1Mobile Edge Communications, Computing, and Caching(MEC3) Technology in the Maritime Communication Network显示文摘With the increasing maritime activities and the rapidly developing maritime economy, the fifth-generation(5G) mobile communication system is expected to be deployed at the ocean. New technologies need to be explored to meet the requirements of ultra-reliable and low latency communications(URLLC) in the maritime communication network(MCN). Mobile edge computing(MEC) can achieve high energy efficiency in MCN at the cost of suffering from high control plane latency and low reliability. In terms of this issue, the mobile edge communications, computing, and caching(MEC3) technology is proposed to sink mobile computing, network control, and storage to the edge of the network. New methods that enable resource-efficient configurations and reduce redundant data transmissions can enable the reliable implementation of computing-intension and latency-sensitive applications. The key technologies of MEC3 to enable URLLC are analyzed and optimized in MCN. The best response-based offloading algorithm(BROA) is adopted to optimize task offloading. 4Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy.Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang 2023Signal Transduction and Targeted Therapy2023,8,4:2

7Virus-inspired surface-nanoengineered antimicrobial liposome:A potential system to simultaneously achieve high activity and selectivity显示文摘Enveloped viruses such as SARS-CoV-2 frequently have a highly infectious nature and are considered effective natural delivery systems exhibiting high efficiency and specificity.Since simultaneously enhancing the activity and selectivity of lipopeptides is a seemingly unsolvable problem for conventional chemistry and pharmaceutical approaches,we present a biomimetic strategy to construct lipopeptide-based mimics of viral architectures and infections to enhance their antimicrobial efficacy while avoiding side effects.Herein,a surface-nanoengineered antimicrobial liposome(SNAL)is developed with the morphological features of enveloped viruses,including a moderate size range,lipid-based membrane structure,and highly lipopeptide-enriched bilayer surface.The SNAL possesses virus-like infection to bacterial cells,which can mediate high-efficiency and high-selectivity bacteria binding,rapidly attack and invade bacteria via plasma membrane fusion pathway,and induce a local“burst”release of lipopeptide to produce irreversible damage of cell membrane.Remarkably,viral mimics are effective against multiple pathogens with low minimum inhibitory concentrations(1.6-6.3μg mL1),high bactericidal efficiency of>99%within 2 h,>10-fold enhanced selectivity over free lipopeptide,99.8%reduction in skin MRSA load after a single treatment,and negligible toxicity.This bioinspired design has significant potential to enhance the therapeutic efficacy of lipopeptides and may create new opportunities for designing next-generation antimicrobials.Yin Shi Xiaoqian Feng Liming Lin Jing Wang Jiaying Chi Biyuan Wu Guilin Zhou Feiyuan Yu Qian Xu Daojun Liu Guilan Quan Chao Lu Xin Pan Jianfeng Cai Chuanbin Wu 2021Bioactive Materials2021,6,10:1

12Human endoderm stem cells reverse inflammation-related acute liver failure through cystatin SN-mediated inhibition of interferon signaling显示文摘Acute liver failure(ALF)is a life-threatening disease that occurs secondary to drug toxicity,infection or a devastating immune response.Orthotopic liver transplantation is an effective treatment but limited by the shortage of donor organs,the requirement for life-long immune suppression and surgical challenges.Stem cell transplantation is a promising alternative therapy for fulminant liver failure owing to the immunomodulatory abilities of stem cells.Here,we report that when transplanted into the liver,human endoderm stem cells(hEnSCs)that are germ layer-specific and nontumorigenic cells derived from pluripotent stem cells are able to effectively ameliorate hepatic injury in multiple rodent and swine drug-induced ALF models.We demonstrate that hEnSCs tune the local immune microenvironment by skewing macrophages/Kupffer cells towards an anti-inflammatory state and by reducing the infiltrating monocytes/macrophages and inflammatory T helper cells.Single-cell transcriptomic analyses of infiltrating and resident monocytes/macrophages isolated from animal livers revealed dramatic changes,including changes in gene expression that correlated with the change of activation states,and dynamic population heterogeneity among these cells after hEnSC transplantation.We further demonstrate that hEnSCs modulate the activation state of macrophages/Kupffer cells via cystatin SN(CST1)-mediated inhibition of interferon signaling and therefore highlight CST1 as a candidate therapeutic agent for diseases that involve over-activation of interferons.We propose that hEnSC transplantation represents a novel and powerful cell therapeutic treatment for ALF.Yilin Xu Jinglin Wang Haozhen Ren Hao Dai Ying Zhou Xiongzhao Ren Yang Wang Sisi Feng Xiaogang Deng Jiaying Wu Tianlong Fu Tengfei Nie Haifeng He Tongkun Wei Bing Zhu Lijian Hui Bin Li Jing Wang Hongyan Wang Luonan Chen Xiaolei Shi Xin Cheng 2023Cell Research2023,33,2:0
13Normalization of Homophonic Words in Chinese Microblogs显示文摘Homophonic words are very popular in Chinese microblog, posing a new challenge for Chinese microblog text analysis. However, to date, there has been very little research conducted on Chinese homophonic words normalization. In this paper, we take Chinese homophonic word normalization as a process of language decoding and propose an n-gram based approach. To this end, we first employ homophonic–original word or character mapping tables to generate normalization candidates for a given sentence with homophonic words, and thus exploit n-gram language models to decode the best normalization from the candidate set.
