Routing valley exciton emission of a WS_(2) monolayer via delocalized Bloch modes of in-plane inversion-symmetry-broken photonic crystal slabs显示文摘The valleys of two-dimensional transition metal dichalcogenides(TMDCs)offer a new degree of freedom for information processing. To take advantage of this valley degree of freedom,on the one hand,it is feasible to control valleys by utilizing different external stimuli, such as optical and electric fields. On the other hand, nanostructures are also used to separate the valleys by near-field coupling. However, for both of the above methods, either the required low-temperature environment or low degree of coherence properties limit their further applications. Here,we demonstrate that all-dielectric photonic crystal(PhC)slabs without in-plane inversion symmetry(C_(2) symmetry)can separate and route valley exciton emission of a WS2 monolayer at room temperature.Coupling with circularly polarized photonic Bloch modes of such PhC slabs, valley photons emitted by a WS_(2) monolayer are routed directionally and are efficiently separated in the far field. In addition,far-field emissions are directionally enhanced and have long-distance spatial coherence properties.Jiajun Wang Han Li Yating Ma Maoxiong Zhao Wenzhe Liu Bo Wang Shiwei Wu Xiaohan Liu Lei Shi Tian Jiang Jian Zi 2020Light(Science & Applications)2020,9,1:4
Special interstitial route can transport nanoparticles to the brain bypassing the blood-brain barrier显示文摘Nowadays, nanoparticles(NPs)are considered to be ideal tools for bioimaging and drug delivery.Although increasing research has focused on NP biodistribution, transportation in the interstitial architecture has been neglected. The entire body is connected by the interstitial architecture, which can provide a long-range and direct pathway for NP biodistribution in a nonvascular system. In this study,we report that 10-nm gold NPs injected directly into the interstitial architecture of the tarsal tunnel of rats(intervaginal space injection(ISI))were delivered to the brain without crossing the blood-brain barrier.Furthermore,NaGdF4 nanoparticles were used to explore the transportation route by magnetic resonance imaging. The results demonstrated that, after ISI,the NaGdF4 nanoparticles were transported through the perivascular interstitial space of the carotid arteries and brain vessels to the brain. This is a special nonvascular transportation route like a stream based on the interstitial architecture that provides an alternative pathway for NP biodistribution.Nan Hu Xiaoli Shi Qiang Zhang Wentao Liu Yuting Zhu Yuqing Wang Yi Hou Yinglu Ji Yupeng Cao Qian Zeng Zhuo Ao Quanmei Sun Xiaohan Zhou Xiaochun Wu Dong Han 2019Nano Research2019,12,11:2

Over 50,000 Metagenomically Assembled Draft Genomes for the Human Oral Microbiome Reveal New Taxa显示文摘The oral cavity of each person is home to hundreds of bacterial species. While taxa for oral diseases have been studied using culture-based characterization as well as amplicon sequencing, metagenomic and genomic information remains scarce compared to the fecal microbiome. Here, using metagenomic shotgun data for 3346 oral metagenomic samples together with 808 published samples,we obtain 56,213 metagenome-assembled genomes(MAGs),and more than 64%of the 3589 species-level genome bins(SGBs)contain no publicly available genomes. The resulting genome collection is representative of samples around the world and contains many genomes from candidate phyla radiation(CPR)that lack monoculture. Also,it enables the discovery of new taxa such as a genus Candidatus Bgiplasma within the family Acholeplasmataceae.Large-scale metagenomic data from massive samples also allow the assembly of strains from important oral taxa such as Porphyromonas and Neisseria. The oral microbes encode genes that could potentially metabolize drugs.Apart from these findings,a strongly male-enriched Campylobacter species was identified.Oral samples would be more user-friendly collected than fecal samples and have the potential for disease diagnosis. Thus, these data lay down a genomic framework for future inquiries of the human oral microbiome.Jie Zhu Liu Tian Peishan Chen Mo Han Liju Song Xin Tong Xiaohuan Sun Fangming Yang Zhipeng Lin Xing Liu Chuan Liu Xiaohan Wang Yuxiang Lin Kaiye Cai Yong Hou Xun Xu Huanming Yang Jian Wang Karsten Kristiansen Liang Xiao Tao Zhang Huijue Jia Zhuye Jie 2022Genomics, Proteomics & Bioinformatics2022,20,2:1
