1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Interaction of hepatitis C virus envelope glycoprotein E2 with the large extracellular loop of tupaia CD81显示文摘AIM: To further analyze the interaction of tupaia CD81 with hepatitis C virus (HCV) envelope protein E2. METHODS: A tupaia CD81 large extracellular loop (CD81 LEL), which binds to HCV E2 protein, was cloned and expressed as a GST-fusion protein, and interaction of HCV E2 protein with a tupaia CD81 LEL was evaluated by enzyme-linked immunosorbent assay (EIA). RESULTS: Although tupaia and human CD81 LEL differed in 6 amino acid changes, tupaia CD81 LEL was strongly recognized by anti-CD81 antibodies against human CD81 LEL conformation-dependent epitopes. Investigating LEL CD81-E2 interactions by EIA, we demonstrated that binding of tupaia CD81 LEL GST fusion protein to recombinant HCV E2 protein was markedly reduced compared to binding of human CD81 LEL GST fusion protein to recombinant HCV E2 protein. CONCLUSION: These data suggest that the structural differences in-between the tupaia and human CD81 may alter the interaction of the large extracellular loop with HCV envelope glycoprotein E2. 3Effects of acid deposition on terrestrial ecosystems and their rehabilitation strategies in China显示文摘South China has become the third largest region associated with acid deposition following Europe and North America, the area subject to damage by acid deposition increased from 1.75 million km 2 in 1985 to 2.8 million km 2 in 1993. Acid deposition has caused serious damage to ecosystem. Combined pollution of acid rain and SO 2 showed the obvious multiple effects on crops. Vegetable was more sensitive to acid deposition than foodstuff crops. Annual economic loss of crops due to acid deposition damage in eleven provinces of south China was 4.26 billion RMB Yuan. Acid deposition caused serious damage to forest. Annual economic loss of wood volume was about 1.8 billion RMB Yuan and forest ecological benefit loss 16.2 billion in eleven provinces of south China. Acid deposition in south China was typical “sulfuric acid type”. According to the thoughts of sustainable development, some strategies were brought forward as follows: (1) enhancing environmental management, specifying acid\|controlling region, controlling and abating the total emission amount of SO 2; (2) selecting practical energy technologies of clean coal, for example, cleansing and selecting coal, sulfur\|fixed\|type industrial briqutting, abating sulfur from waste gas and so on; (3) developing other energy sources to replace coal, including water electricity, atomic energy and the new energy such as solar energy, wind energy and so on; (4) in acid deposition region of south China, selecting acid\|resistant type of crop and tree to decrease agricultural losses, planting more green fertilizer crops, using organic fertilizers and liming, in order to improve buffer capacities of soil.FENG Zong\|wei, MIAO Hong, ZHANG Fu\|zhu, HUANG Yi\|zong (Research Center for Eco\|Environmental Sciences, Chinese Academy of Sciences, Beijing 100085, China. 6Rotational Swashplate Pulse Continuously Variable Transmission Based on Helical Gear Axial Meshing Transmission显示文摘The current research on pulse continuously variable transmission(CVT) is mainly focused on reducing the pulse degree and making pulse degrees a constant value. Current research mainly confined to find out new design parameters by using the method of optimization, and reduce the pulse degree of pulse CVT and its range of variation. But the fact is that the reduction of the pulse degree is not significant. This article presents a new structure of mechanical pulse CVT-the rotational swashplate pulse CVT with driven by helical gear axial meshing. This transmission is simple and compact in structure and low in pulsatile rate (it adopts 6 guide rods), and the pulsatile degree is irrelevant to the transmission ratio. Theoretically, pulsatile rate could be reduced to zero if appropriate curved surface of the swashplate is used. Compared with the connecting rod pulse CVT, the present structure uses helical gear mechanism as transmission part and it avoids unbalanced inertial force in the former