1Cryogenic 3D printing of dual-delivery scaffolds for improved bone regeneration with enhanced vascularization显示文摘Three-dimensional(3D)printing has been increasingly employed to produce advanced bone tissue engineering scaffolds with biomimetic structures and matched mechanical strengths,in order to induce improved bone regeneration in defects with a critical size.Given that the successful bone regeneration requires both excellent osteogenesis and vascularization,endowing scaffolds with both strong bone forming ability and favorable angiogenic potential would be highly desirable to induce improved bone regeneration with required vascularization.In this investigation,customized bone tissue engineering scaffolds with balanced osteoconductivity/osteoinductivity were produced via cryogenic 3D printing ofβ-tricalcium phosphate and osteogenic peptide(OP)containing water/poly(lactic-co-glycolic acid)/dichloromethane emulsion inks.The fabricated scaffolds had a hierarchically porous structure and were mechanically comparable to human cancellous bone.Angiogenic peptide(AP)containing collagen I hydrogel was then coated on scaffold surface to further provide scaffolds with angiogenic capability.A sequential release with a quick AP release and a slow but sustained OP release was obtained for the scaffolds.Both rat endothelial cells(ECs)and rat bone marrow derived mesenchymal stem cells(MSCs)showed high viability on scaffolds.Improved in vitro migration and angiogenesis of ECs were obtained for scaffolds delivered with AP while enhanced osteogenic differentiation was observed in scaffolds containing OP.The in vivo results showed that,toward scaffolds containing both AP and OP,the quick release of AP induced obvious angiogenesis in vivo,while the sustained OP release significantly improved the new bone formation.This study provides a facile method to produce dual-delivery scaffolds to achieve multiple functions.Chong Wang Jiahui Lai Kai Li Shaokui Zhu Bingheng Lu Jia Liu Yujin Tang Yen Wei 2021Bioactive Materials2021,6,1:7

4Molecular Characteristics and Phylogenetic Analysis of N Gene of Human Derived Rabies Virus显示文摘Objective To investigate the relationship between the molecular characteristics and phylogenetic evolution of rabies N gene.Methods Saliva samples were collected from rabies cases,and RT-PCR was used to amplify the N gene of rabies virus with the specific primers.The amplifying product of RT-PCR was cloned to pUCm-T vector and transformed into E.coli XL1-Blue and then the blue-white selection,PCR screening and gene sequencing were carried out to identify the positive clones.Finally,ExPASy and other bioinformatics softwares were used to analyze and predict the structure and biological characteristics of the N genome.Results The amplification product of RT-PCR was 1 353 bp,the recombinant plasmid pUCm-T/N was constructed,the whole length of the N gene open reading frame was composed of 1 353 nucleotide residues to code 450 amino acids (20 kinds),the accession number submitted to the Genbank was HM756692,its sequence homology of nucleotides and amino acids compared with the vaccine strain CTN-1-V was 90% and 99% respectively.The evolutionary analysis showed that the isolated strain belonged to genotype I with certain geographic regionality.Conclusion The characteristics investigation and bioinformatics analysis of Hunan0806 N gene will provide fundament data to reveal the significance of the N gene characteristics for rabies epidemiology and its prevention & control.CAI Liang TAO XiaoYan LIU YunZhi ZHANG Hong GAO LiDong HU ShiXiong LIU FuQiang LI Hao SHEN XinXin LIU JiaHui WANG ShiQing TANG Qing 2011Biomedical and Environmental Sciences2011,24,4:4

6Myristica fragrans promotes ABCA1 expression and cholesterol efflux in THP-1-derived macrophages显示文摘Myristica fragrans is a traditional herbal medicine and has been shown to alleviate the development of atherosclerosis.However,the anti-atherogenic mechanisms of M.fragrans are still to be addressed.In this study,we explored the effect of M.fragrans on lipid metabolism and inflammation and its mechanisms in THP-1-derived macrophages.The quantitative polymerase chain reaction and western blot analysis results showed that M.fragrans promotes cholesterol efflux from THP-1-derived macrophages and reduces intracellular total cholesterol,cholesterol ester,and free cholesterol contents in a dose-and a time-dependent manner.Further study found that liver X receptor alpha(LXRα)antagonist GGPP significantly blocked the upregulation of ABCA1 expression with M.fragrans treatment.In addition,chromatin immunoprecipitation assay confirmed that GATA binding protein 3(GATA3)can bind to the LXRαpromoter,and inhibition of GATA3 led to the downregulation of LXRαand ATP-binding cassette subfamily A member 1 expression.Furthermore,M.fragrans reduced lipid accumulation,followed by decreasing tumor necrosis factor-α,interleukin(IL)-6,and IL-1βand increasing IL-10 produced by THP-1-derived macrophages.Therefore,M.fragrans is identified as a valuable therapeutic medicine for atherosclerotic cardiovascular disease.Shangming Liu Jiahui Gao Linhao He Zhenwang Zhao Gang Wang Jin Zou Li Zhou Xiangjun Wan Shilin Tang Chaoke Tang 2021Acta Biochimica et Biophysica Sinica2021,53,1:2
