| 1 | Geochemistry and regional distribution of ophiolites and associated volcanics in Mianlüe suture, Qinling-Dabie Mountains显示文摘Systematic studies of the ophiolites and associated volcanics stretching more than 1500 km from the Derni-Nanping-Pipasi-Kangxian area in the west to the Qingshuihe area of the south Dabie Mountains in the east indicate the existence of a suture zone (the Mianl黣 suture) and a vanished paleo-ocean basin (the Mianl黣 paleo-ocean basin) in the region. From west to east, ophiolitic mlange associations distribute discontinuously along the suture. Rock assem-blages include ophiolite, island arc and oceanic island rock series. The Mianl黣 paleo-ocean basin experienced its major formation and expending episode during the Carboniferous-Permian period. | 2 | Climate and litter C/N ratio constrain soil organic carbon accumulation显示文摘Soil organic carbon(SOC) plays critical roles in stabilizing atmospheric CO2 concentration, but the mechanistic controls on the amount and distribution of SOC on global scales are not well understood. In turn, this has hampered the ability to model global C budgets and to find measures to mitigate climate change. Here, based on the data from a large field survey campaign with 2600 plots across China’s forest ecosystems and a global collection of published data from forested land, we find that a low litter carbon-to-nitrogen ratio(C/N) and high wetness index(P/PET, precipitation-to-potentialevapotranspiration ratio) are the two factors that promote SOC accumulation, with only minor contributions of litter quantity and soil texture. The field survey data demonstrated that high plant diversity decreased litter C/N and thus indirectly promoted SOC accumulation by increasing the litter quality. We conclude that any changes in plant-community composition, plant-species richness and environmental factors that can reduce the litter C/N ratio, or climatic changes that increase wetness index, may promote SOC accumulation. | 3 | Comparisons between centrifuge and numerical modeling results for slope toppling failure显示文摘This paper presents series studies on the toppling mechanism by centrifuge tests and numerical simulations. Two different discrete element methods, i.e., the continuum-based discrete element method(CDEM) and the discontinuous deformation analysis(DDA), are adopted. The modeling results show that both the methods can accurately capture the failure modes of the centrifuge tests, including three distinct zones and two failure surfaces. Comparisons are made between the physical test and numerical simulation results. The critical inclination angle of the tilting table where the slope models are fixed on can be moderately predicted by the two methods, with different degrees of precision. The error between the test results and the simulated results is within 1% for the slope models without rock-bridges by both CDEM and DDA. However, it is amplified for the staggered-joint models that simulate the rock-bridges. With DDA, the average error is about 5%, and the maximum error is up to 17%. While with CDEM, the errors for the aligned-joint models are ranged from 1% to 6%, and it is from 10% to 29% for the staggered-joint models. The two numerical methods show the capability in simulating toppling failure of blocky rock mass with and without rock-bridges. The model with rock-bridges which provides a certain bending resistance is more stable than the one without any rock-bridge. | 4 | Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. | 5 | Multi-objective optimization of the streamlined head of high-speed trains based on the Kriging model显示文摘As the running speed of high-speed trains increases, aerodynamic drag becomes the key factor which limits the further increase of the running speed and energy consumption. Aerodynamic lift of the trailing car also becomes the key force which affects the amenity and safety of the train. In the present paper, a simplified CRH380A high-speed train with three carriages is chosen as the model in order to optimize aerodynamic drag of the total train and aerodynamic lift of the trailing car. A constrained mul- ti-objective optimization design of the aerodynamic head shape of high-speed trains based on adaptive non-dominated sorting genetic algorithm is also developed combining local function three-dimensional parametric approach and central Latin hypercube sampling method with maximin criteria based on the iterative local search algorithm. The results show that local function parametric approach can be well applied to optimal design of complex three-dimensional aerodynamic shape, and the adaptive non-dominated sorting genetic algorithm can be more accurate and efficient to find the Pareto front. After optimization the aerodynamic drag of the simplified train with three carriages is reduced by 3.2%, and the lift coefficient of the trailing car by 8.24%, the volume of the streamlined head by 2.16%; the aerodynamic drag of the real prototype CRH380A is reduced by 2.26%, lift coefficient of the trailing car by 19.67%. The variation of aerodynamic performance between the simplified train and the true train is mainly concentrated in the deformation region of the nose cone and tail cone. | 6 | Underwater explosion in centrifuge partⅠ:Validation and calibration of scaling laws显示文摘Centrifuge is a promising tool for