1Structural shifts in the intestinal microbiota of rats treated with cyclosporine A after orthotropic liver transplantation显示文摘Understanding the effect of immunosuppressive agents on intestinal microbiota is important to reduce the mortality and morbidity from orthotopic liver transplantation (OLT). We investigated the relationship between the commonly used immunosuppressive agent cyclosporine A (CSA) and the intestinal microbial variation in an OLT model. The rat samples were divided as follows:(1) N group (normal control);(2) I group (isograft LT, Brown Norway [BN] rat to BN);(3) R group (allograft LT, Lewis to BN rat);and (4) CSA group (R group treated with CSA). The intestinal microbiota was assayed by denaturing gradient gel electrophoresis profiles and by using real-time polymerase chain reaction. The liver histopathology and the alanine/aspartate aminotransferase ratio after LT were both ameliorated by CSA. In the CSA group, the numbers of rDNA gene copies of Clostridium cluster I, Clostridium cluster XIV, and Enterobacteriaceae decreased, whereas those of Faecalibacterium prausnitzii increased compared with the R group. Cluster analysis indicated that the samples from the N, I, and CSA groups were clustered, whereas the other clusters contained the samples from the R group. 2Single-center Experience in the Diagnosis and Treatment of Hepatic Perivascular Epithelioid Cell Neoplasm显示文摘Background and Aims:Perivascular epithelioid cell neo-plasms(PEComas)are a rare type of mesenchymal neo-plasm and their preoperative diagnosis is challenging.In this study,we summarized the experience from a single medical center to study the examinations,clinical presen-tations,and pathological and histological characteristics of PEComas in the liver in order to optimize overall un-derstanding of the diagnosis and treatment of these neo-plasms.Methods:We conducted a retrospective analysis to investigate the clinical and pathological characteristics as well as imaging presentations of 75 patients diagnosed with hepatic PEComa in The First Affiliated Hospital of Zhe-jiang University between April 2010 and April 2020.Re-sults:Among the 75 patients,52 were women,and the median age was 48 years.Most patients had no specific symptoms,and two were admitted to the hospital for a second time owing to relapse.All patients underwent surgi-cal resection.Histologically,38 patients had classical angio-myolipoma(AML)and 37 had epithelioid AML.The PECo-mas were accompanied by positive immunohistochemical expression of HMB45,Melan-A,and smooth muscle actin.Follow-up data were obtained from 47 of the total 75 pa-tients,through October 2020.Two patients had metastasis after surgery.Conclusions:AML is the most common type of hepatic PEComa.There are no specific symptoms of he-patic PEComa,and serological examinations and imaging modalities for accurate preoperative diagnosis are lacking.Epithelioid AML should be considered a tumor of uncertain malignant potential;however,the prognosis of PEComa af-ter resection is promising.Junjun Jia Jia Luo Cheng-Gen Pan Guomei Ge Meibao Feng Bei Zou Li Liu Shusen Zheng Jun Yu 2022Journal of Clinical and Translational Hepatology2022,10,1:5
3Reconfigurable photovoltaic effect for optoelectronic artificial synapse based on ferroelectric p-n junction显示文摘Neuromorphic machine vision has attracted extensive attention on wide fields.However,both current and emerging strategies still suffer from power/time inefficiency,and/or low compatibility,complex device structure.Here we demonstrate a driving-voltage-free optoelectronic synaptic device using non-volatile reconfigurable photovoltaic effect based on MoTe_(2)/α-In_(2)Se_(3) ferroelectric p-n junctions.This function comes from the non-volatile reconfigurable built-in potential in the p-n junction that is related to the ferroelectric polarization inα-In_(2)Se_(3).Reconfigurable rectification behavior and photovoltaic effect are demonstrated firstly.Notably,the figure-of-merits for photovoltaic effect like photoelectrical conversion efficiency non-volatilely increases more than one order.Based on this,retina synapse-like vision functions are mimicked.Optoelectronic short-term and long-term plasticity,as well as basic neuromorphic learning and memory rule are achieved without applying driving voltage.Our work highlights the potential of ferroelectric p-n junctions for enhanced solar cell and low-power optoelectronic synaptic device for neuromorphic machine vision.Yanrong Wang Feng Wang Zhenxing Wang Junjun Wang Jia Yang Yuyu Yao Ningning Li Marshet Getaye Sendeku Xueying Zhan Congxin Shan Jun He 2021Nano Research2021,14,11:2