model. This paper analyzes the principle of driving of this transmission, presents its mechanical structure, and discusses its motion characteristics. Experimental prototype of this type of CVT has been manufactured. Tests for the transmission efficiency(when the rotational speed of the output shaft is the maximum) and the angular velocity of the output shaft have been carried out, and data have been analyzed. The experimental results show that the speed of the output shaft for the experimental prototype is slightly lower than the theoretical value, and the transmission efficiency of the experimental prototype is about 70%. The pulse degree of the CVT discussed in this paper is less than the existing pulse CVT of other types, and it is irrelevant to the transmission ratio of the CVT. 7High-resolution microendoscopy for esophageal cancer screening in China: A cost-effectiveness analysis显示文摘AIM:To study the cost-effectiveness of high-resolution microendoscopy(HRME)in an esophageal squamous cell carcinoma(ESCC)screening program in China.METHODS:A decision analytic Markov model of ESCC was developed.Separate model analyses were conducted for cohorts consisting of an averagerisk population or a high-risk population in China.Hypothetical 50-year-old individuals were followed until age 80 or death.We compared three different strategies for both cohorts:(1)no screening;(2)standard endoscopic screening with Lugol’s iodine staining;and(3)endoscopic screening with Lugol’s iodine staining and an HRME.Model parameters were estimated from the literature as well as from GLOBOCAN,the Cancer Incidence and Mortality Worldwide cancer database.Health states in the model included non-neoplasia,mild dysplasia,moderate dysplasia,high-grade dysplasia,intramucosal carcinoma,operable cancer,inoperable cancer,and death.Separate ESCC incidence transition rates were generated for the average-risk and high-risk populations.Costs in Chinese currency were converted to international dollars(I$)and were adjusted to 2012dollars using the Consumer Price Index.RESULTS:The main outcome measurements for this study were quality-adjusted life years(QALYs)and incremental cost-effectiveness ratio(ICER).For the average-risk population,the HRME screening strategy produced 0.043 more QALYs than the no screening strategy at an additional cost of I$646,resulting in an ICER of I$11808 per QALY gained.Standard endoscopic screening was weakly dominated.Among the high-risk population,when the HRME screening strategy was compared with the standard screening strategy,the ICER was I$8173 per QALY.For both the high-risk and average-risk screening populations,the HRME screening strategy appeared to be the most cost-effective strategy,producing ICERs below the willingness-topay threshold,I$23500 per QALY.One-way sensitivity analysis showed that,for the average-risk population,higher specificity of Lugol’s iodine(>40%)and lower specificity of HRME(<70%)could make Lugol’s iodine screening cost-effective.For the high-risk population,the results of the model were not substantially affected by varying the follow-up rate after Lugol’s iodine screening,Lugol’s iodine test characteristics(sensitivity and specificity),or HRME specificity.CONCLUSION:The incorporation of HRME into an ESCC screening program could be cost-effective in China.Larger studies of HRME performance are needed to confirm these findings.Chin Hur Sung Eun Choi Chung Yin Kong Gui-Qi Wang Hong Xu Alexandros D Polydorides Li-Yan Xue Katherine E Perzan Angela C Tramontano Rebecca R Richards-Kortum Sharmila Anandasabapathy 2015World Journal of Gastroenterology2015,21,18:8
