1Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. 2Clinical implications of fatty pancreas:Correlations between fatty pancreas and metabolic syndrome显示文摘AIM:To investigate the clinical implications of lipid deposition in the pancreas(fatty pancreas). METHODS:The subjects of this study were 293 patients who had undergone abdominal computed tomography(CT)and sonography.Fatty pancreas was diagnosed by sonographic findings and subdivided into mild,moderate,and severe fatty pancreas groups comparing to the retroperitoneal fat echogenicity. RESULTS:Fatty pancreas was associated with higher levels for visceral fat,waist circumference,aspartate aminotransferase(AST),alanine aminotransferase (ALT),total cholesterol,triglyceride,high density lipoprotein,free fatty acid,γ-GTP,insulin,and the homeostasis model assessment of insulin resistance (HOMA-IR)than the control group(P<0.05).HOMAIR,visceral fat,triglyceride,and ALT also tended to increase with the degree of fat deposition in the pancreas on sonography.In a multivariate logistic regression analysis,HOMA-IR,visceral fat,and ALT level were independently related to fatty pancreas after adjustment for age,body mass index,and lipid profile.The incidence of metabolic syndrome in the fatty pancreas group was significantly higher than in the control group,and the numbers of metabolic syndrome parameters were significantly higher in the fatty pancreas group(P<0.05).CONCLUSION:Sonographic fatty pancrease showed higher insulin resistance,visceral fat area,triglyceride, and ALT levels than normal pancreases.Fatty pancreas also showed a strong correlation with metabolic syndrome.Jun Seok Lee Sang Heum Kim Dae Won Jun Jee Hye Han Eun Chul Jang Ji Young Park Byung Kwan Son Seong Hwan Kim Yoon Ju Jo Young Sook Park Yong Soo Kim 2009World Journal of Gastroenterology2009,15,15:40
3Environmental changes reflected by n-alkanes of lake core in Nam Co on the Tibetan Plateau since 8.4 kaB.P.显示文摘The n-alkanes are extracted from NMLC-1 core that was drilled in the Nam Co, central Tibet. They are measured by using Gas Chromatography and Mass Spectrometry (GC/MS) for componential and quantitative analyses. According to the constructed depth-age model, the component and concentration of n-alkanes, together with total organic carbon (TOC), total nitrogen (TN) and carbonate are used to elu- cidate palaeoenvironmental changes of Nam Co during the past 8.4 ka. The results indicate that Holo- cene environment performs three stages in the lake area. In the stage of 8.4―6.7 kaB.P., it was warmer while precipitation slightly increased. This stage was ended by an obvious cold/dry event. During 6.7― 5.8 kaB.P., temperature increased rapidly and reached its maximum values at about 6.0 kaB.P. The environments were warm/wet optimum for the blooming of terrestrial plants and submerged aquatic plants. After that, temperature decreased continuously and showed the lowest values at about 3.0 kaB.P. From 2.9 kaB.P. to the present, temperature rose again but alternated with cold and warm. The lake area tended to be dry after 1.4 kaB.P. 4Role of surgical resection for multiple hepatocellular carcinomas显示文摘AIM:To clarify the role of surgical resection for multiple hepatocellular carcinomas(HCCs)compared to transarterial chemoembolization(TACE)and liver transplantation(LT). METHODS:Among the HCC patients who were managed at Yonsei University Health System between January 2003 and December 2008,160 patients who met the following criteria were retrospectively enrolled:(1) two or three radiologically diagnosed HCCs;(2)no radiologic vascular invasion;(3)Child-Pugh class A;(4) main tumor smaller than 5 cm in diameter;and(5) platelet count greater than 50 000/mm3.Long-term outcomes were compared among the following three treatment modalities:surgical resection or combined radiofrequency ablation(RFA)(n=36),TACE(n=107), and LT(n=17).The survival curves were computed using the Kaplan-Meier method and compared with a log-rank test.To identify the patients who gained a survival benefit from surgical resection,we also investigated prognostic factors for survival following surgical resection.Multivariate analyses of the prognostic factors for survival were performed using the Cox proportional hazard model. RESULTS:The overall survival(OS)rate was significantly higher in the surgical resection group than in the TACE group(48.1%vs 28.9%at 5 years,P<0.005). LT had the best OS rate,which was better than that of the surgical resection group,although the difference was not statistically significant(80.2%vs 48.1%at 5 years,P=0.447).The disease-free survival rates were also significantly higher in the LT group than in the surgical resection group(88.2%vs 11.2%at 5 years, P<0.001).Liver cirrhosis was the only significant prognostic factor for poor OS after surgical resection. Clinical liver cirrhosis rates were 55.6%(20/36)in the resection group and 93.5%(100/107)in the TACE group.There were 19 major and 17 minor resections. En bloc resection was performed in 23 patients,multisite resection was performed