4Effect of La_(2)O_(3) addition on the synthesis of tungsten nanopowder via combustion-based method显示文摘In this study, the influences of La_(2)O_(3) added on the phase, morphology and reduction process of tungsten oxide prepared by solution combustion synthesis(SCS) were investigated for the first time. And tungsten nanopowders with different La_(2)O_(3) doping amount(0.5~5.0 wt%) were successfully prepared by SCS and followed hydrogen reduction. The results showed that with the increase of La_(2)O_(3) addition,the product synthesized by SCS changed from needle-like W_(18) O_(49) to irregularly granulated H0.53 WO3 and the complete reduction temperature also increased form 700°C to 850°C. The densification behavior of as-prepared W nanopowders revealed that the densification inhibitory effect of La_(2)O_(3) was enhanced as the La_2O_3 addition increased. 6Robust ultrathin and transparent AZO/Ag-SnOx/AZO on polyimide substrate for flexible thin film heater with temperature over 400℃显示文摘We developed flexible and transparent thin film heaters(TFHs)with a structure of AZO/AgSnOx/AZO/polyimide(PI)which exhibited superior stability under operational conditions.Ultrathin and robust doped silver(Ag)films were produced by introducing a small amount of SnOx during the sputtering process.The AZO/Ag-SnOx/AZO stacks showed the best figure of merit value of 139.9,which was higher than that of ITO counterpart(25.5)with the same thickness.In addition,it exhibited excellent durability,which showed unaffected optical and electrical properties under the heat treatment of 500℃for 30 min in air,highly-accelerated tempe ratu re and humidity stress test(HAST)at 121℃and 97%RH for 36 h,10,000 times of scratching and 1500 times of inner and outer bending test.Furthermore,the TFHs based on AZO/Ag-SnOx/AZO/PI achieved a high temperature of 438.8℃with response time in several seconds,which outperformed most previous studies.The robust high-temperature TFHs in this research hold promising commercial applications in flexible and high temperature occasions.Zhaozhao Wang Jia Li Junjun Xu Jinhua Huang Ye Yang Ruiqin Tan Guofei Chen Xingzhong Fang Yue Zhao Weijie Song 2020Journal of Materials Science & Technology2020,47,13:1

8ACACB is a novel metabolism-related biomarker in the prediction of response to cetuximab therapy in metastatic colorectal cancer显示文摘Cetuximab is one of the most valuable targeted therapy monoclonal antibodies in the treatment of metastatic colorectal cancer(CRC).However,the mechanisms affecting cetuximab resistance in CRC treatment remain unclear.Metabolism,especially fatty acid metabolism,has been reported to play an important role in tumor treatment.The correlation between cetuximab resistance and metabolism and whether it can be a new biomarker to evaluate the sensitivity of cetuximab in CRC treatment still need to be further explored.In this study,we perform a comprehensive analysis to confirm the relationship between fatty acid metabolism and cetuximab resistance,and the differentially expressed genes(DEGs)related to cetuximab drug resistance in CRC are screened by bioinformatics technology.We find that acetyl-CoA carboxylase beta(ACACB),ADH1C,CES1,MGLL,FMO5,and GPT are the hub DEGs,and ACACB is the most important biomarker among them.In addition,we systematically analyze the role of ACACB in the tumorigenesis of CRC,including tissue expression,CRC cell growth,cetuximab sensitivity,and potential downstream pathways,by using