1Inhibition of gasdermin D-dependent pyroptosis attenuates the progression of silica-induced pulmonary inflammation and fibrosis显示文摘Silicosis is a leading cause of occupational disease-related morbidity and mortality worldwide,but the molecular basis underlying its development remains unclear.An accumulating body of evidence supports gasdermin D(GSDMD)-mediated pyroptosis as a key component in the development of various pulmonary diseases.However,there is little experimental evidence connecting silicosis and GSDMD-driven pyroptosis.In this work,we investigated the role of GSDMD-mediated pyroptosis in silicosis.Single-cell RNA sequencing of healthy and silicosis human and murine lung tissues indicated that GSDMD-induced pyroptosis in macrophages was relevant to silicosis progression.Through microscopy we then observed morphological alterations of pyroptosis in macrophages treated with silica.Measurement of interleukin-1βrelease,lactic dehydrogenase activity,and real-time propidium iodide staining further revealed that silica induced pyroptosis of macrophages.Additionally,we verified that both canonical(caspase-1-mediated)and non-canonical(caspase-4/5/11-mediated)signaling pathways mediated silica-induced pyroptosis activation,in vivo and in vitro.Notably,Gsdmd knockout mice exhibited dramatically alleviated silicosis phenotypes,which highlighted the pivotal role of pyroptosis in this disease.Taken together,our results demonstrated that macrophages underwent GSDMD-dependent pyroptosis in silicosis and inhibition of this process could serve as a viable clinical strategy for mitigating silicosis.Meiyue Song Jiaxin Wang Youliang Sun Junling Pang Xiaona Li Yuan Liu Yitian Zhou Peiran Yang Tianhui Fan Ying Liu Zhaoguo Li Xianmei Qi Baicun Li Xinri Zhang Jing Wang Chen Wang 2022Acta Pharmaceutica Sinica B2022,12,3:5
2Pomegranate-inspired porous SnSe/ZnSe@C anode:A stress-buffer nanostructure for fast and ultrastable sodium-ion storage显示文摘Tin selenide(SnSe)is considered as a potential anode for sodium-ion batteries(SIBs)owing to its high theoretical specific capacity.Unfortunately,it suffers from drastic volume expansion/contraction during sodium ions insertion/extraction,resulting in poor cycling stability.Herein,a pomegranate-inspired porous carbon shell wrapped heterogeneous SnSe/ZnSe composite(SnSe/ZnSe@C)is exquisitely designed and fabricated through electrostatic spraying followed by high-temperature selenization.The polyacrylonitrile-derived carbon shell acts as an adhesive to link the porous cubic SnSe/ZnSe and form highly interconnected microcircuits to improve the electron/ion transfer efficiency and inhibit the bulk volume change of internal metallic selenide nanoparticles and polyselenides dissolution during repeated cycling.Moreover,the abundant heterostructure interface of SnSe/ZnSe further significantly accelerates the electrons/ions transport.As a result,the as-prepared SnSe/ZnSe@C electrode exhibits a high specific capacity(508.3 m Ah g^(-1)at 0.05 A g^(-1)),excellent rate performance(177.8 m Ah g^(-1)at 10.0 A g^(-1)),and remarkable cycling stability(195.9 m Ah g^(-1)after 10,000 cycles at 5.0 A g^(-1)).Furthermore,in-situ Xray diffraction(XRD)/Raman,ex-situ transmission electron microscopy,and kinetic analysis clearly reveal a four-step electrochemical reaction process and battery-capacitor dual-mode sodium storage mechanism.This work provides a new perspective for developing commercial SIBs anode materials with high capacity and long lifespan.Zhixin Liang Qinghua Li Wang Zhang Dandan Yu Wei Zhang Jiawei Wu Gaoyu Wang Wenbo Fan Junling Wang Shaoming Huang 2022Journal of Energy Chemistry2022,,12:2
3A large-scale outdoor atmospheric simulation smog chamber for studying atmospheric photochemical processes:Characterization and preliminary application显示文摘Understanding the formation mechanisms of secondary air pollution is very important for the formulation of air pollution control countermeasures in China.Thus,a large-scale outdoor atmospheric simulation smog chamber was constructed at Chinese Research Academy of Environmental Sciences(the CRAES Chamber),which was designed for simulating the atmospheric photochemical processes under the conditions close to the real atmospheric environment.The chamber consisted of a 56-m^(3) fluorinated ethylene propylene(FEP) Teflon film reactor,an electrically-driven stainless steel alloy shield,an auxiliary system,and multiple detection instrumentations.By performing a series of characterization experiments,we obtained basic parameters of the