5Machine learning powered ellipsometry显示文摘Ellipsometry is a powerful method for determining both the optical constants and thickness of thin films.For decades,solutions to ill-posed inverse ellipsometric problems require substantial human-expert intervention and have become essentially human-in-the-loop trial-and-error processes that are not only tedious and time-consuming but also limit the applicability of ellipsometry.Here,we demonstrate a machine learning based approach for solving ellipsometric problems in an unambiguous and fully automatic manner while showing superior performance.The proposed approach is experimentally validated by using a broad range of films covering categories of metals,semiconductors,and dielectrics.This method is compatible with existing ellipsometers and paves the way for realizing the automatic,rapid,high-throughput optical characterization of films.Jinchao Liu Di Zhang Dianqiang Yu Mengxin Ren Jingjun Xu 2021Light(Science & Applications)2021,10,4:1
6Design and fabrication of r-hirudin loaded dissolving microneedle patch for minimally invasive and long-term treatment of thromboembolic disease显示文摘Cardiovascular disease is the leading cause of global mortality,with anticoagulant therapy being the main prevention and treatment strategy.Recombinant hirudin(r-hirudin)is a direct thrombin inhibitor that can potentially prevent thrombosis via subcutaneous(SC)and intravenous(IV)administration,but there is a risk of haemorrhage via SC and IV.Thus,microneedle(MN)provides painless and sanitary alternatives to syringes and oral administration.However,the current technological process for the micro mould is complicated and expensive.The micro mould obtained via three-dimensional(3D)printing is expected to save time and cost,as well as provide a diverse range of MNs.Therefore,we explored a method for MNs array model production based on 3D printing and translate it to micro mould that can be used for fabrication of dissolving MNs patch.The results show that r-hirudin-loaded and hyaluronic acid(HA)-based MNs can achieve transdermal drug delivery and exhibit significant potential in the prevention of thromboembolic disease without bleeding in animal models.These results indicate that based on 3D printing technology,MNs combined with r-hirudin are expected to achieve diverse customizableMNs and thus realize personalized transdermal anticoagulant delivery for minimally invasive and long-term treatment of thrombotic disease.Mengfang Wu Tian Xia Yaran Li Tianfa Wang Shijia Yang Jinchao Yu Qiaoyan Liang Teng Shen Min Yu Bing Zhao 2022Asian Journal of Pharmaceutical Sciences2022,17,2:1

13The effect of drug loading and multiple administration on the protein corona formation and brain delivery property of PEG-PLA nanoparticles显示文摘The presence of protein corona on the surface of nanoparticles modulates their physiological interactions such as cellular association and targeting property.It has been shown that α-mangostin(αM)-loaded poly(ethylene glycol)-poly(l-lactide)(PEG-PLA)nanoparticles(NP-αM)specifically increased low density lipoprotein receptor(LDLR)expression in microglia and improved clearance of amyloid beta(Aβ)after multiple administration.However,how do the nanoparticles cross the blood‒brain barrier and access microglia remain unknown.Here,we studied the brain delivery property of PEG-PLA nanoparticles under different conditions,finding that the nanoparticles exhibited higher brain transport efficiency and microglia uptake efficiency afterαM loading and multiple administration.To reveal the mechanism,we performed proteomic analysis to characterize the composition of protein corona formed under various conditions,finding that both drug loading and multiple dosing affect the composition of protein corona and subsequently influence the cellular uptake of nanoparticles in b.End3 and BV-2 cells.Complement proteins,immunoglobulins,RAB5A and CD36 were found to be enriched in the corona and associated with the process of nanoparticles uptake.Collectively,we bring a mechanistic understanding about the modulator role of protein corona on targeted drug delivery,and provide theoretical basis for engineering brain or microglia-specific targeted delivery system.Yuyun Tang Jinchao Gao Tao Wang Qian Zhang Antian Wang Meng Huang Renhe Yu Hongzhuan Chen Xiaoling Gao 2022Acta Pharmaceutica Sinica B2022,12,4:0

15A highly selective and sensitive upconversion nanoprobe for monitoring hydroxyl radicals in living cells and the liver显示文摘Excessive reactive oxygen species(ROS)would attack living cells and cause a series of oxidative stress related diseases,such as liver damage.Hydroxyl radicals(·OH)are currently known as one of the most toxic and harmful free radicals to organisms.Therefore,studies involving hydroxyl radicals have become important research topics in the fields of biology,biochemistry,and biomedicine.In addition,imaging of analytes using upconversion nanoparticles(UCNPs)possesses significant advantages over that using general fluorescent dyes or nanoparticles due to its high spatial resolution,reduced photodamage,and deep tissue penetration properties.Herein,we designed a highly selective and sensitive hydroxyl radical nanoprobe based on the luminescence resonance energy transfer between upconversion nanoparticles and methylene blue(MB).The concentration of·OH could be determined by the fluorescence recovery of the UCNPs due to the oxidative damage of MB.Using this nanoprobe,the·OH in living cells or in liver tissues could be monitored with high sensitivity and selectivity.Guangshun Yu Na Feng Dan Zhao Hao Wang Yi Jin Dandan Liu Zhenhua Li Xinjian Yang Kun Ge Jinchao Zhang 2021Science China(Life Sciences)2021,64,3:0