bioinformatics techniques,in vitro experiments and clinical cohort validation.Our results confirm that cetuximab resistance is correlated with metabolism.ACACB can lead to decreased sensitivity to cetuximab in CRC,and its mechanism may be related to EGFR phosphorylation,which could affect the activation of the mTOR/Akt signaling pathway and regulation of CDT1-,cyclin D1-,and p21-related cell cycle modulation.Hi-Ju Hong Yanfei Shao Sen Zhang Guang Yang Hongtao Jia Xiao Yang Ling Huang Shuchun Li Batuer Aikemu Luyang Zhang Junjun Ma Lu Zang Jing Sun Minhua Zheng 2022Acta Biochimica et Biophysica Sinica2022,54,11:1
9LRH-1 senses signaling from phosphatidylcholine to regulate the expansion growth of digestive organs via synergy with Wnt/β-catenin signaling in zebrafish显示文摘Liver receptor homolog-1(LRH-1)is an orphan nuclear receptor that is critical for the growth and proliferation of cancer cells and other biological processes,including lipid transportation and metabolism,sexual determination and steroidogenesis.However,because homozygous lrh-1^(-/-) mice die in utero,the regulatory mechanisms involved in embryonic development mediated by this receptor are poorly understood.In the present study,we performed transcription activator-like effector nuclease(TALEN)-mediated loss-of-function assays,taking advantage of zebrafish external fertilization,to investigate the function of lrh-1.The digestive organs were affected by lrh-1 depletion as a result of cellcycle arrest(at the checkpoint of G1 to S phase),but not cell apoptosis.Biochemical analysis revealed that LRH-1 augments the transcriptional activity of b-catenin 1 and 2 via physical interactions.Screening the specific ligand(s)sensed by LRH-1 during organogenesis revealed that phosphatidylcholine(PC),a potential ligand,is the upstream target of LRH-1 during endoderm development.These data provide evidence for the crosstalk between the PC/LRH-1 and Wnt/β-catenin signaling pathways during the expansion growth of endoderm organs.Gang Zhai Jia Song Tingting Shu Junjun Yan Xia Jin Jiangyan He Zhan Yin 2017Journal of Genetics and Genomics2017,44,6:0
10Digestion performance improvement of tributyrin through nano-emulsion preparation technology显示文摘The current research aimed to optimize the preparation technology of tributyrin nano-emulsion(TBNE)to improve its digestion properties, which were investigated through digestion models in vitro and in vivo. The content of each component of TBNE was optimized by response surface methodology(RSM) to improve the z-average particle size and stability of TBNE. The optimized TBNE was evaluated for its digestion properties by in vitro and in vivo models. The preparation conditions of TBNE optimized by RSM were as follows: 61.3% of sorbitol, 32.7% of tributyrin, and 6.0% of modified phospholipid(MP). The predicted z-average particle size of TBNE was(246.02 ± 18.10) nm. The results of the verification test showed that the z-average particle size, zeta potential, conductivity, emulsification activity index, and emulsification stability index of TBNE were(250.02 ± 7.18) nm,(-40.23 ± 0.76) m V,(31.80 ± 2.09) μS/cm,(848.00 ± 84.53) min and(1.14 ± 0.02) m^(2)/g, respectively. The in vitro digestion experiment results showed that the TBNE remained stable in the stomach and was released in the intestine, while the size of TBNE in the gastrointestinal tact was significantly smaller than that of tributyrin(P < 0.05), which made it easier to be digested and absorbed. Compared with tributyrin treatment, TBNE significantly promoted the average body weight at the 7th day, average daily feed intake,average daily gain, feed/gain, ileum weight, and organ index of ileum of Hy-Line BROWN chicks(P < 0.05), and the butyric acid content in the ileal chyme from TBNE and tributyrin treatment were172.18 mg/m L and 100.85 mg/m L(P < 0.01). 