8Toxicity of Graphene Quantum Dots in Zebrafish Embryo显示文摘Objective To evaluate the bio-safety of graphene quantum dots(GQDs), we studied its effects on the embryonic development of zebrafish. Methods In vivo, biodistribution and the developmental toxicity of GQDs were investigated in embryonic zebrafish at exposure concentrations ranging from 12.5-200 μg/m L for 4-96 h post-fertilization(hpf). The mortality, hatch rate, malformation, heart rate, GQDs uptake, spontaneous movement, and larval behavior were examined. Results The fluorescence of GQDs was mainly localized in the intestines and heart. As the exposure concentration increased, the hatch and heart rate decreased, accompanied by an increase in mortality. Exposure to a high level of GQDs(200 μg/mL) resulted in various embryonic malformations including pericardial edema, vitelline cyst, bent spine, and bent tail. The spontaneous movement significantly decreased after exposure to GQDs at concentrations of 50, 100, and 200 μg/mL. The larval behavior testing(visible light test) showed that the total swimming distance and speed decreased dose-dependently. Embryos exposed to 12.5 μg/mL showed hyperactivity while exposure to higher concentrations(25, 50, 100, and 200 μg/mL) caused remarkable hypoactivity in the light-dark test. Conclusion Low concentrations of GQDs were relatively non-toxic. 10Highly sensitive solar-blind deep ultraviolet photodetector based on graphene/PtSe_(2)/β-Ga_(2)O_(3)2D/3D Schottky junction with ultrafast speed显示文摘There is an emerging need for high-sensitivity solar-blind deep ultraviolet(DUV)photodetectors with an ultra-fast response speed.Although nanoscale devices based on Ga_(2)O_(3)nanostructures have been developed,their practical applications are greatly limited by their slow response speed as well as low specific detectivity.Here,the successful fabrication of two-/three-dimensional(2D/3D)graphene(Gr)/PtSe2/β-Ga_(2)O_(3)Schottky junction devices for high-sensitivity solar-blind DUV photodetectors is demonstrated.Benefitting from the high-quality 2D/3D Schottky junction,the vertically stacked structure,and the superior-quality transparent graphene electrode for effective carrier collection,the photodetector is highly sensitive to DUV light illumination and achieves a high responsivity of 76.2 mA/W,a large on/off current ratio of~105,along with an ultra-high ultraviolet(UV)/visible rejection ratio of 1.8×104.More importantly,it has an ultra-fast response time of 12µs and a remarkable specific detectivity of~1013 Jones.Finally,an excellent DUV imaging capability has been identified based on the Gr/PtSe2/β-Ga_(2)O_(3)Schottky junction photodetector,demonstrating its great potential application in DUV imaging systems.Di Wu Zhihui Zhao Wei Lu Lukas Rogée Longhui Zeng Pei Lin Zhifeng Shi Yongtao Tian Xinjian Li Yuen Hong Tsang 2021Nano Research2021,14,6:4

12Efficacy and Safety of Yanggan Jian in Hepatitis B Virusrelated Decompensated Cirrhosis:A Randomized,Doubleblind,Controlled Trial显示文摘Background and Aims:The aim was to evaluate the efficacy and safety of Yanggan Jian(YGJ)in HBV-infected patients with decompensated cirrhosis.Methods:This randomized,double-blind controlled trial enrolled 160 patients with HBV-related decompensated cirrhosis who were already receiving or about to start antiviral therapy.Patients were randomly assigned to receive YGJ or placebo for 24 weeks,and were followed-up to 36 weeks.The primary outcome was the proportion of patients with a≥2 point reduction in Child-Turcotte-Pugh(CTP)score from baseline at week 24.Secondary outcomes were CTP class and score,serum liver function indices,mortality,incidence of hepatocellular carcinoma and variceal bleeding.Results:The proportion of patients with a CTP score reduction≥2 was significantly greater in the YGJ than in the placebo group(p=0.009);the percentage of patients with CTP class C was significantly less than that in the placebo group(p<0.05),and the YGJ group had a significantly greater mean change from baseline in CTP score at week 24(p=0.034).The improvement in measured values and change from baseline of prothrombin time,serum albumin,platelets,cholinesterase,international normalized ratio,and activated partial thromboplastin time were significantly better with YGJ than with placebo.Between-group differences in cumulative rates of variceal bleeding,hepatocellular carcinoma,death,or the frequency of any adverse event(AE),AEs related to treatment,or discontinuation because of AEs were not significant.Conclusions:YGJ significantly improved CTP scores and hepatic synthetic and reserve function in patients with HBV-related decompensated cirrhosis,and was safe and well tolerated.Long Chen Chaoqun Zhao Weili Yao Wei Liu Hua Zhang Yongping Mu Hong Cai Dongying Xue Chengbao Wang Wan’e Wang Yuehong Lin Jiamei Chen Ping Liu 2023Journal of Clinical and Translational Hepatology2023,11,1:3