in 5 patients,and combined resection with RFA was performed in 8 patients. In the TACE group,only 34 patients(31.8%)were recorded as having complete remission after primary TACE.Seventy-two patients(67.3%)were retreated with repeated TACE combined with other therapies. In patients who underwent surgical resection,the 16 patients who did not have cirrhosis had higher 5-year OS and disease-free survival rates than the 20 patients who had cirrhosis(80.8%vs 25.5%5-year OS rate, P=0.006;22.2%vs 0%5-year disease-free survival rate,P=0.048).Surgical resection in the 20 patients who had cirrhosis did not provide any survival benefit when compared with TACE(25.5%vs 24.7%5-year OS rate,P=0.225).Twenty-nine of the 36 patients who underwent surgical resection experienced recur-rence.Of the patients with cirrhosis,80%(16/20) were within the Milan criteria at the time of recurrence CONCLUSION:Among patients with two or three HCCs, no radiologic vascular invasion,and tumor diameters≤ 5 cm,surgical resection is recommended only in those without cirrhosis.Sung Hoon Choi Gi Hong Choi Seung Up Kim Jun Yong Park Dong Jin Joo Man Ki Ju Myoung Soo Kim Jin Sub Choi Kwang Hyub Han Soon Il Kim 2013World Journal of Gastroenterology2013,19,3:12
5Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 6Androgen and prostatic stroma显示文摘Aim:To investigate the effect of androgen on the proliferation,differentiation and regression of canine prostatic stromal cells in vivo and human stromal cells in vitro.Methods:Twenty-two dogs,including 15 normal prostate doge and 7 prostatic hyperplasia dogs,had their serum concentration of testosterone and estrodiol determined by radioimmunoassay before and after castration.The expression of androgen receptor(AR)and estrogen receptor(ER)in the prostate were analysed by immunohistochemistry and semi-quantitative RT-PCR before and after castration.Light microscopy,transmission electron microscopy and TUNEL assay were carried out successively before and after castration to evaluate the prostatic histomorphology.In vitro serum-free cell cultures from human prostatic stroma were established and exposed to dihydrotestosterone(DHT).The proliferation of the cell culture was detected by MTT assay.The expression of TGFβ bFGF,AR,and smooth muscle cell(SMC) specific proteins (myosin and/or smoothelin)were detected using immunohistochemistry and RT-PCR.The differentiation from fibroblasts to smooth muscle cells was deduced by measuring the expression of SMC specific proteins.Results:Before castration,the serum concentrations of testosterone and estrodiol were not statistically different between normal and hyperplasia groups.Following castration,the serum concentration of testerone decreased rapidly in 2 days,and the concentration of estrodiol had no significant change compared with the pre-castration data.In the prostate,AR was presented in both the epithelial and stromal cells and the AR mRNA level was higher in hyperplasia than in normal prostate tissues(P<0.05).While ER predominantly existed in the prostate stromal cells and the ER mRNA had no difference between the hyperplasia and the normal group.Within the early phase of castration(Yuan-Jie NIU, Teng-Xiang MA, Ju ZHANG, Yong XU, Rui-Fa HAN, Guang SUNDepartment of Prostatic Disease, Tianjin Institute of Urologial Surgery, Tianjin Medical University,Tianjin 300211, China 2003Asian Journal of Andrology2003,5,1:11
7The spatial distribution and sedimentary processes of organic matter in surface sediments of Nam Co,Central Tibetan Plateau显示文摘In this study,67 surface sediment samples collected from Nam Co in central Tibet were analyzed for total carbon,total organic carbon and total nitrogen,and 51 of these samples were also analyzed for n-alkanes.The origin and spatial distribution of organic matter were then investigated using these proxies,and the control factors responsible for the spatial distribution patterns and paleolimnological significance were discussed.The results indicated that the origin of organic matter in surface sediment of Nam Co is consistent with the sources of n-alkanes,which were primarily submerged plants,followed by terrestrial plants,and then aquatic algae and bacteria.The organic matter in surface sediments of the lake showed typical spatial variability.Because of the great influence of underwater topography,river inputs and water quality,the spatial distribution of organic matter is enriched from the source to the deposit center.This spatial variability of organic matter in the lake indicates that the sediments in different areas have different sensitivities to environmental changes,which is important to reconstruction of paleoenvironments and paleoclimate using lake sediment cores.WANG Yong ZHU LiPing WANG JunBo JU JianTing LIN Xiao 2012Chinese Science Bulletin2012,57,36:9

