1Salvianolic acid A alleviates renal injury in systemic lupus erythematosus induced by pristane in BALB/c mice显示文摘The purpose of this study was to investigate the effects of salvianolic acid A(SAA) in systemic lupus erythematosus(SLE) induced by pristane in BALB/c mice.Lupus mice were established by confirming elevated levels of autoantibodies and IL-6 after intraperitoneal injection of pristane.Mice were then treated with daily oral doses of SAA for 5 months in parallel with mice treated with prednisone and aspirin as positive controls.The levels of autoantibodies were monitored at monthly intervals and nephritic symptoms observed by hematoxylin and eosin(H&E) and periodic acid-Schiff(PAS) staining.Western blot analysis of renal tissue was also employed.SAA treatment caused a significant reduction in the levels of anti-Sm autoantibodies and reduced renal histopathological changes and pathological effects.SAA treatment also significantly inhibited the phosphorylation of IKK,IκB and NFκB in renal tissues of lupus mice.In conclusion,the results suggest that SAA alleviates renal injury in pristane-induced SLE in BALB/c mice through inhibition of phosphorylation of IKK,IκB and NFκB.Yihuang Lin Yu Yan Huifang Zhang Yucai Chen Yangyang He Shoubao Wang Lianhua Fang Yang Lv Guanhua Du 2017Acta Pharmaceutica Sinica B2017,7,2:8
2Preparation of PE/MMT nanocomposite by monomer intercalation and in situ copolymerization显示文摘The catalyst {[(2-ArN=C(Me))2C5H3N]FeCl3} (Ar = 2-C6H4(i-Pr)) with methylaluminoxane (MAO) as co-catalyst was intercalated between layers of montmorillonite (MMT) for ethylene oligomerization. Metallocene catalyst Me2Si(Ind)2ZrCl2 and MAO was then added to form a dual functional catalytic system. A PE/MMT nanocomposite was prepared by copolymerization of ethylene with α-olefins produced in situ from ethylene over the dual functional catalytic system. The catalytic system was of high polymeric activity. 3First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 6Plant base editing and prime editing:The current status and future perspectives显示文摘Precise replacement of an allele with an elite allele controlling an important agronomic trait in a predefined manner by gene editing technologies is highly desirable in crop improvement.Base editing and prime editing are two newly developed precision gene editing systems which can introduce the substitution of a single base and install the desired short indels to the target loci in the absence of double-strand breaks and donor repair templates,respectively.Since their discoveries,various strategies have been attempted to optimize both base editor(BE)and prime editor(PE)in order to improve the precise editing efficacy,specificity,and expand the targeting scopes.Here,we summarize the latest development of various BEs and PEs,as well as their applications in plants.Based on these progresses,we recommend the appropriate BEs and PEs for both basic plant research and crop improvement.Moreover,we propose the perspectives for further optimization of these two editors.We envision that both BEs and PEs will become the routine and customized precise gene editing tools for both plant biological research and crop improvement in the near future.Jingying Li Chen Zhang Yubing He Shaoya Li Lei Yan Yucai Li Ziwei Zhu Lanqin Xia 2023Journal of Integrative Plant Biology2023,65,2:1

14Rhizoma Chuanxiong regulates vascular endothelial growth factor production in hypoxic human umbilical vein endothelial cells in vitro and in peri-infarct rat brain tissue显示文摘BACKGROUND:Vascular endothelial growth factor(VEGF) acts as'molecular bridge'following ischemic stroke to improve and restore blood supply and reduce infarction volume.Clinical studies have demonstrated the efficacy of Rhizoma Chuanxiong(Chuanxiong) in the treatment of ischemic cerebrovascular diseases.However,whether it promotes endogenous VEGF expression in ischemic stroke remains unknown. OBJECTIVE:To investigate the influence of Rhizoma Chuanxiong on VEGF production in vitro cultured human umbilical vein endothelial cells and on VEGF expression in ischemic cerebral tissues to explore its role in angiogenesis. DESIGN,TIME AND SETTING:In vitro basic comparison of traditional Chinese drug-containing serum pharmacology;in vivo randomized,controlled,animal experiment.This study