Xenopus GLP-1-based glycopeptides as dual glucagon-like peptide 1 receptor/glucagon receptor agonists with improved in vivo stability for treating diabetes and obesity显示文摘Peptide dual agonists toward both glucagon-like peptide 1 receptor(GLP-1R)and glucagon receptor(GCGR)are emerging as novel therapeutics for the treatment of type 2 diabetes mellitus(T2DM)patients with obesity. Our previous work identified a Xenopus GLP-1-based dual GLP-1R/GCGR agonist termed xGLP/GCG-13,which showed decent hypoglycemic and body weight lowering activity. However, the clinical utility of xGLP/GCG-13 is limited due to its short in vivo half-life.Inspired by the fact that O-GlcNAcylation of intracellular proteins leads to increased stability of secreted proteins,we rationally designed a panel of O-GlcNAcylated xGLP/GCG-13 analogs as potential long-acting GLP-1R/GCGR dual agonists. One of the synthesized glycopeptides 1f was found to be equipotent to xGLP/GCG-13 in cell-based receptor activation assays. As expected,O-GlcNAcylation effectively improved the stability of xGLP/GCG-13 in vivo.Importantly, chronic administration of 1f potently induced body weight loss and hypoglycemic effects, improved glucose tolerance, and normalized lipid metabolism and adiposity in both db/db and diet induced obesity(DIO)mice models. These results supported the hypothesis that glycosylation is a useful strategy for improving the in vivo stability of GLP-1-based peptides and promoted the development of dual GLP-1R/GCGR agonists as antidiabetic/antiobesity drugs.LI Qiang YANG Qimeng HAN Jing LIU Xiaohan FU Junjie YIN Jian 2022Chinese Journal of Natural Medicines2022,20,11:1
Mechanical behaviours of sandstone containing intersecting fissures under uniaxial compression显示文摘Predicting rock cracking is important for assessing the stability of underground engineering. The effects of the intersecting angle a and the distribution orientation angleβof intersecting fissures on the uniaxial compressive strength and the failure characteristics of sandstone containing intersecting fissures are investigated through laboratory experiments and two-dimensional particle flow code(PFC2D).The relationship between the mechanical properties of sandstone and the intersecting angle a and the distribution orientation angleβis analysed.Crack initiation forms and the final failure modes are then categorised and determined via empirical methods. In addition, the cracking processes of intersecting fissures with different a andβvalues are discussed. The results show that variations in the peak stress, peak strain, average modulus, and crack initiation stress of sandstone containing intersecting fissures show a“moth”shape in the space of the a-β-mechanical parameters. Two crack initiation forms are identified:inner tip cracking(usually accompanied by one outer tip cracking)and only outer tips cracking. Two failure modes are observed:(1)the main fracture planes are created at the inner tip and one outer tip,and(2)the main fracture planes are formed at the two outer tips. Two main crack evolution processes of sandstone containing intersecting fissures under uniaxial compression are found.Approaches for quickly determining the crack initiation form and the failure mode are proposed. The combination of the determination equations for the crack initiation form and the failure mode can be used to predict the crack evolution. The approach for determining the crack evolution processes is hence proposed with acceptable precision.Fei Xiong Xinrong Liu Xiaohan Zhou Guangyi Lin Dongshuang Liu Yafeng Han Bin Xu Chunmei He Zijuan Wang 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,2:1

