the agglomerated INPs were selectively etched with HNO3 and C1- to yield {100} facet-rich mesoporous Ag nanocubes. The mechanism of Ag-nanocube formation and growth was investigated in detail by elucidating the involved chemical reactions and physical changes at each step during the synthesis. The addition of C1- anions was responsible for facilitating Ag nanoparticle growth by removing surface-adsorbed Ag+ species, thereby eliminating inter-particle repulsive forces. This agglomeration was found crucial for the subsequent selective oxidation of Ag nanoparticles because the protective agent used, polyvinylpyrrolidone (PVP), was the most effective one for adsorption on the surfaces of Ag nanoparticles of size greater than approximately 50 nm. Importantly mesopores were found inside the Ag nanocubes; this can be attributed to the unavoidable imperfect packing during the agglomeration of INPs. 10Progress of Jinping Underground laboratory for Nuclear Astrophysics(JUNA)显示文摘Jinping Underground laboratory for Nuclear Astrophysics(JUNA) will take the advantage of the ultra-low background of CJPL lab and high current accelerator based on an ECR source and a highly sensitive detector to directly study for the first time a number of crucial reactions occurring at their relevant stellar energies during the evolution of hydrostatic stars. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. The experimental setup,which includes an accelerator system with high stability and high intensity, a detector system, and a shielding material with low background, will be established during the above research. 11Acute Pancreatitis Complicated by Sheehan's Syndrome: A Case Report and Literature Review显示文摘A 44-year-old woman was transferred to the ICU of the First Affiliated Hospital of Jinan University for 2 days of persistent epigastric pain and 7 hours of unconsciousness.Her admission diagnosis was severe acute necrotizing pancreatitis(hypertriglyceridemia type)with multiple organ dysfunctions.The results of CT revealed a small area of necrotizing pancreatitis,which was not consistent with the severe clinical manifestations.Considering lack of hair and history of postpartum hemorrhage,hormone examination was carried out.According to the results of the examination,she was further diagnosed as Sheehan's syndrome and pituitary crisis.After hormone replacement therapy,her condition improved rapidly.Dasheng Liu Li Liu Feng Gan Xianlin Wu Gang Ye 2020Chinese Medical Sciences Journal2020,35,1:3
12Petroleum Origins and Accumulation Patterns in the Weixinan Sag in the Beibu Gulf Basin,Using Subsag B as an Example显示文摘Crude oil has been discovered in the Paleogene and Neogene units of the Weixinan Sag in the Beibu Gulf Basin.To determine the source and accumulation mode of this crude oil,12 crude oil samples and 27 source rock samples were collected and an extensive organic geochemical analysis was conducted on them.Based on the geological conditions and the analytical results,the types,origins and accumulation patterns of crude oil in the study area were elucidated.Except for a shallowly-buried and biodegraded crude oil deposit in Neogene rocks,the crude oil samples in the study area were normal.All of the crude oils were derived from lacustrine source rocks.According to biomarker compositions,the crude oils could be divided into two families,A and B,distinctions that were reinforced by differences in carbon isotope composition and spatial distribution.Oil-source correlation analysis based on biomarkers revealed that Family A oils were derived from the mature oil shale at the bottom of the second member of the Liushagang Formation,while the Family B oils formed in the mature shale of the Liushagang Formation.The Family A oils,generated by oil shale,mainly migrated laterally along sand bodies and were then redistributed in adjacent reservoirs above and below the oil shale layer,as well as in shallow layers at high structural positions,occupying a wide distribution range.The Family B oils were generated by other shale units before migrating vertically along faults to form reservoirs nearby,resulting in a narrow distribution range.GAO Gang XU Xinde QU Tong GAN Jun DANG Wenlong ZHOU Xiaoxiao LIU Fengyan 2020Acta Geologica Sinica(English Edition)2020,94,5:3