13Dexamethasone mediates protection against acute pancreatitis via upregulation of pancreatitis-associated proteins显示文摘AIM: To examine the influence of dexamethasone on pancreatitis-associated protein (PAP) gene expression using both in vitro and in vivo models of acute pancreati- tis and to study how PAP gene expression correlates with severity of pancreatitis. METHODS: In vitro, IL-6 stimulated pancreas acinar AR42J cells were cultured with increasing concentrations of dexamethasone and assayed for PAP expression (RT-PCR). In vivo , pancreatitis was induced in rats by retrograde injection of 40 g/L taurocholate into the pancreatic duct. Animals were pretreated with dexamethasone (2 mg/kg) daily or saline for 4 d. Pancreata and serum were harvested after 24 h and gene expression levels of PAPⅠ, Ⅱ and Ⅲ were measured by RT-PCR. Severity of pancreatitis was based on serum amylase, pancreatic wet weight, and histopathological score. RESULTS: In vitro, dexamethasone and IL-6 induced a marked transcription of PAPⅠ, Ⅱ and Ⅲ genes in AR42J cells at 24 h (P < 0.05 for all comparisons). In vivo, pancreas mRNA levels of PAPⅠ, Ⅱ or Ⅲ increased by 2.6-fold, 1.9-fold, and 1.3-fold respectively after dexa- methasone treatment, compared with saline treated ani- mals. Serum amylase levels and edema were significantly lower in the dexamethasone group compared with the saline group. Histopathologic evaluation revealed less inflammation and necrosis in pancreata obtained from dexamethasone treated animals (P < 0.05). CONCLUSION: Dexamethasone significantly decreases the severity of pancreatitis. 14Outcomes after arthroscopic repair of rotator cuff tears in the setting of mild to moderate glenohumeral osteoarthritis显示文摘BACKGROUND Rotator cuff pathology is a very common source of shoulder pain.Similarly,osteoarthritis of the glenohumeral joint can cause shoulder pain and produce similar symptoms.Surgical management can be indicated for both pathologies,however,outcomes data is limited when examining rotator cuff repair(RCR) in the setting of glenohumeral arthritis(GHOA).Thus,this study sought to determine outcomes for patients who undergo RCR in the setting of GHOA.AIM To evaluate if a relationship exists between outcomes of RCR in the setting of GHOA.METHODS This was a retrospective analysis of patients who underwent arthroscopic rotator cuff repair with concurrent glenohumeral osteoarthritis between 2010-2017.Patients were stratified based on rotator cuff tear size and glenohumeral osteoarthritis severity.Cohorts were paired 1:1 with patients without glenohumeral osteoarthritis.Patients included had a minimum two year follow-up.Rate of conversion to total shoulder arthroplasty,complication rates following initial surgery,and patient-reported outcome measures were collected.RESULTS A total of 142 patients were included.The number of patients that required total shoulder arthroplasty within two years after index surgery was low.2/71(2.8%) patients with GHOA,and 1/71(1.4%) without GHOA.Following rotator cuff repair,both groups showed favorable patientreported outcomes.CONCLUSION Patients with glenohumeral osteoarthritis who underwent arthroscopic rotator cuff repair showed comparable outcomes to patients without glenohumeral osteoarthritis.Ian S Hong Allison J Rao Tyler L CarlLee Joshua D Meade Daniel J Hurwit Gregory Scarola David P Trofa Shadley C Schiffern Nady Hamid Patrick M Connor James E Fleischli Bryan Michael Saltzman 2022World Journal of Orthopedics2022,13,7:2
15MODELING OF SEDIMENT AND NONPOINT SOURCE POLLUTANT YIELD显示文摘For water and soil conservation and water pollution control, it is very important to simulate and predict the load of sediment and pollutant during storm-runoff. On the basis of analyzing the simultaneous measurements of flow, sediment and pollutants observed at watershed outlet, a practical sediment yield model is developed by standardizing the load rate. The results show that the standardized pollutant yield equals effective rainfall and the process of effective load yield is the same as effective rainfall hyetograph.
