| 2 | Schisandrol A protects AGEs-induced neuronal cells death by allosterically targeting ATP6V0d1 subunit of V-ATPase显示文摘Diabetes have been shown to cause progressive neuronal injury with pain and numbness via advanced glycation end-products(AGEs)-induced neuronal cell apoptosis;however, the valuable drug targets for diabetic neuropathy have been poorly reported so far. In this study, we discovered a natural small-molecule schisandrol A(SolA) with significant protective effect against AGEs-induced neuronal cell apoptosis. ATP6V0D1, a major subunit of vacuolar-type ATPase(V-ATPase) in lysosome was identified as a crucial cellular target of SolA. Moreover, SolA allosterically mediated ATP6V0D1 conformation via targeting a unique cysteine 335 residue to activate V-ATPase-dependent lysosomal acidification.Interestingly, SolA-induced lysosome pH downregulation resulted in a mitochondrial-lysosomal crosstalk by selectively promoting mitochondrial BH3-only protein BIM degradation, thereby preserving mitochondrial homeostasis and neuronal cells survival. | 3 | Antioxidant,Hypoglycemic and Hypolipidemic Activities of Sambucus williamsii Seed Oil显示文摘[Objectives] To explore the antioxidant activity of Sambucus williamsii seed oil. [Methods]DPPH scavenging method and Prussian blue method( total reducing power) were used. S. williamsii seed oil has antioxidant activity. In the DPPH scavenging free radical experiment,S. williamsii oil showed the ability to scavenge free radicals,and its scavenging ability had linear relationship with oil concentration( R =0. 999 6). Besides,the IC_(50) value of S. williamsii seed oil scavenging DPPH free radicals was 61. 30 ± 0. 88 mg/mL. In the total reducing power measurement experiment,the S. williamsii seed oil has a reducing ability,and its reducing ability is proportional to the oil concentration,and has the concentration dependence. Through studying the inhibitory effect of S. williamsii seed oil on α-glucosidase,the hypoglycemic activity of S. williamsii seed oil was determined. The ability of samples to inhibit α-glucosidase was judged through measuring the absorbance at 400 nm of the reaction system. [Results]In the concentration range of 1. 56-25. 00 mg/mL,the S. williamsii seed oil can effectively inhibit α-glucosidase,and the inhibition rate was 62. 66%-85. 22%. The animal in vivo experiment was used to determine the hypolipidemic activity of the S. williamsii seed oil. First,a high fat modelwas established,S. williamsii seed oil was given through intragastric administration. Then,total cholesterol( TC),triglyceride( TG),high-density lipoprotein cholesterol( HDL-C),and low density lipoprotein cholesterol( LDL-C) were measured. Through comparison of high,medium and low dose groups of S. williamsii seed oil with the high fat modelcontrol group,it was found that the TC,TG,LDL-C levels were significantly decreased( P < 0. 01) and the HDL-C levels were significantly increased( P < 0. 05),indicating that S. williamsii seed oil can effectively reduce the levels of TC,TG,LDL-C in the serum of hyperlipidemic mice,and effectively inhibit the decrease of HDL-C level,thus S. williamsii seed oil can reduce blood lipids in mice,namely,has hypolipidemic effects. In addition,the greater the dose of S. williamsii seed oil,the more obvious the effect of blood lipid levels in mice,indicating that the hypolipidemic effect of S. williamsii seed oil was dose dependent. [Conclusions] S. williamsii seed oil has reducing ability,and there is a significant dose-effect relationship in the concentration range of 2-10 mg/mL. | 6 | Cr(Ⅲ) removal from simulated solution using hydrous magnesium oxide coated fly ash: Optimization by response surface methodology (RSM)显示文摘Hydrous magnesium oxide coated fly ash(MFA) has environmental remediation potential by providing a substrate for the adsorption of aqueous Cr(Ⅲ). Aqueous Cr(Ⅲ) adsorption onto MFA was examined as a function of MFA dosage, p H and initial Cr(Ⅲ) concentration with the Box–Behnken approach used for experimental design and optimization using response surface methodology(RSM). p H and dosage(dosage and concentration) have significant interactive effects on Cr(Ⅲ) adsorption efficiency. Analysis of variance shows that the response surface quadratic model is highly significant and can effectively predict the experimental outcomes. Cr(Ⅲ) removal efficiency of 98% was obtained using optimized conditions of MFA dosage, p H and initial Cr(Ⅲ) concentration of 1.57 g·L^(-1), 4.11 and 126 mg·L^(-1), respectively. Cr(Ⅲ) adsorption onto MFA is mainly attributed to the interaction between Cr(Ⅲ) and the functional group \\OH of the hydrous magnesium oxide, in all probability caused by chemisorptions. | 13 | Anti-psoriasis molecular targets and active components discovery of Optimized Yinxieling Formula via affinity-purified strategy显示文摘Psoriasis,a prevalent inherited skin condition,involves an inflammatory response as a key pathogenic mechanism.The Optimized Yinxieling Formula(OYF),rooted in traditional Chinese medicine,is extensively utilized in clinical settings to treat psoriasis.Although previous studies have demonstrated OYF’s significant anti-inflammatory effects in psoriasis,its potential molecular targets and active components remain unexplored.This study aimed to unveil the anti-psoriasis molecular targets and active components of OYF.Our findings indicated that OYF extract markedly reduced the production of several inflammatory mediators,including IL-23,nitric oxide,TNF-α,and IL-1β,in LPS-induced RAW264.7 cells.We synthesized OYF extract-crosslinked beads to isolate pharmacological targets from RAW264.7 lysates using an affinity purification strategy,known as Target Fishing.The enriched target proteins were subsequently identified via LC-MS/MS,followed by bioinformatics analysis to map the psoriasis-associated pathway-gene network.We identified a total of 76 potential target proteins,which were highly associated with mRNA transcription mechanisms.In particular,pathway-gene network analysis revealed that the IL-23 inflammatory pathway was involved in the anti-psoriasis effect of OYF extract.We further utilized a target protein-based affinity capture strategy,combined with LC-MS and SPR analysis,to globally screen OYF’s active components,focusing on the mRNA transcription regulator,fused in sarcoma(FUS).This process led to the identification of umbelliferone,vanillic acid,protocatechuic acid,gentisic acid,and echinacoside as key compounds targeting FUS to inhibit IL-23 expression.Additionally,we formulated a compound cocktail(CpdC),which significantly reduced psoriasis area and severity index(PASI)scores and the expressions of IL-23 and Ki67 in an imiquimod(IMQ)-induced psoriasis mouse model.Collectively,our study elucidates the primary molecular targets and active components of OYF,offering novel insights for psoriasis treatment. | WANG Wei LIU Lijuan YANG Zhuo LU Chuanjian TU Pengfei ZHAO Ruizhi ZENG Kewu | 2024 | Chinese Journal of Natural Medicines2024,22,2: | 0 |