was performed at the Medical Laboratory of West China Hospital,Sichuan University between December 2002 and April 2004. MATERIALS:Two Chinese rabbits were selected.One was intragastrically perfused with 5.8 g/kg Rhizoma Chuanxiong extract twice per day for three consecutive days to prepare Rhizoma Chuanxiong extract-containing serum.The remaining rabbit was intragastrically perfused with the same volume of normal saline twice per day for three consecutive days.Rhizoma Chuanxiong extract was provided by Beijing Traditional Chinese Medicine Research Institute,predominantly composed of ligustrazine,ligustilide,and ferulic acid.ChemiKine^(TM) human VEGF Kit was purchased from Chemicon,USA;mouse anti-VEGF monoclonal antibody and biotin-goat anti-mouse IgG were purchased from Santa Cruz Biotechnology.Inc.,USA. METHODS:(1) In vitro experiment:in vitro cultured human umbilical vein endothelial cells were separately incubated in rabbit serum with 10%Rhizoma Chuanxiong extract,normal medium without rabbit serum,and rabbit serum without Rhizoma Chuanxiong extract(blank control).In addition,cells from the three groups were incubated under normoxia(5%CO_2,95%air) and hypoxia (1%O_2,5%CO_2,94%N_2),respectively,for 24 hours.(2) In vivo experiment:a total of 4/44 Sprague Dawley rats were selected for the sham-operated group(no occlusion),and the remaining rats were used to establish a cerebral ischemia/reperfusion model by suture occlusion.32 animals with ischemia/reperfusion injury were randomly divided into treatment and model groups,with 16 rats in each group.Both groups were intraperitoneally infused with 0.58 g/kg Rhizoma Chuanxiong extract and normal saline two hours following reperfusion.The sham-operated group was administrated normal saline.Animals were treated with saline or Chuanxiong extracts(0.58 g/kg) twice per day for three consecutive days. MAIN OUTCOME MEASURES:In vitro experiment:VEGF concentration was detected in each group by enzyme-linked immunosorbent assay.In vivo experiment:behavioral alterations of rats were evaluated by neurological function scale;infarct volume was assessed by hematoxylin-eosin staining;VEGF protein expression in the infarct regions was determined by immunohistochemistry. RESULTS:(1) VEGF levels were similar between the three groups under normoxic condition(P>0.05);while hypoxia induced VEGF production(P<0.01).VEGF levels in the drug-containing serum group were particularly higher compared with the other groups(P<0.01).(2) Compared with normal saline treatment,Rhizoma Chuanxiong extract significantly improved the neurological scale and reduced cerebral infarct volumes(P<0.05).The percent of VEGF-positive cells was significantly greater than the model group(P<0.05).The sham-operated group exhibited normal neurological function,with no infarct focus. 16Ni-W Catalysts Supported on Mesoporous SBA-15: Trace W Steering CO_(2) Methanation显示文摘A series of Ni-W catalysts supported on mesoporous SBA-15 with different Ni:W ratios(NixW/SBA-15, Ni-5%, x=1, 10, 50) was prepared and fully characterized by powder X-ray diffraction(PXRD), Brunner-Emmet-Teller(BET), transmission electronic microscopy(TEM), H2-temperature programmed reduction(H2-TPR), and X-ray photoelectron spectroscopy(XPS). High-resolution TEM images, XPS measurements, H2-TPR experiments coupled with PXRD results determined the evolution of Ni and W species. It is found that a trace amount of W from H2WO4 can significantly improve Ni dispersion on SBA-15 (Ni50W/SBA-15) with Ni0 and non-stoichiometric WOx species as small as ca. 3.6 nm. The prepared NixW/SBA-15 was utilized for CO_(2) hydrogenation, which showed that a higher W content restrained the CO_(2) hydrogenation while a lower W ratio promoted both conversion rate and selectivity for methane compared with Ni/SBA-15. The Ni50W/SBA-15 catalyst showed the best performance with a 93.3% CO_(2) conversion rate and 99.7% selectivity for methane at 400 oC under 0.1 MPa and maintained ca. 97% initial performance for 24 h. Tracking product evolution experiments by in-situ Fourier transform infrared spectrascopy(FTIR) indicated that a small amount of W can modify the surface of Ni particles by geometric coverage and electronic modification, which facilitates the adsorption of the CO intermedia and results in the formation of CH4.
