| 1 | Efficacy and safety evaluation of icotinib in patients with advanced non-small cell lung cancer显示文摘Objective: To evaluate the efficacy and safety of icotinib hydrochloride in patients with advanced non-small cell lung cancer (NSCLC). Methods: A total of 89 patients with stage ⅢB or IV NSCLC received icotinib at a dose of 125 mg administered 3 times a day. Icotinib treatment was continued until disease progression or development of unacceptable toxicity. Results: A total of 89 patients were assessable. In patients treated with icotinib, the overall response rate (RR) was 36.0% (32/89), and the disease control rate (DCR) was 69.7% (62/89). RR and DCR were significantly improved in patients with adenocarcinoma versus non-adenocarcinoma (P<0.05). The symptom improvement rate was 57.3% (51/89), and the main symptoms improved were cough, pain, chest distress, dyspnea, and Eastern Cooperative Oncology Group performance status. The main toxic effects were rash [30/89 (33.7%)] and diarrhea [15/89 (16.9%)]. The level of toxicity was typically low. | 2 | Far-field sonic boom prediction considering atmospheric turbulence effects: An improved approach显示文摘Accurate prediction of sonic boom is one of key challenges for the design of a low-boom supersonic aircraft. For most of available far-field prediction methods, the effect of atmospheric turbulence appearing in the planetary boundary layer cannot be considered, which results in remarkable inaccuracy of predicting ground-level sonic boom waveform. Although some efforts have been made to overcome the shortcoming, the turbulence effects are not yet well described so far. This article proposes an improved method by extending the two-dimensional Heterogeneous One-Way Approximation for the Resolution of Diffraction(HOWARD) equation to account for the axial and transverse convections of wind fluctuation as well as the effect of temperature fluctuation. The proposed method is validated by comparing the predictions with the flight-test data of JAXA D-SEND#1 LBM, which shows that the result of the proposed method is in better agreement with the flight-test data than that of the method without considering atmospheric turbulence effects.Then, distortion mechanism of sonic boom waveforms caused by atmospheric turbulence is analyzed by using the proposed method. It is indicated that the effect of turbulent convection makes uniform sonic-boom wavefronts irregular, which creates the condition of diffraction effect to perturb waveforms. Finally, the proposed method is applied to investigate the behavior of two types of waveforms given by the sonic boom minimization theory. | 3 | Preclinical efficacy against acute myeloid leukaemia of SH1573, a novel mutant IDH2 inhibitor approved for clinical trials in China显示文摘Acute myeloid leukaemia(AML) is the most common form of acute leukaemia in adults,with increasing incidence with age and a generally poor prognosis.Almost 20% of AML patients express mutant isocitrate dehydrogenase 2(mIDH2),which leads to the accumulation of the carcinogenic metabolite 2-hydroxyglutarate(2-HG),resulting in poor prognosis.Thus,global institutions have been working to develop mIDH2 inhibitors.SH1573 is a novel mIDH2 inhibitor that we independently designed and synthesised.We have conducted a comprehensive study on its pharmacodynamics,pharmacokinetics and safety.First,SH1573 exhibited a strong selective inhibition of mIDH2 R140 Q protein,which could effectively reduce the production of 2-HG in cell lines,serum and tumors of an animal model.It could also promote the differentiation of mutant AML cell lines and granulocytes in PDX models.Then,it was confirmed that SH1573 possessed characteristics of high bioavailability,good metabolic stability and wide tissue distribution.Finally,toxicological data showed that SH1573 had no effects on the respiratory system,cardiovascular system and nervous system,and was genetically safe.This research successfully promoted the approval of SH1573 for clinical trials(CTR20200247).All experiments demonstrated that,as a potential drug against mIDH2 R140 Q acute myeloid leukaemia,SH1573 was effective and safe. | Zhiqiang Wang Zhibo Zhang Yong Li Li Sun Dezhen Peng Danyu Du Xian Zhang Luwei Han Liwen Zhao Ligong Lu Hongzhi Du Shengtao Yuan Meixiao Zhan | 2021 | Acta Pharmaceutica Sinica B2021,11,6: | 3 |