underwater explosion(UNDEX) research as both Mach and Froude similitudes could be satisfied with hyper-gravity in models, which would result in similarities in both shock wave and bubble oscillation. Scaling laws for UNDEX in centrifuge have been proposed based on dimensional analysis. Two dimensionless numbers, i.e.,π_3 and π_4, are used to characterize shock wave and bubble oscillation, respectively. To validate scaling laws, 17 UNDEX tests are designed by varying accelerations or explosive weights and positions in centrifuge. The tests are classified into different groups to validate scaling laws as well as calibrate coefficients in empirical formulae for both shock wave and bubble oscillation. The results show that changes of gravity acceleration or hydrodynamic pressure almost has no influence on shock wave peak pressure and time constant as long as π3 is constant. The dimensionless bubble period and maximum radius agreed with each other when π4 is constant. | 7 | Metal Sealing Performance of Subsea X-tree Wellhead Connector Sealer显示文摘The metal sealing performance of subsea X-tree wellhead connectors is crucial for the safety and reliability of subsea X-trees. In order to establish the theoretical relation between metal sealing ring's contact stress and its structural parameters and working pressure, a mechanical analysis method for double-cone sealing of high pressure vessels is applied in analyzing the metal sealing ring under the condition of preload and operation. As a result, the formula of the unit sealing load for the metal sealing ring under operation with residual preload is shown in this paper, which ensures that the metal sealing ring has an excellent sealing effect and can prevent the metal sealing ring from yielding. Besides, while analyzing the sealing process of the metal sealing ring, the change rule of contact stress and working pressure is concluded here, putting forward that the structural parameters of the metal sealing ring are the major factors affecting the change rule. Finally, the analytical solution through theoretical analysis is compared with the simulation result through finite element analysis in a force feedback experiment, and both are consistent with each other, which fully verifies for the design and calculation theory on metal sealing ring's contact stress and its structural parameters and working pressure deduced in this paper. | 8 | The constraints of strain partitioning and geochronology in Luonan-Luanchuan tectonic belts on Qinling orogenic belt显示文摘The Luonan-Luanchuan tectonic belt lies between the North China Block and Qinling Mountains, in- cluding the Luonan-Luanchuan fault zone and the strong deformation zone to the north of the fault. The ductile shear zone, imbricate brittle fault and duplex structure in the fault zone now are the expression of the same tectonic event in different depth. Such lineation structure exists in the tectonic belts as mineral lineation, elongation lineation, crenulation lineation, sheath folds and so on, indicating NE-directed plate motion. Fold axes and thrusts in the strong deformation zone are inclined to the Luonan-Luanchuan fault zone at small angles. The structures with different natures show a regular pattern, produced during oblique convergence of plates. The convergence factors are as follows:The direction of plate convergence is 22°, 31° and the angle between the plate convergence direction and plate boundary is 73°, 82° respectively in the west and east segment. | 9 | A more consistent intraluminal rhesus monkey model of ischemic stroke显示文摘Endovascular surgery is advantageous in experimentally induced ischemic stroke because it causes fewer cranial traumatic lesions than invasive surgery and can closely mimic the pathophysiology in stroke patients. However, the outcomes are highly variable, which limits the accuracy of evaluations of ischemic stroke studies. In this study, eight healthy adult rhesus monkeys were randomized into two groups with four monkeys in each group: middle cerebral artery occlusion at origin segment(M1) and middle cerebral artery occlusion at M2 segment. The blood flow in the middle cerebral artery was blocked completely for 2 hours using the endovascular microcoil placement technique(1 mm × 10 cm)(undetachable), to establish a model of cerebral ischemia. The microcoil was withdrawn and the middle cerebral artery blood flow was restored. A reversible middle cerebral artery occlusion model was identified by hematoxylin-eosin staining, digital subtraction angiography, magnetic resonance angiography, magnetic resonance imaging, and neurological evaluation. The results showed that the middle cerebral artery occlusion model was successfully established in eight adult healthy rhesus monkeys, and ischemic lesions were apparent in the brain tissue of rhesus monkeys at 24 hours after occlusion. The rhesus monkeys had symptoms of neurological deficits. Compared with the M1 occlusion group, the M2 occlusion group had lower infarction volume and higher neurological scores. These experimental findings indicate that reversible middle cerebral artery occlusion can be produced with the endovascular microcoil technique in rhesus monkeys. | 13 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |

