1Double detonation drivers for a shock tube/tunnel显示文摘ARecent progress on detonation drivers is reviewed. Performances of the forward detonation driver and backward detonation driver have been observed. To eliminate occurrence of a Taylor wave following the detonation wave in the primary driver and to improve the performance of the detonation driver, an additional backward detonation driver was proposed to attach to the end of the forward detonation driver. When the ratio of the initial pressures between the additional and the primary drivers becomes larger than or equal to a critical value, the Taylor wave will disappear, and thus a homogeneous driving gas with high pressure and high temperature can be generated. 2LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus.Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation.In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional program.These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development.Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang 2022Protein & Cell2022,13,7:4
3Expression of Livin in tissues of lung cancer and its correlation with the expression of caspase-3显示文摘Objective: To study the expressions of two isoforms of Livin in tissues of lung cancer and their relations to histological types and chemotherapy, and to study their correlations to the expression of caspase-3 as well. Methods: Ex-pressions of Livin isoforms α, β and caspase-3 were detected by reverse transcription polymerase chain reaction (RT-PCR) assay in lung cancer tissues as well as in controls. Results: Livin isoforms α and β were expressed in 12 of 27, and 19 of 27 lung cancer tissues respectively, much more than those in lung para-cancerous [both were (0/6)] or benign disease lung tissues (0/12, 1/12; P < 0.01 and P < 0.01). Moreover, they were detected in 7/14, 9 /14 lung adenocarcinomas and 4/12, 9/12 squamocellular and large cell carcinomas, respectively, and both showed expressions in one small cell carcinoma. The levels of these two isoforms in lung cancer were significantly higher than those in controls by Gel imaging system (P < 0.05 and P < 0.05), the former was higher in adenocarcinoma than that in squamocellular carcinoma (P < 0.05), while the latter was the same in both (P > 0.05). Meanwhile, the levels of caspase-3 in lung cancer were significantly lower than those in controls, and it was suggested to be negatively associated with either each of two isoforms or their sum (P < 0.05, P < 0.01 and P < 0.01). Two isoforms of Livin expression seemed to increase after chemotherapy but not related to clinical stages (P > 0.05). Conclusion: Two isoforms of Livin are differently expressed in different histological types of lung cancer and may contribute to corresponding cancerous development; the levels of Livin are negatively associated with those of caspase-3, this may be due to the fact that Livin could resist against apoptosis; high expression of Livin seems to be related to chemotherapy but not clinical stages.Hongru Li Yusheng Chen Gang Chen Baosong Xie Lifang Lin 2008The Chinese-German Journal of Clinical Oncology2008,7,7:3

7A potent PGK1 antagonist reveals PGK1 regulates the production of IL-1βand IL-6显示文摘Glycolytic metabolism enzymes have been implicated in the immunometabolism field through changes in metabolic status. PGK1 is a catalytic enzyme in the glycolytic pathway. Here, we set up a high-throughput screen platform to identify PGK1 inhibitors. DC-PGKI is an ATPcompetitive inhibitor of PGK1 with an affinity of Kd= 99.08 nmol/L. DC-PGKI stabilizes PGK1in vitro and in vivo, and suppresses both glycolytic activity and the kinase function of PGK1. In addition,DC-PGKI unveils that PGK1 regulates production of IL-1β and IL-6 in LPS-stimulated macrophages.Mechanistically, inhibition of PGK1 with DC-PGKI results in NRF2(nuclear factor-erythroid factor 2-related factor 2, NFE2L2) accumulation, then NRF2 translocates to the nucleus and binds to the proximity region of Il-1β and Il-6 genes, and inhibits LPS-induced expression of these genes. DC-PGKI ameliorates colitis in the dextran sulfate sodium(DSS)-induced colitis mouse model. 15Coseismic deformation of the 2021 M_(W)7.4 Maduo earthquake from joint inversion of InSAR, GPS, and teleseismic data显示文摘The M_(W)7.4 Maduo earthquake occurred on 22 May 2021 at 02:04 CST with a large-expansion surface rupture.This earthquake was located in the Bayan Har block at the eastern Tibetan Plateau,where eight earthquakes of M_(S)>7.0 have occurred in the past 25 years.Here,we combined interferometric synthetic aperture radar,GPS,and teleseismic data to study the coseismic slip distribution,fault geometry,and dynamic source rupture process of the Maduo earthquake.We found that the overall coseismic deformation field of the Maduo earthquake is distributed in the NWW-SEE direction along 285°.There was slight bending at the western end and two branches at the eastern end.The maximum slip is located near the eastern bending area on the northern branch of the fault system.The rupture nucleated on the Jiangcuo fault and propagated approximately 160 km along-strike in both the NWW and SEE directions.The characteristic source rupture process of the Maduo earthquake is similar to that of the 2010 M_(W)6.8 Yushu earthquake,indicating that similar earthquakes with large-expansion surface ruptures and small shallow slip deficits can occur on both the internal fault and boundary fault of the Bayan Har block.Chaoya Liu Ling Bai Shunying Hong Yanfang Dong Yong Jiang Hongru Li Huili Zhan Zhiwen Chen 2021Earthquake Science2021,34,5:1

20Design and experiments of 4CJ-1200 self-propelled tea plucking machine显示文摘As agricultural labor is decreasing,famous tea production is nearly hard to sustain and is getting more and more expensive in China.To improve the mechanization of tea plucking,this paper designs a self-propelled track-type tea plucking machine that integrates mechanic,hydraulic and electronics.The main parameters of chassis were optimized based on conditions of tea garden.And,three different kinds of cutter blades were designed and analyzed about vibration performance with finite element software.Also,a rigid-flex mixed motion model was established.Based on the model and plots,it is explicit that:(1)the blades move in a sine law;(2)the blades take a symmetrical deform from middle to end while moving;(3)the deformation increases along the blade from the center to sides.The single factor experiment revealed that both travel speed and speed ratio of cutting and travelling had a significant effect on quality indexes(integrity rate,unpicking rate,stubble unevenness)of plucked tea leaf.By universal rotary combination experiment of quadratic regression,the regression equations of three indexes about two experimental factors were established.And,the comprehensive optimization of three indexes was conducted with software Design expert.The optimal operating parameters of machine are as follow:travel speed is 0.41 m/s,speed ratio of cutting and travelling is 1.06;prediction of three indexes are that integrity rate could be reach to 78.26%,unpicking rate reduced to 0.82%,stubble unevenness reduced to 1.30 mm.The results of verification experiment are that:integrity rate could be reach to 77.41%,unpicking rate reduced to 0.87%,stubble unevenness reduced to 1.23 mm.These test indexes that are very close to the predicted values,all surpass the requests specified in the standard of tea plucking with machine.In all,the optimized parameter set is indeed synthetically optimal one that enables the wide use of this tea plucking machine,which can be used in the tea garden that has a lengthways slope less than 18°,a crosswise slope less than 20°,and a height of tea trees between 50-120 cm.From above,the tea plucking machine investigated in this paper provides an effective solution to present tea picking problem that is the bottleneck of tea industry.Further studies need to do for making this machine more automated and intelligent.Yu Han Hongru Xiao Zhiyu Song Qiaomin Chen Wenqin Ding Song Mei 2021International Journal of Agricultural and Biological Engineering2021,14,6:0
