1Additively manufactured biodegradable porous magnesium implants for elimination of implant-related infections:An in vitro and in vivo study显示文摘Magnesium(Mg)alloys that have both antibacterial and osteogenic properties are suitable candidates for orthopedic implants.However,the fabrication of ideal Mg implants suitable for bone repair remains challenging because it requires implants with interconnected pore structures and personalized geometric shapes.In this study,we fabricated a porous 3D-printed Mg-Nd-Zn-Zr(denoted as JDBM)implant with suitable mechanical properties using selective laser melting technology.The 3D-printed JDBM implant exhibited cytocompatibility in MC3T3-E1 and RAW267.4 cells and excellent osteoinductivity in vitro.Furthermore,the implant demonstrated excellent antibacterial ratios of 90.0% and 92.1% for methicillin-resistant S.aureus(MRSA)and Escherichia coli,respectively.The 3D-printed JDBM implant prevented MRSA-induced implant-related infection in a rabbit model and showed good in vivo biocompatibility based on the results of histological evaluation,blood tests,and Mg2+deposition detection.In addition,enhanced inflammatory response and TNF-α secretion were observed at the bone-implant interface of the 3D-printed JDBM implants during the early implantation stage.The high Mg^(2+)environment produced by the degradation of 3D-printed JDBM implants could promote M1 phenotype of macrophages(Tnf,iNOS,Ccl3,Ccl4,Ccl5,Cxcl10,and Cxcl2),and enhance the phagocytic ability of macrophages.The enhanced immunoregulatory effect generated by relatively fast Mg^(2+)release and implant degradation during the early implantation stage is a potential antibacterial mechanism of Mg-based implant.Our findings indicate that 3D-printed porous JDBM implants,having both antibacterial property and osteoinductivity,hold potential for future orthopedic applications.Kai Xie Nanqing Wang Yu Guo Shuang Zhao Jia Tan Lei Wang Guoyuan Li Junxiang Wu Yangzi Yang Wenyu Xu Juan Chen Wenbo Jiang Penghuai Fu Yongqiang Hao 2022Bioactive Materials2022,7,2:8
2Bamboo charcoal addition enhanced the nitrogen removal of anammox granular sludge with COD:Performance,physicochemical characteristics and microbial community显示文摘The effects of different chemical oxygen demand(COD)concentrations on the anammox granular sludge with Bamboo Charcoal(BC)addition were evaluated in UASB reactor.The results showed that the average total nitrogen(TN)removal efficiency was reduced from 85.9%to 81.4%when COD concentration was increased from 50 to 150 mg/L.However,the TN removal efficiency of BC addition reactors was dramatically 3.1%-6.4%higher than that without BC under different COD concentrations.The average diameter of granular sludge was 0.13 mm higher than that without BC.The settling velocity was increased by elevated COD concentration,while the EPS and VSS/SS were increased with BC addition.The high-throughput Miseq sequencing analyses revealed that the bacterial diversity and richness were decreased under COD addition,and the Planctomycetes related to anammox bacteria were Candidatus Brocadia and Candidatus Kuenenia.The Metagenomic sequencing indicated that the abundance of denitrification related functional genes all increased with elevated COD,while the abundance of anammox related functional genes of decreased.The func-tional genes related to anammox was hydrazine synthase encoding genes(hzsA,hzsB and hzsB).The average relative abundance of hzs genes in the reactor with BC addition was higher than the control at COD concentrations of 50 mg/L and 150 mg/L.The functional genes of denitrification mediated by BC were higher than those without BC throughout the operation phase.It is interesting to note that BC addition greatly enriched the related func-tional genes of denitrification and anammox.Menglei Guo Ying Jiang Junxiang Xie Qianfei Cao Qun Zhang Adams Mabruk Chongjun Chen 2022Journal of Environmental Sciences2022,34,5:5
3Serum semaphorin 4C as a diagnostic biomarker in breast cancer:A multicenter retrospective study显示文摘Background:To date,there is no approved blood-based biomarker for breast cancer detection.Herein,we aimed to assess semaphorin 4C(SEMA4C),a pivotal protein involved in breast cancer progression,as a serum diagnostic biomarker.Methods:We included 6,213 consecutive inpatients from Tongji Hospital,Qilu Hospital,and Hubei Cancer Hospital.Training cohort and two validation cohorts were introduced for diagnostic exploration and validation.A pan-cancer cohort was used to independently explore the diagnostic potential of SEMA4C among solid tumors.Breast cancer patients who underwent mass excision prior to modified radical mastectomy were also analyzed.We hypothesized that increased pretreatment serum SEMA4C levels,measured using optimized in-house enzymelinked immunosorbent assay kits,could detect breast cancer.The endpoints were diagnostic performance,including area under the receiver operating characteristic curve(AUC),sensitivity,and specificity.Post-surgery pathological diagnosis was the reference standard and breast cancer