9Is graphite lithiophobic or lithiophilic?显示文摘Graphite and lithium metal are two classic anode materials and their composite has shown promising performance for rechargeable batteries.However,it is generally accepted that Li metal wets graphite poorly,causing its spreading and infiltration difficult.Here we show that graphite can either appear superlithiophilic or lithiophobic,depending on the local redox potential.By comparing the wetting performance of highly ordered pyrolytic graphite,porous carbon paper(PCP),lithiated PCP and graphite powder,we demonstrate that the surface contaminants that pin the contact-line motion and cause contact-angle hysteresis have their own electrochemical-stability windows.The surface contaminants can be either removed or reinforced in a time-dependent manner,depending on whether the reducing agents(C6→Li C6)or the oxidizing agents(air,moisture)dominate in the ambient environment,leading to bifurcating dynamics of either superfast or superslow wetting.Our findings enable new fabrication technology for Li–graphite composite with a controllable Li-metal/graphite ratio and present great promise for the mass production of Li-based anodes for use in high-energy-density batteries.Jian Duan Yuheng Zheng Wei Luo Wangyan Wu Tengrui Wang Yong Xie Sa Li Ju Li Yunhui Huang 2020National Science Review2020,7,7:8
10Fluidized mining and in-situ transformation of deep underground coal resources: a novel approach to ensuring safe, environmentally friendly, low-carbon, and clean utilisation显示文摘Traditional coal mining and utilisation patterns are severely detrimental to natural resources and environments and significantly impede safe, low-carbon, clean, and sustainable utilisation of coal resources. Based on the idea of in situ fluidized coal mining that aims to transform solid coal into liquid or gas and transports the fluidized resources to the ground to ensure safe mining and low-carbon and clean utilisation, in this study, we report on a novel in situ unmanned automatic mining method. This includes a flexible, earthworm-like unmanned automatic mining machine (UAMM) and a coal mine layout for in situ fluidized coal mining suitable for the UAMM. The technological and economic advantages and the carbon emission reduction of the UAMM-based in situ fluidized mining in contrast to traditional mining technologies are evaluated as well. The development trends and possible challenges to this design are also discussed. It is estimated that the proposed method costs approximately 49% of traditional coal mining costs. The UAMM-based in situ fluidized mining and transformation method will reduce CO2 emissions by at least 94.9% compared to traditional coal mining and utilisation methods. 12Heat Stress Upregulates the Expression of TLR4 and Its Alternative Splicing Variant in Bama Miniature Pigs显示文摘Alternative splicing is a cellular mechanism in eukaryotes that results in considerable diversity of gene products. It plays an important role in several diseases and cellular signal regulation. Heat stress is a major factor that induces immunosuppression in pigs. Little is known about the correlation between alternative splicing and heat stress in pigs. Therefore, this study aimed to clone, sequence and quantify the alternative splicing variant of toll-like receptor 4(TLR4) in Bama miniature pigs(Sus scrofa domestica) following exposure to heat stress. The results showed that the second exon of TLR4 was spliced and 167 bp shorter in the alternative splicing variant, and the protein was putatively identified as a type of truncated membrane protein consisting of extramembrane, transmembrane and intramembrane regions lacking a signal peptide. Further, it was not a nonclassical secretory protein. Five potential reference genes were screened for their potential as reliable standards to quantify the expression of TLR4 alternative spliced variants by real-time quantitative reverse-transcriptase polymerase chain reaction(q RT-PCR). The stability of these reference genes was ranked using the ge Norm and Norm Finder programs, and ribosomal protein L4(RPL4) and TATA box-binding protein(TBP) were found to be the two genes showing the most stable expression in the in vitro cultured peripheral blood mononuclear cells(PBMCs) during heat shock. The m RNA level of the TLR4 gene(both classical and spliced) in stressed pigs increased significantly(P<0.05). Further, the expression levels of the alternative spliced variant of TLR4(TLR4-ASV) showed a 2-3 folds increase in heat-stressed PBMCs as compared to control pigs. 13Pollen-inferred Holocene vegetation and climate histories in Taro Co,southwestern Tibetan Plateau显示文摘A 310-cm-long sediment core,covering the last10,200 years,was collected from Taro Co on the southwestern Tibetan Plateau and analyzed for pollen,grain size and total inorganic carbon content.The pollen data showed that vegetation changed from alpine steppe to alpine meadow during 10,200–8,900 cal a BP,to alpine steppe dominated by Artemisia during 8,900–7,400 cal a BP,to alpine meadow during 7,400–3,300 cal a BP and to alpine steppe after 3,300 cal a BP.Correspondingly,the pollen,grain size and total inorganic carbon content results revealed climatic change in this area over four stages.The initial stage was from 10,200 to 