| 15 | Effects of precipitation on soil organic carbon fractions in three subtropical forests in southern China显示文摘Aims The aim of this study was to investigate the effects of precipitation changes on soil organic carbon(SOC)fractions in subtropical forests where the precipitation pattern has been altered for decades.Methods We conducted field manipulations of precipitation,including ambient precipitation as a control(CK),double precipitation(DP)and no precipitation(NP),for 3 years in three forests with different stand ages(broadleaf forest[BF],mixed forest[MF]and pine forest[PF])in subtropical China.At the end of the experiment,soil samples were collected to assay SOC content,readily oxidizable organic carbon(ROC)and non-readily oxidizable organic carbon(NROC),as well as soil microbial biomass carbon(MBC),pH and total nitrogen content.Samples from the forest floors were also collected to analyze carbon(C)functional groups(i.e.alkyl C,aromatic C,O-alkyl C and carbonyl C).Furthermore,fine root biomass was measured periodically throughout the experiment.Important Findings Among the forests,ROC content did not exhibit any notable differences,while NROC content increased significantly with the stand age.This finding implied that the SOC accumulation observed in these forests resulted from the accumulation of NROC in the soil,a mechanism for SOC accumulation in the mature forests of southern China.Moreover,NP treatment led to significant reductions in both ROC and NROC content and therefore reduced the total SOC content in all of the studied forests.Such decreases may be due to the lower plant-derived C inputs(C quantity)and to the changes in SOC components(C quality)indicated by C functional groups analyses under NP treatment.DP treatment in all the forests also tended to decrease the SOC content,although the decreases were not statistically significant with the exception of SOC and ROC content in PF.This finding indicated that soils in MF and in BF may be more resistant to precipitation increases,possibly due to less water limitations under natural conditions in the two forests.Our results therefore highlight the different responses of SOC and its fractions to precipitation changes among the forests and suggest that further studies are needed to improve our understanding of SOC dynamics in such an important C sink region. | Xiaomei Chen Deqiang Zhang Guohua Liang Qingyan Qiu Juxiu Liu Guoyi Zhou Shizhong Liu Guowei Chu Junhua Yan | 2016 | Journal of Plant Ecology2016,9,1: | 4 |
| 16 | Effects of Cs-substitution and partial reduction on catalytic performance of Keggin-type phosphomolybdic polyoxometalates for selective oxidation of isobutane显示文摘The catalytic performance of Cs-substituted phosphomolybdic salts was studied for selective oxidation of isobutane. The results of activity tests revealed that 360 °C was the optimal reaction temperature. It was demonstrated that oxidizing sites not only took dominating part in the activation of isobutane, but also influenced the product distribution. Besides, appropriate Cs addition led to moderate acidity of catalysts, favoring the selectivity to desired products. Furthermore, to obtain partially reduced catalysts, different calcination atmospheres were investigated and certain proportion of Mo^(5+) produced during calcination was crucial for the redox reaction. The catalyst calcined in N_2 showed the highest yield of MAA(7.0%). | 17 | Human embryonic stem cells-derived endothelial cell therapy facilitates kidney regeneration by stimulating renal resident stem cell proliferation in acute kidney injury显示文摘Endothelial cell therapy has been implicated to enhance tissue regeneration and vascularization in ischemic kidney. However, no published study has yet examined direct effects of endothelial cell treatment in kidney recovery. This study investigated the therapeutic efficacy of endothelial cells in a mouse model with acute kidney injury (AKI). Thus, human embryonic stem cells-derived endothelial cells (hESC-ECs) labeled with a reporter system encoding a double fusion reporter gene for firefly luciferase (Fluc) and green fluorescent protein (GFP) were characterized by Fluc imaging and immunofluoresence staining. Cultured hESC-ECs (1×106) were injected into ischemic kidney shortly after AKI. Survival of the transplanted hESC-ECs was monitored in vivo from day 1 to 14 after endothelial cell transplantation and potential impact of hESC-EC treatment on renal regeneration was assessed by histological analyses. We report that a substantial level of bioluminescence activity was detected 24 h after hESC-EC injection followed by a gradual decline from 1 to 14 d. Human ESC-ECs markedly accelerated kidney cell proliferation in response to ischaemia-induced damage, indicated by an elevated number of BrdU+ cells. Co-expression of Sca-1, a kidney stem cell proliferation marker, and BrdU further suggested that the observed stimulation in renal cell regeneration was, at least in part, due to increased proliferation of renal resident stem cells especially within the medullary cords and arteriole. Differentiation of hESC-ECs to smooth muscle cells was also observed at an early stage of kidney recovery. The results show that the bulge and the interdendritic spacing are responsible for the flow speed of interdendritic melt. At the initial stage of solidification the bulge operates, and at the final stage the interdendritic spacing operates.
