| 2 | PTEN基因甲基化及其表达异常与胃癌的关系(英文)显示文摘Objective:To probe into the relationships between PTEN gene expression,the promoter methylation and gastric cancer and its clinical pathological specific features.Methods:We analyzed the PTEN gene promoter methylation and mRNA expression status in gastric cancer tissues and its adjacent normal tissues by methylation specific PCR(MSP) and reverse transcription-polymerase chain reaction(RT-PCR) techniques.Results:PTEN promoters in 48.2%(27/56) gastric cancer tissues and 3.6(2/56) adjacent normal tissues were methylated and the PTEN promoter methylation rate in carcinoma tissues was obviously higher(P < 0.05).Of the 2 cases where the adjacent gastric tissues were methylated,the gastric cancer tissues were both methylated.Of the 29 gastric cancers with lymph node metastasis,19 had their PTEN gene promoters methylated and the PTEN gene promoter methylation in cases with lymph node metastasis was obviously higher than that without lymph node metastasis(P < 0.05).RT-PCR result showed that no expression of PTEN mRNA existed in any of the methylated gastric cancer tissues.Conclusion:The expression loss of PTEN gene mRNA in gastric cancers is related to their promoter methylation and might be one of the reasons for the generation,development and metastasis of gastric cancers. | Song Liu Jieping Yu Ping Fu Honggang Yu | 2008 | The Chinese-German Journal of Clinical Oncology2008,7,10: | 3 |
| 3 | Extra-copy nifA enhances the nodulation efficiency of Sinorhizobium fredii显示文摘Previous investigations have shown that nifA gene is involved in nodulation and symbiotic nitrogen fixation regulation of Rhizobium. We study the role of nifA on nodulation of leguminous plants. We found that Sinorhizo-bium fredii harboring multi-copy plasmid carrying the con-stitutively expressed Klebsiella pneumoniae nifA exhibited an increase of nodulation activity and nodulation competitiveness on soybean plants. | 9 | Spatial-temporal characteristics of epidemic spread in-out flow——Using SARS epidemic in Beijing as a case study显示文摘For better detecting the spatial-temporal change mode of individual susceptible-infected-symptomatic-treated-recovered epidemic progress and the characteristics of information/material flow in the epidemic spread network between regions,the epidemic spread mechanism of virus input and output was explored based on individuals and spatial regions.Three typical spatial information parameters including working unit/address,onset location and reporting unit were selected and SARS epidemic spread in-out flow in Beijing was defined based on the SARS epidemiological investigation data in China from 2002 to 2003 while its epidemiological characteristics were discussed.Furthermore,by the methods of spatial-temporal statistical analysis and network characteristic analysis,spatial-temporal high-risk hotspots and network structure characteristics of Beijing outer in-out flow were explored,and spatial autocorrelation/heterogeneity,spatial-temporal evolutive rules and structure characteristics of the spread network of Beijing inner in-out flow were comprehensively analyzed.The results show that(1)The outer input flow of SARS epidemic in Beijing concentrated on Shanxi and Guangdong provinces,but the outer output flow was disperse and mainly includes several north provinces such as Guangdong and Shandong.And the control measurement should focus on the early and interim progress of SARS breakout.(2)The inner output cases had significant positive autocorrelative characteristics in the whole studied region,and the high-risk population was young and middle-aged people with ages from 20 to 60 and occupations of medicine and civilian labourer.(3)The downtown districts were main high-risk hotspots of SARS epidemic in Beijing,the northwest suburban districts/counties were secondary high-risk hotspots,and northeast suburban areas were relatively safe.(4)The district/county nodes in inner spread network showed small-world characteristics and information/material flow had notable heterogeneity.The suburban Tongzhou and Changping districts were the underlying high-risk regions,and several suburban districts such as Shunyi and Huairou were the relatively low-risk safe regions as they carried out minority information/material flow.The exploration and analysis based on epidemic spread in-out flow help better detect and discover the potential spatial-temporal evolutive rules and characteristics of SARS epidemic,and provide a more effective theoretical basis for emergency/control measurements and decision-making. | HU BiSong GONG JianHua ZHOU JiePing SUN Jun YANG LiYang XIA Yu Abdoul Nasser IBRAHIM | 2013 | Science China Earth Sciences2013,56,8: | 0 |

| 11 | Overcoming adaptive resistance in AML by synergistically targeting FOXO3A-GNG7-mTOR axis with FOXO3A inhibitor Gardenoside and rapamycin显示文摘Therapeutic targeting FOxO3A(a forkhead transcription factor)represents a prom-ising strategy to suppress acute myeloid leukemia(AML).However,the effective inhibitors that target FOXO3A are lacking and the adaptive response signaling weakens the cytotoxic effect of FOxO3A depletion on AML cells.Here,we show that FOxO3A deficiency induces a compensa-tory response involved in the reactive activation of mTOR that leads to signaling rebound and adaptive resistance.Mitochondrial metabolism acts downstream of mTOR to provoke activa-tion of JNK/c-JUN via reactive oxygen species(ROS).At the molecular level,FOXO3A directly binds to the promoter of G protein gamma subunit 7(GNG7)and preserves its expression,while GNG7 interacts with mTOR and restricts phosphorylated activation of mTOR.Consequently,combinatorial inhibition of FOXO3A and mTOR show a synergistic cytotoxic effect on AML cells and prolongs survival in a mouse model of AML.Through a structure-based virtual screening,we report one potent small-molecule FOxO3A inhibitor(Gardenoside)that exhibits a strong effect of anti-FOXO3A DNA binding.Gardenoside synergizes with rapamycin to substantially reduce tumor burden and extend survival in AML patient-derived xenograft model.These results demonstrate that mTOR can mediate adaptive resistance to FOxO3A inhibition and validate a combinatorial approach for treating AML. | Zhe Chen Qian Guo Shichen Huang Lei Li Feng Wu Zhilong Liu Zhigang Li Tao Chen Guanbin Song Shuangnian Xu Jieping Chen Yu Hou | 2024 | Genes & Diseases2024,11,1: | 0 |