11Enhancing Green Ammonia Electrosynthesis Through Tuning Sn Vacancies in Sn‑Based MXene/ MAX Hybrids显示文摘Renewable energy driven N_(2) electroreduction with air as nitrogen source holds great promise for realizing scalable green ammonia production.However,relevant out-lab research is still in its infancy.Herein,a novel Sn-based MXene/MAX hybrid with abundant Sn vacancies,Sn@Ti_(2)CTX/Ti_(2)SnC–V,was synthesized by controlled etching Sn@Ti_(2)SnC MAX phase and demonstrated as an efficient electrocatalyst for electrocatalytic N2 reduction.Due to the synergistic effect of MXene/MAX heterostructure,the existence of Sn vacancies and the highly dispersed Sn active sites,the obtained Sn@Ti2CTX/Ti_(2)SnC–V exhibits an optimal NH_(3) yield of 28.4μg h^(−1) mg_(cat)^(−1) with an excellent FE of 15.57% at−0.4 V versus reversible hydrogen electrode in 0.1 M Na_(2)SO_(4),as well as an ultra-long durability.Noticeably,this catalyst represents a satisfactory NH3 yield rate of 10.53μg h^(−1) mg^(−1) in the home-made simulation device,where commercial electrochemical photovoltaic cell was employed as power source,air and ultrapure water as feed stock.The as-proposed strategy represents great potential toward ammonia production in terms of financial cost according to the systematic technical economic analysis.This work is of significance for large-scale green ammonia production.Xinyu Dai Zhen‑Yi Du Ying Sun Ping Chen Xiaoguang Duan Junjun Zhang Hui Li Yang Fu Baohua Jia Lei Zhang Wenhui Fang Jieshan Qiu Tianyi Ma 2024Nano-Micro Letters2024,16,5:0

15CTCF organizes inter-A compartment interactions through RYBP-dependent phase separation显示文摘Chromatin is spatially organized into three-dimensional structures at different levels including A/B compartments, topologically associating domains and loops. The canonical CTCF-mediated loop extrusion model can explain the formation of loops. However, the organization mechanisms underlying long-range chromatin interactions such as interactions between A–A compartments are still poorly understood. Here we show that different from the canonical loop extrusion model, RYBP-mediated phase separation of CTCF organizes inter-A compartment interactions. Based on this model, we designed and verified an induced CTCF phase separation system in embryonic stem cells (ESCs), which facilitated inter-A compartment interactions, improved self-renewal of ESCs and inhibited their differentiation toward neural progenitor cells. 16Morphological disruption and visual tuning alterations in the primary visual cortex in glaucoma(DBA/2J)mice显示文摘Glaucoma is a leading cause of irreve rsible blindness wo rldwide,and previous studies have shown that,in addition to affecting the eyes,it also causes abnormalities in the brain.However,it is not yet clear how the primary visual cortex(V1)is altered in glaucoma.This study used DBA/2J mice as a model for spontaneous secondary glaucoma.The aim of the study was to compare the electrophysiological and histomorphological chara cteristics of neurons in the V1between 9-month-old DBA/2J mice and age-matched C57BL/6J mice.We conducted single-unit recordings in the V1 of light-anesthetized mice to measure the visually induced responses,including single-unit spiking and gamma band oscillations.The morphology of layerⅡ/Ⅲneurons was determined by neuronal nuclear antigen staining and Nissl staining of brain tissue sections.Eighty-seven neurons from eight DBA/2J mice and eighty-one neurons from eight C57BL/6J mice were examined.Compared with the C57BL/6J group,V1 neurons in the DBA/2J group exhibited weaker visual tuning and impaired spatial summation.Moreove r,fewer neuro ns were observed in the V1 of DBA/2J mice compared with C57BL/6J mice.These findings suggest that DBA/2J mice have fewer neurons in the VI compared with C57BL/6J mice,and that these neurons have impaired visual tuning.Our findings provide a better understanding of the pathological changes that occur in V1 neuron function and morphology in the DBA/2J mouse model.This study might offer some innovative perspectives regarding the treatment of glaucoma.Yin Yang Zhaoxi Yang Maoxia Lv Ang Jia Junjun Li Baitao Liao Jing’an Chen Zhengzheng Wu Yi Shi Yang Xia Dezhong Yao Ke Chen 2024Neural Regeneration Research2024,19,1:0