13Synergistic impact of cocatalysts and hole scavenger for promoted photocatalytic H2 evolution in mesoporous TiO2–NiSx hybrid显示文摘Photocatalytic solar energy conversion to hydrogen is sustainable and attractive for addressing the global energy and environmental issue. Herein, a novel photocatalytic system (NiS/Ni3S4 cocatalysts modified mesoporous TiO2) with superior photocatalytic hydrogen evolution capability through the synergistic impact of NiS/Ni3S4 (NiSx) cocatalyst and efficient hole scavenger has been demonstrated. The photocatalytic hydrogen evolution of TiO2-NiSx hybrids with the different content of NiSx and upon different organic hole scavengers was both investigated. The hybrid of TiO2 decorated with 3%(mole ratio of Ni^2+) NiSx cocatalyst in methanol solution showed the optimal photocatalytic hydrogen evolution rate of 981.59 μmol h^-1 g^-1 which was about 20 times higher than that of bare mesoporous TiO2. 14Environmentally responsive dual-targeting nanotheranostics for overcoming cancer multidrug resistance显示文摘The development of multiple drug resistance(MDR) to chemotherapy and subsequent treatment failures are major obstacles in cancer therapy. An attractive option for combating MDR is inhibiting the expression of P-glycoprotein(P-gp) in tumor cells. Here, we report a novel chemosensitizing agent, XMD8-92,which can down-regulate P-gp. To enhance the specificity of MDR chemotherapy, a promising nanotheranostic micelle system based on poly(ethylene glycol)-blocked-poly(L-leucine)(PEG-b-Leu) was developed to simultaneously carry the anticancer drug doxorubicin, chemosensitizing agent XMD8-92, and superparamagnetic iron oxide nanoparticles(SPIOs). Featured with MDR environmentally responsive dual-targeting capability, controllable drug delivery, and efficient magnetic resonance(MR) imaging characteristics, the prepared nanotheranostics(DXS@NPs) showed outstanding in vitro cytotoxicity on MDR cells(SCG 7901/VCR) with only 53% of cells surviving compared to 90% of DOX-treated cells.Furthermore, efficient tumor inhibition and highly reduced systemic toxicity were exhibited by MDR tumor-bearing mice treated with DXS@NPs. 16为从变形体 berghei 的氮的氧化物 synthase 和类似的蛋白质的结构和功能的生物信息学分析和预言显示文摘 <正>Objective:To search and analyze nitric oxide synthase(NOS) and similar proteins from Plasmodium berghei(Pb).Methods:The structure and function of nitric oxide synthase and similar proteins from Plasmodium berghei were analyzed and predicted by bioinformatics. Results:P6NOS were not available,but nicotinamide adenine dinucleotide 2’-phosphate reduced tetrasodium(NADPH)-cytochrome p450 reductase(CPR) were gained.PiCPR was in the nucleus of Plasmodium berghei,while 134aa-229aa domain was localize in nucleolar organizer. The amino acids sequence of P6CPR had the closest genetic relationship with Plasmodium vivax showing a 73%homology.The tertiary structure of PbCPR displayed the forcep-shape with wings,but no wings existed in the tertiary structure of its’ host,Mus musculus(Mm).137aa-200aa, 201aa-218aa,220aa-230aa,232aa-248,269aa-323aa,478aa-501aa and 592aa-606aa domains of P6CPR showed no homology with MmCPRs’,and all domains were exposed on the surface of the protein.Conclusions:NOS can’t be found in Plasmodium berghei and other Plasmodium species.PbCPR may be a possible resistance site of antimalarial drug,and the targets of antimalarial drug and vaccine.It may be also one of the mechanisms of immune evasion.This study on Plasmodium berghei may be more suitable to Plasmodium vivax.And137aa-200aa, 201aa-218aa,220aa-230aa,232aa-248,269aa-323aa,478aa-501aa and 592aa-606aa domains of PbCPR are more ideal targets of antimalarial drug and vaccine.Zhigang Fan Gang Lv Lingmin Zhang Xiufeng Gan Qiang Wu Saifeng Zhong Guogang Yan Guifen Lin 2011Asian Pacific Journal of Tropical Medicine2011,4,1:2
