| 3 | Targeting ERK,an Achilles' Heel of the MAPK pathway,in cancer therapy显示文摘The mitogen-activated protein kinases(MAPK) pathway, often known as the RAS-RAFMEK-ERK signal cascade, functions to transmit upstream signals to its downstream effectors to regulate physiological process such as cell proliferation, differentiation, survival and death. As the most frequently mutated signaling pathway in human cancer, targeting the MAPK pathway has long been considered a promising strategy for cancer therapy. Substantial efforts in the past decades have led to the clinical success of BRAF and MEK inhibitors. However, the clinical benefits of these inhibitors are compromised by the frequently occurring acquired resistance due to cancer heterogeneity and genomic instability. This review briefly introduces the key protein kinases involved in this pathway as well as their activation mechanisms. We also generalize the correlations between mutations of MAPK members and human cancers, followed by a summarization of progress made on the development of small molecule MAPK kinases inhibitors. | 7 | Characteristics and Performance of Novel Water-Absorbent Slow Release Nitrogen Fertilizers显示文摘In this article, a research on the characteristics and performance of water-absorbent slow release nitrogen fertilizer (WASRNF) using infrared spectroscopy (IR), scanning electronic microscopy (SEM), differential scanning calorimetry (DSC), and thermogravimetry analysis (TGA), was present. The results indicate that the water absorbency and nitrogen analysis of WASRNF is 103 g g-1 and 30%, respectively, and WASRNF exhibits approximately neutral pH and very low salt index. WASRNF is a copolymer of nitrogen fertilizer and super absorbent polymer (SAP) monomers which is formed through hydrogen bond interaction, and the molecule contains hydrophilic groups, which is responsible for the absorption and water retention capacity of the molecule. WASRNF is a gel that exhibits the ability to swell, but does not dissolve in water. WASRNF shows non-homogenous nature as a whole, but in local zone it is homogenous, the copolymer molecule shows chain network that is the physical structure responsible for absorption and retention of water in WASRNF. The water retained in WASRNF exists as free and nonfreezing bound and freezing bound water states, with the free and the nonfreezing water accounting for more than 95% of water retained in WASRNF, and the nonfreezing bound water for less than 5%. | 13 | Soliton bursts and deterministic dissipative Kerr soliton generation in auxiliary-assisted microcavities显示文摘Dissipative Kerr solitons in resonant frequency combs offer a promising route for ultrafast mode-locking,precision spectroscopy and time-frequency standards.The dynamics for the dissipative soliton generation,however,are intrinsically intertwined with thermal nonlinearities,limiting the soliton generation parameter map and statistical success probabilities of the solitary state.Here,via use of an auxiliary laser heating approach to suppress thermal dragging dynamics in dissipative soliton comb formation,we demonstrate stable Kerr soliton singlet formation and soliton bursts.First,we access a new soliton existence range with an inverse-sloped Kerr soliton evolution—diminishing soliton energy with increasing pump detuning.Second,we achieve deterministic transitions from Turinglike comb patterns directly into the dissipative Kerr soliton singlet pulse bypassing the chaotic states.This is achieved by avoiding subcomb overlaps at lower pump power,with near-identical singlet soliton comb generation over twenty instances.Third,with the red-detuned pump entrance route enabled,we uncover unique spontaneous soliton bursts in the direct formation of low-noise optical frequency combs from continuum background noise.The burst dynamics are due to the rapid entry and mutual attraction of the pump laser into the cavity mode,aided by the auxiliary laser and matching well with our numerical simulations.Enabled by the auxiliary-assisted frequency comb dynamics,we demonstrate an application of automatic soliton comb recovery and long-term stabilization against strong external perturbations.Our findings hold potential to expand the parameter space for ultrafast nonlinear dynamics and precision optical frequency comb stabilization. | Heng Zhou Yong Geng Wenwen Cui Shu-Wei Huang Qiang Zhou Kun Qiu Chee Wei Wong | 2019 | Light(Science & Applications)2019,8,1: | 16 |
