1Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. 2Effects of dietary supplementation of probiotic,Clostridium butyricum,on growth performance,immune response,intestinal barrier function,and digestive enzyme activity in broiler chickens challenged with Escherichia coli K88显示文摘Background:Colibacillosis caused by enterotoxigenic Escherichia coli(E.coli) results in economic losses in the poultry industry.Antibiotics are usually used to control colibacillosis,however,E.coli has varying degrees of resistance to different antibiotics.Therefore the use of probiotics is becoming accepted as an alternative to antibiotics.In this study,we evaluated the effects of Clostridium butyricum(C.butyricum) on growth performance,immune response,intestinal barrier function,and digestive enzyme activity in broiler chickens challenged with Escherichia coli(E.coif) K88.Methods:The chickens were randomly divided into four treatment groups for 28 days.Negative control treatment(NC) consisted of birds fed a basal diet without E.coli K88 challenge and positive control treatment(PC) consisted of birds fed a basal diet and challenged with E.coli K88.C.butyricum probiotic treatment(CB) consisted of birds fed a diet containing 2×10~7 cfu C butyricum/kg of diet and challenged with E.coli K88.Colistin sulfate antibiotic treatment(CS)consisted of birds fed a diet containing 20 mg colistin sulfate/kg of diet and challenged with E.coli K88.Results:The body weight(BW) and average day gain(ADG) in the broilers of CB group were higher(P< 0.05) than the broilers in the PC group overall except the ADG in the 14-21 d post-challenge.The birds in CB treatment had higher(P < 0.05) concentration of tumor necrosis factor-α(TNF-α) at 3 and 7 d post-challenge,and higher(P < 0.05)concentration of interleukin-4(IL-4) at 14 d post-challenge than those in the PC treatment group.The concentration of serum endotoxin in CB birds was lower(P<0.05) at 21 d post-challenge,and the concentrations of serum diamine oxidase in CB birds were lower(P < 0.05) at 14 and 21 d post-challenge than in PC birds.Birds in CB treatment group had higher(P< 0.05) jejunum villi height than those in PC,NC,or CS treatment at 7,14,and 21 d post-challenge.In comparison to PC birds,the CB birds had lower(P < 0.05) jejunum crypt depth during the whole experiment.The birds in CB or CS treatment group had higher(P < 0.05) activities of amylase and protease at 3,7,and 14 d post-challenge,and higher(P< 0.05) activity of lipase at 3,7 d post-challenge than PC birds.Conclusions:In all,these results indicate that dietary supplementation with C.butyricum promotes immune response,improves intestinal barrier function,and digestive enzyme activities in broiler chickens challenged with E.coli K88.There is no significant difference between the C.butyricum probiotic treatment and the colistin sulfate antibiotic treatment.Therefore,the C.butyricum probiotic may be an alternative to antibiotic for broiler chickens.Ling Zhang Lingling Zhang Xiu'an Zhan Xinfu Zeng Lin Zhou Guangtian Cao An'guo Chen Caimei Yang 2016Journal of Animal Science and Biotechnology2016,7,1:43

5Grain-size features of a Miocene loess-soil sequence at Qinan: Implications on its origin显示文摘In this study, grain-size of 507 bulk samples from the QA-I Miocene loess-soil sequence at Qinan were analyzed, and the grain-size features are compared with those of typical Quaternary loess and soil samples, representative lacustrine and fluvial samples. The results indicate that the grain-size distribution pattern of the Miocene loess is essentially similar to that of Quaternary loess, but greatly differs from the lacustrine and fluvial sediments. Loess layers are regularly coarser than soil layers, indicating cyclical climate changes. Median grain-size along the 253.1 m sequence varies from 6 to 13 μm and the >63 μm fraction represents only 5.3% in maximum, 0.9% in average. Long-term grain-size variations are consistent with the loess accumulation rate at Qinan and with the eolian mass accumulation rate in the North Pacific. 