1Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. 2Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders.Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao 2020Cell Research2020,30,9:7
3Achievements in cancer research supported by National Natural Science Foundation of China显示文摘For the past two decades,great achievements have been made in cancer research of China and the majority of them were supported by the National Natural Science Foundation of China(NSFC).In this article,we reviewed the funding history of NSFC including the number of proposals,the amount of grants,distribution of supported program categories during the period of 1987 to 2012 and the representative achievements made by continuous support of NSFC.Also,the impacts of NSFC support on discipline development and talent fostering were analyzed.From 1987 to 2012,a total of 8738 proposals related to cancer research were funded by NSFC.The percentages of the Research Program,Talent-Training Program,and the Research Environment Program were 61.19%,36.32%,and 2.49%,respectively.Under the support of NSFC,cancer research in China has achieved a significantly rapid and balanced development in both disciplines and scientist resources.HONG Wei LI Cui ZHANG LiPing XU YanYing ZHANG Jun HU Hai DONG ErDan 2013Chinese Science Bulletin2013,58,1:5
4The Salt Reduction and Yield Increase Effects of Smashing Ridging Transforming Saline-alkali Land and Its Development Prospects显示文摘Using smashing ridging tillage machine and smashing ridging technology invented by the authors,transformation test of saline-alkali land by smashing ridging was conducted respectively in Xinjiang and Shaanxi during 2015-2016. 5Contribution of ultrasonic to microstructure and mechanical properties of tilt probe penetrating friction stir welded joint显示文摘Tilt probe penetrating friction stir welding(PFSW)was an innovative technology proposed in recent years to avoid the formation of kissing bond in the root of joint.However,with the heat input decreasing,'S'line or zigzag line was easily introduced in the PFSW joint.In this study,ultrasonic enhanced tilt probe penetrating friction stir welding(U-PFSW)was developed to solve this problem and achieve improved joint mechanical properties.The experimental results confirmed that U-PFSW was a potent technology to completely clear the original butt surface,providing a crucial prerequisite for the achievement of highstrength joint.The application of ultrasonic improves the joint tensile strength and fracture elongation from 336 MPa and 4.3%to 359 MPa and 6.8%,respectively.Furthermore,the strength of stir zone was also increased from 391 MPa in PFSW to 420 MPa in U-PFSW.Analyses of texture and precipitate indicated that the dynamic recrystallization(DRX)and precipitation strengthening were both enhanced by the ultrasonic.Ultrasonic-enhanced DRX enabled a complete elimination of the'S'line;the enhanced precipitation strengthening by vacancy-induced mechanism in U-PFSW was the intrinsic reason for the significantly improved mechanical properties.Yanying Hu Huijie Liu Dongrui Li 2021Journal of Materials Science & Technology2021,,26:2
6In Situ Synthesis of Core-Shell Li4Ti5O(12) @ Polyaniline Composites with Enhanced Rate Performance for Lithium-ion Battery Anodes显示文摘The core-shell Li_4Ti_5O_(12) @ polyaniline composites(LP) have been synthesized via an in situ synthesis with different mole ratios(25:1, 50:1 and 100:1, aniline:LTO). As an anode material of lithium-ion batteries,the LP-2 electrodes(50:1) exhibit a high initial reversible capacity of 205 mAh g^(-1)with an initial coulombic efficiency of 97.6% at 0.1 C. Even at a high current density of 10 C, the reversible capacity of the LP-2 electrodes still remains at 102 mAh g^(-1). Moreover, the LP-2 electrodes retain an impressive high capacity of 161 mAh g^(-1)after 100 cycles at 1 C, with 0.11% capacity fading per cycle. 7Dopamine D2 Receptor-Mediated Modulation of Rat Retinal Ganglion Cell Excitability显示文摘Ganglion cells(RGCs) are the sole output neurons of the retinal circuity. Here, we investigated whether and how dopamine D2 receptors modulate the excitability of dissociated rat RGCs. Application of the selective D2 receptor agonist quinpirole inhibited outward K^+ currents, which were mainly mediated by glybenclamide-and 4-aminopyridine-sensitive channels, but not the tetraethylammonium-sensitive channel. In addition,quinpirole selectively enhanced Nav1.6 voltage-gated Na^+ currents. The intracellular c AMP/protein kinase A,Ca^2+/calmodulin-dependent protein kinase Ⅱ, and mitogen-activated protein kinase/extracellular signal-regulated kinase signaling pathways were responsible for the effects of quinpirole on K^+ and Na^+ currents, while phospholipase C/protein kinase C signaling was not involved. Under current-clamp conditions, the number of action potentials evoked by positive current injection was increased by quinpirole. 