1Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5Quantitative response of oil sunflower yield to evapotranspiration and soil salinity with saline water irrigation显示文摘Appropriate application of water-salt-crop function model can optimize agricultural water management in regions with declining water supply,such as the Hetao district.Appropriate use of saline water is also based on the effects of irrigation water demand and water quality on crop growth quantitatively.Therefore,oil sunflower growth testing under both water and salt stress was completed from 2013 to 2014.Water salinity levels at 1.7 ds/m,4 ds/m,6 ds/m and 8 ds/m were used in the experiments.Two water deficit levels were reported,60%and 80%of the irrigation quota,which were considered moderate and mild deficit levels,respectively.All treatments were applied in planting the oil sunflower in critical growing periods,namely,floral initiation,anthesis and maturity.Linear,Cobb-Douglas,quadratic and transcendental function models were used to simulate the relative yield,evapotranspiration(ET)and electrical conductivity(EC).The predictive ability and sensitivity of each model were then evaluated.Compared with salt stress,water stress exerted a more significant effect on the oil sunflower yield;the water parameters(a1 and a3)were most sensitive in the water-salt-crop function model.Oil sunflower was most sensitive to water and salt stress during anthesis.The transcendental function generally showed a relatively high sensitivity coefficient and a relatively small statistical error.Therefore,the transcendental function is the most appropriate model for simulating and predicting the yield of oil sunflower irrigated with saline water.Applying the water-salt-crop function model in planting of oil sunflower can help in the development and utilization of saline water in the Hetao district.He Xin Yang Peiling Ren Shumei Li Yunkai Jiang Guangyu Li Lianhao 2016International Journal of Agricultural and Biological Engineering2016,9,2:4
6Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 12Infection of wild-type mice by SARS-CoV-2 B.1.351 variant indicates a possible novel cross-species transmission route显示文摘COVID-19 is identified as a zoonotic disease caused by SARS-CoV-2,which also can cross・transmit to many animals but not mice.Genetic modifications of SARS-CoV-2 or mice enable the mice susceptible to viral infection.Although neither is the natural situation,they are currently utilized to establish mouse infection models.Here we report a direct contact transmission of SARS-CoV-2 variant B.1.351 in wild-type mice.The SARS-CoV-2(B.1.351)re plicated efficiently and induced significant pathological changes in lungs and tracheas,accompanied by elevated proinflammatory cytokines in the lungs and sera.Mechanistically,the receptor-binding domain(RBD)of SARS-CoV-2(B.1.351)spike protein turned to a high binding affinity to mouse angiotensin-converting enzyme 2(mACE2),allowing the mice highly susceptible to SARS-CoV-2(B.1.351)infection.Our work suggests that SARS-CoV-2(B.1.351)expands the host range and therefore increases its transmission route without adapted mutation.As the wild house mice live with human populations quite closely,this possible transmission route could be potentially risky.In addition,because SARS-CoV-2(B.1.351)is one of the major epidemic strains and the mACE2 in laboratory-used mice is naturally expressed and regulated,the SARS-CoV-2(B.1.351)/mice could be a much convenient animal model system to study COVID-19 pathogenesis and evaluate antiviral inhibitors and vaccines.Ting Pan Ran Chen Xin He Yaochang Yuan Xiaohui Deng Rong Li Haiping Yan Shumei Yan Jun Liu Yiwen Zhang Xiantao Zhang Fei Yu Mo Zhou Changwen Ke Xiancai Ma Hui Zhang 2022Signal Transduction and Targeted Therapy2022,7,1:0

15Enhancement of CAR-T cell activity against cholangiocarcinoma by simultaneous knockdown of six inhibitory membrane proteins显示文摘Background:Existing treatments for cholangiocarcinoma have poor efficacy.However,chimeric antigen receptor-T(CAR-T)cells are emerging as a potential therapeutic strategy.Solid tumors possess multiple adverse factors in an immunosuppressive microenvironment that impair CAR-T cell infiltration and function.This study aimed to improve the function of CAR-T cells through knock down immune checkpoints and immunosuppressive molecular receptors.Methods:We evaluated the expression of epidermal growth factor receptor(EGFR)and B7 homolog 3 protein(B7H3)antigens in cholangiocarcinoma tissues using immunohistochemistry and screened specific immune checkpoints in the cholangiocarcinoma microenvironment via flow cytometry.Subsequently,we engineered CAR-T cells targeting EGFR and B7H3 antigens.We simultaneously knocked down immune checkpoints and immunosuppressive molecular receptors in CAR-T cells by constructing two clusters of small hairpin RNAs and evaluated the engineered CAR-T cells for antitumor activity both in vitro,using tumor cell lines and cholangiocarcinoma organoid models,and in vivo,using humanized mouse models.Results:We observed high expression of EGFR and B7H3 antigens in cholangiocarcinoma tissues.EGFR-CAR-T and B7H3-CAR-T cells demonstrated specific anti-tumor activity.We found an abundance of programmed cell death protein 1(PD-1),T cell immunoglobulin and mucin domain-containing protein 3(Tim-3),and T cell immunoglobulin and ITIM domain(Tigit)on infiltrated CD8^(+)T cells in the cholangiocarcinoma microenvironment.We then decreased the expression of these 3 proteins on the surface of CAR-T cells,named PTG-scFV-CAR-T cells.Furthermore,we knocked-down the expression of transforming growth factor beta receptor(TGFβR),interleukin-10 receptor(IL-10R),and interleukin-6 receptor(IL-6R)of PTG-scFV-CAR-T cells.Those cells,named PTG-T16R-scFVCAR-T cells,potently killed tumor cells in vitro and promoted apoptosis of tumor cells in a cholangiocarcinoma organoidmodel.Finally,the PTG-T16R-scFv-CART cells showed greater inhibitory effect on tumor growth in vivo,and were superior in prolonging the survival of mice.Conclusions:Our results revealed that PTG-T16R-scFV-CAR-T cells with knockdown of sextuplet inhibitory molecules exhibited strong immunity against cholangiocarcinoma and long-term efficacy both in vitro and in vivo.This strategy provides an effective and personalized immune cell therapy against cholangiocarcinoma.Yidan Qiao Jie Chen Xuemei Wang Shumei Yan Jizhou Tan Baijin Xia Yongjian Chen Keming Lin Fan Zou Bingfeng Liu Xin He Yiwen Zhang Xu Zhang Hui Zhang Xiangyuan Wu Lijuan Lu 2023Cancer Communications2023,43,7:0
16Characteristics of S-wave Envelope Broadening in the Changbaishan Tianchi Volcano显示文摘High-frequency S-wave seismogram envelopes of microearthquakes broaden with increasing travel distance,a phenomenon known as S-wave envelope broadening. Multiple forward scattering and diffraction for the random inhomogeneities along the seismic ray path are the main causes of S-wave envelope broadening,so the phenomenon of S-wave envelope broadening is used to study the inhomogeneity of the medium. The peak delay time of an S-wave,which is defined as the time lag from the direct S-wave onset to the maximum amplitude arrival of its envelope,is accepted to quantify S-wave envelope broadening. 204 small earthquake records in Changbaishan Tianchi volcano were analyzed by the S-wave envelope broadening algorithm. The results show that S-wave envelope broadening in the Changbaishan Tianchi volcano is obvious,and that the peak delay time of S-wave has a positive correlation with the hypocenter distance and frequency of the S-wave. The relationships between the S-wave peak delay time and the hypocenter distance for different frequency bands were obtained using the statistics method.