7Silica nanoparticle is a possible safe carrier for gene therapy显示文摘In order to develop a safe and effective gene therapy carrier, some toxicological and biodynamical ex- periments were carried out on silica nanoparticles (SiNPs). First we prepared SiNPs with appropriate portions of cyclo- hexane, deionized water and ethyl silicate, and then trans- fected the modified SiNPs and GFP plasmid DNA complex into the HT1080 cells to test the effectiveness of transfection for gene therapy. At the same time, we injected the SiNPs into a number of mice through tail vein. Then we made the mice crossed to evaluate the acute, long-term and reproduc- tive toxicity. In vivo distribution analysis and pathological examination were made on both adult mice and their off- spring. SiNPs were uniform and had an average diameter of 40 nm, and the modified SiNPs carried exogenous DNA molecules into target cells and the transferred GFP fusion gene was effectively expressed in the cells. The SiNPs injected via tail vein were widely distributed in almost all of tissues, and the injected mice had the ability to reproduce normally. 8Identification and characterization of human neuronal voltage-gated calcium channel gamma 3 subunit gene显示文摘By homologous expressed sequence tag (EST) searching, one EST (GenBank: W29095) was obtained, which shows 75% identity in 435 bp overlap with the coding sequence of mouse Cacng2 gene. A 1 545 bp cDNA fragment was obtained from the nested polymerase chain reaction (PCR) and rapid applification of cDNA end (RACE) reaction in the human brain prefrontal cortex cDNA library and the human brain Ready cDNA with the primers designed on W29095. The fragment contained a 948-bp open reading frame (ORF) encoding 315 amino acids, and was named CACNG3. As it was identical to a BAC clone (GenBank: AC004125) from chromosome 16p12-p13.1, the CACNG3 gene was mapped to human chromosome 16p12-p13.1, and the coding region was composed of 4 exons. Reverse transcription PCR (RT-PCR) analysis showed that the CACNG3 gene expressed in human adult brain and fetal brain. 10Placement of dual capsular tension rings for the combined management of traumatic cyclodialysis cleft and zonular dialysis显示文摘Background:To evaluate the efficacy and safety of placing dual capsular tension rings for the combined management of traumatic cyclodialysis cleft and zonular dialysis.Methods:A modified capsular tension ring was inserted into the ciliary sulcus and a capsular tension ring or modified capsular tension ring was inserted into the capsular bag in 20 eyes in 20 consecutive patients showing signs of ocular hypotony and ectopia lentis.Outcome measures included intraocular pressure,best-corrected visual acuity,and postoperative complications.Results:Dual capsular tension ring placement was performed in 20 patients with a mean age of 48.7 years.The cyclodialysis cleft extended over 2.9 clock hours(range 0.5-6.5).The modified capsular tension ring was successfully inserted into the ciliary sulcus and a capsular tension ring or modified capsular tension ring in the capsular bag in all eyes.At the last follow-up,the cyclodialysis cleft was closed in 16/20(80.0%)eyes.The intraocular lens was stable in all patients postoperatively.Best-corrected visual acuity,in terms of the logarithm of the minimal angle of resolution,improved from 1.3±0.8 before surgery to 0.4±0.3 after surgery(P<0.001).Intraocular pressure increased significantly from 10.6±3.2 mmHg before surgery to 13.0±4.8 mmHg after surgery(P=0.040).Postoperative complications included a painful reversible intraocular pressure spike in four patients(20.0%).Logistic regression revealed no significant factors associated with successful cleft closure and a stable final intraocular pressure of≥10 mmHg.Conclusions:The placement of two capsular tension rings into the ciliary sulcus and the capsular bag is a safe,successful procedure combined for repairing a traumatic cyclodialysis cleft and managing zonular dialysis.Jiahui Chen Lina Lan Yating Tang Yi Lu Yongxiang Jiang 2020Eye and Vision2020,7,1:1

13Waste to wealth: Oxygen-nitrogen-sulfur codoped lignin-derived carbon microspheres from hazardous black liquors for high-performance DSSCs显示文摘Carbon materials are effective substitutes for Pt counter electrodes(CEs) in dye-sensitized solar cells(DSSCs). However, many of these materials, such as carbon nanotubes and graphene, are expensive and require complex preparation process. Herein, waste lignin, recycled from hazardous black liquors,is used to create oxygen-nitrogen-sulfur codoped carbon microspheres for use in DSSC CEs through the facile process of low-temperature preoxidation and high-temperature self-activation. The large number of ester bonds formed by preoxidation increase the degree of cross-linking of the lignin chains, leading to the formation of highly disordered carbon with ample defect sites during pyrolysis. The presence of organic O/N/S components in the waste lignin results in high O/N/S doping of the pyrolysed carbon,which increases the electrolyte ion adsorption and accelerates the electron transfer at the CE/electrolyte interface, as confirmed by density functional theory(DFT) calculations. The presence of inorganic impurities enables the construction of a hierarchical micropore-rich carbon structure through the etching effect during self-activation, which can provide abundant catalytically active sites for the reversible adsorption/desorption of electrolyte ions. Under these synergistic effects, the DSSCs that use this novel carbon CE achieve a quite high power-conversion efficiency of 9.22%. 