| 14 | Binding Activity Difference of Anti-CD20 scFv-Fc Fusion Protein Derived from Variable Domain Exchange显示文摘Two novel engineered antibody fragments binding to antigen CD20 were generated by fusing a murine IgM-type anti-CD20 single-chain Fv fragment (scFv) to the human IgG1 CH2 (i.e., Cγ2) and CH3 (i.e., Cγ3) domains with the human IgG1 hinge (i.e. Hγ). Given the relationship between structure and function of protein, the 3-D structures of the two engineered antibody fragments were modeled using computer-aided homology modeling method. Furthermore, the relationship between 3-D conformation and their binding activity was evaluated theoretically. Due to the change of active pocket formed by CDRs, the HL23 (VH-Linker-VL-Hγ-Cγ2-Cγ3) remained its activity because of its preserved conformation, while the binding activity of the LH23 (VL-Linker-VH-Hγ-Cγ2-Cγ3) was impaired severely. Experimental studies by flow cytometry and fluorescence microscopy showed that HL23 possessed significantly superior binding activity to CD20-expressing target cells than LH23. That is to say, the order of variable regions could influence the binding activity of the fusion protein to CD20+ cell lines, which was in accordance with the theoretical results. | 15 | Soil respiration is driven by fine root biomass along a forest chronosequence in subtropical China显示文摘Aims Soil respiration(Rs)is a major process controlling soil carbon loss in forest ecosystems.However,the underlying mechanisms leading to variation in Rs along forest successional gradients are not well understood.In this study,we investigated the effects of biotic and abiotic factors on Rs along a forest successional gradient in southeast China.Methods We selected 16 plots stratified by forest age,ranging from 20 to 120 years.In each plot,six shallow collars and six deep collars were permanently inserted into the soil.Shallow and deep collars were used to measure Rs and heterotrophic respiration(Rh),respectively.Autotrophic soil respiration(Ra)was estimated as the difference between Rs and Rh.Litter layer respiration(R_(L))was calculated by subtracting soil respiration measured in collars without leaf litter layer(R_(NL))from Rs.Rs was measured every 2 months,and soil temperature(ST)and soil volumetric water content(SVWC)were recorded every hour for 19 months.We calculated daily Rs using an exponential model dependent on ST.Daily Rs was summed to obtain cumulative annual Rs estimates.Structural equation modelling(SEM)was applied to identify the drivers of Rs during forest succession.Important Findings Rs showed significant differences among three successive stages,and it was the highest in the young stage.Ra was higher in the young stage than in the medium stage.Cumulative annual Rs and Ra peaked in the young and old stages,respectively.Cumulative annual Rh and respiration measured from soil organic matter(R_(SOM))decreased,whereas R_(L)increased with forest age.The SEM revealed that cumulative annual Rs was influenced by fine root biomass and SVWC.Our results indicated that the dominant force regulating Rs on a seasonal scale is ST;however,on a successional scale,belowground carbon emerges as the dominant influential factor. | Chao Wang Yinlei Ma Stefan Trogisch Yuanyuan Huang Yan Geng Michael Scherer-Lorenzen Jin-Sheng He | 2017 | Journal of Plant Ecology2017,10,1: | 13 |

| 20 | First mass measurement of short-lived nuclides at HIRFL-CSR显示文摘Recent commissioning of the HIRFL-CSR has demonstrated its ability to perform direct mass measurement for short-lived nuclides. Projectile fragments produced by the 78Kr ions at 481.88 MeV/u were separated with the new radioactive beam line in Lanzhou (RIBLL2), and injected into and stored in the experimental storage ring (CSRe). By operating the CSRe as an isochronous mass spectrometry, a typical mass resolution around 2.0×10?5 has been achieved. The masses for 63Ge, 65As and 67Se were measured for the first time. The measured masses are compared with theoretical predictions and the location of the proton drip-line for As isotopes is discussed.
