| 1 | Salvianolic acid A alleviates renal injury in systemic lupus erythematosus induced by pristane in BALB/c mice显示文摘The purpose of this study was to investigate the effects of salvianolic acid A(SAA) in systemic lupus erythematosus(SLE) induced by pristane in BALB/c mice.Lupus mice were established by confirming elevated levels of autoantibodies and IL-6 after intraperitoneal injection of pristane.Mice were then treated with daily oral doses of SAA for 5 months in parallel with mice treated with prednisone and aspirin as positive controls.The levels of autoantibodies were monitored at monthly intervals and nephritic symptoms observed by hematoxylin and eosin(H&E) and periodic acid-Schiff(PAS) staining.Western blot analysis of renal tissue was also employed.SAA treatment caused a significant reduction in the levels of anti-Sm autoantibodies and reduced renal histopathological changes and pathological effects.SAA treatment also significantly inhibited the phosphorylation of IKK,IκB and NFκB in renal tissues of lupus mice.In conclusion,the results suggest that SAA alleviates renal injury in pristane-induced SLE in BALB/c mice through inhibition of phosphorylation of IKK,IκB and NFκB. | Yihuang Lin Yu Yan Huifang Zhang Yucai Chen Yangyang He Shoubao Wang Lianhua Fang Yang Lv Guanhua Du | 2017 | Acta Pharmaceutica Sinica B2017,7,2: | 8 |

| 3 | Noble gases in corundum megacrysts from the basalts in Changle,Shandong Province,eastern China显示文摘This study presents noble gaseous data of the corundum megacrysts from the Cenozoic basalts in Changle, Shandong Province, eastern China. It is known that no noble gaseous data of corundum megacryst have been documented before. The 3He/4He ratios (1.13-7.37 Ra) of the corundums from Changle vary from atmosphere to MORB values; the 20Ne/22Ne (9.67-10.75) and 21Ne/22Ne (0.0280-0.0372) data define two linear trends on Ne three-isotope diagram, respectively, along the MFL and the correlation line between atmosphere and MORB; the 38Ar/36Ar (0.177-0.194) ratios, the 40Ar/36Ar (280.9 -404.2) ratios and the 128-136Xe/132Xe ration with obvious 129Xe excess are generally higher than at-mospheric component, but the 40Ar/36Ar ratios are much closer to atomospheric ratio. The isotopic compositions of noble gases (particularly for He and Ar) of the corundums are similar to those of py-roxene, anorthoclase megacrysts, and mantle-derived xenoliths from this area, and those of man-tle-derived xenoliths from several areas in eastern China. Therefore, the noble gases trapped in the corundums probably are from mantle source, representing a ‘mixed fluid' produced by the interaction between the lithospheric mantle and fluids releasing from the convective plate. Both the noble gas isotopic compositions and the oxygen isotopic compositions of the solid corundums are not the characteristics of crustal source. | 4 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 5 | A multicenter retrospective cohort study of ketogenic diet therapy in 481 children with infantile spasms显示文摘Background:Ketogenic diet(KD)therapy is one of the main treatments for drug-resistant epilepsy.However,the KD therapy has been applied in only a small number of infantile spasm cases.In this large multicenter study,we investigated the efficacy of KD therapy in the treatment of infantile spasms.Methods:In this retrospective,multicenter cohort study,clinical data from main epilepsy centers were analyzed.Patients were classified into different groups according to age,type of drug and whether glucocorticoid was used before initiation of KD.Results:From October 2014 to March 2020,481 patients(308 males and 173 females)with infantile spasms were treated with the KD therapy.The age of the patients ranged from 2 months to 20 years,with a mean age of 1 year and 10 months.The number of anti-seizure medications(ASMs)used before KD initiation ranged 0-6,with a median of 3.In different time from initiation(1,3,6,and 12 months),the rates of seizure freedom after KD were 6.9,11.6;16.0 