1Experimental far-field imaging properties of high refractive index microsphere lens显示文摘The far-field imaging properties of a high index microsphere lens spatially separated from the object are experimentally studied. Our experimental results show that, for a Blu-ray disk whose spacing is 300 nm, the high index microsphere lens also can discern the patterns of the object sample when the distance between the lens and the object is up to 5.4 μm. When the distance is increased from 0 to 5.4 μm, for the microsphere lens with a diameter of 24 μm, the lateral magnification increases from 3.5× to 5.5×, while the field of view decreases from 5.1 to 3.0 μm. By varying the distance between the lens and the object, the optical image can be optimized. We also indicate that the far-field imaging capability of a high index microsphere lens is dependent on the electromagnetic field intensityprofile of the photonic nanojet under different positions of the microsphere lens.Minglei Guo Yong-Hong Ye Jinglei Hou Bintao Du 2015Photonics Research2015,3,6:3

9Carbon dots based on targeting unit inheritance strategy for Golgi apparatus-targeting imaging显示文摘The Golgi apparatus is one of the important organelles,where the final processing and packaging of cellular secretions(such as proteins)are completed.The disorder of Golgi apparatus structure and function will induce many diseases.Therefore,monitoring the morphological structure of Golgi apparatus is crucial for the diagnosis and treatment of relevant diseases.In order to achieve Golgi apparatus-targeting imaging,the strategy of targeting unit inheritance was adopted and carbon dots(CDs)with Golgi apparatus-targeting ability were synthesized by one-step hydrothermal method with L-ascorbic acid with high reactivity and reducibility as the carbon source and L-cysteine as the targeting unit.CDs have a certain amount of cysteine residues on their surface,and have excitation dependence,satisfactory fluorescence and cysteine residues stability and low toxicity.As an imaging agent,CDs can be used for targeting imaging of Golgi apparatus.Yingying Wei Yuduan Gao Lin Chen Qiang Li Jinglei Du Dongming Wang Fanggang Ren Xuguang Liu Yongzhen Yang 2023Frontiers of Materials Science2023,17,1:0
10Green-emissive carbon quantum dots with high fluorescence quantum yield:Preparation and cell imaging显示文摘High fluorescence quantum yield(QY),excellent fluorescence stability,and low toxicity are essential for a good cellular imaging fluorescent probe.Green-emissive carbon quantum dots(CQDs)with many advantages,such as unique fluorescence properties,anti-photobleaching,low toxicity,fine biocompatibility and high penetration depth in tissues,have been considered as a potential candidate in cell imaging fluorescent probes.Herein,N,S-codoped green-emissive CQDs(QY=64.03%)were synthesized by the one-step hydrothermal method,with m-phenylenediamine as the carbon and nitrogen source,and L-cysteine as the nitrogen and sulfur dopant,under the optimum condition of 200℃ reaction for 2 h.Their luminescence was found to originate from the surface state.In light of the satisfactory photobleaching resistance and the low cytotoxicity,CQDs were used as a cell imaging probe for HeLa cell imaging.The results clearly indicate that cells can be labeled with CQDs,which can not only enter the cytoplasm,but also enter the nucleus through the nuclear pore,showing their broad application prospect in the field of cell imaging.Yingying WEI Lin CHEN Shaoban ZHAO Xuguang LIU Yongzhen YANG Jinglei DU Qiang LI Shiping YU 2021Frontiers of Materials Science2021,15,2:0
11GATA2^(−/−) human ESCs undergo attenuatedendothelial to hematopoietic transition andthereafter granulocyte commitment显示文摘Background: Hematopoiesis is a progressive process collectively controlled by an elaborate network of transcriptionfactors (TFs). Among these TFs, GATA2 has been implicated to be critical for regulating multiple steps of hematopoiesisin mouse models. However, whether similar function of GATA2 is conserved in human hematopoiesis, especially duringearly embryonic development stage, is largely unknown.Results: To examine the role of GATA2 in human background, we generated homozygous GATA2 knockout humanembryonic stem cells (GATA2^(−/−) hESCs) and analyzed their blood differentiation potential. Our results demonstratedthat GATA2^(−/−) hESCs displayed attenuated generation of CD34^(+)CD43^(+) hematopoietic progenitor cells (HPCs), due tothe impairment of endothelial to hematopoietic transition (EHT). Interestingly, GATA2^(−/−) hESCs retained the potentialto generate erythroblasts and macrophages, but never granulocytes. 12TMEM106A transcriptionally regulated by promoter methylation is involved in invasion and metastasis of hepatocellular carcinoma显示文摘Expression of transmembrane protein 106A(TMEM106A)has been reported to be dysregulated in several types of cancers.However,the role of TMEM106A in hepatocellular carcinoma(HCC)is still unknown.In the present study,we demonstrate that TMEM106A is markedly downregulated in HCC compared with normal liver tissue.In particular,tumor-specific DNA methylation of TMEM106A is frequently observed in tumor tissues from HCC patients.Immunohistochemistry and pyrosequencing reveal a significant relationship between TMEM106A methylation and downregulation of protein expression.Receiver operating characteristic(ROC)curve analysis reveals that methylation of TMEM106A in tumor samples is different from that in non-malignant adjacent tissues of HCC patients.Moreover,HCC patients with TMEM106A hypermethylation have a poor clinical prognosis.5-Aza-2′-deoxycytidin treatment of hypermethylated TMEM106A in highly metastatic HCC cells increases the expression of TMEM106A.Functional assays reveal that overexpression of TMEM106A significantly suppresses the malignant behavior of HCC cells in vitro and decreases tumorigenicity and lung metastasis in vivo.Mechanistically,TMEM106A inhibits epithelial mesenchymal transition(EMT)of HCC cells through inactivation of the Erk1/2/Slug signaling pathway.In conclusion,our findings demonstrate that TMEM106A is an inhibitor of HCC EMT and metastasis,and TMEM106A is often transcriptionally downregulated by promoter methylation,which results in reduced levels of TMEM106A protein and predicts poor survival outcomes for HCC patients.Shiming Shi Biao Wang Jinglei Wan Lina Song Guiqi Zhu Junxian Du Luxi Ye Qianqian Zhao Jialiang Cai Qing Chen Kun Xiao Jian He Lei Yu Zhi Dai 2022Acta Biochimica et Biophysica Sinica2022,54,7:0