CRAES chamber,such as the mixing ability,the background reactivity,and the wall loss rates of gaseous compounds(propene,NO,NO_(2),ozone) and aerosols(ammonium sulfate).Oxidation experiments were also performed to study the formation of ozone and secondary organic aerosol(SOA),including α-pinene ozonolysis,propene and 1,3,5-trimethylbenzene photooxidation.Temperature and seed effects on the vapor wall loss and SOA yields were obtained in this work:higher temperature and the presence of seed could reduce the vapor wall loss;SOA yield was found to depend inversely on temperature,and the presence of seed could increase SOA yield.The seed was suggested to be used in the chamber to reduce the interaction between the gas phase and chamber walls.The results above showed that the CRAES chamber was reliable and could meet the demands for investigating tropospheric chemistry.Junling Li Hong Li Xuezhong Wang Weigang Wang Maofa Ge Hao Zhang Xin Zhang Kun Li Yan Chen Zhenhai Wu Fahe Chai Fan Meng Yujing Mu Abdelwahid Mellouki Fang Bi Yujie Zhang Lingyan Wu Yongchun Liu 2021Journal of Environmental Sciences2021,33,4:2

5Paclitaxel liposome for injection (Lipusu) plus cisplatin versus gemcitabine plus cisplatin in the first-line treatment of locally advanced or metastatic lung squamous cell carcinoma: A multicenter, randomized, open-label, parallel controlled clinical study显示文摘Background:Lipusu is the first commercialized liposomal formulation of pacli-taxel and has demonstrated promising efficacy against locally advanced lung squamous cell carcinoma(LSCC)in a small-scale study.Here,we conducted a multicenter,randomized,phase 3 study to compare the efficacy and safety of cis-platin plus Lipusu(LP)versus cisplatin plus gemcitabine(GP)as first-line treat-ment in locally advanced or metastatic LSCC.Methods:Patients enrolled were aged between 18 to 75 years,had locally advanced(clinical stage IIIB,ineligible for concurrent chemoradiation or surgery)or metastatic(Stage IV)LSCC,had no previous systemic chemother-apy and at least one measurable lesion as per the Response Evaluation Criteria in Solid Tumors(version 1.1)before administration of the trial drug.The primary endpoint was progression-free survival(PFS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),overall survival(OS),and safety profiles.To explore the possible predictive value of plasma cytokines for LP treatment,plasma samples were collected from the LP group at baseline and first efficacy evaluation time and were then subjected to analysis by 45-Plex ProcartaPlex Panel 1 to detect the presence of 45 cytokines using the Luminex xMAP technology.The correlation between treatment outcomes and dynamic changes in the levels of cytokines were evaluated in preliminary analyses.Results:The median duration of follow-up was 15.4 months.237 patients in the LP group and 253 patients in the GP group were included in the per protocol set(PPS).In the PPS,the median PFS was 5.2 months versus 5.5 months in the LP and GP group(hazard rtio[HR]:1.03,P=0.742)respectively.The median OS was 14.6 months versus 12.5 months in the LP and GP group(HR:0.83,P=0.215).The ORR(41.8%versus 45.9%,P=0.412)and DCR(90.3%versus 88.1%,P=0.443)were also similar between the LP and GP group.A significantly lower proportion of patients in the LP group experienced adverse events(AEs)leading to treatment interruptions(10.9%versus 26.4%,P<0.001)or treatment termination(14.3%versus 23.1%,P=0.011).The analysis of cytokine levels in the LP group showed that low baseline levels of 27 cytokines were associated with an increased ORR,and 15 cytokines were associated with improved PFS,with 14 cytokines,including TNF-a,IFN-y,IL-6,and IL-8,demonstrating an overlapping trend.Conclusion:The LP regimen demonstrated similar PFS,OS,ORR and DCR as the GP regimen for patients with locally advanced or metastatic LSCC but had more favorable toxicity profiles.The study also identified a spectrum of different cytokines that could be potentially associated with the clinical benefit in patients who received the LP regimen.Jie Zhang Yueyin Pan Qin Shi Guojun Zhang Liyan Jiang Xiaorong Dong Kangsheng Gu Huijuan Wang Xiaochun Zhang Nong Yang Yuping Li Jianping Xiong Tienan Yi Min Peng Yong Song Yun Fan Jiuwei Cui Gongyan Chen Wei Tan Aimin Zang Qisen Guo Guangqiang Zhao Ziping Wang Jianxing He Wenxiu Yao Xiaohong Wu Kai Chen Xiaohua Hu Chunhong Hu Lu Yue Da Jiang Guangfa Wang Junfeng Liu Guohua Yu Junling Li Jianling Bai Wenmin Xie Weihong Zhao Lihong Wu Caicun Zhou 2022Cancer Communications2022,42,1:1