Milk fat globule membrane supplementation protects againstβ-lactoglobul-ininduced food allergy in mice via upregulation of regulatory T cells and enhancement of intestinal barrier in a microbiota-derived short-chain fatty acids manner显示文摘Milk fat globule membrane(MFGM),which contains abundant glycoproteins and phospholipids, exerts beneficial effects on intestinal health and immunomodulation. The aim of this study was to evaluate the protective effects and possible underlying mechanisms of MFGM on cow’s milk allergy(CMA)in aβ-lactoglobulin(BLG)-induced allergic mice model.MFGM was supplemented to allergic mice induced by BLG at a dose of 400 mg/kg body weight. Results demonstrated that MFGM alleviated food allergy symptoms, decreased serum levels of lipopolysaccharide,pro-inflammatory cytokines, immunoglobulin(Ig)E,Ig G1,and Th2 cytokines including interleukin(IL)-4,while increased serum levels of Th1 cytokines including interferon-γand regulatory T cells(Tregs)cytokines including IL-10 and transforming growth factor-β.MFGM modulated gut microbiota and enhanced intestinal barrier of BLG-allergic mice,as evidenced by decreased relative abundance of Desulfobacterota,Rikenellaceae,Lachnospiraceae, and Desulfovibrionaceae, while increased relative abundance of Bacteroidetes,Lactobacillaceae and Muribaculaceae, and enhanced expressions of tight junction proteins including Occludin,Claudin-1 and zonula occludens-1.Furthermore,MFGM increased fecal short-chain fatty acids(SCFAs)levels, which elevated G protein-coupled receptor(GPR)43 and GPR109A expressions. The increased expressions of GPR43 and GPR109A induced CD103+dendritic cells accumulation and promoted Tregs differentiation in mesenteric lymph node to a certain extent. In summary,MFGM alleviated CMA in a BLG-induced allergic mice model through enhancing intestinal barrier and promoting Tregs differentiation, which may be correlated with SCFAs-mediated activation of GPRs.These findings suggest that MFGM may be useful as a promising functional ingredient against CMA.Han Gong Tiange Li Dong Liang Jingxin Gao Xiaohan Liu Xueying Mao 2024Food Science and Human Wellness2024,13,1:1
肝脏移植术后糖尿病患者肠道微生物组的变化显示文摘Post-transplant diabetes mellitus(PTDM)increases the risk of graft failure and death in liver transplant(LT)recipients.Experimental studies have indicated that enteric dysbiosis mediated by immunosuppressive tacrolimus(TAC)could contribute to glucose disorders, but no data on human recipients with PTDM have been reported. Here,by combining high-throughput shotgun metagenomics sequencing and metabolomics profiling,we characterized the intestinal microbiome(IM)in LT recipient cohort with or without PTDM and deciphered the potential relationship among IM,TAC dosage, and diabetes.By comparing with both non-PTDM and classical type 2 diabetes mellitus(T2DM),we identified microbial signatures of PTDM,which was characterized by the enriched Proteobacteria and decreased Bacteroidetes.Additionally, the altered microbes,as well as the microbial metabolomics, correlated with the dosage of TAC.Specifically, the levels of beneficial microbes associated with PTDM were lowered in recipients with the high TAC trough concentrations(>5 ng·mL^(-1))than those with low ones(<5 ng·mL^(-1)),which was accompanied by reduced faecal metabolites involved in the biosynthesis of a-linolenic acid and arachidonic acid-lowering factors of developing T2DM.Moreover,these microbial signatures linked with the extent of glucose disorders in LT recipients. In summary, the faecal microbiome and metabolome differed between PTDM and non-PTDM patients, which were linked with TAC dosage. This study was the first to explore taxonomic alterations and bacterial gene functions to better understand the contribution of the IM to PTDM.Qi Ling Yuqiu Han Yue Ma Xiaosen Wang Zheng Zhu Jingyu Wang Jiaying Cao Lin Xiaohan Jun Wang Baohong Wang 2023Engineering2023,31,12:0