and 16.8%,respectively(x^(2)=27.1772,P<0.0001).There was a significant difference in the rate of seizure freedom between 3 months and 1 month(x^(2)=6.5498,P=0.0105)groups,and 6 months and 3 months(x^(2)=3.8478,P=0.0498)groups,but not between 12 months and 6 months(x^(2)=0.1212,P=0.7278)groups.The rates of effectiveness were 44.7;62.8,49.1 and 32.0%(x^(2)=93.2674,P<0.0001),respectively.The retention rates were 94.0,82.5,55.7 and 33.1%(x^(2)=483.7551,P<0.0001),correspondingly.The rate of effectiveness and the retention rate of KD were significantly different among the 1,3,6 and 12 months.KD treatment was the first choice in 25 patients(5.2%),55 patients(11.4%)started KD after the failure of the first ASM,158 patients(32.8%)started KD after the failure of the second ASM,157 patients(32.6%)started KD after the failure of the third drug,and 86 patients(17.9%)started KD after the failure of the fourth and more.The KD effect was not related to the number of ASMs used before KD startup(P>0.05).Two hundred and eighteen patients(45.3%)failed to respond to corticotropin or glucocorticoid before initiation.There was no significant difference in the effectiveness rate at different time points between the group of KD therapy after glucocorticoid failure and the group after non-hormone failure (x^(2)=0.8613,P=0.8348).The rate of adverse events of KD in 1,3,6,and 12 months after KD initiation were 22.3,21.7,16.8 and 6.9%,respectively.The adverse events mainly occurred during the first 3 months of KD,and the main adverse events were gastrointestinal disturbance and constipation.Conclusions:The efficacy of the KD treatment for infantile spasms was not affected by age,medication,and glucocorticoid use before initiation.KD is one of the effective treatments for infantile spasms. | Yuanzhen Ye Dan Sun Hua Li Jianmin Zhong Rong Luo Baomin Li Dengna Zhu Dan Li Shaoping Huang Yuwu Jiang Nong Xiao Yucai Chen Yuqin Zhang Mei Yu Xiaoyun Shen Li Gao Guo Zheng Congmin Zhao Baoqiang Yuan Jianxiang Liao Jiong Qin CAAE KD group | 2022 | Acta Epileptologica2022,5,1: | 2 |

| 11 | Activating transcription factor 4 protects mice against sepsis-induced intestinal injury by regulating gut-resident macrophages differentiation显示文摘Background: Gut-resident macrophages (gMacs) supplemented by monocytes-to-gMacs differentiation play a critical role in maintaining intestinal homeostasis. Activating transcription factor 4 (ATF4) is involved in immune cell differentiation. We therefore set out to investigate the role of ATF4-regulated monocytes-to-gMacs differentiation in sepsis-induced intestinal injury.Methods: Sepsis was induced in C57BL/6 wild type (WT) mice andAtf4-knockdown (Atf4+/-) mice by cecal ligation and puncture or administration of lipopolysaccharide (LPS). Colon, peripheral blood mononuclear cells, sera, lung, liver, and mesenteric lymph nodes were collected for flow cytometry, hematoxylin and eosin staining, immunohistochemistry, quantitative reverse transcription polymerase chain reaction, and enzyme-linked immunosorbent assay, respectively.Results: CD64, CD11b, Ly6C, major histocompatibility complex-II (MHC-II), CX3CR1, Ly6G, and SSC were identified as optimal primary markers for detecting the process of monocytes-to-gMacs differentiation in the colon of WT mice. Monocytes-to-gMacs differentiation was impaired in the colon during sepsis and was associated with decreased expression of ATF4 in P1 (Ly6Chi monocytes), the precursor cells of gMacs.Atf4 knockdown exacerbated the impairment of monocytes-to-gMacs differentiation in response to LPS, resulting in a significant reduction of gMacs in the colon. Furthermore, compared with WT mice,Atf4+/- mice exhibited higher pathology scores, increased expression of inflammatory factor genes (TNF-α, IL-1β), suppressed expression of CD31 and vascular endothelial-cadherin in the colon, and increased translocation of intestinal bacteria to lymph nodes and lungs following exposure to LPS. | 15 | High drug loading and pH-responsive nanomedicines