8,900 cal a BP,during which the climate changed from cold-dry to warm-humid.The second stage(8,900–7,400 cal a BP)was characterized by a warm and dry climate.However,at approximately7,400 cal a BP,the climate began to become cold and humid,which continued until 3,300 cal a BP.The last stage,from 3,300 cal a BP to present,was characterized as cold and increasingly arid.Climatic events of the early and mid-late Holocene showed that the area was significantly affected by the westerlies.However,the mid-Holocene climate in Taro Co was controlled by the Indian monsoon.The mid-late Holocene depositional environment record of Taro Co was very important to further elaborate the degree of influence by the westerlies or Indian monsoon.Qingfeng Ma Liping Zhu Xinmiao L Yun Guo Jianting Ju Junbo Wang Yong Wang Lingyu Tang 2014Chinese Science Bulletin2014,59,31:7

15Hydrodynamic process of Tibetan Plateau lake revealed by grain size:Case study of Pumayum Co显示文摘Under the background of global warming,some lakes on the Tibetan Plateau(TP) are potentially sensitive to temperature change.With a case study of Pumayum Co,where glacier meltwater is important to supply(we call this a glacier-fed lake hereafter),we analyze the sensitivity of lake sediment grain size to temperature change.This is done by resolving the modern hydrodynamic process,coupled with comparison of paleoclimatic proxies.According to the spatial distributions of parameters,percentage of grain size and the grain size frequency distribution curve,hydrodynamic processes are analyzed.Five clastic sedimentation types are thereby discriminated.In the open lake area,suspended load transport is the main transport agent.Grain Size Trend Analysis(GSTA),a sediment dynamics model,reveals a trend toward eastward transport.This indicates that the largest and glacier-fed river,the Jiaqu River,influences the entire lake(not just the subsurface alluvial fan),and that lake sediment grain size may serve as a temperature indicator.Time series comparison between grain size of a short core from the central lake and meteorological data confirms this temperature indication,which in turn shows reliability of the method discriminating the hydrodynamic process.This case study will improve the ability of paleoclimatic reconstruction using lake sediment in glacier-fed lakes on the Tibetan Plateau.JU JianTing ZHU LiPing FENG JinLiang WANG JunBo WANG Yong XIE ManPing PENG Ping ZHEN XiaoLin LÜXinMiao 2012Chinese Science Bulletin2012,57,19:7
16Toxicity of Multi-Walled Carbon Nanotubes,Graphene Oxide,and Reduced Graphene Oxide to Zebrafish Embryos显示文摘Objective This study was aimed to investigate the toxic effects of 3 nanomaterials, i.e. multi-walled carbon nanotubes(MWCNTs), graphene oxide(GO), and reduced graphene oxide(RGO), on zebrafish embryos. Methods The 2-h post-fertilization(hpf) zebrafish embryos were exposed to MWCNTs, GO, and RGO at different concentrations(1, 5, 10, 50, 100 mg/L) for 96 h. Afterwards, the effects of the 3 nanomateria on spontaneous movement, heart rate, hatching rate, length of larvae, mortality, and malformations ls were evaluated. Results Statistical analysis indicated that RGO significantly inhibited the hatching of zebrafish embryos. Furthermore, RGO and MWCNTs decreased the length of the hatched larvae at 96 hpf. No obvious morphological malformation or mortality was observed in the zebrafish embryos after exposure to the three nanomaterials. Conclusion MWCNTs, GO, and RGO were all toxic to zebrafish embryos to influence embryos hatching and larvae length. 18Alpha-fetoprotein-targeted reporter gene expression imaging in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC) is one of the most common cancers in Eastern Asia, and its incidence is increasing globally. Numerous experimental models have been developed to better our understanding of the pathogenic mechanism of HCC and to evaluate novel therapeutic approaches. Molecular imaging is a convenient and up-to-date biomedical tool that enables the visualization, characterization and quantification of biologic processes in a living subject. Molecular imaging based on reporter gene expression, in particular, can elucidate tumor-specific events or processes by acquiring images of a reporter gene's expression driven by tumor-specific enhancers/promoters. In this review, we discuss the advantages and disadvantages of various experimental HCC mouse models and we present in vivo images of tumorspecific reporter gene expression driven by an alphafetoprotein(AFP) enhancer/promoter system in a mouse model of HCC. The current mouse models of HCC development are established by xenograft, carcinogen induction and genetic engineering, representing the spectrum of tumor-inducing factors and tumor locations. The imaging analysis approach of reporter genes driven by AFP enhancer/promoter is presented for these different HCC mouse models. Such molecular imaging can provide longitudinal information about carcinogenesis and tumor progression. 19Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1).