staging followed the TNM classification.There was no restriction on disease stage for eligibilities.Results:We included 2667 inpatients with breast lesions,2378 patients with other solid tumors,and 1168 healthy participants.Specifically,118 patients with breast cancer were diagnosed with stage 0(5.71%),620 with stage I(30.00%),966 with stage II(46.73%),217 with stage III(10.50%),and 8 with stage IV(0.39%).Patients with breast cancer had significantly higher serum SEMA4C levels than benign breast tumor patients and normal controls(P<0.001).Elevated serum SEMA4C levels had AUC of 0.920(95%confidence interval[CI]:0.900–0.941)and 0.932(95%CI:0.911–0.953)for breast cancer detection in the two validation cohorts.The AUCs for detecting early-stage breast cancer(n=366)and ductal carcinoma in situ(n=85)were 0.931(95%CI:0.916–0.946)and 0.879(95%CI:0.832–0.925),respectively.Serum SEMA4C levels significantly decreased after surgery,and the reduction was more striking after modified radical mastectomy,compared with mass excision(P<0.001).The positive rate of enhanced serum SEMA4C levels was 84.77%for breast cancer and below 20.75%for the other 14 solid tumors.Conclusions:Serum SEMA4C demonstrated promising potential as a candidate biomarker for breast cancer diagnosis.However,validation in prospective settings and by other study groups is warranted.Ya Wang Long Qiao Jie Yang Xiong Li Yaqi Duan Jiahao Liu Shaoqi Chen Huayi Li Dan Liu Tian Fang Jingjing Ma Xiaoting Li Fei Ye Junxiang Wan Juncheng Wei Qin Xu Ensong Guo Ping Jin Mingfu Wu Lin Zhang Yun Xia Yaqun Wu Jun Shao Yaojun Feng Qing Zhang Zongyuan Yang Gang Chen Qinghua Zhang Xingrui Li Shixuan Wang Junbo Hu Xiaoyun Wang Mona P.Tan Kazuaki Takabe Beihua Kong Qifeng Yang Ding Ma Qinglei Gao 2021Cancer Communications2021,41,12:3
4Coupling mechanism of activated carbon mixed with dust for flue gas desulfurization and denitrification显示文摘To clarify the effect of coking dust, sintering dust and fly ash on the activity of activated carbon for various industrial flue gas desulfurization and denitrification, the coupling mechanism of the mixed activated carbon and dust was investigated to provide theoretical reference for the stable operation. The results show that coking dust had 34% desulfurization efficiency and 10% denitrification efficiency;correspondingly, sintering dust and fly ash had no obvious desulfurization and denitrification activities. For the mixture of activated carbon and dust, the coking dust reduced the desulfurization and denitrification efficiencies by blocking the pores of activated carbon, and its inhibiting effect on activated carbon was larger than its own desulfurization and denitrification activity. The sintering dust also reduced the desulfurization efficiency on the activated carbon while enhancing the denitrification efficiency. Fly ash blocked the pores of activated carbon and reduced its reaction activity. The reaction activity of coking dust mainly came from the surface functional groups, similar to that of activated carbon. The reaction activity of sintering dust mainly came from the oxidative property of Fe_2O _3, which oxidized NO to NO_2 and promoted the fast selectively catalytic reduction(SCR) of NO to form N_2. Sintering dust was activated by the joint action of activated carbon, and both had a coupling function. 5Efficient drug delivery and anticancer effect of micelles based on vitamin E succinate and chitosan derivatives显示文摘Nanocarriers have emerged as a promising cancer drug delivery strategy.Multi-drug resistance caused by overexpression of multiple-drug excretion transporters in tumor cells is the major obstacle to successful chemotherapy.Vitamin E derivatives have many essential functions for drug delivery applications,such as biological components that are hydrophobic,stable,water-soluble enhancing compounds,and anticancer activity.In addition,vitamin E derivatives are also effective mitocan which can overcome multi-drug resistance by binding to P glycoproteins.Here,we developed a carboxymethyl chitosan/vitamin E succinate nano-micellar system(O-CMCTS-VES).The synthesized polymers were characterized by Fourier Transform IR,and 1H NMR spectra.The mean sizes of O-CMCTS-VES and DOX-loaded nanoparticles were around 177 nm and 208 nm.The drug loading contents were 6.1%,13.0%and 10.6%with the weight ratio of DOX to O-CMCTS-VES corresponding 1:10,2:10 and 3:10,and the corresponding EEs were 64.3%,74.5%and 39.7%.Cytotoxicity test,hemolysis test and histocompatibility test showed that it had good biocompatibility in vitro and in vivo.Drug release experiments implied good pH sensitivity and sustained-release effect.The DOX/O-CMCTS-VES nanoparticles can be efficiently taken up by HepG2 cancer cells and the tumor inhibition rate is up to 62.57%.In the in vivo study by using H22 cells implanted Balb/C mice,DOX/O-CMCTS-VES reduced the tumor volume and weight efficiently with a TIR of 35.58%.The newly developed polymeric micelles could successfully be utilized as a nanocarrier system for hydrophobic chemotherapeutic agents for the treatment of solid tumors.Xiaotong Chen Junxiang Gu Le Sun Wenya Li Lili Guo Zhiyang Gu Litong Wang Yan Zhang Wangwang Zhang Baoqin Han Jing Chang 2021Bioactive Materials2021,6,10:1

