1Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. 6Effect of copper on magnetostriction and mechanical properties of TbDyFe alloys显示文摘The intrinsic brittleness of the TbDyFe alloy significantly decreases its mach inability and applications.This paper aims to improve the toughness of TbDyFe alloy by adding Cu. Various alloys of the type(Tb_(0.3)Dy_(0.7))_(0.37)Fe_(0.63-χ)Cu_χ(χ=0, 0,01.0.03, 0.05, 0.08, 0.1) were fabricated by an arc melting furnace under a high purity argon atmosphere. The microstructure, magnetostrictive properties and mechanical performance were studied systematically. The results show that the (Tb,Dy)Cu phase forms in these alloys upon the addition of Cu. Correspondingly, their toughness improves, attributed to the formation of a (Tb,Dy)Cu phase. Compared to the Cu-free alloy, the fracture toughness (Kic) increases 2-3 times with increasing Cu content. However, the magnetostriction performance of these alloys declines with Cu addition, due to the low-magnetic performance of the (Tb,Dy)Cu phase at room temperature. 18Multi-omics reveals the mechanisms underlying Lactiplantibacillus plantarum P8-mediated attenuation of oxidative stress in broilers challenged with dexamethasone显示文摘Oxidative stress is a common phenomenon in poultry production.Several molecules,including antioxidant genes,miRNAs,and gut microbiota metabolites,have been reported to participate in redox regulation.Lactiplantibacillus plantarum P8(P8)was shown to improve the antioxidant capacity of chickens,but the specific molecular mechanisms remain unclear.In this study,400 broilers were allocated to 4treatment groups:control diet(Con group),control diet+dexamethasone injection(DEX group),control diet containing 1×108CFU/g P8(P8 group),and control diet containing 1×108CFU/g P8+DEX injection(DEX_P8 group).Integrated analysis of the microbiome,metabolomics,and miRNAomics was conducted to investigate the roles of P8 in oxidative stress in broilers.Results demonstrated that P8supplementation significantly improved growth performance,jejunal morphology,and antioxidant function in DEX-treated broilers.Analysis of the gut microbiota revealed a higher abundance of Barnesiella(P=0.01)and Erysipelatoclostridium(P=0.05)in the DEX_P8 group than in the DEX group.Functional prediction indicated that certain pathways,including the phenylacetate degradation pathway,were enriched in the DEX_P8 group compared to the DEX group.Metabolites in the cecal contents were distinct between the groups.P8 supplementation increased the content of metabolites with antioxidant capacity,e.g.,urobilinogen(P<0.01),and decreased that of metabolites related to oxidative stress,e.g.,genistein(P<0.01).Functional prediction indicated that metabolites that differed between the DEX_P8and DEX groups were enriched in pathways including“tryptophan metabolism”and“primary bile acid biosynthesis”.The miRNAomics analysis further showed that,compared to the DEX group,several miRNAs in the jejunum,such as gga-miR-21-3p(P=0.03),were increased,whereas gga-miR-455-3p(P=0.02)was decreased in the DEX_P8 group.The PI3K-Akt,Ras,and Rap1 signaling pathways were enriched in the DEX_P8 group compared to the DEX group through KEGG analysis.Correlation analysis revealed potential interactions between growth performance,oxidation/antioxidation,jejunal morphology,gut microbiota,cecal content metabolites,and jejunal miRNAs.Overall,our results indicate that P8 supplementation may improve the growth performance,jejunal morphology and antioxidant capacity of DEX-treated broilers by regulating gut microbiota,its metabolites,and intestinal miRNAs.Jinshan Zhao Fan Zhao Xuemin Li Junmeng Yuan Kai Zhang Huawei Liu Yang Wang 2023Animal Nutrition2023,,3:0
19Analysis of the genomic landscape of primary central nervous system lymphoma using whole-genome sequencing in Chinese patients显示文摘Primary central nervous system lymphoma(PCNSL)is an uncommon non-Hodgkin’s lymphoma with poor prognosis.This study aimed to depict the genetic landscape of Chinese PCNSLs.Whole-genome sequencing was performed on 68 newly diagnosed Chinese PCNSL samples,whose genomic characteristics and clinicopathologic features were also analyzed.Structural variations were identified in all patients with a mean of 349,which did not significantly influence prognosis.Copy loss occurred in all samples,while gains were detected in 77.9%of the samples.The high level of copy number variations was significantly associated with poor progression-free survival(PFS)and overall survival(OS).A total of 263 genes mutated in coding regions were identified,including 6 newly discovered genes(ROBO2,KMT2C,CXCR4,MYOM2,BCLAF1,and NRXN3)detected in≥10%of the cases.CD79B mutation was significantly associated with lower PFS,TMSB4X mutation and high expression of TMSB4X protein was associated with lower OS.A prognostic risk scoring system was also established for PCNSL,which included Karnofsky performance status and six mutated genes(BRD4,EBF1,BTG1,CCND3,STAG2,and TMSB4X).Collectively,this study comprehensively reveals the genomic landscape of newly diagnosed Chinese PCNSLs,thereby enriching the present understanding of the genetic mechanisms of PCNSL.Xianggui Yuan Teng Yu Jianzhi Zhao Huawei Jiang Yuanyuan Hao Wen Lei Yun Liang Baizhou Li Wenbin Qian 2023Frontiers of Medicine2023,17,5:0
20Fabrication of lotus-type porous micro-channel copper by single-mold Gasar technique显示文摘A single-mold Gasar technique was developed to produce lotus-type porous micro-channel copper with uniform porous structure. In this paper the effect of withdrawal rate on the solid/liquid interface morphology and the corresponding porous structure was systematically investigated, especially the pore morphology, pore growth direction, porosity, and pore diameter of porous copper ingots. In addition, a temperature fi eld simulation was carried out based on Pro Cast software to investigate the shape and movement velocity of the solidifying solid/liquid interface. The experimental results show that the solidifi cation interface changes from convex to planar, then to concave shape with an increase in withdrawal rate. The average porosities of copper ingots are constant and independent of the withdrawal rate.