12Effect of Ti layer thickness on microstructure and magnetic properties of Ti/Co/Ti films显示文摘TiCoTi granular films were prepared by DC facing-target magnetron sputtering system onto glass substrates and subsequently in situ annealing in vacuum. Structural of Ti (t nm)/Co (40 nm)/Ti (t nm) films were investigated in detail, which shows that the majority Co nanograins are formed as the hexagonal-close-packed (HCP) structure. Vibrating sample magnetometer (VSM) and scanning probe microscope (SPM) were applied to study the magnetic properties, morphologies and domain structures of these samples. It has been found that the structure and magnetic properties of the Ti/Co/Ti films depend strongly on the Ti layer thickness. The out-of-plane coercivities (Hc) of the film is maximum about 78.8 kA·m-1 when t=5 nm with annealing at 300 ℃; the distributing of grains of the sample is uniformity; and the average size of particles is about 13 nm. 13Proteomic profiling of cotton fiber developmental transition from cell elongation to secondary wall deposition显示文摘Cotton fiber developmental transition from elongation to secondary cell wall biosynthesis is a critical growth shifting phase that affects fiber final length,strength,and other properties.Morphological dynamic analysis indicated an asynchronous fiber developmental pattern between two most important commercial cotton species,Gossypium hirsutum(Gh)and G.barbadense(Gb).Using isobaric tags for relative and absolute quantitation techniques,we examined the temporal changes of protein expression at three representative development periods(15-19,19-23,and 23-27 dpa)in both species.Strikingly,a large proportion of differentially expressed proteins(DEPs)were identified at 19-23 dpa in Gh and at 23-27 dpa in Gb,corresponding to their fiber developmental transition timing.To better understand fiber transitional development,we comparatively analyzed those DEPs in 19-23 dpa of Gh vs.in 23-27 dpa of Gb,and noted that these cotton species indeed share fundamentally similar fiber developmental features under the biological processes.We also showed that there are limited overlaps in both specific upregulated and downregulated proteins between the two species,suggesting species-specific protein regulations in the development process.Proteomic profiling results revealed dynamic changes of several key proteins and biological processes that are potentially correlated with fiber developmental transition.During the transition,upregulated proteins are mainly involved in carbohydrate/energy metabolism,oxidation-reduction,cytoskeleton,protein turnover,Ca2+signaling,etc.,whereas important downregulated proteins are mostly involved in phenylpropanoid and flavonoid secondary metabolism pathways.The gene expressions of several changed proteins in this key stage were also examined by quantitative reverse transcription polymerase chain reaction.Overall,the present study provides accurate pictures of the regulatory networks of functional proteins during the fiber developmental transition,therefore highlighting candidate genes/proteins and related pathways for the cotton fiber improvement.Xiaoyun Zhou Wenran Hu Bo Li Yang Yang Yong Zhang Kieran Thow Ling Fan Yanying Qu 2019Acta Biochimica et Biophysica Sinica2019,51,11:1

15The binding epitope of sintilimab on PD-1 revealed by AbMap显示文摘PD-1 plays an important role as an immune checkpoint.Sintilimab is a newly approved PD-1 antibody for cancer immunotherapy with an unknown binding epitope on PD-1.In this study,to elucidate the molecular mechanism by which sintilimab blocks PD-1 activation,we applied Antibody binding epitope Mapping(AbMap)to identify the binding epitope of sintilimab.An epitope was successfully identified,i.e.SLAPKA,aa 127-132.By constructing a series of point mutations,the dominant residues S127,L128,A129,P130,and A132 of PD-1 were further validated by western blot analysis,biolayer interferometry,and flow cytometry.Structural analysis showed that the epitope is partially within the binding interface of PD-1 and PD-L1,and this epitope also partially overlaps with that of nivolumab and pembrolizumab.These results demonstrate that sintilimab can attenuate PD-1 activation by directly competing with the interaction between PD-1 and PD-L1 through binding with the key residues of the FG loop on PD-1.This study also demonstrates the high efficiency and accuracy of AbMap for determining the binding epitope of therapeutic antibodies.Mingliang Ma Huan Qi Chuansheng Hu Zhaowei Xu Fanlin Wu Nan Wang Danyun Lai Yang Li Hainan Zhang Hewei Jiang Qingfeng Meng Shujuan Guo Yani Kang Xiaodong Zhao Hua Li Sheng-ce Tao 2021Acta Biochimica et Biophysica Sinica2021,53,5:1

20In Vitro Antibacterial Effect of Traditional Chinese Medicine on Main Pathogenic Bacteria of Recessive Mastitis in Dairy Cows显示文摘The inhibition zone diameter,minimal inhibitory concentration( MIC) and minimum bactericidal concentration( MBC) of 20 kinds of traditional Chinese medicine on Staphylococcus aureus and Streptococcus agalactiae isolated from recessive mastitis were studied using plate test and tube test.
