4Life prediction and test period optimization research based on small sample reliability test of hydraulic pumps显示文摘Hydraulic pumps belong to reliable long-life hydraulic components. The reliability evaluation includes characters such as long test period,high cost,and high power loss and so on. Based on the principle of energy-saving and power recovery,a small sample hydraulic pump reliability test rig is built,and the service life of hydraulic pump is predicted,and then the sampling period of reliability test is optimized. On the basis of considering the performance degradation mechanism of hydraulic pump,the feature information of degradation distribution of hydraulic pump volumetric efficiency during the test is collected,so an optimal degradation path model of feature information is selected from the aspect of fitting accuracy,and pseudo life data are obtained. Then a small sample reliability test of period constrained optimization search strategy for hydraulic pump is constructed to solve the optimization problem of the test sampling period and tightening end threshold,and it is verified that the accuracy of the minimum sampling period by the non-parametric hypothes is tested. 7QKI-5 regulates the alternative splicing of cytoskeletal gene ADD3 in lung cancer显示文摘Accumulating evidence indicates that the alternative splicing program undergoes extensive changes during cancer develop-ment and progression.The RNA-binding protein QKI-5 is frequently downregulated and exhibits anti-tumor activity in lung cancer.Howeve-r,little is known about the functional targets and regulatory mechanism of QKI-5.Here,we report that upregulation of exon 14 inclusion of cytoskeletal gene Adducin 3(ADD3)significantly correlates with a poor prognosis in lung cancer.QKI-5 inhibits cell proliferation and migration in part through suppressing the splicing of ADD3 exon 14.Through genome-wide mapping of QKI-5 binding sites in vivo at nucleotide resolution by iCLIP-seq analysis,we found that QKI-5 regulates alternative splicing of its target mRNAs in a binding position-dependent manner.By binding to multiple sites in an upstream intron region,QKI-5 represses the splicing of ADD3 exon 14.We also identified several QKI mutations in tumors,which cause dysregulation of the splicing of QKI targets ADD3 and NUMB.Taken together,our results reveal that QKI-mediated alternative splicing of ADD3 is a key lung cancer-associated splicing event,which underlies in part the tumor suppressor function of QKI.Jin-Zhu Wang Xing Fu Zhaoyuan Fang Hui Liu Feng-Yang Zong Hong Zhu Yan-Fei Yu Xiao-Ying Zhang Shen-Fei Wang Ying Huang jingyi Hui 2021Journal of Molecular Cell Biology2021,13,5:4

10Controlling plasmon‐exciton interactions through photothermal reshaping显示文摘We investigated the plasmon-exciton interactions in an individual gold nanorod(GNR)with monolayer MoS2 at room temperature with the single-particle spectroscopy technique.To control the plasmon-exciton interaction,we tuned the local surface plasmon resonance of an individual GNR in-situ by employing the photothermal reshaping effect.The scattering spectra of the GNR-MoS2 hybrids exhibited two dips at the frequencies of the A and B excitons of monolayer MoS2,which were caused by the plasmon-induced resonance energy transfer effect.The resonance energy transfer rate increased when the surface plasmon resonance of the nanorod matched well with the exciton transition energy.Also,we demonstrated that the plasmon-enhanced fluorescence process dominated the photoluminescence of the GNR-MoS2 hybrid.These results provide a flexible way to control the plasmon-exciton interaction in an all-solid-state operating system at room temperature.Aiqin Hu Shuai Liu Jingyi Zhao Te Wen Weidong Zhang Qihuang Gong Yongqiang Meng Yu Ye Guowei Lu 2020Opto-Electronic Advances2020,3,1:2

15Macrophage polarization is involved in liver fibrosis induced byβ_(1)-adrenoceptor autoantibody显示文摘Accumulating evidence suggests that liver injury can be induced by the over-expression ofβ_(1)-adrenergic receptors(β_(1)-ARs).High titers of autoantibodies specific toβ_(1)-adrenergic receptors(β_(1)-AA)are detected in the sera of heart failure patients,potentially playing agonist-like roles.However,the role ofβ_(1)-AA in liver function has not been characterized.In this study,we collect the sera of primary biliary cholangitis(PBC)patients,a condition which easily develops into liver fibrosis,and analyze the relationship between PBC andβ_(1)-AA.A passive immunization model is established to assess the effect ofβ_(1)-AA on the liver.Subsequently,the effect ofβ_(1)-AA on macrophages is investigated in vitro.Results show that PBC patients have a high titer and ratio ofβ_(1)-AA,compared to controls.Liver injury and fibrosis are induced byβ_(1)-AA.In vitro experiments with ROS probe demonstrate thatβ_(1)-AA induces macrophages to produce ROS and secrete TNFα.These effects can be partially reversed by metoprolol,a blocker forβ_(1)-AR.Results from the transwell and phagocytosis assays show thatβ_(1)-AA promotes macrophage migration and phagocytosis.FCM tests suggest thatβ_(1)-AA induces the alteration of M1 rather than M2 markers in macrophages.Finally,the Annexin V/PI assay indicates that macrophage culture supernatants stimulated byβ_(1)-AA cause hepatocyte apoptosis.Overall,these results suggest thatβ_(1)-AA is involved in PBC.Theβ_(1)-AA-induced activation,phagocytosis and phenotypic modification of macrophages may play an important role in the development of hepatic fibrosis and injury.Ye Wu Xiongxiong Fan Haicun Yu Jingyi Liu Yanru Duan Suli Zhang Li Yan Yunhui Du Huirong Liu 2022Acta Biochimica et Biophysica Sinica2022,54,8:1

17Feasibility of sentinel lymph node biopsy omission after integration of ^(18)F-FDG dedicated lymph node PET in early breast cancer: a prospective phase II trial显示文摘Objective:Sentinel lymph node biopsy(SLNB)is currently the standard of care in clinically node negative(cN0)breast cancer.The present study aimed to evaluate the negative predictive value(NPV)of 18F-FDG dedicated lymph node positron emission tomography(LymphPET)in cN0 patients.Methods:This was a prospective phase II trial divided into 2 stages(NCT04072653).In the first stage,cN0 patients underwent axillary LymphPET followed by SLNB.In the second stage,SLNB was omitted in patients with a negative preoperative axillary assessment after integration of LymphPET.Here,we report the results of the first stage.The primary outcome was the NPV of LymphPET to detect macrometastasis of lymph nodes(LN-macro).Results:A total of 189 patients with invasive breast cancer underwent LymphPET followed by surgery with definitive pathological reports.Forty patients had LN-macro,and 16 patients had only lymph node micrometastasis.Of the 131 patients with a negative LymphPET result,16 patients had LN-macro,and the NPV was 87.8%.After combined axillary imaging evaluation with ultrasound and LymphPET,100 patients were found to be both LymphPET and ultrasound negative,9 patients had LN-macro,and the NPV was 91%.Conclusions:LymphPET can be used to screen patients to potentially avoid SLNB,with an NPV>90%.The second stage of the SOAPET trial is ongoing to confirm the safety of omission of SLNB according to preoperational axillary evaluation integrating LymphPET.Junjie Li Jingyi Cheng Guangyu Liu Yifeng Hou Genghong Di Benglong Yang Yizhou Jiang Liang Huang Feilin Qu Sheng Chen Yan Wang Keda Yu Zhimin Shao 2022Cancer Biology & Medicine2022,19,7:1