Structure and immunomodulatory activity of Lentinus edodes polysaccharides modified by probiotic fermentation显示文摘Plant-based fermentations provide an untapped source for novel biotechnological applications. In this study,a probiotic named Lactobacillus fermentum 21828 was introduced to ferment Lentinus edodes.Polysaccharides were extracted from fermented and non-fermented L.edodes and purified via DEAE-52 and Sephadex G-100.The components designated F-LEP-2a and NF-LEP-2a were analyzed by FT-IR,HPGPC,HPAEC,SEM,GC-MS and NMR.The results revealed that probiotic fermentation increased the molecular weight from 1.16×10^(4) Da to 1.87×10^(4) Da and altered the proportions of glucose, galactose and mannose,in which glucose increased from 45.94%to 48.16%.Methylation analysis and NMR spectra indicated that F-LEP-2a and NF-LEP-2a had similar linkage patterns.Furthermore, their immunomodulatory activities were evaluated with immunosuppressive mice.NF-LEP and F-LEP improved immune organ indices, immunoglobulin(Ig G and Ig M)and cytokines concentrations;restored the antioxidation capacity of liver;and maintained the balance of gut microbiota.F-LEP displayed better moderating effects on the spleen index, immunoglobulin, cytokines and the diversity of gut microbiota than NF-LEP(200,400 mg/kg).Our study provides an efficient and environment-friendly way for the structural modification of polysaccharides, which helps to enhance their biological activity and promote their wide application in food, medicine and other fields.Jingjing Liang Meina Zhang Xiaohan Li Yuan Yue Xiaowei Wang Mengzhen Han Tianli Yue Zhouli Wang Zhenpeng Gao 2024Food Science and Human Wellness2024,13,1:0
Tailoring spatiotemporal dynamics of plasmonic vortices显示文摘Plasmonic vortices confining orbital angular momentums to surface have aroused wide research interest in the last decade.Recent advances of near-field microscopes have enabled the study on the spatiotemporal dynamics of plasmonic vortices, providing a better understanding of optical orbital angular momentums in the evanescent wave regime. However, these works only focused on the objective characterization of plasmonic vortex and have not achieved subjectively tailoring of its spatiotemporal dynamics for specific applications.Herein,it is demonstrated that the plasmonic vortices with the same topological charge can be endowed with distinct spatiotemporal dynamics by simply changing the coupler design.Based on a near-field scanning terahertz microscopy, the surface plasmon fields are directly obtained with ultrahigh spatiotemporal resolution, experimentally exhibiting the generation and evolution divergences during the whole lifetime of plasmonic vortices. The proposed strategy is straightforward and universal, which can be readily applied into visible or infrared frequencies, facilitating the development of plasmonic vortex related researches and applications.Xinyao Yuan Quan Xu Yuanhao Lang Xiaohan Jiang Yuehong Xu Xieyu Chen Jie Han Xueqian Zhang Jiaguang Han Weili Zhang 2023Opto-Electronic Advances2023,6,4:0
Diel dynamics of multi-omics in elkhorn fern provide new insights into weak CAM photosynthesis显示文摘Crassulacean acid metabolism(CAM)has high water-use efficiency(WUE)and is widely recognized to have evolved from C3 photosynthesis.Different plant lineages have convergently evolved CAM,but the molecular mechanism that underlies C3-to-CAM evolution remains to be clarified.Platycerium bifurcatum(elkhorn fern)provides an opportunity to study the molecular changes underlying the transition from C3 to CAM photosynthesis because both modes of photosynthesis occur in this species, with sporotrophophyll leaves(SLs)and cover leaves(CLs)performing C3 and weak CAM photosynthesis, respectively. Here,we report that the physiological and biochemical attributes of CAM in weak CAM-performing CLs differed from those in strong CAM species. We investigated the diel dynamics of the metabolome, proteome, and transcriptome in these dimorphic leaves within the same genetic background and under identical environmental conditions. We found that multi-omic diel dynamics in P.bifurcatum exhibit both tissue and diel effects. Our analysis revealed temporal rewiring of biochemistry relevant to the energy-producing pathway(TCA cycle),CAM pathway, and stomatal movement in CLs compared with SLs.We also confirmed that PHOSPHOENOLPYRUVATE CARBOXYLASE KINASE(PPCK)exhibits convergence in gene expression among highly divergent CAM lineages.Gene regulatory network analysis identified candidate transcription factors regulating the CAM pathway and stomatal movement.Taken together, our results provide new insights into weak CAM photosynthesis and new avenues for CAM bioengineering.Cheng Li Wenjie Huang Xiaoxu Han Guohua Zhao Wenyang Zhang Weijun He Bao Nie Xufeng Chen Taijie Zhang Wenhui Bai Xiaopeng Zhang Jingjing He Cheng Zhao Alisdair RFernie Timothy JTschaplinski Xiaohan Yang Shijuan Yan Li Wang 2023Plant Communications2023,4,5:0