7Genetic Variation in Rhizome Lotus(Nelumbo nucifera Gaertn.ssp.nucifera) Germplasms from China Assessed by RAPD Markers显示文摘To estimate genetic variation in rhizome lotus(Nelumbo nucifera Gaertn.ssp.nucifera) germplasms in China,a total of 94 rhizome lotus germplasms collected from 18 provinces in China were assessed.The RAPD(randomly amplified polymorphic DNA) marker was employed.The selected 17 random primers detected 139 polymorphic alleles out of a total 207(67.15%).Nei's gene diversity statistics and region differentiation parameters indicated that all germplasms had a relatively high level of genetic diversity with ne = 1.3202,h = 0.1937,I = 0.2982 and the gene flow among all regions was Nm = 5.5742.The UPGMA dendrogram clustered all 94 germplasms into two clusters:One contained eight commercial cultivars and major landraces,and the other included the wild and some special landraces from five regions,and the PCA analysis exhibited the similar result.Those germplasms from southwestern and eastern China had higher genetic diversity than those from the southern,northern and central China.Predominant proportion of genetic variation(95.61%) was found significant within rather than among(4.39%) regions,as revealed by AMOVA analysis.The data analysis also revealed that the genetic diversity of rhizome lotus germplasms among different regions is positively related to their geographic distances,though it is ambiguous to find the trend from the UPGMA dendrogram and the PCA analysis.A relatively high genetic diversity and gene flow resided in the root lotus germplasms;about 96% of the variation was found within region;accessions from southwest and eastern China have higher genetic diversity than those from the southern,northern and central China.AN Na GUO Hong-bo KE Wei-dong 2009Agricultural Sciences in China2009,8,1:27
8Compound Danshen injection improves endotoxin-induced microcirculatory disturbance in rat mesentery显示文摘AIM: To investigate the effect of compound Danshen injection on lipopolysaccharide (LPS)-induced rat mesenteric microcirculatory dysfunctions and the underlying possible mechanism by an inverted intravital microscope and high-speed video camera system. METHODS: LPS was continuously infused through the jugular artery of male Wistar rats at the dose of 2 mg/kg per hour. Changes in mesenteric microcirculation,such as diameters of arterioles and venules,velocity of RBCs in venules,leukocyte rolling,adhesion and emigration,free radicals released from post-capillary venules,FITC- albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope assisted with CCD camera and SIT camera. Meanwhile,the expression of adhesion molecules CD11b/CD18 and the production of free radical in neutrophils,and the expression of intercellular adhesion molecule 1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) were quantified by flow cytometry (FACS) in vitro. RESULTS: The continuous infusion with LPS resulted in a number of responses in microcirculation,including a significant increase in the positive region of venulestained with Monastral blue B,rolling and adhesion of leukocytes,production of oxygen radical in venular wall,albumin efflux and enhanced mast cell degranulation in vivo,all of which,except for the leukocyte rolling,were attenuated by the treatment with compound Danshen injection. Experiments performed in vitro further revealed that the expression of CD11b/CD18 and the production of oxygen free radical in neutrophils,and the expression of ICAM-1 in HUVECs were increased by exposure to LPS,and they were attenuated by compound Danshen injection. 12Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. 13Inhibition of 12-lipoxygenase reduces proliferation and induces apoptosis of hepatocellular carcinoma cells in vitro and in vivo显示文摘BACKGROUND:12-lipoxygenase(12-LOX) has been reported to be an important gene in cancer cell proliferation and survival,and tumor metastasis.However,its role in hepatocellular carcinoma(HCC) cells remains unknown.METHODS:Expression of 12-LOX was assessed in a diethylnitrosamine-induced rat HCC model,and in SMMC-7721,HepG2 and L-02 cells using immunohistochemical staining and reverse transcriptase-polymerase chain reaction(RT-PCR).GST-π and Ki-67 were determined in vivo by immunohistochemical staining.Apoptosis was evaluated by TUNEL assay.Cell viability and apoptosis were determined by MTT assay and flow cytometry,respectively.Apoptosis-related proteins in SMMC-7721 and HepG2 cells were detected by Western blotting.RESULTS:Immunohistochemical staining and RT-PCR