| 4 | Exposure measurement, risk assessment and source identification for exposure of traffic assistants to particle-bound PAHs in Tianjin, China显示文摘To investigate the levels of exposure to particulate-bound polycyclic aromatic hydrocarbon(PAH) and to estimate the risk these levels pose to traffic assistants(TAs) in Tianjin(a megacity in North China), a measurement campaign(33 all-day exposure samples, 25 occupational-exposure samples and 10 indoor samples) was conducted to characterize the TAs' exposure to PAHs, assess the cancer risk and identify the potential sources of exposure. The average total exposure concentration of 14 PAHs was approximately 2871 ± 928 ng/m3(on-duty), and 1622 ± 457 ng/m3(all-day). The indoor PAHs level was 1257 ± 107 ng/m3. After 8000 Monte Carlo simulations, the cancer risk resulting from exposure to PAHs was found to be approximately 1.05 × 10-4. A multivariate analysis was applied to identify the potential sources, and the results showed that, in addition to vehicle exhaust, coal combustion and cooking fumes were also another two important contributors to personal PAH exposure. | 7 | Development and application of digital assistive teaching system for anatomy显示文摘Background Anatomy is a required course for all medicine-related industries.In recent decades,the teaching quality and effect of anatomy have been compromised by factors including a decrease in human body specimens,dampened enthusiasm for the discipline,reduced teaching hours of anatomy,scale expansion of medical education,and obstacles in performing field autopsies and observations.Methods Based on China's digitalized visible human research achievements,this article extracts the boundary information of anatomic structures from tomographic images,constructs three dimensional(3D)digital anatomical models with authentic texture information,and develops an anatomy assistive teaching system for teachers and students based on the knowledge points of anatomy,to meet the anatomy teaching requirements of different majors at various levels.Results This scientific,complete,and holistic system has produced over 60003D digital anatomical models,5000 anatomy knowledge points,50 anatomical operation videos,and 150 micro demonstration classes,with teaching contents for different majors and levels,such as systematic anatomy,topographic anatomy,sectional anatomy,anatomy of motion,and virtual anatomical operation table.Ranging from network terminals,desktops,touchscreen 3D displays,desktops,and projection 3D volumetric displays to augmented reality,its diversified interactive forms meet the requirements for a learning environment in different settings.Conclusions With multiple teaching and learning links covered,such as teaching environment,teaching resources,instructional slides,autonomous learning,and learning effect evaluation,this novel teaching system serves as a vital component and a necessary resource in anatomy teaching and functions as an important supplement to traditional anatomy teaching.Applied and promoted in most medical colleges and schools in China,this system has been recognized and approved by anatomy teachers and students,and plays a positive role in guaranteeing the effect and quality of anatomy teaching. | Na ZHANG Liwen TAN Fengying LI Bing HAN Yifa XU | 2021 | Virtual Reality & Intelligent Hardware2021,3,4: | 1 |

| 12 | Effect of longitudinal lift distribution on sonic boom of a canard-wing-stabilator-body configuration显示文摘After the last flight of the Concorde in 2003,sonic boom has been one of the obstacles to the return of a supersonic transport aircraft to service.To reduce the sonic boom intensity to an acceptable level,it is of great significance to study the effect of lift distribution on far-field sonic boom,since lift is one of the most important contributors to an intense sonic boom.Existing studies on the longitudinal lift distribution used low-fidelity methods,such as Whitham theory,and in turn,only preliminary conclusions were obtained,such as that extending the lift distribution can reduce sonic boom.This paper uses a newly developed high-fidelity prediction method to quantitatively study the effect of longitudinal lift distribution on the sonic boom of a Canard-Wing-Stabilator-Body(CWSB)configuration.This high-fidelity prediction method combines near-feld CFD simulation with far-field propagation by solving the augmented Burgers equation.A multipole analysis method is employed for the