6Arabidopsis F-BOX STRESS INDUCED 4 is required to repress excessive divisions in stomatal development显示文摘During the terminal stage of stomatal development,the R2 R3-MYB transcription factors FOUR LIPS(FLP/MYB124) and MYB88 limit guard mother cell division by repressing the transcript levels of multiple cell-cycle genes. In Arabidopsis thaliana possessing the weak allele flp-1, an extra guard mother cell division results in two stomata having direct contact.Here, we identified an ethylmethane sulfonatemutagenized mutant, flp-1 xs01 c, which exhibited more severe defects than flp-1 alone, producing gianttumor-like cell clusters. XS01 C, encoding F-BOX STRESS-INDUCED 4(FBS4), is preferentially expressed in epidermal stomatal precursor cells.Overexpressing FBS4 rescued the defective stomatal phenotypes of flp-1 xs01 c and flp-1 mutants. The deletion or substitution of a conserved residue(Proline166) within the F-box domain of FBS4 abolished or reduced, respectively, its interaction with Arabidopsis Skp1-Like1(ASK1), the core subunit of the Skp1/Cullin/F-box E3 ubiquitin ligase complex. Furthermore, the FBS4 protein physically interacted with CYCA2;3 and induced its degradation through the ubiquitin-26 S proteasome pathway. 8The protective effects of Xuebijing injection on intestinal injuries of mice exposed to irradiation显示文摘Background:Gastrointestinal(GI)injury is one of the most common side effects of radiotherapy.However,there is no ideal therapy method except for symptomatic treatment in the clinic.Xuebijing(XBJ)is a traditional Chinese medicine,used to treat sepsis by injection.In this study,the protective effects of XBJ on radiation-i nduced intestinal injury(RⅢ)and its mechanism were explored.Methods:The effect of XBJ on survival of irradiated C57BL/6 mice was monitored.Histological changes including the number of crypts and the length of villi were evaluated by H&E.The expression of Lgr5^(+)intestinal stem cells(ISCs),Ki67^(+)cells,villin and lysozymes were examined by immunohistochemistry.The expression of cytokines in the intestinal crypt was detected by RT-PCR.DNA damage and apoptosis rates in the small intestine were also evaluated by immunofluorescence.Results:In the present study,XBJ improved the survival rate of the mice after 8.0and 9.0 Gy total body irradiation(TBI).XBJ attenuated structural damage of the small intestine,maintained regenerative ability and promoted proliferation and differentiation of crypt cells,decreased apoptosis rate and reduced DNA damage in the intestine.Elevation of IL-6 and TNF-α was limited,but IL-1,TNF-β and IL-10 levels were increased in XBJ-treated group after irradiation.The expression of Bax and p53 were decreased after XBJ treatment.Conclusions:Taken together,XBJ provides a protective effect on RⅢby inhibiting inflammation and blocking p53-related apoptosis pathway.Yinping Dong YuanYang Zhang Xinyue Wang Wenxuan Li Junling Zhang Lu Lu Hui Dong Saijun Fan Aimin Meng Deguan Li 2022Animal Models and Experimental Medicine2022,5,6:0
9Kinetic and mechanism of the reaction between Cl and several mono-methyl branched alkanes显示文摘Branched alkanes are ubiquitous in the troposphere and play an important role in the chemical processes.In this work,the rate constants and products for the reaction of Cl atoms with 3-methylhexane and 2-methylheptane were measured at room temperature(298±0.2K)and atmospheric pressure using a conventional relative rate method.The rate constants of 3-methylhexane and 2-methylheptane in units of cm~3/(mol·sec)are(3.09±0.31)×10^(-10)and(3.67±0.40)×10^(-10),respectively.Furthermore,the corresponding atmospheric lifetime of the studied branched alkanes with Cl was 6.92-89.90 hours and 5.82-75.69 hours,respectively.The estimated atmospheric lifetimes indicated that the reaction with Cl atoms could be the most important atmospheric degradation pathway for 3-methylhexane and2-methylheptane.Primary gas-phase products of the reactions were identified and quantified,and particle-phase products were also obtained.The atmosphere oxidation mechanism of Cl atoms with 3-methylhexane and 2-methylheptane is proposed.The SOA yields of 3-methylhexane and 2-methylheptane from the reaction of Cl atoms were determined to be7.96%±0.89%and 13.35%±1.50%respectively.Overall,the results reveal that the primary loss process of branched alkanes is the reaction with Cl atoms,which impacts its degradation on a regional scale.Yan Chen Weigang Wang Junling Li Li Zhou Bo Shi Cici Fan Ke Wang Hao Zhang Hong Li Maofa Ge 2024Journal of Environmental Sciences2024,,1:0