Preoperative Prediction of Lymph Node Metastasis in Colorectal Cancer with Deep Learning显示文摘Objective. To develop an artificial intelligence method predicting lymph node metastasis(LNM)for patients with colorectal cancer(CRC).Impact Statement.A novel interpretable multimodal AI-based method to predict LNM for CRC patients by integrating information of pathological images and serum tumor-specific biomarkers.Introduction.Preoperative diagnosis of LNM is essential in treatment planning for CRC patients.Existing radiology imaging and genomic tests approaches are either unreliable or too costly.Methods.A total of 1338 patients were recruited, where 1128 patients from one centre were included as the discovery cohort and 210 patients from other two centres were involved as the external validation cohort. We developed a Multimodal Multiple Instance Learning(MMIL)model to learn latent features from pathological images and then jointly integrated the clinical biomarker features for predicting LNM status. The heatmaps of the obtained MMIL model were generated for model interpretation.Results.The MMIL model outperformed preoperative radiology-imaging diagnosis and yielded high area under the curve(AUCs)of 0.926,0.878,0.809,and 0.857 for patients with stage T1,T2,T3,and T4 CRC,on the discovery cohort. On the external cohort,it obtained AUCs of 0.855,0.832,0.691,and 0.792,respectively(T1-T4),which indicates its prediction accuracy and potential adaptability among multiple centres.Conclusion.The MMIL model showed the potential in the early diagnosis of LNM by referring to pathological images and tumor-specific biomarkers, which is easily accessed in different institutes. We revealed the histomorphologic features determining the LNM prediction indicating the model ability to learn informative latent features.Hailing Liu Yu Zhao Fan Yang Xiaoying Lou Feng Wu Hang Li Xiaohan Xing Tingying Peng Bjoern Menze Junzhou Huang Shujun Zhang Anjia Han Jianhua Yao Xinjuan Fan 2022Biomedical Engineering Frontiers2022,3,1:0
Thoracic interstitial injection of drug-liposomes in mice for treating atherosclerosis显示文摘Intervaginal space injection(ISI)is a novel mode of administration investigated over the last decade. After injecting nanoparticles into the intervaginal space, they can be transported along low flow resistance channels into the interstitial space. This transport has a certain delivery direction, and site-specific injection can work on specific organs or tissues. In this study, the thorax,a new ISI site in the interstitial surrounding the internal thoracic artery named the thoracic interstitial injection(tISI)was investigated. To prove the targeting ability of the tISI,two sizes of gold nanoparticles(AuNPs)(47 and 87 nm)were administered to mice. After 1 h,the biodistribution of AuNPs in the tissues was measured via single particle inductively coupled plasma mass spectrometry(spICP-MS).The results showed that the concentration of AuNPs in the aorta after tISI injection was significantly higher than that after intravenous injection. Moreover, fewer nanoparticles with larger particle sizes were observed to have entered the blood and were better targeted to the aorta.Thereafter, tanshinone IIa sodium sulfonate liposomes were administered for the treatment of aortic atherosclerosis. The proportion of aortic plaques in atherosclerotic Apoe-/-mice administered via tISI was significantly lower than that in other model animals(P<0.001).Furthermore,the proteoglycan content and CD68-positive cell count in the plaques were significantly reduced. The vascular elastic fibers at the plaque site were thickened, and fractures were reduced.tISI was, therefore, determined to be an effective strategy for the treatment of atherosclerotic aortic plaques.Tun Yan Huizhen Sun Yahong Shi Ya Gao Xi Lu Kai Li Yuting Zhu Qiang Zhang Tingting Li Zhongxian Li Xiaohan Zhou Yuting Guo Yinglu Ji Xiaoli Shi Dong Han 2023Nano Research2023,16,4:0