extraction of near-field waveform in order to reduce computational cost.Seven configurations with the same total lift but different distributions are studied,and the quantitative relationship between the longitudinal lift distribution and far-field sonic boom intensity is investigated.It is observed that a small lift generated by the stabilator can prevent aft-stabilator and aft-fuselage shocks from merging,while the balanced lift generated by the canard and wing can effectively keep the corresponding shocks further apart,which is beneficial for reducing both the on-track and off-track sonic boom.In turn,the acoustic level perceived at the ground can be reduced by 5.9 PLdB on-track and 5.4 PLdB off-track,on average. | Liwen ZHANG Zhonghua HAN Jianling QIAO Wenping SONG Yulin DING Xuehe WANG | 2023 | Chinese Journal of Aeronautics2023,36,6: | 1 |

| 15 | HTPdb and HTPtools:Exploiting maize haplotypetag polymorphisms for germplasm resource analyses and genomics-informed breeding显示文摘Along with rapid advances in high-throughput-sequencing technology,the development and application of molecular markers has been critical for the progress that has been made in crop breeding and genetic research.Desirable molecular markers should be able to rapidly genotype tens of thousands of breeding accessions with tens to hundreds of markers.In this study,we developed a multiplex molecular marker,the haplotype-tag polymorphism(HTP),that integrates Maize6H-60K array data from 3,587 maize inbred lines with 6,375 blocks from the recombination block map.After applying strict filtering criteria,we obtained 6,163 highly polymorphic HTPs,which were evenly distributed in the genome.Furthermore,we developed a genome-wide HTP analysis toolkit,HTPtools,which we used to establish an HTP database(HTPdb)covering the whole genomes of 3,587 maize inbred lines commonly used in breeding.A total of 172,921 non-redundant HTP allelic variations were obtained.Three major HTPtools modules combine seven algorithms(e.g.,chain Bayes probability and the heterotic-pattern prediction algorithm)and a new plotting engine named“BCplot”that enables rapid visualization of the background information of multiple backcross groups.HTPtools was designed for big-data analyses such as complex pedigree reconstruction and maize heterotic-pattern prediction.The HTP-based analytical strategy and the toolkit developed in this study are applicable for high-throughput genotyping and for genetic mapping,germplasm resource analyses,and genomics-informed breeding in maize. | Yikun Zhao Hongli Tian Chunhui Li Hongmei Yi Yunlong Zhang Xiaohui Li Han Zhao Yongxue Huo Rui Wang Dingming Kang Yuncai Lu Zhihao Liu Ziyue Liang Liwen Xu Yang Yang Ling Zhou Tianyu Wang Jiuran Zhao Fengge Wang | 2022 | Plant Communications2022,3,4: | 0 |

| 19 | Loss of chromosome 9p21 is associated with a poor prognosis in adenosquamous carcinoma of the pancreas显示文摘Adenosquamous carcinoma of the pancreas(ASCP)is a rare histological subtype of pancreatic cancer with a poor prognosis and a high metastasis rate.However,little is known about its genomic landscape and prognostic biomarkers.A total of 48 ASCP specimens and 98 pancreatic ductal adenocarcinoma(PDAC)tumour specimens were sequenced to explore the genomic landscape and prognostic biomarkers.The homozygous deletion of the 9p21.3 region(including CDKN2A,CDKN2B,and MTAP)(9p21 loss)occurred in both ASCP and PDAC,and a higher frequency of 9p21 loss was observed in ASCP(12.5%vs 2.0%,P=0.022).Notably,9p21 loss was significantly associated with poor disease-free survival(DFS)in ASCP patients(mDFS(Median DFS)=4.17 vs 7.33 months,HR(Hazard Ratio)=3.70,P=0.009).The most common gene alterations in patients with ASCP were KRAS(96%),TP53(81%),CDKN2A(42%),SMAD4(21%),CDKN2B(13%),and FAT3(13%).The mutation rates of ACVR2A(6.25%vs 0%),FANCA(6.25%vs 0%),RBM10(6.25%vs 0%),and SPTA1(8.33%vs 1.02%)were significantly higher in ASCP than in PDAC.In conclusion,we have comprehensively described the genomic landscape of the largest cohort of ASCP patients to date and highlight that 9p21 loss may be a promising prognostic biomarker for ASCP,which provides a molecular basis for prognosis prediction and new therapeutic strategies for ASCP. | Yina Jiang Yinying Wu Liwen Zhang Yan Wang Guiping Xu Yuan Deng Liang Han Enxiao Li Qingyong Ma Mian Xu Zheng Wu Zheng Wang | 2023 | Precision Clinical Medicine2023,6,4: | 0 |
