| 5 | Transcriptional repression by androgen receptor: roles in castration-resistant prostate cancer显示文摘Androgen receptor (AR), a hormonal transcription factor, plays important roles during prostate cancer progression and is a key target for therapeutic interventions. While androgen-deprivation therapies are initially successful in regressing prostate tumors, the disease ultimately comes back as castration-resistant prostate cancer (CRPC) or at the late stage as neuroendocrine prostate cancer (NEPC). CRPC remains largely dependent on hyperactive AR signaling in the milieu of low androgen, while NEPC is negative of AR expression but positive of many AR-repressed genes. Recent technological advances in genome-wide analysis of transcription factor binding sites have revealed an unprecedented set of AR target genes. In addition to its well-known function in activating gene expression, AR is increasingly known to also act as a transcriptional repressor. Here, we review the molecular mechanisms by which AR represses gene expression. | 6 | Surface-grafted block copolymer brushes with continuous composition gradients of poly(poly(ethylene glycol)-monomethacrylate) and poly(N-isopropylacrylamide)显示文摘Surface-grafted block copolymer brushes with continuous composition gradients containing poly(poly(ethylene glycol) monomethacrylate) (P(PEGMA)) and poly(N-isopropylacrylamide) (PNIPAAm) chains were fabricated by integration of the surface-initiated atom transfer radical polymerization (SI-ATRP) and continuous injection method.Three types of copolymer gradients were prepared: (1) a uniform P(PEGMA) layer was block copolymerized with a gradient PNIPAAm layer (PP1);(2) a gradient P(PEGMA) layer was block copolymerized with a uniform PNIPAAm layer (PP2);and (3) a gradient P(PEGMA) layer was inversely block copolymerized with a gradient PNIPAAm layer (PP3).The as-prepared gradients were characterized by ellipsometry,water contact angle and atomic force microscopy (AFM) to determine their alterations in thickness,surface wettability and morphology,confirming the gradient structures.In vitro culture of HepG2 cells was implemented on the gradient surfaces,revealing that the cells could adhere at 37 °C and be detached at 20 °C.Introduction of the PEG chains as an underlying layer on the PNIPAAm grafting surfaces resulted in faster cell detachment compared with the PNIPAAm grafting surface. | LI LinHui WU JinDan GAO ChangYou | 2011 | Science China Chemistry2011,54,2: | 3 |
| 7 | Reconfiguring wireless environments via intelligent surfaces for 6G:reflection,modulation,and security显示文摘The reconfigurable intelligent surface(RIS)has been recognized as an essential enabling technology for sixth-generation(6G)mobile communication networks.An RIS comprises a large number of small and low-cost reflecting elements whose parameters can be dynamically adjusted with a programmable controller.Each of these elements can effectively reflect a phase-shifted version of the incident electromagnetic wave.By configuring the wave phases in real time,the propagation environment of the information-bearing signals can be dynamically manipulated to enhance communication reliability,boost transmission rate,expand cellular coverage,and strengthen communication security.In this study,we provide an overview on RIS-assisted wireless communications.Specifically,we elaborate on the state-of-the-art enabling techniques for the RIS technology as well as their corresponding substantial benefits from the perspectives of RIS reflection and RIS modulation.With these benefits,we envision the integration of RISs into emerging applications for 6G.In addition,communication security is of unprecedented importance in future 6G networks with ubiquitous wireless services in multifarious verticals and areas.We highlight potential contributions of RISs to physical-layer security,in terms of secrecy rate and secrecy outage probability,exemplified by a typical case study from both theoretical and numerical aspects.Finally,we discuss challenges and opportunities on the deployment of RISs in practice to motivate future research. | Jindan XU Chau YUEN Chongwen HUANG Naveed UL HASSAN George CALEXANDROPOULOS Marco DI RENZO Mérouane DEBBAH | 2023 | Science China(Information Sciences)2023,66,3: | 3 |