Saturation efficiency for detecting 1550 nm photons with a 2×2 array of Mo0.8Si0.2 nanowires at 2.2 K显示文摘Amorphous materials are attractive candidates for fabricating the superconducting nanowire single-photon detectors(SNSPDs) due to their superior tolerance and scalability over crystalline niobium nitride. However, the reduced superconducting transition temperature degenerates both operating temperature and saturation efficiency. Herein, the SNSPD(6.5 nm thickness and 50 nm width) based on the amorphous Mo0.8Si0.2 film with a high optical absorption coefficient demonstrates close-to-unity intrinsic detection efficiency for 1550 nm photons from 75 m K to 2.2 K. Further, a high-performance array SNSPD with optimized 90 nm-width wires is also demonstrated. As-fabricated uniform 4-pixel SNSPD exhibits a saturation plateau for the photon counts at 2.2 K,which overcomes the limitation of operation at low temperature(< 1 K) for traditional amorphous SNSPDs.Coupled with superior intrinsic quantum efficiency, highly efficient photon counts, and low dark count ratio, this detector paves a way for achieving high efficiency and superior yield for large array systems.Feiyan Li Hang Han Qi Chen Biao Zhang Han Bao Yue Dai Rui Ge Shuya Guo Guanglong He Yue Fei Shuchao Yang Xiaohan Wang Hao Wang Xiaoqing Jia Qingyuan Zhao Labao Zhang Lin Kang Peiheng Wu 2021Photonics Research2021,9,3:0
Survival outcomes of conversion surgery for metastatic pancreatic ductal adenocarcinoma after neoadjuvant therapy显示文摘Objective:To evaluate the survival outcomes of patients who underwent conversion surgery for metastatic pancreatic ductal adenocarcinoma(mPDAC)after neoadjuvant therapy(NAT)and to identify potential candidates that may benefit from this treat-ment strategy. Background:The role and eligibility population of conversion surgery for mPDAC remains controversial in the era of NAT.Methods:A consecutive cohort of patients diagnosed with mPDAC and treated with NAT followed by conversion surgery be-tween 2019 and 2021 were confirmed from a prospective database maintained by the Department of Pancreatic Hepatobiliary Surgery of Changhai Hospital. In accordance with residual metastases and technical resectability after NAT,patients were classi-fied as the complete pathological response of metastases(ypM0)resection group, residual metastases(ypM1)resection group, and exploration group.Median overall survival(mOS)was calculated using the Kaplan-Meier method,uni-and multivariable cox regression was performed to identify clinicopathological predictors of OS.Results:A total of 244 patients with mPDAC were identified from the prospective database, with 19(7.8%)patients who un-derwent ypM0 resection,22(9.0%)underwent ypM1 resection, and 23(9.4%)underwent explorative laparotomy. The mOS was 32.6 months for ypM0 resected patients,15.1 months for ypM1 resected patients, and 13.4 months for those who underwent explorative laparotomy(P<.001).Univariable and multivariable Cox regression analyses confirmed that ypM0 resection,normal-ization of preoperative CA19-9 levels, and continued adjuvant therapy were independent prognostic factors of conversion surgery for mPDAC after NAT.Subgroup analyses revealed that oligometastases and continued adjuvant therapy were associated with improved prognosis in the ypM1 resection group. Conclusion:In patients with mPDAC who underwent NAT followed by conversion surgery, the complete pathological response of metastases, normalization of preoperative CA19-9 levels, and continued adjuvant therapy were independent risk factors for prognosis.Patients with residual oligometastases after treatment were expected to prolong survival through resection. These patients may benefit from conversion surgery and should be potential candidates for this treatment strategy.Lingyu Zhu Suizhi Gao Xinqian Wu Bo Li Xiaohan Shi Xiaoyi Yin Huan Wang Meilong Shi Penghao Li Yikai Li Chaoliang Zhong Chuanqi Teng Jiawei Han Yiwei Ren Jian Wang Zhendong Fua Xinyu Liu Kailian Zheng Shiwei Guo Gang Jin 2023Journal of Pancreatology2023,6,3:0