17Downregulation of MEIS1 mediated by ELFN1-AS1/EZH2/DNMT3a axis promotes tumorigenesis and oxaliplatin resistance in colorectal cancer显示文摘Oxaliplatin is widely used in the frontline treatment of colorectal cancer(CRC),but an estimated 50%of patients will eventually stop responding to treatment due to acquired resistance.This study revealed that diminished MEIS1 expression was detected in CRC and harmed the survival of CRC patients.MEIS1 impaired CRC cell viabilities and tumor growth in mice and enhanced CRC cell sensitivity to oxaliplatin by preventing DNA damage repair.Mechanistically,oxaliplatin resistance following MEIS1 suppression was critically dependent on enhanced FEN1 expression.Subsequently,we confirmed that EZH2-DNMT3a was assisted by lncRNA ELFN1-AS1 in locating the promoter of MEIS1 to suppress MEIS1 transcription epigenetically.Based on the above,therapeutics targeting the role of MEIS1 in oxaliplatin resistance were developed and our results suggested that the combination of oxaliplatin with either ELFN1-AS1 ASO or EZH2 inhibitor GSK126 could largely suppress tumor growth and reverse oxaliplatin resistance.This study highlights the potential of therapeutics targeting ELFN1-AS1 and EZH2 in cell survival and oxaliplatin resistance,based on their controlling of MEIS1 expression,which deserve further verification as a prospective therapeutic strategy.Yimin Li Yaqi Gan Jiaxin Liu Juanni Li Zhengwei Zhou Ruotong Tian Ruizheng Sun Jiaqi Liu Qing Xiao Yuanyuan Li Pengyan Lu Yulong Peng Yuqian Peng Guang Shu Gang Yin 2022Signal Transduction and Targeted Therapy2022,7,4:2
18Diagnosis,treatment,and prevention of severe acute hepatitis of unknown etiology in children显示文摘Background Severe acute hepatitis of unknown etiology in children has recently exhibited a global trend of concentrated occurrence.This review aimed to summarize the current available information regarding the outbreak of severe acute hepatitis and introduce our hospital’s previous experiences with the diagnosis and treatment of severe acute hepatitis for reference.Data sources Websites including the UK Health Security Agency,European Centre for Disease Prevention and Control,CDC,WHO,and databases including PubMed/Medline,Cochrane Library,Embase and Web of Science were searched for articles on severe acute hepatitis in children.Results As of May 26,2022,a total of 650 cases have been reported in 33 countries;at least 38(6%)children required liver transplantation,and nine(1%)died.Cases are predominantly aged between 3 and 5 years old,and there are no epidemiological links among them.The common manifestations are jaundice,vomiting and pale stools.Adenovirus tested positive in most cases,and SARS-CoV-2 and other viruses were detected in a few cases,but virus particles were not found in liver tissue.Adenovirus immunohistochemistry showed immunoreactivity in the intrasinusoidal lumen from some liver samples.The hierarchical treatment includes symptomatic and supportive therapy,management of coagulation disorders and hepatic encephalopathy,artifcial liver support,and liver transplantation(approximately 6%–10%of cases require liver transplant).Conclusions The etiology of this severe acute hepatitis in children is not clear.The clinical features are severe acute hepatitis with signifcantly elevated liver enzymes.Clinicians need to be alert to children with hepatitis.Ying‑Hu Chen Jin‑Gan Lou Zi‑Hao Yang Qing‑Jiang Chen Chun‑Zhen Hua Sheng Ye Chen‑Mei Zhang Jie Chen Zong‑Wei Huang Jin‑Dan Yu Zhi‑Gang Gao Qiang Shu 2022World Journal of Pediatrics2022,18,8:2
19Synthesis and Antitumor Activities In Vitro of Solanesylpiperazinotriamine显示文摘A series of novel antitumor agents—the solanesylpiperazinotriamine derivatives were designed and synthesized, their structures were confirmed by IR, 1H-NMR, MS, and element analysis.