| 8 | Regional Metabolic Patterns of Abnormal Postoperative Behavioral Performance in Aged Mice Assessed by XH-NMR Dynamic Mapping Method显示文摘Abnormal postoperative neurobehavioral performance(APNP)is a common phenomenon in the early postoperative period.The disturbed homeostatic status of metabolites in the brain after anesthesia and surgery might make a significant contribution to APNP.The dynamic changes of metabolites in different brain regions after anesthesia and surgery,as well as their potential association with APNP are still not well understood.Here,we used a battery of behavioral tests to assess the effects of laparotomy under isoflurane anesthesia in aged mice,and investigated the metabolites in 12 different sub-regions of the brain at different time points using proton nuclear magnetic resonance('H-NMR)spectroscopy.The abnormal neurobehavioral performance occurred at 6 h and/or 9 h,and recovered at 24 h after anesthesia/surgery.Compared with the control group,the altered metabolite of the model group at 6 h was aspartate(Asp),and the difference was mainly displayed in the cortex;while significant changes at 9 h occurred predominantly in the cortex and hippocampus,and the corresponding metabolites were Asp and glutamate(Glu).All changes returned to baseline at 24 h.The altered metabolic changes could have occurred as a result of the acute APNP,and the metabolites Asp and Glu in the cortex and hippocampus could provide preliminary evidence for understanding the APNP process. | Taotao Liu Zhengqian Li Jindan He Ning Yang Dengyang Han Yue Li Xuebi Tian Huili Liu Anne Manyande Hongbing Xiang Fuqiang Xu Jie Wang Xiangyang Guo | 2020 | Neuroscience Bulletin2020,36,1: | 2 |
| 9 | Microwave triggered multifunctional nanoplatform for targeted photothermal-chemotherapy in castration-resistant prostate cancer显示文摘Lacking a precise targeting strategy,castration-resistant prostate cancer(CRPC)is still hard to be treat effectively.Exploring treatment options that can accurately target CPRC is an important issue with urgent need.In this study,a novel nanotechnologybased strategy had been developed for the precise target treatment of CRPC.By combining microwaves and photothermal therapy(PTT),this nanoplatform,cmHSP70-PL-AuNC-DOX,targets tumor tissues with outstanding precision and achieves better anti-tumor activity by simultaneously eliciting photothermal and chemotherapeutic effects.From nanotechnology,cmHSP70-modified and thermo-sensitive liposome-coated AuNC-DOX were prepared and used for CRPC-targeted photothermal ablation and chemotherapy.Doxorubicin(DOX)was selected as the chemotherapeutic agent for cytotoxicity.In terms of the curative scheme,prostate tissues were firstly pre-treated with microwaves to induce the expression of heat shock protein 70(HSP70)and its migration to the cell membrane,which was then targeted by HSP70 antibody(cmHSP70)coated on the nanoparticles to achieve accurate drug delivery.The nanoplatform then achieved precise ablation and controlled release of DOX under external near-infrared(NIR)irradiation.Through the implementation,the targeting,cell killing,and safety of this therapeutical strategy had been verified in vivo and in vitro.This work establishes an accurate,controllable,efficient,non-invasive,and safe treatment platform for targeting CRPC,provides a rational design for CRPC’s PTT,and offers new prospects for nanomedicines with great precision. | Pengyuan Liu Yajun Wu Xiaogang Xu Xudong Fan Chuan Sun Xiaojie Chen Jindan Xia Shiting Bai Li Qu Huasong Lu Jing Wu Jun Chen Ji-Gang Piao Zhibing Wu | 2023 | Nano Research2023,16,7: | 2 |
| 10 | Down-regulation of lncRNA LUADT1 suppresses cervical cancer cell growth by sequestering microRNA-1207-5p显示文摘Emerging evidence has proved the essential roles of long non-coding RNAs(lncRNAs)in cervical carcinoma(CC).LncRNA lung adenocarcinoma-associated transcript 1(LUADT1)is overexpressed and plays an oncogenic role in various cancers;however,the function and clinical values of LUADT1 in CC remain unclear.In this study we found that LUADT1 is highly expressed in CC tissues and cells.Up-regulated LUADT1 is significantly correlated with the more aggressive status and poorer survival of CC patients.In vitro studies show that LUADT1 depletion suppresses CC proliferation,and leads to cell apoptosis and cell cycle arrest.Furthermore,the in vivo xenograft mouse assay demonstrates that LUADT1 knockdown remarkably suppresses tumor growth.Mechanistically,LUADT1 binds to miR-1207-5p and inhibits miR-1207-5p expression in CC cells.Septin 9(SEPT9)is identified as a miR-1207-5p target which is negatively regulated by LUADT1.Overexpression of SEPT9 abrogates the suppressed proliferation of CC cells induced by LUADT1 knockdown.These results demonstrate that LUADT1 sponges miR-1207-5p and consequently modulates SEPT9 expression in CC.Our study suggests the possible application of LUADT1 as a prognostic and therapeutic target to inhibit CC. | Qian Zhang Jindan Zheng Lili Liu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,3: | 2 |

Results showed that 5 ×106 MRA markedly reduced the in vitro invasiveness and adhesiveness to the HABM, and the migration of the CCL229 cells. In addi-tion, to elucidate the relation between expression of epider-mal growth factor receptor (EGFR) and the invasiveness of the colorectal carcinoma cells, two well-differentiated, but with different invasiveness colorectal cancer cell lines were compared at mRNA level for expressioll of EGFR by using EGFR cDNA probe labeled with digoxigenin (DIG). Expression of EGFR was showll to be markedly higher in the highly invassive CCL229 cells than that in the low in- vasive CX-1 cells.