11Host protection against Omicron BA.2.2 sublineages by prior vaccination in spring 2022 COVID-19 outbreak in Shanghai显示文摘The Omicron family of SARS-CoV-2 variants are currently driving the COVID-19 pandemic.Here we analyzed the clinical laboratory test results of 9911 Omicron BA.2.2 sublineages-infected symptomatic patients without earlier infection histories during a SARS-CoV-2 outbreak in Shanghai in spring 2022.Compared to an earlier patient cohort infected by SARS-CoV-2 prototype strains in 2020,BA.2.2 infection led to distinct fluctuations of pathophysiological markers in the peripheral blood.In particular,severe/critical cases of COVID19 post BA.2.2 infection were associated with less pro-inflammatory macrophage activation and stronger interferon alpha response in the bronchoalveolar microenvironment.Importantly,the abnormal biomarkers were significantly subdued in individuals who had been immunized by 2 or 3 doses of SARS-CoV-2 prototypeinactivated vaccines,supporting the estimation of an overall 96.02% of protection rate against severe/critical disease in the 4854 cases in our BA.2.2 patient cohort with traceable vaccination records.Furthermore,even though age was a critical risk factor of the severity of COVID-19 post BA.2.2 infection,vaccination-elicited protection against severe/critical COVID-19 reached 90.15% in patients aged≥60 years old.Together,our study delineates the pathophysiological features of Omicron BA.2.2 sublineages and demonstrates significant protection conferred by prior prototype-based inactivated vaccines.Ziyu Fu Dongguo Liang Wei Zhang Dongling Shi Yuhua Ma Dong Wei Junxiang Xi Sizhe Yang Xiaoguang Xu Di Tian Zhaoqing Zhu Mingquan Guo Lu Jiang Shuting Yu Shuai Wang Fangyin Jiang Yun Ling Shengyue Wang Saijuan Chen Feng Liu Yun Tan Xiaohong Fan 2023Frontiers of Medicine2023,17,3:1
12Study on Difference in Cold Tolerance Between Weedy Rice and Cultivated Rice in Heilongjiang Province of China显示文摘A long-red awn weedy rice and rice cultivars named Ken99004(ZC1), Crossing-503(ZC2), Ken Sticky Rice(ZC3), Shashani(ZC4), Long-Grain Aromatic Rice(ZC5) were used to study typical wild traits of weedy rice and to identify the cold resistance of all genotypes. The results showed that the stem of long-red awn weedy rice was the weakest. No difference was found in the early stage of rice growth, e.g. the vegetable growth stage, between long-red awn weedy rice and other rice cultivars in the stem rigidity, but in the later stage, or reproductive growth stage, their leaves and stems were senescent rapidly because of the speedy supply of nutrients for panicle growth, meanwhile the rigidity of stem was reduced sharply just as withered weeds. The germination rate of longred awn weedy rice was the highest in cold condition, and in turn were ZC1, ZC2, ZC3, ZC4, ZC5, respectively. 15Influence of calcium preconditioning and streptomycin on ventricular dilation-induced arrhythmias in isolated rat hearts显示文摘Objective To investigate the mechanism of ventricular dilation-induced arrhythmias by dilating isolated rat hearts. Methods Isolated rat hearts were perfused by Langerdorff method. After equilibration,80 hearts were randomly divided into four groups as follows:(1) control group(n=20) ,(2) Ca2+ preconditioning(CPC) group(n=20) ,(3) streptomycin group(n=20) ,and(4) CPC + streptomycin group(n=20) . A latex balloon which can be filled with fluid was anchored in the left ventricle through the left atrium and mitral valve. Epicardial ECG of the left ventricle,left ventricular pressure,coronary flow and heart rate were recorded before and during ventricular dilation by injecting fluid into the latex balloon. The rate and duration of ventricular dilation-induced arrhythmias were recorded. Results Under the same increase in ventricular end-diastolic pressure made by inflation of the balloon,the rate of arrhythmias was 100% and duration of arrhythmias was 2. 56±0.46s in the control group. Both the rates of premature ventricular beat(90%) and ventricular tachycardia 70%) were high. Compared with the control group,the total rate(60%) of arrhythmias was lower,and duration(1.67±0.61s) of arrhythmias was shorter in the CPC group. Both the rates of premature ventricular beat(60%) and ventricular tachycardia(40%) were low comparatively. The rate of arrhythmias(45%) was lower and duration(1.64±0.42s) of arrhythmias was shorter,and the rates of premature ventricular beat(30%) or ventricular tachycardia(35%) were lower in the streptomycin group than in the control one. The least ventricular dilation-induced arrhythmias occurred in the CPC + streptomycin group. The rate of arrhythmias(10%) was the lowest and duration(1.01±0.37s) of arrhythmias was the shortest;both the rates of premature ventricular beat(5%) and ventricular tachycardia(10%) were the lowest. Conclusions Ventricular dilation may induce arrhythmias in isolated rat hearts.