driven by dynamic boronate covalent chemistry for potent cancer immunotherapy显示文摘Tumor cells undergoing immunogenic cell death (ICD) have emerged as an in situ therapeutic vaccine helping to activate a persistent anti-tumor response. Several chemotherapeutic agents have been demonstrated to induce ICD, however accompanied with severe adverse effects in the clinic, weakening its immune responses. Herein, to elicit an intensive ICD while minimizing the systemic toxicity, we introduce a tumor targeting peptide modified bortezomib (BTZ) loading nanomedicine (i-NPBTZ) for the efficient delivery and controlled release of BTZ in tumors. This system is constructed by conjugating BTZ to PEGylated polyphenols via a pH-sensitive covalent boronate-phenol bond that allows them to self-assemble into nanovesicles in neutral condition with high drug loading efficiency. Once accumulated in acidic environment, BTZ-phenolic network is disassembled and thereby accelerates the release of BTZ from nanocarriers. The released BTZ selectively kill tumor cells with a concomitant evocation of tumor-specific cytotoxic T cells by triggering ICD in vivo. This can finally lead to an extended tumor ablation and prevention of distant metastasis in a syngeneic tumor mouse model, while reducing the systemic toxicity of BTZ. MATERIALS:Two Chinese rabbits were selected.One was intragastrically perfused with 5.8 g/kg Rhizoma Chuanxiong extract twice per day for three consecutive days to prepare Rhizoma Chuanxiong extract-containing serum.The remaining rabbit was intragastrically perfused with the same volume of normal saline twice per day for three consecutive days.Rhizoma Chuanxiong extract was provided by Beijing Traditional Chinese Medicine Research Institute,predominantly composed of ligustrazine,ligustilide,and ferulic acid.ChemiKine^(TM) human VEGF Kit was purchased from Chemicon,USA;mouse anti-VEGF monoclonal antibody and biotin-goat anti-mouse IgG were purchased from Santa Cruz Biotechnology.Inc.,USA. METHODS:(1) In vitro experiment:in vitro cultured human umbilical vein endothelial cells were separately incubated in rabbit serum with 10%Rhizoma Chuanxiong extract,normal medium without rabbit serum,and rabbit serum without Rhizoma Chuanxiong extract(blank control).In addition,cells from the three groups were incubated under normoxia(5%CO_2,95%air) and hypoxia (1%O_2,5%CO_2,94%N_2),respectively,for 24 hours.(2) In vivo experiment:a total of 4/44 Sprague Dawley rats were selected for the sham-operated group(no occlusion),and the remaining rats were used to establish a cerebral ischemia/reperfusion model by suture occlusion.32 animals with ischemia/reperfusion injury were randomly divided into treatment and model groups,with 16 rats in each group.Both groups were intraperitoneally infused with 0.58 g/kg Rhizoma Chuanxiong extract and normal saline two hours following reperfusion.The sham-operated group was administrated normal saline.Animals were treated with saline or Chuanxiong extracts(0.58 g/kg) twice per day for three consecutive days. MAIN OUTCOME MEASURES:In vitro experiment:VEGF concentration was detected in each group by enzyme-linked immunosorbent assay.In vivo experiment:behavioral alterations of rats were evaluated by neurological function scale;infarct volume was assessed by hematoxylin-eosin staining;VEGF protein expression in the infarct regions was determined by immunohistochemistry. RESULTS:(1) VEGF levels were similar between the three groups under normoxic condition(P>0.05);while hypoxia induced VEGF production(P<0.01).VEGF levels in the drug-containing serum group were particularly higher compared with the other groups(P<0.01).(2) Compared with normal saline treatment,Rhizoma Chuanxiong extract significantly improved the neurological scale and reduced cerebral infarct volumes(P<0.05).The percent of VEGF-positive cells was significantly greater than the model group(P<0.05).The sham-operated group exhibited normal neurological function,with no infarct focus.