18Gut microbiota links with cognitive impairment in amyotrophic lateral sclerosis: A multi-omics study显示文摘Recently, cognitive impairments(CI) and behavioral abnormalities in patients with amyotrophic lateral sclerosis(ALS) have been reported. However, the underlying mechanisms have been poorly understood. In the current study, we explored the role of gut microbiota in CI of ALS patients. We collected fecal samples from 35ALS patients and 35 healthy controls. The cognitive function of the ALS patients was evaluated using the Edinburgh Cognitive and Behavioral ALS Screen. We analyzed these samples by using 16S rRNA gene sequencing as well as both untargeted and targeted(bile acids) metabolite mapping between patients with CI and patients with normal cognition(CN). We found altered gut microbial communities and a lower ratio of Firmicutes/Bacteroidetes in the CI group, compared with the CN group. In addition, the untargeted metabolite mapping revealed that 26 and 17 metabolites significantly increased and decreased, respectively, in the CI group, compared with the CN group. These metabolites were mapped to the metabolic pathways associated with bile acids. We further found that cholic acid and chenodeoxycholic acid were significantly lower in the CI group than in the CN group. In conclusion, we found that the gut microbiota and its metabolome profile differed between ALS patients with and without CI and that the altered bile acid profile in fecal samples was significantly associated with CI in ALS patients. 19Phenotype and metabolism alterations in PCB-degrading Rhodococcus biphenylivorans TG9^(T) under acid stress显示文摘Environmental acidification impairs microorganism diversity and their functions on substance transformation.Rhodococcus is a ubiquitously distributed genus for contaminant detoxification in the environment,and it can also adapt a certain range of pH.This work interpreted the acid responses from both phenotype and metabolism in strain Rhodococcus biphenylivorans TG9^(T)(TG9)induced at pH 3.The phenotype alterations were described with the number of culturable and viable cells,intracellular ATP concentrations,cell shape and entocyte,degradation efficiency of polychlorinated biphenyl(PCB)31 and biphenyl.The number of culturable cells maintained rather stable within the first 10 days,even though the other phenotypes had noticeable alterations,indicating that TG9 possesses certain capacities to survive under acid stress.The metabolism responses were interpreted based on transcription analyses with four treatments including log phase(LP),acid-induced(PER),early recovery after removing acid(RE)and later recovery(REL).With the overview on the expression regulations among the 4 treatments,the RE sample presented more upregulated and less downregulated genes,suggesting that its metabolism was somehow more active after recovering from acid stress.In addition,the response mechanism was interpreted on 10 individual metabolism pathways mainly covering protein modification,antioxidation,antipermeability,H+consumption,neutralization and extrusion.Furthermore,the transcription variations were verified with RT-qPCR on 8 genes with 24-hr,48-hr and 72-hr acid treatment.Taken together,TG9 possesses comprehensive metabolism strategies defending against acid stress.Consequently,a model was built to provide an integrate insight to understand the acid resistance/tolerance metabolisms in microorganisms.Aili Li Jiahui Fan Yangyang Jia Xianjin Tang Jingwen Chen Chaofeng Shen 2023Journal of Environmental Sciences2023,,5:0
20A new modeling approach for stress-strain relationship taking into account strain hardening and stored energy by compacted graphite iron evolution显示文摘Compacted graphite iron(CGI)is considered to be an ideal diesel engine material with excellent physical and mechanical properties,which meet the requirements of energy conservation and emission reduction.However,knowledge of the microstructure evolution of CGI and its impact on flow stress remains limited.In this study,a new modeling approach for the stress–strain relationship is proposed by considering the strain hardening effect and stored energy caused by the microstructure evolution of CGI.The effects of strain,strain rate,and deformation temperature on the microstructure of CGI during compression deformation are examined,including the evolution of graphite morphology and the microstructure of the pearlite matrix.The roundness and fractal dimension of graphite particles under different deformation conditions are measured.Combined with finite element simulation models,the influence of graphite particles on the flow stress of CGI is determined.The distributions of grain boundary and geometrically necessary dislocations(GNDs)density in the pearlite matrix of CGI under different strains,strain rates,and deformation temperatures are analyzed by electron backscatter diffraction technology,and the stored energy under each deformation condition is calculated.Results show that the proportion and amount of low-angle grain boundaries and the average GNDs density increase with the increase of strain and strain rate and decreased first and then increased with an increase in deformation temperature.The increase in strain and strain rate and the decrease in deformation temperature contribute to the accumulation of stored energy,which show similar variation trends to those of GNDs density.The parameters in the stress–strain relationship model are solved according to the stored energy under different deformation conditions.The consistency between the predicted results from the proposed stress–strain relationship and the experimental results shows that the evolution of stored energy can accurately predict the stress–strain relationship of CGI.Jiahui NIU Chuanzhen HUANG Zhenyu SHI Hanlian LIU Zhengyi TANG Binghao LI Zhen CHEN Guoyan JIANG 2023Frontiers of Mechanical Engineering2023,18,4:0