showed that 12-LOX was over-expressed in rat HCC and two HCC cell lines,while the expression was inhibited by baicalein,a specific inhibitor of 12-LOX.Baicalein inhibited cell proliferation and induced apoptosis in rat HCC and both cell lines in a dose-and time-dependent manner.Our in vivo study demonstrated that baicalein also reduced neoplastic nodules.Mechanistically,baicalein reduced Bcl-2 protein expression coupled with a slight increase of the expression of Bax and activation of caspase-3.Furthermore,baicalein inhibited the activation of ERK-1/2(phosphorylated).Interestingly,the effects of baicalein were reversed by 12(S)-HETE,a metabolite of 12-LOX.CONCLUSIONS:Inhibition of 12-LOX leads to reduced numbers of HCC cells,partially caused by increased apoptosis.12-LOX may be a potential molecular target for HCC prevention and treatment.Xi-Ming Xu,Guang-Jin Yuan,Jun-Jian Deng,Hong-Ting Guo,Miao Xiang,Fang Yang,Wei Ge and Shi-You Chen Cancer Center, Renmin Hospital of Wuhan University, Wuhan 430060, China Cancer Center, the 82nd Hospital of the Chinese PLA, Huai’an 223001, China Department of Physiology, Medical College of Wuhan University, Wuhan 430071, China Department of Physiology & Pharmacology, University of Georgia, Athens, GA 30602, USA 2012Hepatobiliary & Pancreatic Diseases International2012,11,2:20
14Effect of L-NAME on nitric oxide and gastrointestinal motility alterations in cirrhotic rats显示文摘AIM: To invsstigare the effect of L-NAME on nitric oxide andgastriubtestubal motility alterations in cirrhotic ratsMETHODS: Rats with cirrhosis induced by carbontetrachloride were randomly divided into two groups, one( n= 13) receiving 0. 5 mg@ kg-1 per clay of NG-nitro-L-argininemethyl ester (L-NAME), a nitric oxide synthase inhibitor,for 10 days, whereas the other group ( n = 13) and control( n = 10) rats were administrated the same volume of 9 g@ L-1saline.Half gastric emptying time and 2 h residual rate weremeasured by SPECT, using 99m Tc-DTPA-labeled bariumsuifate as test meal. Gastrointestinal transition time wasrecorded simultaneously. Serum concentration of nitrcoxide (NO) was determined by the kinetic cadmiunreduction and colorimetric methods. ImmunohistochemicalSABC method was used to observe the expression anddistribution of three types of nitric oxide synthase (NOS)isoforms in the mt gastrointestinal tract. Western blot wasused to detect expression of gastrointestinal NOS isoforms.RESULTS: Half gastric emptying time and trans-gastrointestinal time were significantly prolonged( 124.0 ± 26.4min; 33.7± 8.9min;72.1 ± 15.3 min; P<0.01), (12.4±0.5h; 9.5±0.3 h; 8.2±0.8 h; P<0.01), 2h residual rate wasraised in cirrhotic rots than in controls and cirrhotic ratstreated with L-NAME(54.9± 7.6 % ,13.7 ± 3.2 %, 34.9± 10.3%, P< 0.01). Serum concentration of NO was significantlyincreased in cirrhotic rots than in the other groups (8.20 ± 2.48)μmol@L-1, (5.94± 1.07) μmol@L-1 ,and control (5.66± 1.60) tμmol@L-1, P< 0.01. NOS staining intensities which weremainly located in the gastrointestinal tissues were markedlylower in cirrhotic rats than in the controls and cirrhotic ratsafter treated with L- NAME.CONCLUSION: Gastrointestinal motility was remarkablyinhibited in cirrhotic rats, which could he alleviated by L-NAME. 16Relation among p130Cas,E-cadherin and β-catenin expression,clinicopathologic significance and prognosis in human hepatocellular carcinoma显示文摘BACKGROUND:p130Cas(p130Crk-associated substance) is a junction protein that is important to the adhesion between cytoskeleton and extracellular matrix.Also the adhesion molecules E-cadherin andβ-catenin play important roles in the invasiveness of carcinoma.This study was undertaken to investigate the effects of p130Cas E-cadherin andβ-catenin on the invasion,metastasis and prognosis of hepatocellular carcinoma(HCC). METHODS:Immunohistochemistry was used to evaluate the expression of p130Cas,E-cadherin,andβ-catenin in 40 patients with HCC.All patients were followed up postoperatively,and the relationship between expression and clinicopathological prognostic parameters was analyzed RESULTS:The positive expression rates of p130Cas and E-cadherin in HCC tissue(n=40)were 62.50%and 55.00% but in normal liver tissue 10%,and 100%,respectively (P<0.05).The abnormal expression rate ofβ-catenin in HCC tissue was 70%,while in normal liver tissue it was 13.33%(P<0.05).The positive rate of p130Cas was correlated with lymph node invasion,pathological stage TNM stage,and a worse prognosis,but not with gender age,HBV infection,hepatic cirrhosis,alpha-fetoprotein (AFP)level before operation,and tumor diameter Similarly,the expression of E-cadherin andβ-catenin was correlated with lymph node invasion,pathological stage,TNM stage,and worse prognosis,but not with gender,age,HBV infection,hepatic cirrhosis,AFP level before operation,and tumor size.Correlations were found between p130Cas and abnormal E-cadherin/β-catenin expression(P<0.001 and<0.05,respectively).CONCLUSIONS:In HCC,there is a negative correlation between the positive expression of p130Cas and the normal expression of the adhesion molecules E-cadherin/β-catenin, and p130Cas plays important roles in the invasion, metastasis and prognosis of HCC.p130Cas may be involved in alterating the structure and function of E-cadherin/ β-catenin,by regulating tyrosine phosphorylation via the p130Cas-Src signal pathway.Cheng Guo,Qing-Guang Liu,Wei Yang,Zheng-Liang Zhang and Ying-Min Yao Department of Hepatobiliary Surgery,First Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710061, China 2008Hepatobiliary & Pancreatic Diseases International2008,7,5:15
17Rapid report of the 8 January 2022 M_(S)6.9 Menyuan earthquake,Qinghai,China显示文摘The M_(S)6.9 Menyuan earthquake in Qinghai Province,west China is the largest earthquake by far in 2022.The earthquake occurs in a tectonically active region,with a background b-value of 0.87 within 100 km of the epicenter that we derived from the unified catalog produced by China Earthquake Networks Center since late 2008.Field surveys have revealed surface ruptures extending 22 km along strike,with a maximum ground displacement of 2.1 m.We construct a finite fault model with constraints from In SAR observations,which showed multiple fault segments during the Menyuan earthquake.The major slip asperity is confined within 10 km at depth,with the maximum slip of 3.5 m.Near real-time back-projection results of coseismic radiation indicate a northwest propagating rupture that lasted for~10 s.Intensity estimates from the back-projection results show up to a Mercalli scale of IX near the ruptured area,consistent with instrumental measurements and the observations from the field surveys.Aftershock locations(up to January 21,2022)exhibit two segments,extending to~20 km in depth.The largest one reaches M_(S)5.3,locating near the eastern end of the aftershock zone.Although the location and the approximate magnitude of the mainshock had been indicated by previous studies based on paleoearthquake records and seismic gap,as well as estimated stressing rate on faults,significant surfacebreaching rupture leads to severe damage of the high-speed railway system,which poses a challenge in accurately assessing earthquake hazards and risks,and thus demands further investigations of the rupture behaviors for crustal earthquakes.Hongfeng Yang Dun Wang Rumeng Guo Mengyu Xie Yang Zang Yue Wang Qiang Yao Chuang Cheng Yanru An Yingying Zhang 2022Earthquake Research Advances2022,2,1:14
18Growth induction of hepatic stimulator substance in hepatocytes through its regulation on EGF receptors显示文摘The cytosolic liver-specific growth factor-hepatic stimulator substance (HSS) has been shown to be able to amplify the rat hepatocyte proliferation responded to EGF. In order to get more insight into the mechanism, the regulatory effect of HSS on EGF-receptor(EGF-R) and the receptor phosphorylation at molecular level was studied. HSS partially purified from weanling rat liver was given to cultured hepatocytes and its influence on EGF-R specific binding and internalization as well as mRNA expression were investigated. The results showed that preincubation of hepatocytes with HSS could lead to an increase in [125I]-EGF binding to its receptors and inhibit EGFinduced receptor down-regulation. Furthermore, the overexpression of EGF-R mRNA stimulated by HSS was seen during 2-12 h after the incubation. Additionally, it was demonstrated with human hepatoma sMMC-7721 cells in Western blot that the EGF-R expression and the receptor autophosphorylation were increased with dose/timedependency after HSS treatment.
